    Characteristics of reproductive performance of Garut rams

    Basic information on reproductive potency of Garut rams is necessary in order to identify the capacity of rams in producing chilled or frozen semen. Eight Garut rams (three to five years old) were used in this study. The male sexual behaviors were observed and semen was collected once a week using artificial vagina. Semen quality was evaluated and its potency to produce frozen semen was calculated. Results of this study indicated that first, second, and third ejaculations were at the 29, 87 and 176th seconds, respectively. Fresh semen volume, sperm concentration, motility, intact acrosomal cap, and intact plasma membrane were 0.99 ml, 3224 million/ml; 76.67; 86.13 and 87.73%, respectively. Protein value, fructose, vitamin C, vitamin E, sodiu, potassium, calcium, magnesium, phosphor, chloride, and mangan in seminal plasma of fresh semen were 4140, 180, 3.2, 24, 180, 117, 9, 6.12, 60, 104, and 5 mg/ml, respectively. Measurement of head length, width, and length of sperm tail were 6.59, 3.99, and 42.65 μm, respectively. Length and width measurement of right and left testes, and scrotal circumference were 12.71, 6.5, and 32.36 cm, respectively. Capacity of each Garut rams to produce frozen semen from three consecutive ejaculations are 35.88 mini straw with the cencentration of 200 million motile sperm per 0.25 ml.   Key words: Reproductive characteristics, Garut rams

    Preservation of Bacillus pumilus PU4-2 xylanases by immobilization technique into pollard and cation addition

    Utilization of by-product from agriculture as alternative source of feedstuff has been widely practiced. However their usage is limited due to high fiber content and low nutrient digestibility. The use of specific hydrolizing enzymes, xylanases are gaining importance because of their wide application in various industrial sectors especially in bioconversion of hemicellulosic material. This experiment was done to evaluate the effect of cation addition and immobilization of enzyme into pollard on stability of B. pumilus xylanase. The enzyme extract was purified by precipitation with 75% ammonium sulphate. Four kinds of cation (Ca2+, Fe3+, Mg2+, Zn2+) were added to the purified enzyme, at concentration of 1m M and stored at 4 and 27˚C. For immobilization process, the optimum enzyme concentration that will be added to pollard has been evaluated by analysis of xylanase activity and their recovery. The specific activity of enzyme after precipitation increased 1.8 times, from 420.3 to 765.2 U/mg protein. All cations act as activator which relative activity become 130.6; 139.0; 103.8 and 163.5% respectively. Concentration of 0.5mM Ca2+ and Fe3+ were most able to keep xylanases activity stable at 4˚C. The optimum composition of enzymes and pollard was 1.5 ml for 5 gram of pollard with recovery of xylanases activity of 82.2%. In immobilized enzyme, the activity of enzyme without cation addition is higher than that with addition of Ca2+ and Fe3+. Activity of enzyme stored at 4˚C is more stable than that at 27˚C. Immobilized enzyme is more stable for storage, which lasted for 7 weeks at 27˚C and 12 weeks at 4˚C compared to liquid enzyme which lasted for only 7 days at 27˚C and 13 days at 4˚C.     Utilization of Probiotic and Organic-Cn on Ruminal Ecosystem In Vitro

