    Comparative systeomics to elucidate physiological differences between CHO and SP2/0 cell lines

    Omics-based tools were coupled with bioinformatics for a systeomics analysis of two biopharma cell types: Chinese hamster ovary (M-CHO and CHO-K1) and SP2/0. Exponential and stationary phase samples revealed more than 10,000 transcripts and 6000 proteins across these two manufacturing cell lines. A statistical comparison of transcriptomics and proteomics data identified downregulated genes involved in protein folding, protein synthesis and protein metabolism, including PPIA-cyclophilin A, HSPD1, and EIF3K, in M-CHO compared to SP2/0 while cell cycle and actin cytoskeleton genes were reduced in SP2/0. KEGG pathway comparisons revealed glycerolipids, glycosphingolipids, ABC transporters, calcium signaling, cell adhesion, and secretion pathways depleted in M-CHO while retinol metabolism was upregulated. KEGG and IPA also indicated apoptosis, RNA degradation, and proteosomes enriched in CHO stationary phase. Alternatively, gene ontology analysis revealed an underrepresentation in ion and potassium channel activities, membrane proteins, and secretory granules including Stxbpt2, Syt1, Syt9, and Cma1 proteins in M-CHO. Additional enrichment strategies involving ultracentrifugation, biotinylation, and hydrazide chemistry identified over 4000 potential CHO membrane and secretory proteins, yet many secretory and membrane proteins were still depleted. This systeomics pipeline has revealed bottlenecks and potential opportunities for cell line engineering in CHO and SP2/0 to improve their production capabilities.1FEB 281

    Ovarian reserve parameters and IVF outcomes in 510 women with poor ovarian response (POR) treated with intraovarian injection of autologous platelet rich plasma (PRP)

    The aim of the current study was to characterize ovarian reserve parameters and IVF outcomes in women with a history of poor ovarian response (POR) treated with intraovarian injection of autologous platelet rich plasma (PRP). Reproductive age women (N=510age range 30-45yo) diagnosed with POR based on Poseidon criteria were included in the study. PRP treatment resulted in higher AFC, higher serum AMH, lower serum FSH, and a higher number of mature oocytes and cleavage and blastocyst stage embryos. After PRP injection, 22 women (4.3\%) conceived spontaneously, 14 (2.7\%) were lost to follow up, and 474 (92.9\%) attempted IVF. Among women who attempted IVF, 312 (65.8\%) generated embryos and underwent embryo transfer, 83 (17.5\%) achieved a pregnancy, and 54 (11.4\%) achieved sustained implantation/live birth (SI/LB). In total, of the 510 women with POR and mean age of 40.3, PRP resulted in improvement of ovarian reserve parameters, a pregnancy rate of 20.5\% and SI/LB rate of 12.9\%. Our findings suggest that PRP treatment may be considered in women with POR. For wider clinical application, its clinical efficacy will need to be demonstrated in prospective randomized clinical trials.6MAR 312513-25231

    Glenoid Morphology and Related Parameters in Turkish Society

    Introduction Glenoid morphology may vary in different ethnic groups. Detection of these differences may be important in preoperative planning, especially in reverse shoulder arthroplasty. In this study, we investigated the mean glenoid size and retroversion in Turkish society and their relationship with the dominant side, gender, height, weight, and body mass index (BMI). Materials and methods Between 2019 and 2021, 102 shoulders of 51 patients (24 females/27 males, 51 left/51 right) who were included in shoulder joint imaging during thorax CT scanning were examined. Those with glenoid fracture, arthrosis, shoulder surgery or deformity, or younger than 18 years of age were not included in the study. The mean age was 41.69 (range: 18-73) years. Glenoid anterior-posterior diameter (D) and glenoid version (GV) were measured in axial slices, and glenoid height (H) was measured in coronal slices. The correlation of these parameters with gender, height, weight, and dominant side was examined. Results Mean D was 25.79 +/- 4.44 mm, mean H was 29.08 +/- 4.08 mm, and mean GV was-0.99 degrees +/- 0.92 degrees. The mean height of the patients was 162 +/- 16.23 cm and the mean weight was 71.9 +/- 15.36 kg. The glenoid diameter and height were smaller in females, however, no statistically significant difference was found in the glenoid version (p<0.01, p<0.01, and p=0.92). The glenoid on the dominant side was statistically significantly more retroverted, whereas D and H were not associated with dominance (p<0.01, p=0.9, and p=0.98). It was found that the glenoid sizes were very highly correlated with the patient's height, and it was highly correlated with the patient's weight (p<0.01 and p<0.01). On the other hand, height and weight were not correlated with the glenoid version (p=0.47 and p=0.81, respectively). There was no statistically significant relationship between BMI and glenoid sizes and glenoid version (p=0.14 and p=0.46, respectively). Conclusions Females in Turkish society had small glenoid sizes. Male gender, height, and weight were positively correlated with large glenoids. The glenoid was more retroverted on the dominant side. These findings should be considered in preoperative planning in Turkish society.8AUG 131

