    Diversity of methanogenic archaea in the natural subsurface sediments revealed by culture-independent genomic approaches

    北九州市立大学博士(工学)This study identified methanogenic communities in estuary and wetland ecosystems based on 16S rRNA gene sequencing and molecular cloning of the functional mcrA gene. Well-known methanogenic archaea (mainly Methanomicrobiales) and uncultivated putative methanogenic archaea (Verstraetearchaeota and Methanofastidiosales) were detected in the estuarine sediments, indicating the CH₄ production potentials from hydrogenotrophic and H₂-dependent methylotrophic methanogenesis. For wetland peat soils, the “Candidatus Bathyarchaeota” was highly observed (up to 96% of the archaeal abundance) based on microbial gene quantification. Furthermore, partial fragments of the mcrA gene, belonging to the Bathyarchaeota, was detected based on the functional gene calling and metagenomic reconstruction. These results firstly demonstrated that Bathyarchaeota that possess the mcrA gene and its potential in methane production could be observed in a natural wetland ecosystem. The discovery of novel Mcr and Mcr-like genes provides a great discussion and interpretation of the origin and evolutionary history of the mcrA and Mcr complex.doctoral thesi

    Investigation and Evaluation of Thermal Environment for Living Space of Infants in Nursery Schools

    北九州市立大学博士(工学)The indoor environment has a great influence on the physical and mental efficiency of the resident. On the other hand, infants lack autonomy in terms of thermal adaptive comfort behavior. Hence, to provide a suitable indoor environment for children, the evaluation of indoor thermal comfort and the cognition of children’s thermal comfort by teachers engaged in nursing work are more important. Through a questionnaire survey of teachers, this research analyzed and compared the relationship between teachers’ thermal adaptation behavior and children’s thermal sensation. Compared with the traditional fixed-point measurement method, a method of wearable sensors for children was proposed to measure the indoor temperature distribution in summer and winter. Finally, this research proposes measures to improve the architectural design of the nursery to create a comfortable thermal environment for young children.doctoral thesi

    Improvement of Indoor Environment based on Cooking Heating Wall and Sunlight Room in Traditional Chinese Housing

    北九州市立大学博士(工学)The research aim of this paper was to put forward a cooking heating wall system with an additional sunlight room, focusing on the analysis of the impact of this system on improving the indoor temperature. It was verified that this system is effective for indoor heating in residential buildings. The ratio of walls to other elements is also an important factor affecting the heating of the system. In future research, this viewpoint can be used as the main research direction. First, the main elements affecting the indoor temperature can be determined based on the local geographical environment and climatic conditions to further determine the proportion of walls and major elements, so as to optimize the system. 1. By comparing the heating conditions of traditional residential buildings with or without the new passive system, the heat load reduction when adopting the new system in winter was estimated. 2. In the absence of solar radiation, the heating effects of the ordinary heating system and the new passive system were analyzed. The purpose was to calculate the heating rate, the time required, and the decrease in the annual heat load of the building. 3. Considering the presence or absence of solar radiation, the new system was compared to the ordinary heating of residential buildings to simulate and calculate the change in indoor temperature, heating efficiency, heating time, and the value of the annual building heat load.doctoral thesi
