    Screening Antimicrobial Activity of Actinomycetes Isolated from Raja Ampat, West Papua, Indonesia

    In the framework of exploitation of antimicrobial activity of Actinomycetes in Papua, one hundred isolates of Actinomycetes isolated from soil and leaf litter samples from various ecosystems in Batanta and Salawati Island, Raja Ampat, West Papua were screened. We obtained 200 crude extracts from 100 isolates based on two extraction phases. Nonpolar metabolites were extracted by ethyl acetate : methanol (4:1) solvent while the polar metabolites were concentrated using a freeze-drying method. Based on the agar dilution method, a total of 43 from 200(21.5%) crude extracts have antimicrobial activity against bacteria and yeasts (Escherichia coli NBRC 14237, Bacillus subtilis NBRC 3134, Staphylococcus aureus NBRC 13276, Micrococcus luteus NBRC 1367, Candida albicans NBRC 1594 and Saccharomyces cerevisiae NBRC 10217). Some crude extracts showed anti-Gram negative (1.5%), anti-Gram positive (17%) and antifungal (17%) activities. Crude metabolites which were extracted using ethyl acetate : methanol were more effective on antimicrobial activity (35%) compared with water extraction (17%). Five most potential isolates (BL-13-5, BL-06-5, BL-14-2, BL-22-3, and Sl-36-1) were identified based on 16S rRNA gene sequence data. Sequence similarity search by BLAST program revealed that they show sequence similarities to Streptomyces kanamyceticus (92%), Streptomyces verne (92%), Streptomyces narbonensis (92%), Streptomyces malachitofuscus (98%), and Streptomyces hygroscopicus (96%), respectively.     The Genetic Diversity of Endophytic and Phyllosphere Bacteria from Several Indonesian Herbal Plants

    Herbal plants have been believed by Indonesians to be an alternative medicine to treat illnesses. The bioactive compounds in the plant can be derived from secondary metabolites or from endophytic and phyllosphere bacteria which coexist within medicinal plants. A total of 18 endophytic bacteria and 32 phyllosphere bacteria were isolated from the herbal plants of Citrus sp., Pluchea indica, Curcuma longa, Nothopanax scuttelarium, Piper crocatum, and Andrographis paniculata. About 72% of endophytic bacteria isolates have proteolytic activity and about 11% have lipolytic activity. On the other hand, about 59% of phyllosphere bacteria isolates have proteolytic activity and about 19% have lipolytic activity. Phylogenetic diversity analysis was conducted by using the amplified ribosomal DNA restriction analysis (ARDRA) method and the sequence of 16S rDNA was digested with endonuclease restriction enzymes: MspI, RsaI, and Sau961. The diversity of endophytic and phyllosphere bacterium from the samples of herbal plants was high. Bacteria isolated from the same herbal plant does not always have a close genetic relationship except for the bacteria isolated from the P. indica plant which showed a close genetic relationship with each other.     Combination of Depot Medroxy Progesterone Acetate and Javanese Long Pepper Extract on Body Weight, Hematology, and Blood Biochemistry as a Safe Contraception Model

    The development of male hormonal contraception is based on a decrease in sperm concentration without affecting libido and sexual potency. The combination of depot medroxy progesterone acetate (DMPA) + extract of Javanese long pepper (JLP) with dosages of 0.94 mg and 1.88 mg decreases the concentration of spermatozoa. However, it remains unknown whether the combination influences body weight, hematology, and blood biochemistry. Therefore, it is necessary to investigate the effect of DMPA + JLP extracts on the body weight, hematology, and blood biochemistry of male rats (Rattusnorvegicus L.) using Sprague-Dawley strains. The research uses a completely randomized design (CRD); one group control and two treatment groups. In the first group, the castrated rats were given oral administration extracts of JLP (CJ) with doses of 0, 0.94, 1.88, 2.82, and 3.76 mg. In the second group, the rats were injected with 1.25 mg DMPA and given an oral administration extract of JLP. Injection was given in week-0 and 12. Administration was conducted every day from week 7-18. Analysis of the normality and homogeneity of data is done before the ANOVA test. Data that is abnormal and not homogeneous are tested with non-parametric statistical Kruskal-Wallis. This study shows that the combination of minimal doses of DMPA and administration variousdoses of extracts of JLP does not affect body weight and hematology (erythrocyte, hemoglobin, hematocrite), and the blood biochemistry of rats, such as the values of SGPT, SGOT, HDL, and triglycerides (p < 0.05), but rather the total cholesterol and LDL (p < 0.05). Furthermore, it is concluded that the combination of the minimal dosage of DMPA and weaned various dosages of JLP extracts affect the total value and LDL cholesterol but do not influence body weight, nor hematology and blood biochemistry. Such combinations can be drawn on for asafe male contraceptive model t by taking into account the value of the total cholesterol and LDL during its use

