    Evaluation of the influence of in vitro human digestion simulation on the chemical composition and bioactivities of Ziziphus jujuba Mill.

    Fruit of Ziziphus jujuba Mill. are used as functional foods for centuries due to their rich content and bioactivities. Although in vitro antioxidant and hypoglycaemic activity of jujube fruit were investigated previously, the bioavailability phenomenon has been disregarded so far. For this study, 80\% ethanol extract of Ziziphus jujuba fruit (ZJE) was investigated for its in vitro hypoglycaemic and antioxidant potentials, before and after the interaction with simulated human digestion. DPPH scavenging activity, FRAP, CUPRAC, and TOAC assays were used for this purpose. Moreover, inhibition potentials of a-amylase and a-glucosidase enzymes and advanced glycation end products (AGE) were examined for the hypoglycaemic effect. Results indicated that ZJE showed significant antioxidant and dose dependent enzyme and AGE inhibition activity. Nonetheless, subsequent to simulated human digestion in vitro bioactivities of ZJE were significantly lowered for bioavailable fraction (IN). Protocatechuic acid (PA) (major phenolic compound of the fruit) contents of the extract and fractions were measured via HPTLC for more accurate understanding of the effects of human digestion and bioavailability profile.1MAR105-1145

    Background Magnetic resonance-guided radiotherapy (MRgRT) utilization is rapidly expanding, driven by advanced capabilities including better soft tissue imaging, continuous intrafraction target visualization, automatic triggered beam delivery, and the availability of on-table adaptive replanning. Our objective was to describe patterns of 0.35 Tesla (T)-MRgRT utilization in Europe and Asia among early adopters of this novel technology. Methods Anonymized administrative data from all 0.35T-MRgRT treatment systems in Europe and Asia were extracted for patients who completed treatment from 2015 to 2020. Detailed treatment information was analyzed for all MR-linear accelerators (linac) and -cobalt systems. Results From 2015 through the end of 2020, there were 5796 completed treatment courses delivered in 46,389 individual fractions. 23.5\% of fractions were adapted. Ultra-hypofractionated (UHfx) dose schedules (1-5 fractions) were delivered for 63.5\% of courses, with 57.8\% of UHfx fractions adapted on-table. The most commonly treated tumor types were prostate (23.5\%), liver (14.5\%), lung (12.3\%), pancreas (11.2\%), and breast (8.0\%), with increasing compound annual growth rates (CAGRs) in numbers of courses from 2015 through 2020 (pancreas: 157.1\%prostate: 120.9\%lung: 136.0\%liver: 134.2\%). Conclusions This is the first comprehensive study reporting patterns of utilization among early adopters of a 0.35T-MRgRT system in Europe and Asia. Intrafraction MR image-guidance, advanced motion management, and increasing adoption of on-table adaptive RT have accelerated a transition to UHfx regimens. MRgRT has been predominantly used to treat tumors in the upper abdomen, pelvis and lungs, and increasingly with adaptive replanning, which is a radical departure from legacy radiotherapy practices.1AUG 221

    Expression of vascular endothelial growth factor in follicular cell-derived lesions of the thyroid: Is NIFTP benign or precancerous?

    Objective: Vascular endothelial growth factor (VEGF) is an angiogenic factor that plays an important role in physiological and pathological angiogenesis of the thyroid. The aim of the current study was to determine the expression characteristics of VEGF in follicular cell-derived lesions of the thyroid and to assess whether a new entity noninvasive follicular thyroid neoplasm with papillary-like nuclear features (NIFTP) is precancerous. Material and Methods: Patients diagnosed with 33 follicular adenomas (FA), 41 invasive follicular variant papillary thyroid cancer (IN-FVPTC), and 40 NIFTP in surgical resection materials were evaluated retrospectively. Immunostaining was performed on 5-mu m paraffin tissue sections. The percentages of immunostaing for VEGF were evaluated on pathological materials. We used a percentage of labeled thyrocytes score (0, no labeling1, 60\%) and an intensity score (0, no staining1, weak2, intermediate3, strong). The sum of two scores were accepted as the total score. Results: Mean ages of the FA, IN-FVPTC, and NIFTP groups were 44.7 +/- 11.7 years, 46.9 +/- 13.6 years, 43.2 +/- 15.4 years, respectively and the mean VEGF immunostaining scores were 44.7 +/- 29.3, 50.2 +/- 32.54, 4 +/- 26.3 respectively. Although there was no statistically significant difference (p= 0.347), the total score of the NIFTPs was higher than the scores of the FA (mean= 3.9 +/- 1.8) and IN-FVPTC(mean= 4.3 +/- 1.9) groups with a mean value of 4.6 +/- 1.7. This result was remarkable. There was no statistically significant difference between tumor diameters and staining percentages (p= 0.750). Conclusion: Even if there were no statistical differences for VEGF immunostaining, it was high in NIFTPs. Since we know the role of VEGF in tumorigenesis, we can hypothesize that NIPTP can be precancerous. Our argue should be corroborated by a large prospective study.1MAR60-663

