Fermentation quality and nutritive value of rice crop residue based silage ensiled with addition of epiphytic lactic acid bacteria
Silage is the feedstuff resulted from the preservation of forages through lactic acid fermentation. The aim of this study was to evaluate nutritive value, fermentation characteristics and nutrients digestibility of rice crop residue based silage ensiled with epiphytic lactic acid bacteria (LAB). The mixture of rice crop residue (RC), soybean curd residue (SC) and cassava waste (CW) in a 90: 5: 5 (on dry matter basis) ratio was used as silage material. Three treatments silage were (A) RC + SC + CW as a control; (B) RC + SC + CW + LAB inoculums from rice crop residue; (C) RC + SC + CW + LAB inoculums from king grass. Silage materials were packed into plastic silo (1.5 kg capacity) and stored for 30 days. The results showed that crude protein content in B and C silage was higher than that of silage A, but NDF content in silages B and C was lower than that of silage A. Lactic acid concentration was higher (P 0.01) in silage C compared to silage B and A, thus pH value of silage C was lower (P 0.01) than silage B and A. Silage C had the highest Fleigh point than that of other silages. Dry matter and organic matter digestibilities were higher in silages B and C (P 0.01) than that of control silage. It was concluded that the addition of LAB inoculums from king grass to rice crop residue based silage resulted a better fermentation quality compared to LAB inoculums from rice crop residue. Effectivity of Aloe vera bioactives as feed additive for broilers reared on deep litter
There are plenty of Indonesian plants contain usefull bioactive components. One of them is Aloe vera. Previous experiment showed that Aloe vera bioactives reduced aerob bacteria in the intestinal and improved feed efficiency in broilers reared in cages. The results however, gave some variations, may be due to variation in rearing the chickens. Two experiments were carried out to study the effectivity of Aloe vera bioactives as feed additives for broilers reared on deep litter. In the first study, six experimental diets in mash form were formulated, i.e.: Control (K); K + antibiotic; K + 0.50 g/kg dry Aloe vera (LBK); K + 1.00 g/kg LBK; K + anthraquinone; K + Aloe vera in semi-liquid form. The amount of semi liquid Aloe vera and the anthraquinone were equally to 1.00 g LBK/kg. Results showed that antibiotic improved body weight gain 6.10% and feed efficiency 5.50% better than the control, although statistically not significant (P0.05). Aloe vera bioactives in low doses (0.50 g/kg) also improved weight gain (6.30%) and feed efficiency (5.20%) similar to the antibiotic. However, Aloe vera in high doses and anthraquinone (equal to 1.00 g/kg diet) did not improve performance of broilers. There were no significant changes on carcass yield, abdominal fat levels, weight of liver, gizard and gastro intestinal tract due to any feed additives tested. The second experiment were carried out to study the effectivity of feed additives when included in crumble diets. Six experimental diets, i.e.: Control (K), K + antibiotic, K + Semi-liquid Aloe vera (equal to 1.00g dry Aloe vera/kg), K + 0.50 g dry Aloe vera/kg, K + 1.00 g dry Aloe vera/kg, K + 0.50 g dry Aloe vera + 0.50 g Curcuma xanthorrhiza meal/kg. All diets were fed in crumble form. Results showed that chickens fed with feed additives (antibiotic or Aloe vera bioactives) have a significantly (P0.05) higher body weight gain and feed efficiency than those fed with control diet. The best weight gain was achieved by low dose dry Aloe vera (1342 g/bird) and the lowest was the control (1039 g/bird). The best feed conversion ratio was achieved by antibiotic treatment (1.756) followed by low dose dry Aloe vera + curcuma meal (1.758) and the worst was showed by the control (1.908). It is concluded that Aloe vera bioactives at low dose (0.50 g/kg diet) could improve body weight gain and feed efficiency of broilers reared on deep litter. The improvement achieved was similar as the antibiotic. The bioactive is more effective when fed in crumble diets. Key words: Broilers, bioactives, Aloe ver
Detection of Brugia malayi microfilaria/Larvae in mosquito using Polimerase Chain Reaction.