    European Biological Variation Study (EuBIVAS): within- and between-subject biological variation estimates for serum thyroid biomarkers based on weekly samplings from 91 healthy participants

    Objectives Thyroid biomarkers are fundamental for the diagnosis of thyroid disorders and for the monitoring and treatment of patients with these diseases. The knowledge of biological variation (BV) is important to define analytical performance specifications (APS) and reference change values (RCV). The aim of this study was to deliver BV estimates for thyroid stimulating hormone (TSH), free thyroxine (FT4), free triiodothyronine (FT3), thyroglobulin (TG), and calcitonin (CT). Methods Analyses were performed on serum samples obtained from the European Biological Variation Study population (91 healthy individuals from six European laboratories21-69 years) on the Roche Cobas e801 at the San Raffaele Hospital (Milan, Italy). All samples from each individual were evaluated in duplicate within a single run. The BV estimates with 95\% CIs were obtained by CV-ANOVA, after analysis of variance homogeneity and outliers. Results The within-subject (CVI) BV estimates were for TSH 17.7\%, FT3 5.0\%, FT4 4.8\%, TG 10.3, and CT 13.0\%, all significantly lower than those reported in the literature. No significant differences were observed for BV estimates between men and women. Conclusions The availability of updated, in the case of CT not previously published, BV estimates for thyroid markers based on the large scale EuBIVAS study allows for refined APS and associated RCV applicable in the diagnosis and management of thyroid and related diseases.4MAR 28523-5326

    Benzodithiophene bearing conjugated polymer-based surface anchoring for sensitive electrochemical glucose detection

    An amino-functionalized, conjugated polymer (P(BDBT)) modified glassy carbon electrode (GCE) was employed as an immobilization platform for glucose oxidase (GOx) enzyme to assemble a novel glucose biosensor. Amino groups available on the polymer backbone served as bioconjugation sites for GOx via glutaraldehyde (GA). The biosensor response to the reduction in oxygen amount because of the enzyme reaction was monitored at ???0.7 V potential versus Ag/AgCl. The biosensor displayed a broad linear range between 0.1???1.0 mM glucose with a detection limit of 0.17 mM. The values of the apparent Michaelis-Menten constant (KMapp) and sensitivity were determined as 1.74 mM, and 28.17 ??A/(mM??cm2), respectively. GOx immobilized P(BDBT) film displayed high stability, selectivity, and reproducibility. Cyclic voltammetry (CV) and Scanning electron microscopy (SEM) techniques were utilized for the characterization of surface modifications. The fabricated biosensor was adept at determining the amount of glucose in a commercial beverage. The simple electrochemical method for the construction of P(BDBT)/GOx biosensors could pave the way to new perspectives in developing profitable biosensors.10OCT1012-10211

    Stool Microbiota Diversity Analysis of Blastocystis-Positive and Blastocystis-Negative Individuals

    Blastocystis is a unicellular eukaryote found in the gastrointestinal tract of both human and other animal hosts. The clinical significance of colonic Blastocystis colonization remains obscure. In this study, we used metabarcoding and bioinformatics analyses to identify differences in stool microbiota diversity between Blastocystis-positive and Blastocystis-negative individuals (n = 1285). Alpha diversity was significantly higher in Blastocystis carriers. At phylum level, Firmicutes and Bacteroidetes were enriched in carriers, while Proteobacteria were enriched in non-carriers. The genera Prevotella, Faecalibacterium, Flavonifracter, Clostridium, Succinivibrio, and Oscillibacter were enriched in carriers, whereas Escherichia, Bacteroides, Klebsiella, and Pseudomonas were enriched in non-carriers. No difference in beta diversity was observed. Individuals with Blastocystis-positive stools appear to have gut microbiomes associated with eubiosis unlike those with Blastocystis-negative stools, whose gut microbiomes are similar to those associated with dysbiosis. The role of Blastocystis as an indicator organism and potential modulator of the gut microbiota warrants further scrutiny.2FEB1

    The Impact of Sex Hormone-Binding Globulin Levels on Thromboembolic Events at Patients with Advanced Stage Adenocarcinoma

