The Australian Microbiome Initiative for accurate detection and ecological modelling of fungi
This repository contains processed topsoil ITS data from the Australian Microbiome Initiative, covering over 2000 plots in Australia and Antarctica. Our bioinformatic analysis followed a conservative approach and best practices in metabarcoding. We paired this dataset with over 100 predictor variables to fast-track data exploration. The code use to produce this dataset can be found at GitHub.</p
Lifestyle Intervention in Reducing Insulin Resistance and Preventing type 2 Diabetes in Asia Pacific Region: A Systematic Review and Meta-Analysis
Purpose of Review: To update the evidence of lifestyle interventions for the prevention of type 2 diabetes mellites (T2DM) in adults, particularly in the Asia Pacific region. The key questions to ask are: 1) How effective are lifestyle interventions in preventing T2DM among at-risk adults in the Asia Pacific Region? 2)What are the key characteristics of the implementation of lifestyle interventions for diabetes prevention? Recent Findings: Lifestyle interventions for the prevention of T2DM have been suggested to be effective. There is evidence of ethnic differences in some glycaemic and anthropometric outcomes. Summary: The meta-analysis suggested a significant result in reducing waist circumference (standardised mean difference − 019, 95%CI (-0.31, -0.06)), and no significant effects in other outcomes. However, the implementation outcomes suggested lifestyle intervention might be a cost-effective and sustainable approach in T2DM particularly in countries in the Asia Pacific Region. The focus of lifestyle intervention in the Asia Pacific Region should not only lie in the effectiveness of the trial but a thorough evaluation of the implementation outcomes, as well as cultural adaptations, with the support of all stakeholders through all stages of the implementation
Serum and CSF metabolomics analysis shows Mediterranean Ketogenic Diet mitigates risk factors of Alzheimer’s disease
Alzheimer's disease (AD) is influenced by a variety of modifiable risk factors, including a person's dietary habits. While the ketogenic diet (KD) holds promise in reducing metabolic risks and potentially affecting AD progression, only a few studies have explored KD's metabolic impact, especially on blood and cerebrospinal fluid (CSF). Our study involved participants at risk for AD, either cognitively normal or with mild cognitive impairment. The participants consumed both a modified Mediterranean Ketogenic Diet (MMKD) and the American Heart Association diet (AHAD) for 6 weeks each, separated by a 6-week washout period. We employed nuclear magnetic resonance (NMR)-based metabolomics to profile serum and CSF and metagenomics profiling on fecal samples. While the AHAD induced no notable metabolic changes, MMKD led to significant alterations in both serum and CSF. These changes included improved modifiable risk factors, like increased HDL-C and reduced BMI, reversed serum metabolic disturbances linked to AD such as a microbiome-mediated increase in valine levels, and a reduction in systemic inflammation. Additionally, the MMKD was linked to increased amino acid levels in the CSF, a breakdown of branched-chain amino acids (BCAAs), and decreased valine levels. Importantly, we observed a strong correlation between metabolic changes in the CSF and serum, suggesting a systemic regulation of metabolism. Our findings highlight that MMKD can improve AD-related risk factors, reverse some metabolic disturbances associated with AD, and align metabolic changes across the blood-CSF barrier.</p
Organisational culture in ‘better’ group homes for adults with intellectual and developmental disabilities in England: A qualitative study
Background: Previous research identifies organisational culture as one of a number of factors associated with the quality of life outcomes of group home residents' with intellectual and developmental disabilities. This study aims to elaborate on the dimensions of group home culture in settings in England. Method: Participant observations and semi-structured interviews with staff were carried out in two group homes. Field-notes, interview notes and transcripts were analysed using inductive thematic analysis by a researcher naïve to the project and the previous literature. Initial coding was re-examined after sensitisation to theorised models in previous literature to identify the most parsimonious fit. The two settings were rated and compared using a five-point Likert scale for each of the dimensions. Results: The findings describe group home culture across seven dimensions. There were mixed ratings across the different dimensions reflecting inconsistencies in culture that were reflected in staff practice. The challenge in assigning a global rating of culture in group homes, which includes interactions across multiple staff and multiple residents over time, was highlighted. Conclusion: The development of an observational measure of culture is highlighted as potentially helpful in understanding and responding to culture in services for individuals with intellectual and developmental disabilities
Development and validation of the polycultural identity scale
Abstract: While globalisation brings opportunities for cultural enrichment, it may also present challenges as individuals attempt to make sense of and integrate fragmented and often conflicting cultural influences into a coherent self‐concept. A qualitative inquiry into contemporary Australians' cultural identity experiences indicated that some people perceive themselves as a product of multiple influences from diverse contacts and learnings, and they mix and reconcile these influences into their self‐concept. Building on this prior work, the current study developed and tested a new quantitative measure of polycultural identity. Using two diverse Australian samples (N = 301; N = 525) recruited from Prolific, exploratory and confirmatory factor analyses supported a single‐factor structure and a high Cronbach's α among the final 10 items. Correlational analyses revealed that, as expected, the Polycultural Identity Scale (PIS) was more strongly linked with the endorsement of polycultural ideology than multicultural ideology and was uncorrelated with colourblind ideology. Also, as expected, the PIS correlated moderately with generalised identity blendedness, interest in diverse contact and endorsement of global citizenship but not with identity compartmentalisation or generalised identity harmony. The PIS has the potential to provide valuable insights into the dynamics of multifaceted cultural identity and how individuals organise and synthesise a breadth of cultural influences into a meaningful self‐concept.</p
ChemsexPH: The association between chemsex, HIV status and adherence to antiretroviral therapy among men who have sex with men in the Philippines