Lymphathic filariasis that is also known as elepanthiasis is caused by infestation of 3 species nematode Wuchereria bancrofti, Brugia malayi and Brugia timori. In Indonesia 70% filariasis case caused by Brugia malayi. Mosquito species from genus Anopheles, Aedes, Culex, Mansonia and Armigeres are known as vector of this disease. Microfilaria detection on mosquito is one methode to know infection rate in vector population in endemic area.The objectives of the research were to study the ability of Hha1 repeat applicable to detect microfilaria/larvae in a pool of mosquitoes and to get description of adult mosquito night biting population lived in endemic area of filariasis brugian. Mosquito as positive control used in this research come from laboratory of parasitology of FKUI. Mosquito sample from the field was from Binawara and Kolam Kiri villages, South Kalimantan province. Mosquito were trapped then identified by its species. DNA of mosquitoes was extracted and then run by the PCR using Hha 1 repeat primer. Result of the research indicated that adult mosquitoes night biting from Binawara village consist of Culex, Mansonia, Anopheles genus and from Kolam Kiri village only from Mansonia genus. Hha 1 repeat primer is applicable to detect 1 mosquito infected with microfilaria/larvae in a pool of negative mosquitoes. Mosquito samplesfrom the two villages showing negative PCR. Seroepidemiology of Canine parvovirus infection in dogs
Canine parvovirus is an acute and fatal viral disease in dogs. A total of 209 local, cross breed and breed dogs sera from Kodya Bogor, Kabupaten Bogor, Sukabumi, and Jakarta, had been tested using Haemagglutination Inhibition Test (HI) with pig red blood cells. A total of 64 breed and cross breed dogs from Sukabumi and Kodya Bogor, were used as a sentinel dogs to study the epidemiology of Canine parvovirus (CPV) infection and its immunological responses caused by vaccination. The results indicated that 78% (95) breed and cross bred dogs and 59% (51) local dogs had antibody to CPV. Sentinel dogs results indicated that dogs had been vaccinated showed antibody response with the varied titre dependant upon prevaccination titre. Low prevaccinated titre gave better response than protective level titre. From 19 puppies observed, Maternal antibodi were still detected until 5 weeks old puppies. First vaccination given at less than 3 months old, should be boosted after 3 months old puppied. Antibodi titre produced by natural infection will keep untill 2 years. Feeding of Aspergillus oryzae fermentation culture (AOFC) to growing sheep: 2. Growth rate and feed efficiency
Utilization of yeast and or filamentous fungi as feed additive to ruminants has been of interest since the late 1980’s. Two fungi species have been commercially produced in the United States, (1) Yea-Sacc containing living cells of Saccharomyces cerevisiae and (2) Amaferm bearing Aspergillus oryzae fermentation extract. It has been demonstrated and proven that the cultures can enhance rumen development and function in young ruminants. This paper concerns the use of Aspergillus oryzae fermentation culture (AOFC) as feed additive for young-growing male ‘Garut’ sheep. The A. oryzae was cultured in a media made of mineral-enriched ‘onggok’ flour, a material of tapioca processing waste. The AOFC was prepared gradually by incubating the fungus at room temperature (26 – 300C) for 5 days, dried at 400C and ground. The AOFC was added to a commercial concentrate (GT-03) at 0, 5 and 10% (w/w) levels, as treatment C0 (control), C1, and C2, respectively. Fifteen growing ‘Garut’ sheep were used and the concentrate feed treatments were randomly allotted based on a randomized block design. Drinking water was available at all time. The amount of feed offered (chopped King grass and concentrates) and their refusals were weighed daily and live-weight of sheep was measured once a week in the morning. Daily feces was collected and weighed in the last 10 days of the 14-week experimental period. All feed and fecal samples were analyzed for dry matter, crude protein, total fiber (NDF), and ash. AOFC supplementation resulted in higher weight gains (P0.05), which were 94.81; 122.08; and 140.52 g/d for C0, C1, and C2 treatments, respectively. Dry and organic matter, as well as protein intake was also significantly increased by inclusion of AOFC into concentrate diet (P0.05). The increment in nutrient intake was from increased consumption of concentrates, and not from King grass, however, there was no effect of AOFC supplementation on feed efficiency. Characteristic of cow milk dadih using starter of probiotic of lactic acid bacteria
