Metabolism of 2,2ʼ,3,4,4ʼ,5,6ʼ-Heptachlorobiphenyl (CB182) by Rat, Guinea Pig and Human Liver Microsomes
The in vitro metabolism of 2,2ʼ,3,4,4ʼ,5,6ʼ-heptachlorobiphenyl (CB182) by rat, guinea pig and human liver microsomes was compared and the effects of cytochrome P450 (CYP) inducers, phenobarbital (PB) and 3-methylcholanthrene (MC), on CB182 metabolism were examined. Only one metabolite was produced by rat, guinea pig and human liver microsomes and the order of the activity was rat (PB-treated)>> guinea pig (PB-treated)> guinea pig (untreated, MC-treated)> human > rat (untreated). Pretreatment of PB resulted in the remarkable increase of the metabolite in rats (1,370 pmol/hr/mg protein) and a slight increase in guinea pigs (27 pmol/hr/mg protein). In contrast, MC treatment to rats and guinea pigs decreased M-1. By comparison of GC-MS data of the methylated M-1 with a synthesized authentic sample, M-1 was determined to be 3ʼ-hydroxy (OH)-CB182. These results suggest that 3ʼ-OH-CB182 is a major metabolite formed by PB-inducible CYP2B enzymes in both animals and rat CYP2B enzymes possess much higher activity to hydroxylate CB182 than guinea pig and human CYP2B enzymes
Inter-Laboratory Cross-Comparison Study of Dioxins and PCBs Analyses in Human Blood Samples (Part 2)
We developed an analytical method for accurately determining the concentrations of dioxins and polychlorinated biphenyls (PCBs) in a blood volume of 5 g. Inter-laboratory cross-comparison is available for maintaining the quality of dioxins and PCBs analyses. We carried out the cross-comparison of dioxin and PCBs analyses in the blood with domestic analysis organizations. Two cross-comparison studies each of dioxins and PCBs analyses were carried out from 2012 through 2015. As a result, the findings of our analysis accorded well with the analyses of the other organizations. It was found that our analytical method is sufficiently reliable

Thyroid Function in Patients with Yusho
To investigate chronic effects of polychlorinated biphenyls (PCBs) and polychlorinated dibenzofurans (PCDFs) on thyroid functions, serum concentrations of thyroid stimulating hormone (TSH), triiodothyronine (T_3), thyroxine (T_4) and free T_4 were studied in serum of 197 patients with Yusho and 42 control subjects in 2015. Serum levels of T_3 were decreased in patients with Yusho compared with controls. A significant relationship between concentrations of PCB in the blood and serum levels of T_3 was observed, while there was no correlation between blood PCB concentrations and TSH, T_4 or free T_4 levels. Serum levels of T_3 were significantly decreased in the subjects with high PCB concentrations compared with those with low PCB concentrations. Furthermore, serum levels of T_3 were significantly decreased in the subjects with high 2,3,4,7,8-pentachlorodibenzofuran (PeCDF) concentrations compared with those with low 2,3,4,7,8-PeCDF concentrations. We conclude that serum T_3 levels decrease in patients with Yusho, and it may be associated with PCBs and 2,3,4,7,8-PeCDF in the blood
Peripheral Membrane Interactions Boost the Engagement by an Anti-HIV-1 Broadly Neutralizing Antibody.
The 4E10 antibody displays an extreme breadth of HIV-1 neutralization and therefore constitutes a suitable model system for structure-guided vaccine design and immunotherapeutics against AIDS. In this regard, the relevance of autoreactivity with membrane lipids for the biological function of this antibody is still a subject of controversy. To address this dispute, herein we have compared the membrane partitioning ability of the 4E10 antibody and several of its variants, which were mutated at the region of the paratope surface in contact with the membrane interface. We first employed a physical separation approach (vesicle flotation) and subsequently carried out quantitative fluorescence measurements in an intact system (spectroscopic titration), using 4E10 Fab labeled with a polarity-sensitive fluorescent probe. Moreover, recognition of epitope peptide in membrane was demonstrated by photo-cross-linking assays using a Fab that incorporated the genetically encoded unnatural amino acid p-benzoylphenylalanine. The experimental data ruled out that the proposed stereospecific recognition of viral lipids was necessary for the function of the antibody. In contrast, our data suggest that nonspecific electrostatic interactions between basic residues of 4E10 and acidic phospholipids in the membranes contribute to the observed biological function. Moreover, the energetics of membrane partitioning indicated that 4E10 behaves as a peripheral membrane protein, tightening the binding to the ligand epitope inserted in the viral membrane. The implications of these findings for the natural production and biological function of this antibody are discussed
Development of junction temperature measurement system for blue LED in operation using pulsed-laser Raman scattering
A novel method by using a pulsed-laser Raman scattering technique was employed to estimate junction temperatures of a phosphor-less blue-LED module. Observation of GaN-E_2^H Raman spectra was done successfully and temperature dependent Raman spectra of GaN were detected. This technique was applied to simultaneous estimation of the junction temperatures of the several chips in the phosphor-less blue LED module in operation. It is concluded that this technique would be a useful method for remoteness and simultaneous multipoint measurement of the junction temperature of a LED module in which several chips are integrated