    Terpenoids from the Stem Bark of Jatropha Plants and Their Biological Activities

    Three terpenoids, including two diterpenes (curcusone B and jatrophone) and a triterpene (stigmasterol) have been isolated from the stem bark of Jatropha plants. Curcusone B and stigmasterol were isolated from J. curcas, meanwhile jatrophone and stigmasterol were from J. gossypifolia. The biological activities of these compounds have been evaluated toward bacteria, fungi and tumour cells. Isolation was carried out in vacuum liqiud cromatography (VLC) technique with silica gel as an adsorben and some solvents as eluents. The compound structures were determined by spectroscopic methodes i.e. UV-vis, FTIR, NMR (1-D, 2-D) and were then compared based on their spectroscopic data with similiar data from literatures. The biological properties of these compounds were evaluated against four strains of bacteria (Acetobacter sp., Eschericia coli, Staphylococcus aureus, and Streptococcus sp.), 4 strains of fungi (Aspergilus niger, Penicillium sp. (grey), Penicillium sp. (white) and Rhizopus sp.) and murine leukemia P-388 cells. The results showed that cytotoxic property of curcusone B towards murine leukemia P-388 cells is better than jatrophone and stigmasterol which are IC50 = 0.57 μg/mL (1.93 μM) for curcusone B and IC50 > 100 μg/mL for jatrophone and stigmasterol. Meanwhile, activities against bacteria, jatrophone is better than curcusone B and stigmasterol. Jatrophone is the most active against S. aureus (bacteria) with growth inhibition zone 36 mm and A.niger (fungi) is 44 mm. Further study indicated that jatrophone was bacteriostatic against S. aureus.&nbsp

    Secondary Metabolite of Aspergillus fumigatus, Endophytic Fungi of the Medicinal Plant Garcinia griffithii

    The endophytic fungi Aspergillus fumigatus was isolated from the tissues of the fruits of Garcinia griffithii. The fungal strain was identified from the colony, and it was characteristic of cell morphology. The ethyl acetate extracts derived from fungus cultures showed major spots on TLC under UV light, which was continued to the isolation of the secondary metabolites. The structure of the isolated compound was elucidated on the basis of NMR analyses (1H-NMR, 13C-NMR, HMQC, HMBC and H-H COSY). The compounds were identified as: 4,6-dihydroxy, 3,8a-dimethyl-1-oxo- 5-(3’-oxobutan-2’-yl)-1,4,4a,5,6,8a-hexahydronaphthalen-2-yl-1”,2”-dimethyl-5”-(2”’-methylprop 1”’-enyl)cyclopentanecarboxylate

    Solvent Extraction of Ginger oleoresin using Ultrasound

    The use of ultrasound in extraction process creates novel and interesting methodologies, which are often complementary to conventional extraction methods. In the present study, the use of ultrasound to extract oleoresin from ginger (Zingiber officinale R.) was investigated. The extraction was performed by using ethanol as solvent in the presence of ultrasonic irradiations operating at frequency of 42 kHz at extraction temperature of 60 oC. The oleoresin extracted was in the form of dark thick liquid with specific ginger flavor. Based on GC-MS analysis, the use of ultrasound was not give an effect on alteration of main component in ginger oleoresin. The main component in extracted ginger oleoresin was zingerone. Gingerol as one of the pungent principle of the ginger oleoresin was not detected due to decomposition of gingerol at a temperature above 45 oC. Extraction rate of ultrasound-assisted extraction was about 1.75 times more rapid than a conventional system based on soxhlet. The scanning electron microscopy images provided more evidence for the mechanical effects of ultrasound, mainly appearing on cells’ walls and shown by the destruction of cells, facilitating the release of their contents

    Sequential Isolation of Saturated, Aromatic, Resinic and Asphaltic Fractions Degrading Bacteria from Oil Contaminated Soil in South Sumatera