Dadih is an original dairy product from West Sumatera processed traditionally. It is a spontaneous fermentation of buffalo milk at room temperature for 48 hours in a bamboo tube, has no standard of processing and quality. Dadih is potentially to be develop into probiotic products (functional food) that can be enjoyed by the public widely. Development of cow's milk dadih is necessary since buffalo milk is available only in certain area. Product and characteristic information of cow milk dadih using probiotic of lactic acid bacteria starter has not been known. The research objective was to determine the characteristics of cow milk dadih that used starter of probiotic lactic acid bacteria during storage at room temperature (27oC) and cold temperature (4oC). The study was designed using a factorial randomized block design pattern 12x3 at room temperature and 12X4 at cold temperatures, with the number of repeatation of 3 times. Treatment consisted of: (i) starter formula (A) using a single bacterium or a combination of Lactobacillus acidophilus, Lactobacillus casei and Bifidobacterium longum, and (ii) storage time (B). Observed parameters included pH value, titrable acidity, the total plate count, and in-vitro probiotic testing (bacterial resistance to bile salts and low pH) of cow milk dadih. The results showed that L. acidophilus early exponential phase was at the hour 3rd, L. casei at the hour 4th and B. longum on the 3rd of which is used as the optimum time of mixing two or more bacteria in the manufacture of cow milk dadih. The volume of starter used was 3% with time fermentation of 48 hours at room temperature (27-30oC). Cow milk dadih that was stored for 7 days at room temperature (27-30oC) and for 21 days at cold temperatures (4-10oC) was able to maintain viability of bacteria to bile salts and low pH at 1010-1012 cfu/ml with percentage resistance varied. The cow milk dadih using a combination starter of B. longum with other probiotics on the composition of 1:1 had a resistance to bile salts which were relatively constant after the dadih was kept for 21 days at cold temperature. Cow milk dadih with a starter of probiotic bacteria still qualify as a probiotic product with good quality that storaged up to 4 days at room temperature and 21 days in cold temperatures. The effect of aloe vera bioactive level as feed additive on the egg performances of laying hens
A study on the use of aloe vera bioactives as feed additive in layer ration was conducted. One hundred and twenty pullets strain Isa Brown were allocated into 5 treatments with 6 replicates and 4 birds/replicate. The treatments were: control,control+antibiotic (50 ppm zinc bacitracin), and control+ aloe vera at three levels (0.25; 0.50 and 1.00 g/kg). The treatments were conducted in a completely randomized design. Parameter measured were first initial body weight, age at 1st lay, feed intake, egg weight, hen day (%HD) and feed conversion ratio. The results showed that antibiotic and aloe vera used as additive for 9 months production did not significantly (P0.05) affect all parameter measured, except feed intake of hens fed diet containing 0.5 g/kg aloe vera was significantly (P0.05) higher than control. The addition of aloe vera at 1.0 g/kg significantly (P0.05) reduced the feed intake as compared with the control, aloe vera 0.25 and 0.50 g/kg. The used of aloe vera (1.00 g/kg) produced egg weight significantly (P0.05) higher than the control, and feed conversion ratio was significantly (P0.05) better than the control and aloe vera (0.25 g/kg). It is concluded that the best treatment was the diet with aloe vera level at 1.00 g/kg. This treatment improved feed efficiency 8.40%. Key Words: Bioactive, Egg Production, Laye
Molecular characterization of six sub population Indonesian local goats based on mitochondrial DNA D-loop
Indonesian local goats were spread in some region, but there was still limited data’s known about the characteristics of its genetic diversity and origin. The Mitochondrial DNA D-loop sequences were used to study the genetic diversity and relationships of six sub population Indonesian local goats, namely, Kacang, Marica, Samosir, Jawarandu, Muara and Bengali goats. From 539 blood samples and DNA extraction collections were selected about 60 samples (10 samples each sub populations) analyzed by PCR-RFLP methods, followed sequence analyzed about 5 PCR products each sub population. The results of the sequence analyses were edited and acquired about 957 bp of nucleotides length. After the alignment analyses were found 50 polymorphic sites which divided into 19 haplotype groups of mtDNA D-loop region. The value of nucleotide diversity was 0.014 ± 0.002. Analysis of Neighbour Joining with Kimura 2 Parameter methods and bootstrap test with 1000 replication indicated that each sub population groups was significantly different between one groups to the others. The maternal lineages origin of six breeds of Indonesian local goats was included to the group of lineage B. The Lineage B was the maternal origin of the haplogroup of goats in the region of East Asia, South Asia, China, Mongolia, North and South Africa, Malaysia, Indonesia, Pakistan and India. Growth and carcass development of Kedu x Arab cross under two feeding regimes
