    Facial and mandibular landmark tracking with habitual head posture estimation using linear and fiducial markers

    This study compared the accuracy of facial landmark measurements using deep learning-based fiducial marker (FM) and arbitrary width reference (AWR) approaches. It quantitatively analysed mandibular hard and soft tissue lateral excursions and head tilting from consumer camera footage of 37 participants. A custom deep learning system recognised facial landmarks for measuring head tilt and mandibular lateral excursions. Circular fiducial markers (FM) and inter-zygion measurements (AWR) were validated against physical measurements using electrognathography and electronic rulers. Results showed notable differences in lower and mid-face estimations for both FM and AWR compared to physical measurements. The study also demonstrated the comparability of both approaches in assessing lateral movement, though fiducial markers exhibited variability in mid-face and lower face parameter assessments. Regardless of the technique applied, hard tissue movement was typically seen to be 30% less than soft tissue among the participants. Additionally, a significant number of participants consistently displayed a 5 to 10° head tilt

    Role reversals: non-canonical roles for immune and non-immune cells in the gut

    The intestine is home to an intertwined network of epithelial, immune, and neuronal cells as well as the microbiome, with implications for immunity, systemic metabolism, and behavior. While the complexity of this microenvironment has long since been acknowledged, recent technological advances have propelled our understanding to an unprecedented level. Notably, the microbiota and non-immune or structural cells have emerged as important conductors of intestinal immunity, and by contrast, cells of both the innate and adaptive immune systems have demonstrated non-canonical roles in tissue repair and metabolism. This review highlights recent works in the following two streams: non-immune cells of the intestine performing immunological functions; and traditional immune cells exhibiting non-immune functions in the gut

    Nursing home managers' descriptions of multi-level barriers to leading person-centred care: A content analysis

    Background: Research suggests that person-centred care can be beneficially implemented and sustained, even though barriers remain that prevent uptake in clinical practice. Understanding barriers to person-centred care seems important, as this has an impact on care practices and resident outcomes. Moreover, there is limited knowledge about nursing home managers' descriptions of barriers when leading person-centred care. Objectives: To explore barriers to leading person-centred care as narrated by nursing home managers. Methods: A descriptive qualitative design was used to collect data using individual interviews with 12 nursing home managers in highly person-centred nursing homes. Data were analysed using content analysis. Results: Multi-level barriers to leading person-centred care were identified on the (1) person level, (2) team level and (3) organisational level. Placing professional and family considerations ahead of resident considerations was described as a barrier on the personal level (1). Also, staff's divergent care values, processes, and priorities together with turnover and low foundational knowledge were identified as barriers on the team level (2). On an organisational level (3), constrained finances, functional building design and group level rostering were identified as barriers. Conclusion: Multi-level barriers influence nursing home managers' ability to lead and promote person-centred care. Promoting the development of person-centred practices requires efforts to eliminate barriers on person, team and organisational level. Implications for Practice: Identifying and overcoming barriers at various levels in nursing home care has the potential to promote person-centred practices. This study can inform stakeholders and policymakers of challenges and complexities in person-centred practices. Multi-level strategies are needed to target challenges at person-, team- and organisational level when striving to develop person-centred care

    Fungi associated with the ambrosia beetle Xyleborus perforans (Coleoptera: Curculionidae: Scolytinae) on drought-stressed Pinus in New South Wales, Australia

    Ambrosia beetles have co-evolved symbiotic relationships with an array of fungal partners. Mutualistic fungal partners are often highly successful in vertical transmission between beetle generations. These persisting relationships can alter beetle behaviour, resulting in the opportunity to occupy new ecological niches and to spread geographically. In Australia, ambrosia beetles are not currently considered a significant pest in commercial Pinus plantations, where the bark beetle Ips grandicollis is known as the primary invader of stressed trees. However, in 2019, ambrosia beetles Xyleborus perforans and X. bispinatus, co-occurring with I. grandicollis, were found to have colonised a large proportion of drought-stressed trees in commercial Pinus plantations in north-east New South Wales. In this study, X. perforans (the most prevalent of two Xyleborus spp.) was collected from infested dead and dying trees in two NSW Pinus plantations. Fungal isolates of suspected Pinus pathogens were recovered from beetle mycangia and exoskeletons as well as ambrosia beetle galleries. Morphological examination and multilocus sequence analysis identified five fungi associated with X. perforans: Fusarium parceramosum, Fusarium aff. solani, Ophiostoma ips, Raffaelea deltoideospora and Sporothrix pseudoabietina. For Australia, this is the first report of F. parceramosum, as well as the first records of O. ips, R. deltoideospora and S. pseudoabietina being vectored by Xyleborus. Pathogenicity tests were performed on seedlings of three Pinus spp., with O. ips producing significantly longer lesions than the other fungi. This study demonstrates the potential for seemingly harmless ambrosia beetles to vector plant pathogens in Australian forests, providing a mode of disease transmission that should be considered in plantation management and forest biosecurity

