    Emotion regulation difficulties in binge eating disorder with and without the overvaluation of weight and shape

    The objective of this study was to examine the relationship between overvaluation of weight/shape ('overvaluation') and emotion regulation (ER) difficulties among women with binge eating disorder (BED) symptoms. Four groups of women were recruited from a community-based sample and compared on ER difficulties: individuals with probable BED with (n=102) and without (n=72) overvaluation, and non-binge eating obese (n=40) and healthy-weight (n=40) control participants. Data for patients with a formal diagnosis of BED receiving treatment from a previous study were included for numerical comparative purposes. Women with probable BED and overvaluation reported significantly greater ER difficulties than all other groups and had similar levels of ER difficulties to BED patients. Women with probable BED in the absence of overvaluation were comparable to the obese control group on total ER difficulties and the majority of the ER difficulties subscales. The findings provide further evidence for the clinical significance of overvaluation among individuals with BED symptomatology. BED in the absence of overvaluation does not appear to align with current models of the disorder in which ER difficulties are viewed as a core etiological mechanism. Further research is needed to elucidate the status of this presentation.7 page(s

    One protein, multiple pathologies : multifaceted involvement of amyloid β in neurodegenerative disorders of the brain and retina

    Accumulation of amyloid β (Aβ) and its aggregates in the ageing central nervous system is regarded synonymous to Alzheimer’s disease (AD) pathology. Despite unquestionable advances in mechanistic and diagnostic aspects of the disease understanding, the primary cause of Aβ accumulation as well as its in vivo roles remains elusive; nonetheless, the majority of the efforts to address pathological mechanisms for therapeutic development are focused towards moderating Aβ accumulation in the brain. More recently, Aβ deposition has been identified in the eye and is linked with distinct age-related diseases including age-related macular degeneration, glaucoma as well as AD. Awareness of the Aβ accumulation in these markedly different degenerative disorders has led to an increasing body of work exploring overlapping mechanisms, a prospective biomarker role for Aβ and the potential to use retina as a model for brain related neurodegenerative disorders. Here, we present an integrated view of current understanding of the retinal Aβ deposition discussing the accumulation mechanisms, anticipated impacts and outlining ameliorative approaches that can be extrapolated to the retina for potential therapeutic benefits. Further longitudinal investigations in humans and animal models will determine retinal Aβ association as a potential pathognomonic, diagnostic or prognostic biomarker.19 page(s

    Characterizing twitter discussions about HPV vaccines using topic modeling and community detection

    Background: In public health surveillance, measuring how information enters and spreads through online communities may help us understand geographical variation in decision making associated with poor health outcomes. Objective: Our aim was to evaluate the use of community structure and topic modeling methods as a process for characterizing the clustering of opinions about human papillomavirus (HPV) vaccines on Twitter. Methods: The study examined Twitter posts (tweets) collected between October 2013 and October 2015 about HPV vaccines. We tested Latent Dirichlet Allocation and Dirichlet Multinomial Mixture (DMM) models for inferring topics associated with tweets, and community agglomeration (Louvain) and the encoding of random walks (Infomap) methods to detect community structure of the users from their social connections. We examined the alignment between community structure and topics using several common clustering alignment measures and introduced a statistical measure of alignment based on the concentration of specific topics within a small number of communities. Visualizations of the topics and the alignment between topics and communities are presented to support the interpretation of the results in context of public health communication and identification of communities at risk of rejecting the safety and efficacy of HPV vaccines. Results: We analyzed 285,417 Twitter posts (tweets) about HPV vaccines from 101,519 users connected by 4,387,524 social connections. Examining the alignment between the community structure and the topics of tweets, the results indicated that the Louvain community detection algorithm together with DMM produced consistently higher alignment values and that alignments were generally higher when the number of topics was lower. After applying the Louvain method and DMM with 30 topics and grouping semantically similar topics in a hierarchy, we characterized 163,148 (57.16%) tweets as evidence and advocacy, and 6244 (2.19%) tweets describing personal experiences. Among the 4548 users who posted experiential tweets, 3449 users (75.84%) were found in communities where the majority of tweets were about evidence and advocacy. Conclusions: The use of community detection in concert with topic modeling appears to be a useful way to characterize Twitter communities for the purpose of opinion surveillance in public health applications. Our approach may help identify online communities at risk of being influenced by negative opinions about public health interventions such as HPV vaccines.12 page(s

    Are people with whiplash associated neck pain different to people with non-specific neck pain?