    OBJECTIVE Several studies have shown that increased risk of venous thromboembolism (VTE) with hormonal contraceptives is mediated through the sex hormone-binding globulin (SHBG) through different pathways. Assuming SHBG as surrogate marker for hormonal stimulus as an increasing risk factor for VTE, we investigated if background SHBG has any impact on VTE in patient with advanced stage adenocarcinoma. Blood drawn from patients with VTE (n=45) and from patients without VTE (non-VTEn=23), as a control group without a history of VTE, and SHBG levels were compared between goups. There was no difference in SHBG levels between VTE (59.7 +/- 41.5) and non-VTE (60 +/- 49) (p=0.71) group, but gastric cancer patients had statistically higher SHBG levels (73.7 +/- 39.9range from 36.6 to 176.2) than rest of the cohort (56.54 +/- 44.4range from 9.6 to 246.6) (p=0.033), although it was out of scope of this study. Although our study did not show any impact of SHBG levels on risk of VTE at patients with advanced stage adenocarcinoma, it revealed high levels of SHBG at gastric cancer patients which deserve further research.     Turkey Prostate Cancer Map 2021: Turkish Urooncology Association Prostate Cancer Database Report

    Objective: This study aimed to present the data of patients with prostate cancer (PCa) whose detailed information was stored in the Urologic Cancer Database-Prostate, Urooncology Association, Turkey with the title of ``Turkey Prostate Cancer Map 2021.{''} Materials and Methods: Patient data between 1995 and 2020 were retrospectively scanned. The age of the patients, their distribution according to age groups, symptoms during diagnosis, examination findings {[}digital rectal examination (DRE)], prostate-specific antigen (PSA) values, biopsy methods in the diagnosis, metastatic disease rates, treatment methods, and progression rates at follow-up were examined. These results were compared with the results of the previous report, namely ``Prostate Cancer Incidence (Incidence) in Turkey,{''} by the Urooncology Association in Turkey in 2009. Results: This study analyzed the data of 5040 patients from 19 different centers. The mean patient age was 63.6 (37-97) years. The age distribution examination revealed that most patients (49.8\%) were aged 60-69 years. Of the patients, 51.8\% were symptomatic at the time of diagnosis. The presence of symptoms was determined in 88.6\% in 2009 data. The ORE of patients revealed that 25\% of patients had malignancy findings. The PSA distribution examination revealed a >10 ng/mL PSA value in 37.5\% of patients. With the increasing use of magnetic imaging resonance (MRI) in PCa diagnosis over the years, increased MR-fusion biopsy rates have been observed. Considering the biopsy data, 91\% of patients were diagnosed with a classical transrectal ultrasound-guided biopsy, whereas 9\% were diagnosed with MR-Fusion biopsy. Fusion biopsies revealed that 23\% of patients with Prostate Imaging-Reporting and Data System (PI-RADS) 4 lesion and 57\% with PI-RADS 5 lesion were diagnosed with cancer. Of the patients, 8.9\% of patients had metastases during the initial diagnosis. This rate was 17\% in 2009 data. The treatment methods examination after the diagnosis revealed that 73.9\% of patients had undergone radical prostatectomy. This rate was 51.8\% in 2009. Robotic and laparoscopic approaches, which are among the surgical modalities, have increased over the years. However, the most frequently applied modality in our country was open radical prostatectomy with 62.6\%. Considering the follow-up data after treatment, 8.9\% of patients had progression, of which 62.6\% was biochemical, 30.2\% was radiological, and 6.9\% was a clinical progression. Conclusion: Technological advancements for PCa diagnosis (MRI and MR-guided biopsies) are becoming a routine part of daily practice compared to the results of the ``Prostate Cancer Incidence in Turkey{''} project in 2009. The comparative study results revealed that the rate of symptomatic and metastatic disease decreases at the time of diagnosis, and laparoscopic and robotic surgery methods are used at increasing rates for localized disease.1MAR1-42

    Critical appraisal and meta-analysis of biological variation estimates for kidney related analytes

    Objectives Kidney markers are some of the most frequently used laboratory tests in patient care, and correct clinical decision making depends upon knowledge and correct application of biological variation (BV) data. The aim of this study was to review available BV data and to provide updated BV estimates for the following kidney markers in serum and plasmaalbumin, creatinine, cystatin C, chloride, potassium, sodium and urea. Content Relevant studies were identified from a historical BV database as well as by systematic literature searches. Retrieved publications were appraised by the Biological Variation Data Critical Appraisal Checklist (BIVAC). Meta-analyses of BIVAC compliant studies with similar design were performed to deliver global estimates of within-subject (CVI) and between-subject (CVG) BV estimates. Out of the 61 identified papers, three received a BIVAC grade A, four grade B, 48 grade C, five grade D grade and one was not appraised as it did not report numerical BV estimates. Most studies were identified for creatinine (n=48). BV estimates derived from the meta-analysis were in general lower than previously reported estimates for all analytes except urea. For some measurands, BV estimates may be influenced by age or states of health, but further data are required. This review provides updated global BV estimates for kidney related measurands. For all measurands except for urea, these estimates were lower than previously reported. Outlook For the measurands analyzed in this review, there are sufficient well-designed studies available to publish a trustworthy estimate of BV. However, for a number of newly appearing kidney markers no suitable data is available and additional studies are required.4MAR 28469-4786