    Improvement of nutrient intake was needed to increase cattle productivity. Probiotics and organic Cr, as feed additives, might have beneficially improve ruminant microbial ecosystem, and optimize digestion process by rumen microbes. The objective of this study was to examine the effects of combination between probiotic or mixed probiotcs and organic Cr on rumen fermentation activity. An in vitro study was held based on randomized block design with 5 treatments and 3 replications. The treatments were (A) Control ration, (B) Control ration + probiotic TSD 10, (C) Control ration + probiotic TSD 10 + organic Cr, (D) Control ration + mixed probiotics, and (E) Control ration + mixed probiotics + organic Cr. Organic Cr concentration was supplemented 2 ppm, whereas probiotics supplement contained 109 cfu/g. Supplementation of probiotic and organic Cr increased dry matter and organic matter digestibility but decreased total VFA and NH3 concentration. Treatment E (combination between mixed probiotics and organic Cr) gave the highest dry matter (52.45 %) and organic matter (51.96 %) digestibilities. It also tended to increase NH3 and total VFA production. Supplementation of single or mixed probiotics showed no difference for dry matter and organic matter digestibilities, and proportion of individual VFA. Mixed probiotics gave higher VFA and NH3 concentration compared to single probiotic. Probiotics supplementation resulted in a tendency for higher acetate proportion, while organic Cr supplementation resulted in a tendency of higher propionate proportion.     Growth of reproductive organ and its effect on laying performance of medium ype layer due to different levels of dietary protein in growing period

    The purpose of this experiment was to study the optimal of dietary protein level of medium type layer in growing period associated with the growth of reproductive organ and the effect on laying performance. The experiment used 480 pullets of 10 weeks old grouped into three treatments of three protein levels, 12, 15 and 18. In laying period all of the treatments used the same ration with 15% protein. The results showed that dietary protein level in growing period significantly affected the growth of reproduction organ, growth performance and eggs quality. Effects of treatment in laying period was significantly different on laying performance only in early laying period, and subsequent period was not differently different. The conclusion of this experiment was the level of 12% adequate for minimal growth, level of 15% adequate for optimal growth and level 18% showed better performances during laying period. The results suggest that in growing period the dietary protein level is not below 15%.     Key Words: Protein, Reproduction Organ, Laying Performanc

    The utilization of alfalfa that planted at Tobasa highland, North Sumatra for growing Boerka goat feed

    Alfalfa (Medicago sativa L.) is a herbaceus legume which is potential to be used as goat feed for it’s high production and nutritive value. This research was aimed to study the adaptation of alfalfa planted at highland-moderate climate and it’s utilization for goat feed. This research concists of two activities, ie 1) Agronomy of alfalfa that adapted to highland as a goat feed resource, and 2) The alfalfa usage technology as goat feed. On the first activity alfalfa was planted on highland-moderate climate Gurgur, Tobasa District, North Sumatra Province. Data was collected included: growth percentage, morphology and production aspects, and nutritive value. The harvesting was conducted for three times, where the first cutting had done at 100 days after planting. Investigation of morphology characterirtics was done before alfalfa harvesting. The utilization of alfalfa as goat feed was carried out on the second activity which was arranged in a completely randomized design. Twenty male Boer x Kacang crossbred  (Boerka) goats of 5-6 months of age with average body weight 14.2±0.8 kg were randomly assigned to four feed treatments where each treatment consited of five replications. The four feed treatments were: P1 = 100% grass + 0% alfalfa; P2 = 90% grass + 10% alfalfa, P3 = 80% grass + 20% alfalfa, and P4 = 70% grass + 30% alfalfa. Data observation included dry matter intake, average daily gain, feed efficiency, and income over feed cost. Results showed that alfalfa growth percentage was 65% with good growth and high either production or nutritive value. The average shoot dry matter production was 438.6 g/m2 which was equivalent to 26.3 t/ha/yr. The crude protein content of alfalfa on the first, second and third harvesting were 17.93; 21.89 and 17.73 per cent, respectively. The utilization of alfalfa that had been processed to be crude-meal can be applied as goat feed. Supplementation of 70% grass and 30% alfalfa meal showed the best results: ADG 59.17 g/d, feed efficiency 0.14, and IOFC Rp 736/h/d. Key Words: Alfalfa, Herbage, Production, Meal, Feed, Goa

    Effect of urea or coconut cake supplementation on nutrient intake and digestion of Bali cows maintained on tropical grass hay

