Heterologous expression of genes involved in biosynthesis of polyunsaturated fatty acids in Streptomyces rimosus
Bakterije iz rodu Streptomyces so znane po sintezi različnih sekundarnih metabolitov. Zaradi razvitih številnih genskih orodij se vrste iz rodu Streptomyces postavljajo kot odlični gostitelji za proizvodnjo heterolognih sekundarnih metabolitov. Namen te raziskave je bil vstaviti dva različna plazmidna vektorja v gensko spremenjen sev Streptomyces rimosus, ki vsebuje 145 kbp veliko delecijo, ki nosi zapis dveh glavnih sekundarnih metabolitov, oksitetraciklina in rimocidina, zaradi česar ta sev predstavlja odličenega gostitelja za proizvodnjo heterolognih produktov. Večkrat nenasičene maščobne kisline (PUFA) igrajo ključno vlogo v različnih fizioloških in parafizioloških procesih v človeškem telesu. Med njimi izstopajo omega-3 maščobne kisline, kot sta eikozapentaenojska kislina (EPA) in dokozaheksaenojska kislina (DHA). Te kisline so povezane s preprečevanjem in zdravljenjem bolezni srca in ožilja, debelosti in diabetesa. V študiji smo uporabili biosintezno gensko gručo za sintezo večkrat nenasičenih maščobnih kislin, ki je sestavljena iz treh genov: pfa1, pfa2 in pfa3. Ti geni izvirajo iz miksobakterije Aetherobacter fasciculatus. Biosintezna genska gruča kodira multifunkcionalni encim, ki katalizira biosintezo nenasičenih maščobnih kislin, kot sta EPA in DHA. Uporabljena sta bila dva plazmidna vektorja, pAB04ErmE*PUFA in pAB04ErmE*PUFA-ppt. Oba vektorja vsebujeta gensko gručo miksobakterije A. fasciculatus, odgovorno za sintezo PUFA. Poleg tega plazmid pAB04ErmE*PUFA-ppt vsebuje tudi gen za fosfopanteteinil transferazo, ki naj bi povečala sintezo PUFA. Vektorji so bili sestavljeni in preneseni v bakterijo S. rimosus OTC 145. Sevi S. rimosus ki vsebujejo vse tri gene pfa1, pfa2 in pfa3, so proizvajali DHA. Koncentracija DHA je bila 1-4 mg/L. Z ustrezno optimizacijo gojišča, s spreminjanjem koncentracij virov ogljika, dušika, fosforja in rastlinskega olja, je bil dosežen titer DHA 21,03 mg/L, kar predstavlja 17-kratno povečanje glede na osnovno produkcijsko gojišče GOTC-P.Bacteria from the genus Streptomyces are known for production of variety of secondary metabolites. Steptomyces species also serve as excellent host for production of heterologous secondary metabolites and a number of gene tools have been developed. The aim of this study was to insert two different plasmid vectors into the engineered strain of Streptomyces rimosus, which contains 145 kbp deletion encoding two main antibiotics oxytetracicline and rimocidine, which represent excellent heterologous host for production of diverse metabolites. Polyunsaturated fatty acids (PUFAs) play a crucial role in various physiological and paraphysiological processes in the human body. Among them, omega-3 fatty acids, such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), hold a prominent position. These acids are associated with the prevention and treatment of cardiovascular diseases, obesity, and diabetes. In the study, biosynthetic gene cluster was used for the synthesis of polyunsaturated fatty acids. The cluster was composed of three genes: pfa1, pfa2, and pfa3. This pathway originates from the myxobacterium Aetherobacter fasciculatus. The biosynthetic gene cluster encodes a multifunctional enzyme catalysing synthesis of the unsaturated fatty acids such as EPA and DHA. Two plasmid vectors were used pAB04ErmE*PUFA and pAB04ErmE*PUFA-ppt. Both vectors used in the study carry gene cluster from the myxobacterium A. fasciculatus, encoding PUFAs. Additionally, the pAB04ErmE*PUFA-ppt plasmid also contains a gene for phosphopantetheinyl transferase, which is expected to enhance the synthesis of PUFAs. Vectors were constructed and transferred into the bacterium S. rimosus OTC 145. The S. rimosus strains containing the genes pfa1, pfa2 and pfa3 had the ability to produce DHA. The final concentration of DHA ranged from 1-4 mg/L. By a simple media optimization, such as adjusting the concentrations of carbon, nitrogen, phosphorus sources, and vegetable oil, a DHA titer of 21.03 mg/L was achieved, representing a 17-fold increase compared to the baseline production medium GOTC-P