    Molecular insights into the HLA-B35 molecules' classification associated with HIV control

    Human leukocyte antigen (HLA) class I molecules have been shown to influence the immune response to HIV infection and acquired immunodeficiency syndrome progression. Polymorphisms within the HLA-B35 molecules divide the family into two groups, namely, Px and PY. The Px group is associated with deleterious effects and accelerated disease progression in HIV+ patients, whereas the PY group is not. The classification is based on the preferential binding of a tyrosine at the C-terminal part of the peptide in the PY group, and a nontyrosine residue in the Px group. However, there is a lack of knowledge on the molecular differences between the two groups. Here, we have investigated three HLA-B35 molecules, namely, HLA-B*35:01 (PY), HLA-B*35:03 (Px) and HLA-B*35:05 (unclassified). We selected an HIV-derived peptide, NY9, and demonstrated that it can trigger a polyfunctional CD8+ T-cell response in HLA-B*35:01+/HIV+ patients. We determined that in the complex with the NY9 peptide, the PY molecule was more stable than the Px molecule. We solved the crystal structures of the three HLA molecules in complex with the NY9 peptide, and structural similarities with HLA-B*35:01 would classify the HLA-B*35:05 within the PY group. Interestingly, we found that HLA-B*35:05 can also bind a small molecule in its cleft, suggesting that small drugs could bind as well

    Acceptability and Feasibility of a Smartphone-Based Real-Time Assessment of Suicide Among Black Men: Mixed Methods Pilot Study

    Background: Suicide rates in the United States have increased recently among Black men. To address this public health crisis, smartphone-based ecological momentary assessment (EMA) platforms are a promising way to collect dynamic, real-time data that can help improve suicide prevention efforts. Despite the promise of this methodology, little is known about its suitability in detecting experiences related to suicidal thoughts and behavior (STB) among Black men. Objective: This study aims to clarify the acceptability and feasibility of using smartphone-based EMA through a pilot study that assesses the user experience among Black men. Methods: We recruited Black men aged 18 years and older using the MyChart patient portal messaging (the patient-facing side of the Epic electronic medical record system) or outpatient provider referrals. Eligible participants self-identified as Black men with a previous history of STB and ownership of an Android or iOS smartphone. Eligible participants completed a 7-day smartphone-based EMA study. They received a prompt 4 times per day to complete a brief survey detailing their STB, as well as proximal risk factors, such as depression, social isolation, and feeling like a burden to others. At the conclusion of each day, participants also received a daily diary survey detailing their sleep quality and their daily experiences of everyday discrimination. Participants completed a semistructured exit interview of 60-90 minutes at the study’s conclusion. Results: In total, 10 participants completed 166 EMA surveys and 39 daily diary entries. A total of 4 of the 10 participants completed 75% (21/28) or more of the EMA surveys, while 9 (90%) out of 10 completed 25% (7/28) or more. The average completion rate of all surveys was 58% (20.3/35), with a minimum of 17% (6/35) and maximum of 100% (35/35). A total of 4 (40%) out of 10 participants completed daily diary entries for the full pilot study. No safety-related incidents were reported. On average, participants took 2.08 minutes to complete EMA prompts and 2.72 minutes for daily diary surveys. Our qualitative results generally affirm the acceptability and feasibility of the study procedures, but the participants noted difficulties with the technology and the redundancy of the survey questions. Emerging themes also addressed issues such as reduced EMA survey compliance and diminished mood related to deficit-framed questions related to suicide. Conclusions: Findings from this study will be used to clarify the suitability of EMA for Black men. Overall, our EMA pilot study demonstrated mixed feasibility and acceptability when delivered through smartphone-based apps to Black men. Specific recommendations are provided for managing safety within these study designs and for refinements in future intervention and implementation science research

    Combined absence of TRP53 target genes ZMAT3, PUMA and p21 cause a high incidence of cancer in mice

    Transcriptional activation of target genes is essential for TP53-mediated tumour suppression, though the roles of the diverse TP53-activated target genes in tumour suppression remains poorly understood. Knockdown of ZMAT3, an RNA-binding zinc-finger protein involved in regulating alternative splicing, in haematopoietic cells by shRNA caused leukaemia only with the concomitant absence of the PUMA and p21, the critical effectors of TRP53-mediated apoptosis and cell cycle arrest respectively. We were interested to further investigate the role of ZMAT3 in tumour suppression beyond the haematopoietic system. Therefore, we generated Zmat3 knockout and compound gene knockout mice, lacking Zmat3 and p21, Zmat3 and Puma or all three genes. Puma −/− p21 −/− Zmat3 −/− triple knockout mice developed tumours at a significantly higher frequency compared to wild-type, Puma −/− Zmat3 −/− or p21 −/− Zmat3 −/−deficient mice. Interestingly, we observed that the triple knockout and Puma −/− Zmat3 −/− double deficient animals succumbed to lymphoma, while p21 −/− Zmat3 −/− animals developed mainly solid cancers. This analysis suggests that in addition to ZMAT3 loss, additional TRP53-regulated processes must be disabled simultaneously for TRP53-mediated tumour suppression to fail. Our findings reveal that the absence of different TRP53 regulated tumour suppressive processes changes the tumour spectrum, indicating that different TRP53 tumour suppressive pathways are more critical in different tissues