    Study Design: Secondary analysis of a prospective cohort study with cross-sectional and longitudinal analyses. Background: The clinical importance of a history of whiplash-associated disorder (WAD) in people with neck pain remains uncertain. Objective: To compare people with WAD to people with nonspecific neck pain, in terms of their baseline characteristics and pain and disability outcomes over 1 year. Methods: Consecutive patients with neck pain who presented to a secondary-care spine center answered a comprehensive self-report questionnaire and underwent a physical examination. Patients were classified into a group of either those with WAD or those with nonspecific neck pain. We compared the outcomes of baseline characteristics of the 2 groups, as well as pain intensity and activity limitation at follow-ups of 6 and 12 months. Results: A total of 2578 participants were included in the study. Of these, 488 (19%) were classified as having WAD. At presentation, patients with WAD were statistically different from patients without WAD for almost all characteristics investigated. While most differences were small (1.1 points on an 11-point pain-rating scale and 11 percentage points on the Neck Disability Index), others, including the presence of dizziness and memory difficulties, were substantial. The between-group differences in pain and disability increased significantly (P<.001) over 12 months. At 12-month follow-up, the patients with WAD had on average approximately 2 points more pain and 17 percentage points more disability than those with nonspecific neck pain. Conclusion: People referred to secondary care with WAD typically had more self-reported pain and disability and experienced worse outcomes than those with nonspecific neck pain. Caution is required when interpreting the longitudinal outcomes due to lower-than-optimal follow-up rates.8 page(s

    Children's interpretation of conjunction in the scope of negation in English and Mandarin : new evidence for the semantic subset maxim

    We tested 3- to 5-year-old English- and Mandarin-speaking children on their interpretation of sentences like The elephant didn't eat both the carrot and the capsicum. These sentences are scopally ambiguous. Adult English speakers favor a weak interpretation of such sentences, with negation taking scope over conjunction (i.e., the elephant probably ate one of the vegetables, but not both). In contrast, adult Mandarin speakers favor a strong interpretation of the corresponding Mandarin sentences, with conjunction taking scope over negation (i.e., the elephant ate neither vegetable). The semantic subset maxim (Notley, Zhou, Jensen, & Crain, 2012) predicts that children acquiring all human languages should initially prefer the strong (subset) reading of such sentences. In contrast, the question–answer requirement model (Gualmini, Hulsey, Hacquard, & Fox, 2008; Hulsey, Hacquard, Fox, & Gualmini, 2004) predicts that children should initially prefer the scope reading that constitutes a good true answer to a question under discussion in the context. We designed a task in which the weak reading of our sentences corresponded to a good true answer to the question under discussion. We found that children across languages nonetheless preferred to assign a strong interpretation to our test sentences, providing empirical support for the semantic subset maxim.34 page(s

    Uncertainty in the school playground : shifting rationalities and teachers’ sense-making in the management of risks for children with disabilities

    Recent theoretical debates highlight the competing risk logics and varying rationalities mobilised in response to dangers and approaches to risk management. Yet the concept of uncertainty, and how it informs perceived risks, is relatively less well understood. Debates of this kind are illuminated in contexts where risks are managed as part of everyday practice. The school setting provides an example of a context in which risks are continuously negotiated amidst dominant protectionist concerns about children’s well-being and safety. Such protectionist concerns are particularly pronounced for children with disabilities, as assumptions about limited capabilities complicate and structure the everyday play experiences for children. Drawing on findings from the Sydney Playground Project, in this article we aim to unpack the felt discomfort experienced by school staff in their responses to uncertain moments in children’s play. We report qualitative data collected from two schools between October 2014 and September 2015 using video observations of children’s play and teachers’ responses to an online Tolerance of Risk in Play Scale. Our findings point to the competing logics and forms of sense-making operationalised by teachers to manage the unknown. Our analysis explored the ways in which risk strategies were (re)framed by school staff and such reframing explained their action (or inaction) in the playground and how these were underpinned by concerns about professional accountabilities. Their responses located risks within the child with disabilities, rather than the play activity itself. Another approach to uncertainty can be achieved by mobilising a discourse of trust in which ‘letting-go’ offers children opportunities to reflexively engage in risk-taking.17 page(s