Spectral arbitrariness for trees fails spectacularly
Given a graph G, consider the family of real symmetric matrices with the property that the pattern of their nonzero off-diagonal entries corresponds to the edges of G. For the past 30 years a central problem has been to determine which spectra are realizable in this matrix class. Using combinatorial methods, we identify a family of graphs and multiplicity lists whose realizable spectra are highly restricted. In particular, we construct trees with multiplicity lists that require a unique spectrum, up to shifting and scaling. This represents the most extreme possible failure of spectral arbitrariness for a multiplicity list, and greatly extends all previously known instances of this phenomenon, in which only single linear constraints on the eigenvalues were observed

This study focuses on the challenging problem of labeling a collection of objects with inherent morphological and positional dependencies, where instances may be missing or duplicated. We integrate principles of assignment theory in the design of a convolutional neural network to find the optimal label set given pairwise geometrical features extracted from the candidate objects. The objective function aims to minimize the distance between the one-hot encoded labels of the objects and the scores produced by the model, with added emphasis on the scores corresponding to the optimal assignment plan. We tested our solution in the dental domain on the task of finding the teeth labels given a set of candidate instances. The study database included 1200 dental casts of upper and lower jaws from 600 patients. The model reached identification accuracies of 0.952 and 0.968 for the lower and upper jaws, respectively. Moreover, we presented a solution for generating teeth candidates using a multi-step pipeline consisting of coarse and fine segmentations. The algorithm was tested on a database consisting of 600 dental casts, reaching an F1 score of 0.968
Speech-language pathologists’ job satisfaction across various workplaces
Logoped je strokovnjak na področju komunikacije, govora, jezika, sluha, glasu, hranjenja in požiranja. Po zaključenem izobraževanju se lahko zaposli na različnih delovnih mestih v javnem (v zdravstveni dejavnosti, izobraževalni dejavnosti in dejavnosti socialnega varstva) ali zasebnem sektorju. Zaposlitev v različnih okoljih prinaša različne vloge, naloge in pogoje dela za logopeda, zaradi česar se lahko med posamezniki na različnih delovnih mestih pojavijo razlike v delovni motivaciji in delovnem zadovoljstvu.
V magistrskem delu smo raziskali, kakšna je stopnja delovnega zadovoljstva logopedov na različnih delovnih mestih v Sloveniji in kateri dejavniki vplivajo nanjo. Oblikovali smo deset raziskovalnih hipotez, ki smo jih preverili z uporabo vprašalnika o delovnem zadovoljstvu (Job Descriptive Index – JDI), ki je namenjen široki populaciji zaposlenih. Preverjali smo zadovoljstvo slovenskih logopedov s petimi področji delovnega zadovoljstva, tj. z delom, vodjo, sodelavci, plačo in možnostmi napredovanja. V raziskavi je sodelovalo 89 logopedov, zaposlenih predvsem v zdravstvu in šolstvu, nekaj udeležencev je bilo zaposlenih tudi v socialnem sektorju, zasebnem sektorju in akademskem okolju.