    Morphology of the cornea and iris in the Australian lungfish Neoceratodus forsteri (Krefft 1870) (Dipnoi): Functional and evolutionary perspectives of transitioning from an aquatic to a terrestrial environment

    The Australian lungfish, Neoceratodus forsteri (Krefft 1870), is the sole extant member of the Ceratodontidae within the Dipnoi, a small order of sarcopterygian (lobe-finned) fishes, that is thought to be the earliest branching species of extant lungfishes, having changed little over the last 100 million years. To extend studies on anatomical adaptations associated with the fish–tetrapod transition, the ultrastructure of the cornea and iris is investigated using light and electron (transmission and scanning) microscopy to investigate structure–function relationships and compare these to other vertebrate corneas (other fishes and tetrapods). In contrast to previous studies, the cornea is found to have only three main components, comprising an epithelium with its basement membrane, a stroma with a Bowman's layer and an endothelium, and is not split into a dermal (secondary) spectacle and a scleral cornea. The epithelial cells are large, relatively low in density and similar to many species of non-aquatic tetrapods and uniquely possess numerous surface canals that contain and release mucous granules onto the corneal surface to avoid desiccation. A Bowman's layer is present and, in association with extensive branching and anastomosing of the collagen fibrils, may be an adaptation for the inhibition of swelling and/or splitting of the stroma during its amphibious lifestyle. The dorsal region of the stroma possesses aggregations of pigment granules that act as a yellow, short wavelength-absorbing filter during bright light conditions. Desçemet's membrane is absent and replaced by an incomplete basement membrane overlying a monocellular endothelium. The iris is pigmented, well-developed, vascularised and contractile containing reflective crystals anteriorly. Based upon its ultrastructure and functional adaptations, the cornea of N. forsteri is more similar to amphibians than to other bony fishes and is well-adapted for an amphibious lifestyle.</p

    Morphological Strategies in Ant Communities along Elevational Gradients in Three Mountain Ranges

    Species traits often vary in a coordinated manner, making up an ecological strategy comprised of suites of interrelated traits. Environmental gradients, such as those along elevational gradients, provide an ideal venue in which to examine variation in ecological strategies with the environment. We examined variation in the morphological strategies of ants along elevational gradients on thirteen mountains across three mountain ranges in central and south-eastern Australia. We pitfall-trapped ants, counted and identified workers and measured morphological traits. Most species showed a hump-shaped relationship between occurrence and elevation, and several responded to microhabitat variables. Morphological traits varied along two key axes: “gracility”, where high values indicated longer-legged species with dorsally positioned eyes and smooth, bare cuticles; and “size and darkness”, where species with high values were larger and darker. Analysis of assemblage-weighted means revealed that gracility decreased with temperature and increased with precipitation, suggesting links with desiccation tolerance. Size and darkness increased with UV-B, declined with increasing canopy cover and peaked at mid-temperatures. We thus detected strong shifts in dominant morphological strategies along our elevational gradients. However, the multifunctionality and interrelatedness of traits and the covariance of climatic factors may make isolation of the function of individual traits difficult. Further, the predictive power of our models may be limited in the context of novel environments predicted under global change

    Caring Self-Efficacy of Personal Care Attendants From English-Speaking and Non-English-Speaking Countries Working in Australian Residential Aged Care Settings

    Objectives: This study compared the caring self-efficacy between personal care attendants (PCAs) from English-speaking and non-English-speaking countries, controlling for potential sociodemographic and work-related covariates. PCAs’ perceptions of their caring self-efficacy were further explored. Methods: An independent samples t-test was used to determine the mean difference in the caring self-efficacy score between the two groups. A multivariate analysis was conducted to adjust for covariates. Thematic analysis was conducted on open-ended responses. Results: The results showed that caring self-efficacy was significantly influenced by whether participants primarily spoke English at home rather than where they were born. Younger age and everyday discrimination experiences were negatively associated with caring self-efficacy. Both groups perceived that inadequate resources and experiencing bullying and discrimination reduced their caring self-efficacy. Discussion: Access to organisational resources and training opportunities and addressing workplace bullying and discrimination against PCAs, particularly younger PCAs and those from non-English-speaking backgrounds, could improve their caring self-efficacy.</p
