Effect of Repeated Exposure to Low Oxygen on Respiratory Metabolism and Vertical Movements in the Pen Shell Atrina lischkeana
We exposed pen shells (Atrina lischkeana) to repeated periods of low–oxygen for 30 d and measured their survival, behavior, glycogen content, and organic acid concentration. Exposure to low–oxygen had no effect on survival. Animals moved upward above the sediment under low–oxygen conditions and downward under aerobic conditions, repeating these movements for 19 d. The difference in shell height above the sediment before and after low–oxygen exposure gradually decreased; after 20 d there was no significant difference. Low–oxygen exposure also resulted in a significant decrease in glycogen content and in concentrations of malate and fumarate. Thus, repeated exposure of pen shells to low–oxygen conditions reduces glycogen content, alters metabolism, and affects vertical movements, all of which may cause ecological dysfunction. We suggest that hypoxia might be one factor contributing to the collapse of pen shell resources

Development of a Quantification Method of Harmful Raphidophytes Chattonella marina and Heterosigma akashiwo by the quantitative PCR assay
はじめに / 材料と方法 / 結果 / 考察 / 謝辞 / 要約定量PCR を用いて,海水中に存在する有害ラフィド藻Chattonella marinaおよびHeterosigma akashiwo遺伝子の検出法の検討を行った.プライマーおよびプロー ブ(TaqMan プローブ) は,C. marinaについてはmtDNA のcyt c 領域に,H. akashiwo についてはrRNA遺伝子のITS 領域に設定した.標的遺伝子の希釈系列を鋳型に用いた定量PCR の結果,高い増幅効率(C.marina:80.0 %; H. akashiwo:82.6 %)が確認され,さらにCt 値と遺伝子コピー数との間に高い正の相関(C. marina:R^2 = 0 . 991; H. akashiwo:R^2 = 0 . 998)が認められた.また,他種藻類(渦鞭毛藻,珪藻等)からDNeasy Plant Mini Kit(QIAGEN)を用いて抽出したDNA に対して非特異的増幅を示さなかった.さらに両種を対数増殖期から衰退期まで培養し,検鏡および定量PCR による細胞密度の定量を行った結果,両定量値間には有意(p<0.05)な正の相関(C. marina:R^2 =0.84;H. akashiwo:R^2 =0.96)が観察された.以上の結果から,DNA 抽出から定量PCR に至る本法の有用性が強く示唆された.We developed a quantification method of harmful raphidophytes Chattonella marina and Heterosigma akashiwo using quantitative PCR from seawater sample. Primers and probes were designed from cyt c region of mtDNA in C. marina and ITS region of rRNA in H. akashiwo as target genes for quantitative PCR. High PCR amplification efficiency (80.0% in C. marina and 82.6% in H. akashiwo) and R^2 value (0.991 in C. marina and 0.998 in H. akashiwo) between Ct value and gene copy number were observed in quantitative PCR using dilution series of the target gene fragments from 103 to 109 copies. Then, we confirmed the species selectivity of the designed primers and probes by quantitative PCR using DNA samples extracted from other algal species. In addition, we checked the accuracy of quantitative value of present method including extraction using DNeasy Plant Mini Kit (QIAGEN) using cultured algal cells. As a result, non-specific gene amplification was not detected in DNA samples extracted from other algal species tested. Also, there were significant relation between quantitative values of cell densities by microscopic observation and quantitative PCR in C. marina (R^2 = 0 . 84 ; p< 0 . 05 ) and H. akashiwo (R^2 =0.96; p<0.05) through the growth phase. These results strongly suggested the application of the present method to field monitoring of C. marina and H. akashiwo
A Nonlinear Analysis on the Euro Exchange Rate Using MF-DFA

Future researchThe objective of this study is to examine the nonlinear characteristics of the Euro exchange rate. Many nonlinear approaches have been applied in the finance field. Multifractal detrended fluctuation analysis (MF-DFA) is one of the popular methodologies employed to detect the nonlinear properties of a time series. The MF-DFA analysis is executed by removing the trend of a time series and abstracting its multifractality. This paper examines the features of the entire Euro exchange rate series and the sub-periods during the Global Financial Crisis and European Sovereign Debt Crisis. The overall pattern of the Euro exchange rate is determined. Further, the degrees of complexity during the two crisis periods are detected
Effects of Sucrose Monopalmitate under Microbubble Generation and/or Slightly Acidic Hypochlorous Water on Salmonella Enteritidis Adsorbed on the Surface of Various Materials