Ugotovili smo, da je splošna stopnja delovnega zadovoljstva med slovenskimi logopedi visoka in da so logopedi bolj zadovoljni z delom, vodjo, sodelavci, plačo in možnostmi napredovanja kot normativna populacija. Zadovoljstvo z delom, vodjo, sodelavci in možnostmi za napredovanje se med sektorji ne razlikuje. S plačo so najbolj zadovoljni logopedi, ki so zaposleni kot samostojni podjetniki, najmanj pa logopedi v zdravstvu brez klinične specializacije. Logopedi z večjim številom obravnav na teden so manj zadovoljni z delom kot logopedi z manjšim številom obravnav. Logopedi z najnižjim številom oseb v obravnavi so najbolj zadovoljni z delom, medtem ko so bili mobilni logopedi z najvišjim številom oseb v obravnavi najmanj zadovoljni. Rezultati magistrskega dela prispevajo k boljšemu razumevanju delovnega zadovoljstva logopedov v Sloveniji in dejavnikov, ki vplivajo nanj. Služijo lahko kot podlaga za izboljšanje delovnih pogojev logopedov v različnih sektorjih.A speech-language pathologist is a professional in the fields of communication, speech, language, hearing, voice, feeding, and swallowing. After completing their education, speech-language pathologists may be employed in various workplaces within the public sector (healthcare, education, and social care) or in the private sector. Employment in different settings entails diverse roles, tasks, and working conditions, which may lead to differences in work motivation and job satisfaction.
In this master’s thesis, we examined the level of job satisfaction among speech-language pathologists across different workplaces in Slovenia and the factors influencing it. We formulated ten research hypotheses, which were tested using the Job Descriptive Index (JDI), a job satisfaction questionnaire designed for the general working population. We assessed the job satisfaction of Slovenian speech-language pathologists across five dimensions: work itself, supervisor, coworkers, pay, and opportunities for promotion. The study included 89 speech-language pathologists employed primarily in healthcare and education. A smaller number of participants were also employed in the social sector, the private sector, and academia.
The results showed that the overall level of job satisfaction among Slovenian speech language pathologists is high and that they report higher satisfaction with their work, supervisor, coworkers, pay, and opportunities for promotion compared to the normative population. Job satisfaction related to work itself, supervisors, coworkers, and opportunities for promotion does not differ across sectors. Satisfaction with pay is highest among self-employed speech-language pathologists and lowest among those working in healthcare without clinical specialization. Speech-language pathologists with a higher number of therapy sessions per week are less satisfied with their work than those with fewer sessions. Speech-language pathologists with the lowest caseload report the highest job satisfaction, whereas mobile speech-language pathologists with the highest caseload report the lowest satisfaction. The findings of this master’s thesis contribute to a better understanding of job satisfaction among speech-language pathologists in Slovenia and the factors influencing it. Our findings may serve as a basis for improving working conditions for speech-language pathologists across different sectors

This paper analyses the function of the myriad things (wanwu 萬物) in the Zhuangzi 莊子, arguing that the text’s conception of the “myriad things’ transformation (wanwu zhi hua 萬物之化)” destabilises anthropocentrism and reconfigures the human-nature relationship. Through close analysis of key passages, I elucidate the ways in which the Zhuangzi’s non-hierarchical framing of wanwu subverts materialist paradigms that privilege human agency over the nonhuman world. Central to my thesis is the paradox that the absence of environmental preoccupations in the text enables its radical decentring of anthropocentrism. This aligns with decolonial critiques of the nature/culture binary, shifting ethical responsibility away from preservationist approaches towards refusal of self-harm through unobstructive action (wuwei 無為). Such attunement prevents injury both to the self and the relational continuum of wanwu, fostering adaptation to constant transformation. Consequently, the Zhuangzi offers a pluralistic framework for rethinking human-world relations beyond anthropocentric paradigms. It grounds