    Physical properties and cytotoxicity of Cu(ii) and Zn(ii) complexes with a TMS-substituted indolo[2,3-c]quinoline-derived Schiff base

    The incorporation of non-native chemical elements, such as silicon, into drug molecules has gained significant attention as a strategy to broaden the chemical space in medicinal chemistry and develop novel drug candidates. Traditionally, research has focused on the isosteric replacement of a carbon atom with silicon ("silicon switch") in known drug structures or the attachment of a trimethylsilyl (TMS) group to biologically active scaffolds. In this study, a TMS-substituted indoloquinoline-based Schiff base (HLTMS) and its corresponding metal complexes, Cu(HLTMS)Cl2 (1) and Zn(HLTMS)Cl2 (2), were synthesized and comprehensively characterized using elemental analysis, spectroscopic techniques (IR, UV-vis, 1H and 13C NMR for HLTMS and 2), ESI mass spectrometry and single-crystal X-ray diffraction (SC-XRD) for 1 and electron diffraction (ED) for 2. The attachment of the TMS group enhanced the lipophilicity of HLTMS, while complex formation with Cu(ii) substantially improved the antiproliferative activity. Exploitation of their intrinsic fluorescence to investigate cellular uptake and intracellular localization in cancer cells was impeded by limited solubility. Both HLTMS and 2 were found to generate reactive oxygen species under cell-free conditions in accord with their redox activity established by cyclic voltammetry. The photochemical activity of the indolo[2,3-c]quinoline-based proligand HLTMS and its complexes 1 and 2 has been disclosed. The compounds exhibited significant toxicity on various human cancer cells and disrupted the mitochondrial membrane potential, suggesting the contribution of mitochondrial dysfunction, triggered by HLTMS and its metal complexes, to their toxic effects. These findings highlight the potential of TMS-substituted Schiff bases as promising anticancer drug candidates

    Addressing White Collar Crime with Situational Crime Prevention Approach: What’s Promising and How It Works?

    The study aims to demonstrate that white collar crimes can be prevented by limiting organizational opportunities for such crimes. By employing a situational approach to crime prevention, the research seeks to highlight practical measures businesses can adopt to reduce these opportunities. It explores the 25 techniques proposed by Cornish and Clarke, discussing their implementation within the context of organizational crime prevention. The findings reveal that while situational crime prevention measures are effective in controlling white collar crime, they cannot eradicate it entirely. These measures help to maintain white collar crime within an acceptable range, as complete elimination is unattainable due to the complexity and organized nature of these crimes. This research underscores the importance of a situational approach in white collar crime prevention, providing a systematic framework for reducing crime opportunities. It emphasizes the need for businesses to adopt comprehensive internal controls, ethical guidelines, and awareness programs to mitigate the risk of white collar crimes, contributing to both academic discourse and practical crime prevention strategies in corporate settings

    Evaluation of the risk of heart failure with tumour necrosis factor inhibitors: A large-scale meta-analysis in immune-mediated inflammatory diseases

    Current therapeutic guidelines for immune-mediated inflammatory diseases (IMIDs) contraindicate the administration of tumour necrosis factor inhibitors (TNFis) in advanced heart failure (HF) and highlight the potential risk for new-onset HF. The current evidence has low certainty, and the results of studies in the past two decades have even challenged the recommendations regarding the impact of TNFis on the risk of HF.The objective was to systematically synthesize data on the risk of HF in TNFi-treated groups compared to non-treated controls in IMIDs.A systematic search was conducted in August 2023. Randomized controlled trials (RCTs) and non-randomized observational studies of IMID patients comparing groups receiving TNFis to non-TNFi-exposed controls were included. The outcome was the incidence of worsening, de novo, and composite HF. Random-effects meta-analysis was conducted using risk ratios (RR) with 95% confidence intervals (CIs) to pool data.The systematic search identified 49 studies, with 45 included in the quantitative analysis. For the worsening of HF, the pooled results of non-randomized studies showed no statistically significant risk-increasing effect of TNFis (RR = 1.18, 95% CI: 0.69-2.00). Analyses from both RCTs and non-randomized data indicated no increased risk of de novo HF in the TNFi-group compared to controls (RR = 0.87, 95% CI: 0.60-1.25 and RR = 0.86, 95% CI: 0.64-1.14, respectively). Similarly, no increased risk was found for composite (worsening and de novo) HF in the TNFi-treated group versus controls, pooling non-randomized data.Our findings indicate that TNFi-treated IMID patients do not have an increased risk for developing de novo HF, and no statistically significant risk enhancement could be observed in the risk of worsening HF with TNFis. Updating IMID guidelines should be considered regarding TNFis' non-risk increasing effect on de novo HF, whereas further validating data on the risk of worsening HF in TNFi-treated IMID patients is needed

    Counter-Terrorism Strategies in Pakistan: Addressing the Menace of Terrorism

    This research aims to analyze fatalities caused by terrorism in Pakistan and assess the role of the National Action Plan (NAP) in countering terrorism and extremism. Terrorism has posed a severe threat to Pakistan’s security, integrity, and socio-economic development, placing the country at the forefront of the global fight against terrorism. Data was collected from secondary sources, and analyzed through content analysis. From 2001 to 2020, terrorism events were categorized into three phases: low casualties (2001-2006), intensified counter-terrorism operations (2007-2015), and reduced fatalities (2017-2020). In total, 15,397 incidents resulted in 20,636 civilian and 7,318 security personnel deaths, while 33,236 terrorists were eliminated. The National Action Plan, anti-terrorist financing measures, and strengthening NACTA were pivotal strategies. Addressing the root causes of extremism requires education reforms, economic development, stricter anti-terror financing controls, enhanced inter-agency coordination, and sustained implementation of counter-narrative campaigns. These measures are crucial for minimizing terrorism and fostering lasting peace