    Three rumen cannulated Bali cows were fed basal diet consisting of about 2 kg DM of medium quality grass hay (CP = 7.47%) together with ad libitum amount of low quality grass hay (CP = 3.5%) (HH). In the supplemented rations, this basal ration was supplemented with isonitrogenous amount of urea (HU) or coconut cake (HC). The experimental was on design 3 x 3 latin square aiming to study effects of different source of nitrogen supplementation on the intake of poor quality hay (PQH), total diet, nutrients supply and nitrogen utilisation by the animals. Changes in rumen environment and dry matter and protein degradation after supplementation were also investigated. Neither urea or coconut cake (CC) supplementation increased hay intake but the total dry matter intake was significantly higher (P 0.05) in HC than in HH and HU. Digestibility of EE, CHO and GE but not NDF were also improved with CC supplementation. In contrast, CC supplementation tended to depress CF apparent digestibility. Rumen pH was significantly elevated (P 0.05) with urea but it was reduced (P 0.05) with CC supplementation. Both supplements increased rumen ammonia concentration, while total as well as individual VFAs remained unaffected. Dry matter and NDF rumen degradability tended to increase with urea but not with CC supplementation.     The effectivity of Annona squamosa L seeds extracted by diverse organic solvents: water, methanol and hexane against mortality of tick larvae, Boophilus microplus in vitro

    Boophilus microplus is the most important pest in livestock industries. The use of synthetic chemical acaricides is the main method of tick control, however, chemical acaricides are expensive, and they are harmful to environment and cause strain resistance. The aim of study was to investigate the affectivity of Annona squamosa L seeds extracted by diverse organic solvents such as water, methanol and hexane against mortality of Boophilus microplus larvae in vitro. Five hundred and fifty larvae were used in this study and divided into three groups e.g. water (3, 4 and 5% concentration), methanol and hexane extract groups (0.25, 0.50, and 0.75% concentration). Coumaphos (0.50%) was used as a positive control. The larvae were dipped into extract solution for 10 seconds and dried using filter paper. Their mortality was observed from one to five hours. The mortality data were transformed to Abbot formula and analyzed using probit analysis with 95% significant level. This study showed that the active compound of Annona squamosa L seeds had effectively contact toxic property for B. microplus larvae at 5, 0.50, and 0.75% for water, methanol and hexane extractions, respectively. The lethal concentrations of methanol extract (LC50, LC90, and LC95) were lower than hexane extract e.g. 0.32, 0.86, and 1.13%, respectively and for hexane extract were 0.35, 1.11, and 1.54%, respectively at fifth hour. The lethal times of methanol extract on 0.50% concentration were shorter than hexane extract e.g. 3.12 hours (LT50), 5.86 hours (LT90), and 7.00 hours (LT95) and for hexane extract on 0.75% concentration were 3.26 hours (LT50), 6.21 hours (LT90), and 7.45 hours (LT95). Water extract of 5% concentration was effective for traditional farmer in rural area due to easy and cheap method. The lethal concentrations of water extract on fifth hour were 2.02% (LC50), 4.00% (LC90), and 4.85% (LC95) and the lethal time on 5% concentration were 2.54 hours (LT50), 4.13 hours (LT90), and 4.75 hours (LT95).     Key Words: Annona squamosa, Boophilus microplus, Water Extract, Methanol, Hexan

    Viability of Timor deer stag (Cervus timorensis) spermatozoa extended in tris egg yolk diluent with different sources of carbohydrate and storage at room temperature