    A novel practical approach for monitoring the crosslink density of an ethylene propylene diene monomer compound: Complementary scanning acoustic microscopy and FIB-SEM-EDS analyses

    Tuning of the crosslink density (CLD) in the rubber compounds is very crucial for optimizing the physical and mechanical properties of the ultimate rubber products. Conventionally, CLD can be measured via rheological methods such as moving die rheometer (MDR), via mechanical tests such as temperature scanning stress relaxation analysis (TSSR), or via direct swelling experiments using Flory-Rehner approach. In the current study, two novel techniques, focused ion beam - scanning electron microscopy (FIB-SEM) processing, with simultaneous energy dispersive X-ray spectrometry (EDS) mapping analysis and scanning acoustic microscopy (SAM) were combined and correlated to conventional methods on a model recipe of ethylene propylene diene monomer (EPDM) compound having different sulphur contents. Depending on the applied technique, the increase in the crosslink density with sulphur content was found to be 1.7 fold for the Flory-Rehner approach and 1.2 fold for both TSSR and MDR. It is directly monitored from the FIB-SEM-EDS analysis that the sulphur distribution and agglomeration behavior increased in line with ZnO content, which is an indirect indication of the rise in crosslink density. The impedance maps of the crosslinked samples obtained through SAM analysis revealed that the impedance of the samples increased with the increasing sulphur content, which can be attributed to higher level of crosslink density. A quantified correlation was obtained between SAM images and the crosslink density of the samples. It was shown that SAM is a promising tool for practical and non-destructive analysis for determining the formation of crosslink density of the rubbers.JAN 173

    Skin Biopsy Results of Geriatric Patients Over a 5-year Period and the Frequency of Skin Diseases Before and After COVID-19 Pandemic

    Objective: The number of older adults has increased throughout the world. Aging affects all the organs and creates psychological, physiologic and anatomic changes. One of the most important organs of the human body is the skin, which shows the effects of aging the most. This study aims to determine whether age, gender, and season of biopsy play a significant role in skin biopsy results. Additionally, the study investigates whether the frequency of skin diseases differs before and after the coronavirus disease-2019 (COVID-19) pandemic. Methods: We conducted a retrospective study on the histopathology results of patients over 65 years old between June 2016 and September 2021. The histopathology results were obtained from the Acibadem Pathology Department. Results: Of the 677 patients, 310 (45.8\%) were male and 367 (54.2\%) were female. The most common disease in all patients were benign cutaneous neoplasms (23\%), followed by eczematous disease (18.5\%) and epithelial cutaneous cancers (16.8\%). We divided the results into 12 groups: group 1: Urticaria, erythema and purpuras, group 2: Papulosquamous and eczematous diseases, group 3: Infectious diseases, group 4: Rheumatologic diseases and alopecia, group 5: Benign cutaneous neoplasms, group 6: Precancerous lesions, group 7: Basal cell carcinoma, squamous cell carcinoma, group 8: Cutaneous metastasis and other skin cancers, group 9: Pigmentation disorders, group 10: Pschycology related dermatological disorders, group 11: Granulomatous dermatitis, group 12: Bullous dermatitis. Before the COVID-19 pandemic, the most prevalent results were group 2 (21.3\%), followed by group 5 (20.4\%) and group 7 (16.7\%) whereas, after the COVID-19 pandemic, the most frequent results were group 5 (28.4\%) followed by group 7 (17.1\%), and group 6 (14.9\%). In terms of seasons, the most common diseases were group 5 (24.1\%) in winter, group 2 (21.6\%) in spring, group 5 (30.0\%) in summer, and group 2 (18.9\%) in autumn. Before the COVID-19 pandemic, the most common result was group 2 (21.3\%), followed by group 5 (20.4\%) and group 7 (16.7\%), and during the COVID-19 pandemic, the most common result was group 5 (28.4\%), followed by group 7 (17.1\%), group 6 (14.9\%). Conclusion: Many skin diseases affect the geriatric population. Geriatric patients face challenges such as multiple drug use, comorbidities, mobility problems and cognitive disorders. In our study, the most common diseases in all patients were benign cutaneous neoplasms (23\%), followed by eczematous diseases (18.5\%), and epithelial cutaneous cancers (16.8\%). Knowing about the frequency of skin diseases is critical for the early detection of precancerous and cancerous lesions.2JUN189-1941

