    Effect of immune stress on growth performance and immune functions of livestock: Mechanisms and prevention

    Immune stress markedly affects the immune function and growth performance of livestock, including poultry, resulting in financial loss to farmers. It can lead to decreased feed intake, reduced growth, and intestinal disorders. Studies have shown that pathogen-induced immune stress is mostly related to TLR4-related inflammatory signal pathway activation, excessive inflammatory cytokine release, oxidative stress, hormonal disorders, cell apoptosis, and intestinal microbial disorders. This paper reviews the occurrence of immune stress in livestock, its impact on immune function and growth performance, and strategies for immune stress prevention

    Interactions of treated municipal wastewater with native plant species

    Potentially, the restoration of native ecosystems could be combined with the land application of treated municipal wastewater (TMW), reducing TMW discharge into waterbodies. High levels of nutrients, pathogens, and other contaminants from TMW can degrade water quality. The land application of TMW onto native vegetation reduces the nutrient load in water bodies and may create zones of ecological value. However, establishing native plants may be challenging if the species are not adapted to highly fertile environments, such as those resulting from TMW irrigation. There is a critical knowledge gap about the response of native plant species to irrigation with TMW. We aimed to determine the distribution and speciation of nutrients in the soil- plant system following application of TMW onto 11 species of native plants in a long-term field trial on Banks Peninsula, New Zealand (NZ). TMW was irrigated at a rate of 1000 mm per annum, equivalent to N, and P loading rates of 194 and 110 kg ha yr¯¹, respectively. We determined physicochemical properties from soil profiles (0–65 cm) under selected species as well as the growth and chemical composition of the plants. Despite the site receiving 950 kg ha¯¹ yr¯¹ of Na, there was no evidence of impaired soil structure following TMW irrigation. Nitrogen did not accumulate in the soil, and it is likely to have been taken up by plants or lost through denitrification and nitrate leaching. The accumulation rate of P indicated that soil P concentrations will remain within the range found in NZ agricultural soils for at least 50 years. TMW irrigation increased plant height by 10% compared to the control after 3.5 years of growth. Plant species significantly affected the concentrations of total C, total N, nitrate (NO‾₃), and Na in the soil. TMW application had negligible effects on the elemental composition of plant foliage. NZ native vegetation can facilitate the land application of TMW. Future work should elucidate the maximum rates that can be applied as well as the effect of TMW on the soil microbiota

    Purification of high molecular-weight antibacterial proteins of insect pathogenic Brevibacillus laterosporus isolates

    Brevibacillus laterosporus (Bl) is a Gram-positive and spore-forming bacterium belonging to the Brevibacillus brevis phylogenetic cluster. Globally, insect pathogenic strains of the bacterium have been isolated, characterised, and some activities patented. Two isolates, Bl 1821L and Bl 1951, exhibiting pathogenicity against the diamondback moth and mosquitoes, are under development as a biopesticide in New Zealand. However, due to the suspected activity of putative antibacterial proteins (ABPs), the endemic isolates often grow erratically. Various purification methods including size exclusion chromatography, sucrose density gradient centrifugation, polyethylene glycol precipitation, and ammonium sulphate precipitation employed in this study enabled the isolation of two putative antibacterial proteins of ~30 kD and ~48 kD from Bl 1821L and one putative antibacterial protein of ~30 kD from Bl 1951. Purification of the uninduced cultures of Bl 1821L and Bl 1951 also yielded the protein bands of ~30 kD and ~48 kD on SDS-PAGE which indicated their spontaneous induction. Disc diffusion assay was used to determine the antagonistic activities of the putative ABPs. Subsequent transmission electron microscope (TEM) examination of purified putative antibacterial protein-containing solution showed the presence of encapsulin (~30 kD) and polysheath (~48 kD) like structures. Although only the ~30 kD protein was purified from Bl 1951, both structures were seen in this strain under TEM. Furthermore, while assessing the antibacterial activity of some fractions of Bl 1951 against Bl 1821L in size exclusion chromatography method, population of Bl 1821L persister cells was noted.     Comammox Nitrospira Clade B is the most abundant complete ammonia oxidizer in a dairy pasture soil and inhibited by dicyandiamide and high ammonium concentrations

    The recent discovery of comammox Nitrospira, a complete ammonia oxidizer, capable of completing the nitrification on their own has presented tremendous challenges to our understanding of the nitrification process. There are two divergent clades of comammox Nitrospira, Clade A and B. However, their population abundance, community structure and role in ammonia and nitrite oxidation are poorly understood. We conducted a 94-day microcosm study using a grazed dairy pasture soil amended with urea fertilizers, synthetic cow urine, and the nitrification inhibitor, dicyandiamide (DCD), to investigate the growth and community structure of comammox Nitrospira spp. We discovered that comammox Nitrospira Clade B was two orders of magnitude more abundant than Clade A in this fertile dairy pasture soil and the most abundant subcluster was a distinctive phylogenetic uncultured subcluster Clade B2. We found that comammox Nitrospira Clade B might not play a major role in nitrite oxidation compared to the role of canonical Nitrospira nitrite-oxidizers, however, comammox Nitrospira Clade B is active in nitrification and the growth of comammox Nitrospira Clade B was inhibited by a high ammonium concentration (700 kg synthetic urine-N ha¯¹) and the nitrification inhibitor DCD. We concluded that comammox Nitrospira Clade B: (1) was the most abundant comammox in the dairy pasture soil; (2) had a low tolerance to ammonium and can be inhibited by DCD; and (3) was not the dominant nitrite-oxidizer in the soil. This is the first study discovering a new subcluster of comammox Nitrospira Clade B2 from an agricultural soil