ethics in participatory responsiveness to wanwu’s irreducible plurality, positioning adaptability—rather than domination—as the cornerstone of coexistence.V tem članku avtor analizira funkcijo desettisočih stvari(wanwu萬物) v Zhuangziju莊子 in zagovarja tezo, da pojem »preobrazbe neštetih stvari« (wanwu zhi hua 萬物之化) v tem besedilu spodkopava antropocentrizem ter ponuja drugačno razmerje med človekom in naravo. Z natančno analizo ključnih odlomkov pokaže, kako nehierarhič-na zasnova wanwu v Zhuangziju negira materialistične paradigme, ki dajejo prednost človeškemu delovanju pred nečloveškim svetom. Osrednje izhodišče razprave je navi-dezno protislovje, ki se v Zhuangziju kaže v tem, da prav odsotnost okoljevarstvenih skr-bi omogoča radikalno razsrediščenje antropocentrizma. To se sklada s postkolonialnimi kritikami binarne opozicije narava/kultura, saj preusmerja etično odgovornost od ohran-janja k zavrnitvi samopoškodovanja prek nedelovanja (wuwei無為). Takšna uglašenost preprečuje poškodbo človeškega sebstva kot relacijskega kontinuuma wanwu in hkrati spodbuja fleksibilno prilagajanje nenehnim preobrazbam. V tem okviru Zhuangzi ponuja pluralistični okvir za ponovni razmislek o odnosih med človekom in svetom onkraj antro-pocentričnih paradigem. Tovrstno etiko utemeljuje v takšnem odzivanju na nerešljivo mnogoterost wanwu, ki temelji na zavesti dejstva, da smo vsi njegov del. Kompleksni adaptivni sistemi in kitajska filozofija: plodno sozvočje
In this paper, a connection is made between complex adaptive systems (CAS) theory and Chinese philosophy. Some central features of CAS are presented, grouped under a complexity and an adaptivity set. Under the complexity-set the following characteristics are discussed: complexity, connectedness, modularity, scales, boundaries, embeddedness, heterogeneity, perspectivism, implication centres, distributedness, robustness, and patterns. And under the adaptivity-set the following notions are discussed: adaptivity, dynamics, openness, contextuality, non-equilibrium, non-linearity, sensitivity to initial conditions, threshold effects, path dependency, emergence, self-organization, and temporality. Corresponding notions from the Chinese traditions are also presented, from different times and texts. This suggests that Chinese philosophical traditions could offer good ontologies for CAS. Conversely, CAS could be used to resist certain ossifying and mechanizing tendencies in the Chinese traditionfor instance, the idea that yin-yang and five phases can be tied to fixed entities and qualities. No homogeneous Chinese tradition is presupposed, and rather it is claimed that the ideas underlying CAS can be found across traditions and in several thinkers.Članek vzpostavlja povezavo med teorijo kompleksnih adaptivnih sistemov (KAS) in kitajsko filozofijo. Predstavljene so nekatere osrednje značilnosti KAS, razvrščene v sklop kompleksnosti in sklop adaptivnosti. V sklopu kompleksnosti članek obravnava naslednje značilnosti: kompleksnost, povezanost, modularnost, merila, meje, vpetost, heterogenost, perspektivizem, implikacijska središča, razpršenost, robustnost in vzorci. V sklopu adaptivnosti pa obravnava naslednje pojme: prilagoditvena zmožnost, dina-mika, odprtost, kontekstualnost, neravnovesje, nelinearnost, občutljivost na začetne raz-mere, pragovni učinki, odvisnost od poti, emergenca, samoorganizacija in temporalnost. Članek prav tako predstavi ustrezne pojme iz kitajskih tradicij iz različnih obdobij in besedil. To nakazuje, da bi kitajske filozofske tradicije lahko ponudile dobre ontologije za KAS. Obratno pa bi KAS lahko uporabili za odpor proti določenim togim in mehan-icističnim težnjam v kitajski tradicijina primer proti ideji, da je mogoče yin-yang ter pet faz vezati na fiksne entitete in lastnosti. Članek ne predpostavlja homogene kitajske tradicije, temveč trdi, da je mogoče ideje, ki so v temelju KAS, najti v različnih tradicijah in pri več mislecih
Electric wheelchair simulator using Stewart platform and virtual reality
V tem magistrskem delu je predstavljena zasnova, implementacija in eksperimentalna evalvacija simulatorja elektromotornega invalidskega vozička, ki združuje dinamično modeliranje, realnočasovno vodenje in tridimenzionalno virtualno okolje. Simulator povezuje fizikalno utemeljen model vzmetenja s sistemom gibanja Stewartove poščadi, kar omogoča realistično poustvarjanje odzivov el. vozička na vsakodnevne ovire, kot so klančine, stopnice in neravne površine.