    National Multi-Center Observational Retrospective Study to Understand Treatment Patterns and Outcomes for Stage III Non-Small Cell Lung Cancer Patients in Turkey: Turkish Society for Radiation Oncology Study, STONE Trial

    This study investigated treatment patterns and outcomes in patients with inoperable stage III non-small cell lung cancer (NSCLC) treated with radiotherapy (RT) in Turkey. We included 492 patients with stage III NSCLC in this multi-center retrospective study. Pa-tient demographics, clinical characteristics, and clinical treatment patterns from the time of the initial diagnosis to disease progression were recorded. Additionally, the prognostic factors predicting overall survival (OS) and progression-free survival (PFS) were analyzed. For the initial treatment, 429 patients (89.2\%) received chemotherapy and RT, whereas 53 patients (10.8\%) were treated only with RT. The first disease progression occurred in 288 patients (58.4\%) at 9.3 months (median) after the initial treatment, and 64.6\% re-ceived treatment after first progression. The second disease progression occurred in 30 patients, and 20 patients (66.7\%) received treatment. Median OS and PFS were 27.0 months and 13.4 months, respectively. Age (p< 0.001), stage (p= 0.04), poor performance score (PS) (p= 0.03) and RT doses (p= 0.002) were independent predictors for OS and PFS in our multivariate analysis. Additional significant predictors for OS in the multivariate analysis were gender (p= 0.004), treatment period (0.02), and irradiation technique (p= 0.02). Disease progression occurred in nearly 58\% of the patients, and one-third of these patients remained untreated during the disease progression. These findings indicate a need for additional treatment options in patients with unresectable stage III NSCLC with high-risk features, namely older age, stage IIIB disease, poor PS, and lower RT doses.123-343

    Managing advanced prostate cancer in the Asia Pacific region: ``Real-world'' application of Advanced Prostate Cancer Consensus Conference 2019 statements

    Aim The second Asia-Pacific Advanced Prostate Cancer Consensus Conference (APAC APCCC 2020) gathered insights into the real-world application in the Asia-Pacific (APAC) region of consensus statements from the 3rd Advanced Prostate Cancer Consensus Conference (APCCC 2019). Methods The 4-h our virtual meeting in October 2020 brought together 26 experts from 14 APAC countries to discuss APCCC 2019 recommendations. Presentations were prerecorded and viewed prior to the meeting. A postmeeting survey gathered views on current practice. Results The meeting and survey highlighted several developments since APAC APCCC 2018. Increased access and use in the region of PSMA PET/CT imaging is providing additional diagnostic and staging information for advanced prostate cancer and influencing local and systemic therapy choices. Awareness of oligometastatic disease, although not clearly defined, is increasing. Novel androgen receptor pathway antagonists are expanding treatment options. Cost and access to contemporary treatments and technologies continue to be a significant factor influencing therapeutic decisions in the region. With treatment options increasing, multidisciplinary treatment planning, shared decision making, and informed choice remain critical. A discussion on the COVID-19 pandemic highlighted challenges for diagnosis, treatment, and clinical trials and new service delivery models that will continue beyond the pandemic. Conclusion APAC-specific prostate cancer research and data are important to ensure that treatment guidelines and recommendations reflect local populations and resources. Facilitated approaches to collaboration across the region such as that achieved through APAC APCCC meetings continue to be a valuable mechanism to ensure the relevance of consensus guidelines within the region.6DEC686-6951