    Barriers to coastal planning and policy use of environmental research in Aotearoa-New Zealand

    Identifying barriers to the effective use of science in coastal management of Aotearoa-New Zealand is easy, due to the present lack of complicated governance and management structures, coupled with an emphasis on funding science that includes pathways to implementation. This opinion piece discusses four areas that still hinder effective use of science, all of which are likely to be problematic for other countries. We initially focus on why the science may not be used related to: misunderstandings (linguistic and conceptual differences including indigenous world views); timing of information delivery; uncertainty surrounding the information (knowledge limitations and funding); and top-down constraints (legal systems, politics and institutional objectives). We use Aotearoa-New Zealand examples to demonstrate the barriers operating within each area and discuss three potential solutions. Importantly our analysis indicates that researchers alone cannot transcend these barriers; rather, we need to work as part of an ecosystem, requiring commitment from all society, extending beyond the usual suspects (management agencies). We believe that ecological and systems education from junior school levels through to universities have an important role to play in setting the context to overcome current barriers

    Effects of whole body vibration exercise combined with weighted vest in older adults: A randomized controlled trial

    Background: To evaluate the training effects of whole body vibration (WBV) combined with weighted vest (WV) in older adults. Methods: This randomized controlled trial study was conducted in healthy older adults living in the community. Fifty-one participants were randomly allocated into 3 groups: group 1 (n = 17), WBV alone, training on WBV at a frequency 30 Hz, amplitude 2 mm, 10 sets of 1 min squats, with 60 s rest, group 2 (n = 15), WV alone, squat exercise, 10 sets of 1 min, with 60 s rest, while WV loaded with 10% body weight and group 3 (n = 19), WBV + WV, combining WBV exercise with the addition of a WV. All groups completed training 3 times per week for 8 weeks. The outcomes were total muscle mass, muscle thickness, maximal isometric strength, single-leg-stance and timed-up-and-go evaluated at baseline and after training. Results: As a result of training all groups improved their isometric muscle strength with little difference between groups. The single-leg-stance significantly improved only in WBV + WV group 25.1 ± 10.8 s (mean ± 95% CI, p <  0.01). The timed-up-and-go improved in all groups, but the improvement was significantly greater in the WBV + WV group (17.5 ± 6.9%) compared to the WV (8.5 ± 3.2%) and WBV groups (9.2 ± 5.4%, p = 0.043, 0.023 respectively). Rectus femoris muscle thickness and total muscle mass were significantly increased in all groups equally with little difference between groups. Conclusion: The combined WBV + WV had a greater effect on the single-leg-stance and the timed-up-and-go compared to WV or WBV alone.     A single fungal strain was the unexpected cause of a mass aspergillosis outbreak in the world's largest and only flightless parrot

    Kākāpō are a critically endangered species of parrots restricted to a few islands off the coast of New Zealand. Kākāpō are very closely monitored, especially during nesting seasons. In 2019, during a highly successful nesting season, an outbreak of aspergillosis affected 21 individuals and led to the deaths of 9, leaving a population of only 211 kākāpō. In monitoring this outbreak, cultures of aspergillus were grown, and genome sequenced. These sequences demonstrate that, very unusually for an aspergillus outbreak, a single strain of aspergillus caused the outbreak. This strain was found on two islands, but only one had an outbreak of aspergillosis; indicating that the strain was necessary, but not sufficient, to cause disease. Our analysis provides an understanding of the 2019 outbreak and provides potential ways to manage such events in the future

    Running demands in New Zealand club, semi-professional and professional rugby union competitions

    Game demands of professional rugby players are well documented, however, there is minimal research on the running demands in amateur and semi-professional competitions. To the authors knowledge, no research has compared the running demands in club, national (division 1 (NPC) and division 2 (Heartland)) or international provincial (Super rugby) competitions. Information on the running demands of each competition would enable trainers to develop training programs specific to player’s needs and assist in the transition of players between competitions. Players from a team in each competition level wore a 10 Hz GPS unit during all games in a rugby season. Total distance (m), running distance (≥7 km.h-1(m)) and high intensity running (≥16 km.h-1(m)) were recorded. The research was approved by an Institutional Ethics Board and the NZ Rugby Union. Lower competition (club and division 2) players covered greater distances (150m to 400m) per game than higher level players. As in total distance, division 2 players ran (≥7 km.h-1) more in games (120m) compared to higher level players. However, club players typically covered less running distance than higher level players (3102m vs 3237 to 3319m, p=0.10 to 0.00). In contrast, Super rugby players typically ran greater distances (962m) at higher speeds (≥16 km.h-1) per game. This information could assist in the development of competition-specific training programs, and the monitoring of training loads during a season and when a player returns from an injury. Further analysis will investigate running demands of rugby positions in each competition to enable the development of position-specific-training programs

    Reducing vulnerability to forced labour and trafficking of women migrant workers from South- to West-Asia

    Millions of short-term, low-skilled women migrant workers from South-Asia to West-Asia experience exploitative and unsafe conditions. We review evidence from literature and interview 18 key informants to assess the exploitation migrant women face, and highlight the impacts of past interventions to determine their potential, and realised effectiveness, in reducing forced labour and trafficking. We find that women face varied precarious situations along the migration pathway, including interactions with recruiters in the home country, incurring debt, pre-departure formalities and training, withheld wages and mobility restrictions. We discuss varying degrees of success of mechanisms that aim to reduce vulnerability to forced labour