One hundred unsexed chicks of each group of Kedu x Arab cross, of Arab x Arab and of Kedu x Kedu were kept under intensive husbandry for about 52 days (from 32 to 84 days of age). Each group was allocated to 10 cages of 10 unsexed birds. Experimental rations consisted of main ingredients of ground yellow corn, soybean meal, rice bran and fishmeal. Experimental ration were R1 containing energy of 2920 kkal ME/kg with crude protein of 15.17%. R2a contained energy of 3087 kkal ME/kg with crude protein of 7.36% and R2b contained energy of 2336 kkal ME/kg with crude protein of 42,51%. R1 ration was given ad libitum for 24 hours and R2a ration was given from 08.00 a.m.–12.00 p.m. and continued with R2b ration was given up to 8.00 a.m. in the following day. Results showed that live bodyweight and bodyweight gain were not significantly different among the three groups of chicken. Feed consumption of Kedu x Arab cross was significantly lower than that of Kedu x Kedu nor Arab x Arab. Feed conversion ratio of Kedu x Arab cross was also significantly lower (3.74) than its comparative groups. Birds on R1 ration grew faster (804 g/bird) than that on R2a-R2b ration (738 g/bird) with feed consumption of 2135 g bird-1 52 day-1 on R1 ration versus 2338 g bird-1 52 day-1 on R2a-R2b ration. FCR of the birds were lower on R1 ration (3.44) than on R2a-R2b ration (4.26). Energy consumption, protein consumption, protein efficiency ratio, energy efficiency ratio did not show significant different among the three groups of chicken, but for all those variables under R1 ration were significantly higher than for those under R2a-R2b ration. Whole carcass and carcass cuts were not significantly affected by neither the kind of chickens nor dietary treatments. Development of liver was the slowest among development of other carcass cuts and organs, whilst development of breast meat was 4.4–6.0 times, and drumstick meat was 4.6–6.6 times. Cacao pod based ration supplemented with organic Zn: Growth responce on sheeps
Cacao pod can be used as feed to replace grass. The aims of study was to evaluate the growth response of sheep fed on cacao pod based rations to replace grass and supplemented with organic Zn. Fresh cacao pod was chopped to the size of 1-2 cm. In untreated cacao pod (without ammoniation), chopped cacao pod was dried then milled, while in ammoniated cacao pod, the chopped cacao pod was mixed with urea, 1.5% (w/w) and kept in anaerobic conditions for 7 days. After 7 days it was dried and milled. Both, without ammoniated and ammoniated cacao pod were used for ration formulation. The rations were formulated in iso protein and iso energy (16% CP and 70% TDN). The study used 25 male lambs Sumatera Composite breed at age 7-8 months old with average body weight 18.8 ± 2.86 kg. Experiments were conducted using a randomized block design with number of animal in each group was 5. Five treatment rations evaluated were: R = grass + concentrate (positive control); K = cacao pod + concentrate; KZ = cacao pod + concentrate + organic Zn; KU = cacao pod ammoniated + concentrate; KUZ = cacao pod ammoniated + concentrate + organic Zn. The treatment rations were offered for 10 weeks. The results showed that the consumption of cacao pod based rations was higher (P 0.05) than the grass-basal ration (75.19 g/kg BW0,75 vs 70.22 g/kg BW0,75). Suplementation of Zn organic to ammoniated or untreated cacao pod resulted in no significant difference in DM, and CP intake. DM, OM, CP digestibility and N retention were relatively similar among the fifth rations from 53.13 to 63.91%, from 54.69 to 66.49%, from 66.0 to 79.28% and from 9.5 to 13.4 g/h respectively. R ration had similar NH3 levels with KU ration (21.57 mM vs 22.94 mM) but higher than the K, KZ and KUZ ration (16.45 mM, 16.62 mM and 13.61 mM). VFA concentration showed no difference among treatments (from 140.32 to 179.76 mM). Sheep fed ration R had similar ADG with K and KZ (92.19 g/h vs 83.13 g/h; 93.44 g/h) but higher than the ration KU and KZ (73.44 g/h and 61.72 g/h). From this study can be concluded that grass-based rations can be replaced by the cacao pod based rations supplemented with organic Zn and resulted in relatively similar ADG of 92.19 g/h vs 93.44 g/h.