    Gendered labour and media : histories and continuities

    On the evening of 5 September 1975, 150 women occupied the offices of the Canberra Times, protesting about an editorial hostile to participants in a national conference on 'Women and Politics'. This action, at the production site of the Australian capital's only broadsheet newspaper, provides a context for this themed issue's focus on gendered labour and media. We review recent perspectives on contemporary labour, and note that a persistent theme of this research is that recent changes in the media industries have seen the devaluation of professional work cultures as work in such industries has become more precarious. These changes are set against legacies of the devaluation of women’s work within the media, and negotiations of spaces for women to carve out media careers, which are explored by contributors to this issue. The article concludes by drawing out the need for a historically informed position on the gendering of media labour.12 page(s

    Recombinant human heterodimeric IL-15 complex displays extensive and reproducible N- and O-linked glycosylation

    Human interleukin 15 (IL-15) circulates in blood as a stable molecular complex with the soluble IL-15 receptor alpha (sIL-15Rα). This heterodimeric IL-15:sIL-15Rα complex (hetIL-15) shows therapeutic potential by promoting the growth, mobilization and activation of lymphocytes and is currently evaluated in clinical trials. Favorable pharmacokinetic properties are associated with the heterodimeric formation and the glycosylation of hetIL-15, which, however, remains largely uncharacterized. We report the site-specific N- and O-glycosylation of two clinically relevant large-scale preparations of HEK293-derived recombinant human hetIL-15. Intact IL-15 and sIL-15Rα and derived glycans and glycopeptides were separately profiled using multiple LC-MS/MS strategies. IL-15 Asn79 and sIL-15Rα Asn107 carried the same repertoire of biosynthetically-related N-glycans covering mostly α1-6-core-fucosylated and β-GlcNAc-terminating complex-type structures. The two potential IL-15 N-glycosylation sites (Asn71 and Asn112) located at the IL-2 receptor interface were unoccupied. Mass analysis of intact IL-15 confirmed its N-glycosylation and suggested that Asn79-glycosylation partially prevents Asn77-deamidation. IL-15 contained no O-glycans, whereas sIL-15Rα was heavily O-glycosylated with partially sialylated core 1 and 2-type mono- to hexasaccharides on Thr2, Thr81, Thr86, Thr156, Ser158, and Ser160. The sialoglycans displayed α2-3- and α2-6-NeuAc-type sialylation. Non-human, potentially immunogenic glycoepitopes (e.g.N-glycolylneuraminic acid and α-galactosylation) were not displayed by hetIL-15. Highly reproducible glycosylation of IL-15 and sIL-15Rα of two batches of hetIL-15 demonstrated consistent manufacturing and purification. In conclusion, we document the heterogeneous and reproducible N- and O-glycosylation of large-scale preparations of the therapeutic candidate hetIL-15. Site-specific mapping of these molecular features is important to evaluate the consistent large-scale production and clinical efficacy of hetIL-15.17 page(s

    The Coral Trait Database, a curated database of trait information for coral species from the global oceans

    Trait-based approaches advance ecological and evolutionary research because traits provide a strong link to an organism's function and fitness. Trait-based research might lead to a deeper understanding of the functions of, and services provided by, ecosystems, thereby improving management, which is vital in the current era of rapid environmental change. Coral reef scientists have long collected trait data for corals; however, these are difficult to access and often under-utilized in addressing large-scale questions. We present the Coral Trait Database initiative that aims to bring together physiological, morphological, ecological, phylogenetic and biogeographic trait information into a single repository. The database houses species- and individual-level data from published field and experimental studies alongside contextual data that provide important framing for analyses. In this data descriptor, we release data for 56 traits for 1547 species, and present a collaborative platform on which other trait data are being actively federated. Our overall goal is for the Coral Trait Database to become an open-source, community-led data clearinghouse that accelerates coral reef research.21 page(s