Realnočasovno izvajanje je zagotovljeno z uporabo okolja Beckhoff TwinCAT 3, pri čemer vodenje in izmenjavo parametrov omogoča komunikacija po protokolu TwinCAT ADS. Virtualni prostor, razvit v okolju Unity, zagotavlja poglobljeno in prilagodljivo okolje za generiranje ponovljivih profilov terena in morebitnih interakcijskih scenarijev. Model vzmetenja vozička, prirejen po modelu avtomobilskega vzmetenja (četrtinski, polovični, celotni model), je bil ustvarjen in večstopenjsko analiziran v okolju Simulink.
Preizkus simulatorja je bil izveden s tremi testnimi osebami, ki so simulator upravljale pod enakimi pogoji. Za oceno realističnosti in uporabniško pogojenega obnašanja sistema so bili analizirani kazalniki delovanja, kot so lateralni odmik od idealne poti, navpični odziv vzmetene mase ter uporabniška izkušnja testiranih oseb. Rezultati prikazujejo dinamične odzive, razlike v načinu in stabilnosti vožnje med udeleženci, kjer smo se osredotočili na delovanje simulatorja.
Končna zasnova simulatorja predstavlja ugoden sistem za preučevanje dinamike invalidskih vozičkov, interakcije med uporabnikom in sistemom ter razvoj strategij vodenja, obenem pa ponuja potencial za prihodnje aplikacije na področju usposabljanja uporabnikov in rehabilitacijskih raziskav.In the following master’s thesis the design, implementation, and experimental evaluation of an electric wheelchair simulator that integrates dynamic modeling, real-time control and a three-dimensional virtual environment, is presented. The simulator combines a physics-based suspension model with a Stewart platform motion system, enabling realistic reproduction of the electric wheelchair’s responses to everyday obstacles such as ramps, steps, and uneven surfaces.
Real-time execution is achieved using the Beckhoff TwinCAT 3 environment, where control and parameter exchange are enabled through communication based on the TwinCAT ADS protocol. The virtual environment, developed in Unity, provides an immersive and flexible space for generating repeatable terrain profiles and potential interaction scenarios. The wheelchair suspension model, adapted from automotive suspension models (quarter-car, half-car, full-car model), was developed and subjected to multi-stage analysis in the Simulink environment.
The simulator was operationally tested through experiments involving three test subjects who operated the system under identical conditions. To assess fidelity and user-dependent behavior of the system, performance indicators such as lateral deviation from the ideal path, vertical response of the sprung mass, and the user experience of the test subjects were analyzed. The results demonstrate realistic dynamic responses and reveal differences in driving behavior and stability among participants, with a focus on the performance of the simulator.
The final simulator design represents a reliable concept for investigating wheelchair dynamics, user–system interaction, and control strategy development, while also offering potential for future applications in user training and rehabilitation research
α‑enolase and γ‑enolase expression in enriched S- and N-type SH-SY5Y cells
Enolase is well-known for its role in glycolysis but also plays other roles in the central nervous system, including neuronal survival, differentiation, and axonal regeneration. Here, we investigated α- and γ-enolase expression patterns and their association with cathepsin X in distinct SH-SY5Y cell phenotypes. Enriched substrate-adherent S-type cells are characterized by large, flat morphology with extensive cytoplasm and higher expression of vimentin, while neuroblastic N-type are recognized by neurite extensions and higher expression of B-cell lymphoma 2 (Bcl-2) and growth-associated protein-43. We demonstrated that γ-enolase expression was specific to N-type cells, whereas α-enolase expression was not phenotype-specific. Moreover, a shift from ubiquitously expressed α-enolase to neuron-specific γ-enolase was observed during the enrichment and differentiation. Additionally, cathepsin X exhibited higher proteolytic activity in S-type cells. Inhibition of cathepsin X with AMS36 promoted differentiated cell morphology and increased expression of the active form of γ-enolase. Furthermore, AMS36 altered the expression of vimentin and Bcl-2, indicating a regulatory role in neuronal differentiation. Furthermore, AMS36 activated extracellular signal-regulated kinase 1/2 in N-type cells and enhanced the association between γ-enolase and tyrosine receptor kinase in both, suggesting a link between cathepsin X/γ-enolase and the key signaling pathways of differentiation. Our findings underscore the multifaceted role of enolase isoforms in SH-SY5Y cell differentiation, with α-enolase and γ-enolase showing distinct expression patterns in S- and N-type cells. The expression and activity of cathepsin X in S-type cells, along with its regulatory impact on γ-enolase in N-type cells, highlight the importance of these proteins in neuronal differentiation
Needs of Children Who Are Adopted to Explore Their Biological Roots
Posvojitev je poseben socialnovarstveni ukrep države, katerega namen je delati v največjo korist otrok. Od strokovnih služb se pričakuje, da bodo ugotovile otrokove potrebe, na osnovi katerih bodo potem za otroka poiskale ustrezne posvojiteljske starše.