緒言 / 材料と方法 / 結果 / 考察 / 要約ショ糖パルミチン酸エステルおよびマイクロバブルと微酸性次亜塩素酸水(SAHW)の併用効果を,付着状態と浮遊状態のSalmonella Enteritidis に対して調べた.さらにタンパク質保持力の異なるニトロセルロース膜,PVDF 膜,Hybond- N +膜に付着させたS.Enteritidis に対する併用効果をキャベツに付着させた場合と比較検討した.浮遊状態では,マイクロバブルを発生させた100 mg/L のショ糖パルミチン酸エステルで処理してもS. Enteritidis はほとんど損傷しないことが示された.これに対して0.5 mg/L SAHW で単独処理した場合にはTSA による生菌数が3 log と大きく低下したが,ショ糖パルミチン酸エステル処理後にSAHW 処理した場合には,TSA による生菌数の低下は,1.6 log で,SAHW 単独処理よりも少なかった.キャベツの葉,ニトロセルロース膜,PVDF 膜,およびHybond-N+ 膜に付着させたS. Enteritidis に対するショ糖パルミチン酸エステルおよびSAHW 処理の効果を調べた結果,キャベツに付着させたS. Enteritidis ではショ糖パルミチン酸エステル単独処理,SAHW 単独処理および併用処理で,TSA による生菌数がコントロールに比べ,0.3,1.3,1.3 log それぞれ低下した.しかし,同様の処理後には,TSA による生菌数が,ニトロセルロース膜付着菌では,0.7,3.2,4.2 log,PVDF 膜に付着させた場合には,0.6,1.2,2.3 log,Hybond-N+ 膜付着菌では0.7,1.0,0.9 log それぞれ低下した.PVDF 膜などを用いた付着細菌のモデル系は,常在菌が存在しないため付着させる菌数をコントロールしやすく,また表面の観察がしやすいといった利点があり,付着菌に対する殺菌機構の解明に有用であると考えられる.Effects of sucrose monopalmitate under microbubble generation and/or slightly acidic hypochlorous water(SAHW)on Salmonella Enteritidis were investigated in suspension and on the surface of various materials. In suspension, S. Enteritidis was not injured by the treatment with sucrose palmitate at 100 mg/L under generation of microbubble, though the cells decreased the viable counts by 3.0 log by the treatment with SAHW at 30 mg/L. In contrast, the viable counts decreased only by 1.6 log when S. Enteritidis was treated with sucrose palmitate under microbubble generation and subsequent treatment with SAHW. The viable counts of S. Enteritidis adsorbed on the surface of cabbage leaf decreased by 0 . 3 , 1 . 3 , and 1 . 3 log by the treatment with sucrose monopalmitate under microbubble generation, SAHW and sucrose monopalmitate under microbubble generation followed by SAHW. By the same treatments, the viable counts of S. Enteritidis decreased by 0.7, 3.2, and 4.2 log in cells adsorbed on nitrocellulose membrane, 0.6, 1.2, and 2.3 log in cells on PVDF membrane, 0.7, 1.0, and 0.9 log in cells on Hybond-N+membrane, respectively. It seems that the bacteria adsorbed on the surface of PVDF membrane provide a model system to investigate the efficacy of detergents and sterilizers on decontamination of vegetables
データキュレーションへの期待と課題 : 自然科学から人文科学まで
ライブラリーサイエンス専攻シンポジウム「オープンデータとデジタルヒューマニティーズ」 : 2017年1月18日(水) 10:00-17:00 : 九州大学(箱崎)中央図書館 4F視聴覚ホール : 主催 九州大学統合新領域学府ライブラリーサイエンス専攻 : 共催 九州大学附属図書館 : 協賛 九州大学つばさプロジェクト「デジタル人文学における研究資源オープン化と研究プラットフォーム構築に向けた基盤的研究」「データキュレーション」とは何か。講演者自身の定義は、データの整理・編集・展示などを通してデータを利活用しやすい形で広く公開する活動、というものである。この概念には、科学研究に関連するデータの循環を通して、オープンイノベーションや超学際的研究などを進め、さらに研究の透明性を高めることへの期待が込められている。一方、これほど幅広い知識とスキルを有する概念に対して、それを実際に推進する人材をどう育成してどう評価するかが重大な課題として残っている。そこで、地球環境データプラットフォーム(DIAS)や人文学データプラットフォーム(COD同など、講演者が推進するプロジェクトを事例として取り上げ、自然科学から人文科学に及ぶ多様な研究データに対するキュレーシヨンのニーズと役割を論じてみたい
An Efficient Protocol of Protocorm–Like Bodies Regeneration from Callus Cultures of Gastrodia elata Blume and the Further Associations with Mycorrhizal Fungi
In this study, we reported a reliable protocol of PLBs regeneration from embryogenic callus cultures of Gastrodia elata, a fully mycoheterotrophic orchid and an endangered herbal medicinal plant. The continuous immersion cultures with TDZ (0.5 and 1 mg · L^〈–1〉) and BA (10 mg · L^〈–1〉) improved PLBs regeneration frequency and rate. Histological studies indicated that starch grains were abundant within in the cytoplasm of embryogenic callus and developing PLBs. The formation of PLBs from cell aggregates in the periphery region of embryogenic callus was supposed to be the multicellular origin pathway. After inoculated with suitable Armillaria isolates, PLBs enlarged further and resulted in the formation of tubers. The results of phylogenetic analysis and inoculation experiments indicated that the asymbiotic PLBs could establish the appropriate association with Armillaria mellea complex, including A. mellea and A. gallica. Thus, this protocol of PLBs regeneration from callus cultures is beneficial for the multiplication of selected elites, and could be used as an experimental system for the molecular investigations of the mycoheterotrophic orchid – mycorrhizal fungi interactions