Introduction: Chemsex, the use of psychotropic drugs before or during sexual intercourse, is associated with various HIV risk factors, including condomless sex and reduced adherence to pre-exposure prophylaxis or antiretroviral therapy (ART). In the Philippines, there are still limited studies exploring the associations between chemsex, HIV status and ART adherence. This study aims to compare recent and lifetime chemsex engagement in association with self-reported HIV status among Filipino men who have sex with men (MSM). We further explored the association between chemsex and ART adherence among people living with HIV engaged in chemsex. Methods: A cross-sectional online survey of 479 Filipino MSM was conducted from 3 August to 1 December 2019. Demographic profiles, sexual behaviours, drug use, history of sexually transmitted infections (STIs), chemsex engagement and HIV status were collected and analysed. Bivariable and multivariable logistic regression were employed to assess the association between self-reported HIV status and chemsex engagement. Results: Among the 479 respondents, Filipino MSM engaged in drug use and chemsex were generally older compared to those not engaged in drug use and chemsex (average age 31−33 vs. 29 years old; p<0.05). Methamphetamine was the most common drug for people who reported using drugs. An HIV-positive status was associated with recent chemsex engagement (aOR = 5.18, p<0.05) and a history of STIs (aOR = 2.09, p<0.05). The subgroup analysis showed that 79% (166/200) of persons living with HIV were adherent to ART. There was no significant association found between chemsex and ART adherence in the logistic regression analyses. Conclusions: Chemsex behaviour, particularly recent chemsex engagement, is significantly associated with self-reported HIV status. The emerging data on MSM engaged in chemsex require integration of a more person-centred, comprehensive and robust harm reduction programmes into the existing combination prevention strategies in the country. Health education for Filipino MSM engaged in chemsex should prioritize raising awareness about methamphetamine effects and overdose risks, alongside proper medical management
Single-extracellular vesicle (EV) analyses validate the use of L1 Cell Adhesion Molecule (L1CAM) as a reliable biomarker of neuron-derived EVs
Isolation of neuron-derived extracellular vesicles (NDEVs) with L1 Cell Adhesion Molecule (L1CAM)-specific antibodies has been widely used to identify blood biomarkers of CNS disorders. However, full methodological validation requires demonstration of L1CAM in individual NDEVs and lower levels or absence of L1CAM in individual EVs from other cells. Here, we used multiple single-EV techniques to establish the neuronal origin and determine the abundance of L1CAM-positive EVs in human blood. L1CAM epitopes of the ectodomain are shown to be co-expressed on single-EVs with the neuronal proteins β-III-tubulin, GAP43, and VAMP2, the levels of which increase in parallel with the enrichment of L1CAM-positive EVs. Levels of L1CAM-positive EVs carrying the neuronal proteins VAMP2 and β-III-tubulin range from 30% to 63%, in contrast to 0.8%–3.9% of L1CAM-negative EVs. Plasma fluid-phase L1CAM does not bind to single-EVs. Our findings support the use of L1CAM as a target for isolating plasma NDEVs and leveraging their cargo to identify biomarkers reflecting neuronal function
Evaluating the risk to Australia's flora from Phytophthora cinnamomi
Context: Phytophthora cinnamomi Rands is a destructive pathogen of Australian native vegetation, often causing permanent damage to ecosystems and threatening the survival of rare, susceptible species. Despite that, much information about the effects of P. cinnamomi on plant species remains unpublished and the risk of extinction to most species is unknown. Aims: We aimed to classify the risk of extinction from P. cinnamomi to Australian native plants. Methods: We used available data and personal knowledge about P. cinnamomi effects on plants, spatial data on plant species distribution and habitat suitability of P. cinnamomi to assign an extinction-risk category of low, moderate, high or very high. Key results: There are currently 65 plant species at a very high risk of extinction in Australia as a result of P. cinnamomi infection. The genera Andersonia, Banksia, Darwinia, Daviesia, Epacris, Gastrolobium, Grevillea, Hibbertia, Isopogon, Lambertia, Latrobea, Leucopogon, Phebalium and Styphelia have multiple species at a very high risk of extinction, most of which occur in south-western Western Australia. Conclusions: The available data confirmed the high risk to the Australian flora from P. cinnamomi and identified species in plant families not previously known to be affected, highlighting data gaps (e.g. lack of knowledge about effects and risk in orchids and grasses). Implications: Much more work is required to fully understand the risk from P. cinnamomi (and other Phytophthora species) to the Australian flora
Introducing virtual reality to older adults: A qualitative analysis of a co-design innovation with care staff
Previous studies of Virtual Reality (VR) in aged care settings have demonstrated that the benefits can be multiple, including improved social connection and engagement and reduced social isolation in later life. However, there remains a lack of widespread uptake of VR in aged care facilities. This prompts an important question: Given the potential benefits, why is there such poor engagement in VR by aged care facilities? The aim of this qualitative study is to investigate the experience of introducing VR into an aged care facility. Our innovative approach supported care staff to introduce VR into aged care facilities. Fieldwork diaries and focus group discussions were used to document experiences of introducing VR, including the obstacles, challenges and benefits and the adaptations to aged care environments that were required to accommodate new VR activities. Thematic analysis of the data revealed that VR can be an important medium to support the wellbeing of older residents. However, our findings also indicate that there are significant challenges associated with introducing VR, including substantial costs in time, money and institutional resources and attention. This study concludes that, to be successful, VR requires substantive care and relational resources from both staff and residents that are only visible when paying attention to the contextual adjustments required to introduce the technology to a new setting. This suggests that other research on gerontechnologies would likely also benefit from further attention to the role of the broader social context – including care and relational resources – in ensuring their successful design and implementation