V diplomskem delu sem raziskovala potrebe otrok, ki so posvojeni, po spoznavanju in raziskovanju bioloških korenin ter kako strokovne službe razumejo in ravnajo s temi potrebami. V raziskavi se je pokazalo, da so potrebe posvojenih otrok po raziskovanju bioloških korenin močne, saj želijo razumeti svoj izvor, biološke starše in genetsko dediščino. Te potrebe so pogosto povezane z iskanjem identitete, občutkom pripadnosti, radovednostjo o zdravstveni zgodovini in genetskih značilnostih. Z raziskavo se je pokazalo tudi, da strokovne službe slabo razumejo potrebe oseb, ki so posvojene. Te potrebe niso pomembne samo v otroštvu, temveč tudi v obdobju odraščanja, ko se srečujejo s številnimi pomembnimi vprašanji, kot so: komu sem podoben, od kod izhajam, zakaj je prišlo do posvojitve.
Vzorec je zajemal deset posvojenih oseb, ki so bile posvojene v Sloveniji in so bile ob posvojitvi dovolj mlade, da se svojih bioloških staršev oziroma biološke družine ne spominjajo. V raziskavi me je zanimalo, kakšni so njihovi odnosi v posvojiteljski družini, kako ti odnosi vplivajo na potrebo po raziskovanju biološkega izvora in na pogled na biološke starše. Zanimalo me je tudi, kakšna so pričakovanja oseb, ki so posvojene, do strokovnih delavcev, ki delajo na področju posvojitev. Prav tako me je zanimalo, kaj bi osebe, ki so posvojene, še potrebovale od stroke in kako močna je dejanska potreba po spoznavanju bioloških staršev.
Rezultati raziskave so pokazali, da so potrebe po spoznavanju bioloških staršev zelo močne. Potrebe po raziskovanju bioloških korenin niso odvisne od odnosov v posvojiteljski družini, a se v primeru slabih odnosov poveča stiska otroka, ki je posvojenpojavijo se lahko občutki krivde ob raziskovanju. Potreba po odprtem komuniciranju in sprejetosti v družini je ključna potreba otrok, ki so posvojeni. Pri osebah, kjer so se doma odprto pogovarjali o posvojitvi, se kaže manj težav v odnosih in večja sproščenost pri pogovoru o posvojitvi.
Raziskava kaže, da osebe, ki so sodelovale v intervjuju, večinoma nimajo dobrih izkušenj s socialnimi delavci na področju posvojitev. Želele bi si več podpore in večje razumevanje strokovnih služb. Prav tako se jim zdi pomembno, da so socialni starši na prihod otroka dobro pripravljeni ter da bi bil posvojenim osebam na voljo tudi program, kjer bi se lahko družile in med seboj delile svoje izkušnje.
Vse intervjuvane osebe se strinjajo, da je pravica vedeti za svoj izvor osnovna človekova pravica in bi morala biti omogočena vsem.Adoption is a specific social welfare measure implemented by the state with the aim of acting in the best interests of the child. Professional services are expected to assess the child’s needs, on the basis of which they then seek suitable adoptive parents.