    The successful  sperm preservation, influenced by the capability of its extender on the maintenance the sperm quality during storage. The carbohydrate such as glucose and fructose were the common sugar added on the mammalian sperm extender to support their live and motility. The sucrose was the main carbohydrate in  Timor deer stag seminal plasma. The experiment was conducted to evaluate the effect of carbohydrates  in Tris egg yolk (TEY) extender on the motility and viability of stag sperm, stored in room temperature (27-28 oC). The semen was collected using electro ejaculator from five Timor deer stags at hard antler stage, 3-5 years old, body weight of 64-102 kg with normal testes. The semen was than evaluated macro-and microscopically and divided into 3 aliquots.  Each of them was diluted with TEY-glucose (TEYG), TEY-fructose (TEYF) and TEY-Sucrose (TEYS) with the concentration of spermatozoa 100 x 106 ml-1. The extended semen was than stored at room temperature. The sperm motility and viability were evaluated every 3 hours. Result of the experiment showed that the semen volume was 2.06 ± 0.63 ml, pH 7.03±0.13, yellow white until creamy in color and the consistency ranged from normal to thick. The mass movement between ++ to +++ and the sperm motility was 68.67 ± 7.4%. The average of sperm concentration was 842.35 ± 258.14x106 ml-1, the viable sperm was 78.11 ± 3.61%, the sperm abnormality was 7.31 ± 2.98%. The percentages of sperm motility on TEYG (18.00 ± 17.63%) and TEYS (21.83 ± 15.92%) were higher compare to TEYF (4,00 ± 0,00%) extender in 24 hours observation. The percentage of sperm viability showed the same pattern. The sperm viability in TEYG (28.17 ± 20.06) and TEYS (24.00 ± 22.59%) (P0.05) were significantly higher compare to TEYF (4.00 ± 0.00%).  It is concluded that the deer stag sperm can use the three sugars for their nutrition source. The diluted sperm still can be used  for artificial insemination after 12 hour storage. Key Words: Liquid Semen, Deer, Room Temperature, Carbohydrat

    Substitution of commercial concentrate with soy bean meal protected by tannin from banana stem juice for lambs

    Study was conducted to evaluate the optimal utilization of protected soy bean meal with secondary compound from banana stem juice in ration for sheep and its effect on sheep performance. Soy bean meal was mixed with banana stem juice at ratio 1:1 (w/v) then dried in the oven at temperature 90oC. This protected soy bean meal was used to substitute commercial concentrate in sheep ration. The experiment used 24 head male lamb Sumatera composite breed. The lambs were grouped into six group based on its body weight and was assigned to one of 4 diets treatment. The sheep was fed on grass basal diet and supplemented with commercial concentrate. Data recorded were feed consumption, nutrient digestibility, average daily gain, feed efficiency and nitrogen utilization. Study was conducted in randomized complete block design and data obtained were analyzed using general linier model from SAS program.  Results show that dry matter intake (DMI) significantly (P 0.05) increased with concentrate substitution by protected soy bean meal, however, there was no significant different (P 0.05) between R10, R20 and R30. The increasing in DMI is followed by the increasing crude protein (CP) from 8.75 (R0) to 10.64; 11.68 and 12.32 g/BB0.75 respectively for R10; R20 and R30. Commercial concentrate substitution by protected soy bean meal significantly increased DM and CP digestibility at all levels. However, this substitution did not significantly affected organic matter (OM), neutral detergent fiber (NDF) and acid detergent fiber (ADF) digestibility. Nitrogen excretion in urine was only increased at this level 30% substitution but nitrogen retention increased at substitution levels 20 and 30%. From this study can be concluded that commercial concentrate substitution with protected soy bean meal in the diet only increased CP consumption and CP digestibility but not average daily gain of lamb.     The Effect of sheep serum and estrus sheep serum on in vitro maturation and fertility rate of ewe oocyte

    The main objective of this research was to obtain the effect of sheep serum (SS) and estrus sheep serum (ESS) on in vitro maturation oocyte and ovine fertilization. This study was carried out in experimental laboratory in animal reproduction laboratory, Faculty of Animal Husbandry, Padjadjaran University. Results showed that treatments significantly (P0,05) influenced on maturation rate for germinal vesicle (GV), germinal vesicle breakdown (GVBD), metaphase-I (M-I) and metaphase-II (M-II), but no significantly (P0.05) results observed on ovine in vitro fertilization (1, 2 and 2 pronuclei). Concentration of 10-20% ESS in CR1aa media were significantly (P0,05) better than that of SS on maturation rate of ovine oocyte.       Key Words: Sheep Serum, Estrus Sheep Serum, Maturation, Fertilizatio