    Sequential isolation has been conducted to obtain isolates of saturated, aromatic, resin, and asphaltene fractions degrading bacteria from oil contaminated sites. Five soil samples were collected from South Sumatera. These were analyzed using soil extract medium enriched with oil recovery or Remaining-Oil recovery Degradated (ROD) as sole carbon and energy sources according to the isolation stage. ROD at the end of every isolation stage analyzed oil fractions by use of the SARA analysis method. Six isolates of bacteria have been selected, one isolate was fraction saturates degrading bacteria that are Mycobacterium sp. T1H2D4-7 at degradation rate 0.0199 mgs/h with density 8.4x106 cfu/g from stage I. The isolate T2H1D2-4, identified as Pseudomonas sp. was fraction aromatics degrading bacteria at accelerate 0.0141 mgs/h with density 5.1x106 cfu/g are obtained at stage II. Two isolates namely Micrococcus sp. T3H2D4-2 and Pseudomonas sp. T1H1D5-5 were fraction resins degrading bacteria by accelerate 0.0088 mgs/h at density 5.6x106 cfu/g and 0.0089 mgs/h at density 5.7x106 cfu/g are obtained at stage III. Isolation of stage IV has been obtained two isolates Pseudomonas sp. T4H1D3-1and Pseudomonas sp. T4H3D5-4 were fraction asphaltenes degrading bacteria by accelerate 0.0057 mgs/h at density 5.6x106 cfu/g and accelerate 0.0058 mgs/h at density 5.7x106 cfu/g.     Preparation and Characterization of Co-Processed Excipient Carrageenan-Pregelatinized Cassava Starch Propionate as a Matrix in The Gastroretentive Dosage Form

    The gastroretentive dosage form is designed to prolong the gastric residence time of the drug delivery system which also results in the development of an appropriate excipient. The purpose of this study is to develop and characterize coprocessed excipient made from carrageenan (kappa-iota = 1:1) and pregelatinized cassava starch propionate (PCSP) in ratios of 1:1, 1:2, and 1:3. PCSP was prepared with propionic anhydride in an aqueous medium. The product was mixed with carrageenan (kappa-iota = 1:1), as well as characterized physicochemical and functional properties. The coprocessed excipient was then used as a mucoadhesive granule and floating tablet. The USP Basket was selected to perform the dissolution test of the granules in HCl buffer (pH 1.2) and distilled water for 8 hours each. Mucoadhesive properties were evaluated using bioadhesive through a vitro test and wash-off test. As for the floating tablet, the USP Paddle was selected to perform the dissolution test of the tablets in 0.1 N HCl for 10 hours. The floating lag time and floating time were tested in 0.1 N HCl for 24 hours. The result of these studies indicated that co-processed excipient carrageenan-PCSP can retard dosage form in gastric and drug controlled release, thus making it a suitable material for the gastroretentive dosage form

    Isolation of Antioxidant Compound from Endophytic Fungi Acremonium sp. from the Twigs of Kandis Gajah

    The endophytic fungi Chrisonilia sitophila, Acremonium sp., and Penicillium sp. have been isolated from the tissues of the twigs of kandis gajah. All of the fungi strains were grown in 3 L potatoe dextrose broth medium (PDB) at a room temperature for 28 days. To extract the antioxidant compounds, the culture broth were filtered for mycelia removal followed by extraction and evaporation. All of the extracts were evaluated for their antioxidant activities by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. The extract of Acremonium sp. have strong activity with IC50 value of 10.3 μg/mL, which is equivalent to ascorbic acid activity with IC50 value of 9.8 μg/mL. The extract was subjected to column chromatography on Si gel twice to obtain a high purity antioxidant compound in the form of yellow oil. The molecular structure was determined based on spectroscopic data, including 1H-NMR, 13C-NMR, HMQC, HMBC, and COSY. The compound was determined as sesquiterpene 3,5-dihydroxy-2,5-dimethyltrideca-2,9,11-triene-4,8-dione.     Characterization of Cr/Bentonite and HZSM-5 Zeolite as Catalysts for Ethanol Conversion to Biogasoline

    In this research it has been done characterization on Cr/Bentonit and Zeolit HZSM-5 catalysts for ethanol catalytic process to biogasoline (equal to gasoline). Cr/Bentonit has high acidity and resistant to a lot of moisture, so in addition to being able to processing feed which a lot of moisture (>15%) from ethanol-water mixture, also it is not easy deactivated. Cr/Bentonit which is then used as the catalyst material on the process of ethanol conversion to be biogasoline and the result was compared with catalyst HZSM-5 zeolite. Several characterization methods: X-ray diffraction, Brunauer Emmett Teller (BET), thermogravimetry analysis (TGA), and catalyst activity tests using catalytic Muffler instrument and gas chromatography-mass spectrometry (GC-MS) for product analysis were performed on both catalysts. From acidity measurement, it is known that acidity level of Cr/Bentonit is the highest and also from XRD result, it is known there is shift for 2theta in Cr/Bentonit, which indicates that Cr-pillar in the Bentonite can have interaction. It is also supported by BET data that shows the addition of specific surface are in Cr/Bentonite compared with natural Bentonite before pillarization. Futhermore catalyst activity test produced the results, analyzed by GC-MS, identified as butanol and also possibly formed hexanol, decane, dodecane, undecane, which are all included in gasoline range (C4 until C12).