In my diploma thesis, I explored the needs of adopted children regarding their wish to learn about and explore their biological roots, as well as how professional services understand and respond to these needs. The research showed that the needs of adopted children to explore their biological origins are strong, as they wish to understand their background, biological parents, and genetic heritage. These needs are often connected to the search for identity, a sense of belonging, and curiosity about health history and genetic characteristics. The findings also revealed that professional services often have a limited understanding of the needs of adopted individuals. These needs are important not only in childhood but also during adolescence, when individuals face significant questions such as whom they resemble, where they come from, and why the adoption took place.
My sample consisted of ten adopted individuals who were adopted in Slovenia and were young enough at the time of adoption that they do not remember their biological parents or biological family. I investigated the relationships within their adoptive families, how these relationships influence their need to explore their biological origins, and their perceptions of their biological parents. I was also interested in their expectations of professionals working in the field of adoption, what further support adopted individuals would need from these services, and how strong the need to learn about their biological parents truly is.
The results showed that the need to learn about one’s biological parents is very strong. The need to explore biological roots is not dependent on the quality of relationships within the adoptive family, although poor relationships may increase the distress of the adopted child, who may experience feelings of guilt when exploring these questions. The need for open communication and acceptance within the family emerged as a key need of adopted children. Among individuals whose families openly discussed adoption, fewer relational difficulties and greater ease in talking about adoption were observed.
The research indicated that the interview participants generally do not have positive experiences with social workers in the field of adoption. They would like more support and greater understanding from professional services. They also consider it important that social parents are well prepared for the child’s arrival and that adopted individuals have access to programmes where they can meet and share their experiences with one another.
All interviewees agreed that the right to know one’s origins is a fundamental human right and should be available to everyone
Using artificial intelligence for schedule generation in health institutions
Zadovoljstvo zaposlenih je zelo pomemben vidik v delovnem okolju. K temu prispevajo tudi obvezna dežurstva, za katera želimo, da so skladna z željami osebja. Ker trenuten sistem izbiranja in dodeljevanja dežurstev v določeni zdravstveni ustanovi ne izpolnjuje pričakovanj, smo se odločili razviti sistem, ki bo to izboljšal. Namesto priprave lastnega algoritma smo pri generiranju urnika preizkusili uporabnost umetne inteligence, kjer smo pozornost namenili produktom podjetja OpenAI, in sicer modelom GPT-3.5-turbo, GPT-4.1 in GPT-5 ter pogovornemu robotu ChatGPT. Modele smo testirali na enakih vhodnih podatkih in jih ovrednotili na podlagi njihovega izhoda, kjer smo preverjali enakomernost razporeditve dela in upoštevanje želja ter dnevov počitka. Pogovorni robot je imel drugačno nalogo, saj je moral pripraviti kodo, ki generira urnik. Ustreznost kode smo ocenili z enakimi vhodnimi podatki kot pri modelih. Ugotovili smo, da se pri generiranju urnika najbolj izkaže model GPT-5. Končni izdelek je spletna aplikacija, ki vsakemu članu osebja omogoča izbiro preference dežurstva po dnevih v mesecu in pri mesečnem razporedu poskusi izpolniti vse zahteve.The contentment of employees is a very import aspect in working environment. Mandatory shifts, which we want to be in accordance with staff\u27s wishes, also contribute to that. Because the current selection and assignment system in particular healthcare facility does not reach expectations, we decided to develop a system, which will provide improvements in that department. Instead of writing our own algorithm, we have tried to use the capabilities of artificial intelligence in solving the problem of scheduling. We monitored closely the products of company OpenAI, specifically models GPT-3.5-turbo, GPT-4.1 and GPT-5 and chatbot ChatGPT.We tested the models with the same input data and evaluated them based on their output, where we checked the evenness of the workload and the consideration of doctors\u27 preferences and rest days.The chatbot had a different assignment, because it had to prepare the code, which then generates the schedule.We assessed the suitability of the code with the same input as the models.We found out that the model GPT-5 performs best in generating the schedule.The final products is a web application, that allows each staff member to select their shift preferences by day of the month and attempts to meet all requirements for created monthly schedule
