    Productivity of Local Goats Supplemented with Acacia villosa and Coripha gebanga

    Most feed for goats in the villages of West Timor, Indonesia, came from communal grazing areas, consist of native grass during wet season, crop residues and tree leaves (Sesbania and Acacia species) in dry season. This study was aiming at examining the growth of goat supplemented with local feed. Twenty four goats were used, the average initial live weight was 12.5 kg. Four feeding treatments were applied: T1-200 g cut grass; T2-200 g Acacia villosa; T3-200 g Coripha gebanga; T4-100 g A. villosa + 100 g  C. gebanga. Live weights and feed consumption were analyzed using repeated measures, analysis of variance. The average live weight showed a small increase, as well as daily weight gain of goats of which different significantly (PA. villosa, C. gebanga, and A. villosa plus C. gebanga, respectively. Goats given C. gebanga and mixed Acacia and C. gebanga gave higher average weight gain, but also consumed more feed than those given cut grass or A.villosa (P < 0.01). Supplementing feed to maintain growth of goats during dry seasons was better on A. villosa than on palm pith and its combination

    Supplementation of Sapindus rarak and Garlic Extract in Feed Containing Adequate Cr, Se, and Zn on Rumen Fermentation

    The objective of the study was to evaluate the effect of Sapindus rarak extract (SRE) with or without garlic extract (GE) on in vitro ruminal fementation. This research was conducted experimentally with a randomized block design, with 7 treatments and 5 blocks. The treatments were: R0:  dairy cow feed; R1: R0 + 1.5 ppm Cr + 0.3 ppm Se + 40 ppm Zn; R2: R1 + 1.8 g/kg methanol extract of lerak fruit  meal (SRE); R3: R2 + 0.25 ppm of garlic extract (GE); R4: R2 + 0.50 ppm of GE; R5: R2 + 0.75 ppm of GE; R6: R2 +1.0 ppm of GE. The results showed that the supplementation of SRE alone or without GE did not affect the pH, however, it decreased crude fiber digestibility. The supplementations of SRE and GE, decreased crude fibre digestibility as much as 13.01% up to 16.6%. The supplementation of 1.8 g/kg SRE + 0.25 ppm GE in the dairy cattle diet was able to decrease ace-tate, protozoal population and increase propionate. The supplementation of 1.8 g/kg SRE and 0.25 ppm garlic represents the best combination for dairy cattle feed in improving ruminal fermentation based on feed digestibility, fermentation products, and rumen bacterial population.

    The Incorporation of Chromium in Rice Straw Fermented with Ganoderma lucidum

    The research was designed to evaluate chromium incorporation by Ganoderma lucidum in rice straw substrate supplemented with chromium chloride. Treatments were combination of Cr level (0, 500, 1000, 1500, 2000, 25000, and 3000 ppm) and fermentation time (0, 2, 3, and 4 wk). The treatments were arranged in a factorial 7 x 4 and allocated in completely randomized design with three replications.  G. lucidum was grown in potato dextrose agar (PDA) medium for 10 days and was inoculated to the substrate which have been sterilized and mixed with CrCl3.6H2O. The moisture of substrate was maintained at 70%. Fiber and protein components of growth media of G. lucidum was determined and analyzed for their Cr content. The finding result showed that addition of Cr up to 3000 ppm into the substrate stimulated the G. lucidum growth.  Chromium was incorporated into the fiber and protein components of the growth substrate of G. lucidum during fermentation. Incorporation of Cr into the protein of substrate containing3000 ppm Cr  was highest when fermented for 4 wk.  Protein component of substrate contained 9.29% Cr while in NDF and ADF was 27.20% and 10.55% Cr, respectively. It is concluded that Cr was incorporated into the G. lucidum cells during fermentation.

    In Vitro Digestibility of Ration Containing Different Level of Palm Oil Frond Fermented with Phanerochaetae chrysosporium

    Phanerochaete chrysosporium was widely used to delignify agricultural waste product and improve biodegradation of the substrate as animal feed.  The experiment was carried out to increase the use of palm oil fronds as a substitute material for napier grass through biodegradation process with P. chysosporium. A completely randomized design with four treatments and four replications was used. The treatments were ration containing 60% napier grass (R1), ration containing 40% napier grass and 20% fermented palm oil frond (R2), ration containing 20% napier grass and 40% fermented palm oil frond (R3), ration containing 60% fermented palm oil frond (R4). Fourty percent concentrate was included in all treatment rations. Parameters measured were in vitro digestibilities of dry matter, organic matter, crude fiber, NDF, ADF, NH3, TVFA, and ruminal cellulolitic bacteria. Results showed that increasing level of fermented palm oil frond in the ration reduced (P<0.05) digestibilities of dry matter, organic matter, crude fiber, NDF, ADF, N-NH3, TVFA concentration and number of ruminal cellulolytic bacteria. It is concluded that fermentation of palm oil frondwith P. chysosporium decrease lignin content by 47.79%, but increasing the fermented palm oil frond in the ration reduces nutrient digestibilities, N-NH3 and TVFA concentrations and rumen cellulolytic bacteria counts. Fermented palm oil frond up to 40% could be used as a substitute for forages in ruminant rations

    Preferences of Rusa timorensis to Grasses and Their Body Morphometry and Velvet Antler Characteristics

    Timor deer (Rusa timorensis) is a wild native animal to Indonesia. The stags produce a high economic value of  velvet antler (VA). This study was designed to evaluate the feed preference, nutrient consumption, age and morphometric of deer in relation to their velvet antler product (weigth and length) and quality (mineral and amino acids). Five stags with three ages classes were offered 3 combinations of 4 grasses in a captive area. Grasses were offered for 3 periods of 12 consecutive days during velvet antler growth period. Velvet antlers were harvested at 36 days old and analysed for their chemical content. The result showed that Timor deer had different preference to the grasses offered. Deer had a high preference on the combination of Pennisetum purpureum, Commelina benghalensis, Hierochloe horsfieldii, and Sorghum caudatum. High intake of grasses increased body weight. The regresion equation between the weight or length of velvet antler withage (year) and girth size (cm) were: Y (velvet antler weight)= 0.43 + 0.03 (age), r2= 0.99, P<0.05, and Y (length of velvet antler) = 5.7 + 0.31 (girth size), r2= 0.99, P<0.05. Aspartic acid, glutamic acid, and proline content of velvel antler were high in the older deer. The older deer having high girth size had the higher quality of velvet antler

    Microbial and Oligosaccharides Treatments of Feces and Slurry in Reducing Ammonia of the Poultry Farm

    This study was conducted to investigate the effectiveness of Lactobacillus sp and fructooligosaccaride (FOS) to reduce the volatile ammonia formation from chicken excreta and layer slurry. For each treatment-replication, 150 g of fecal material were collected from the poultry farm and placed in 500 ml beaker glass. The fecal sample was then treated with 2% Lactobacillus sp (2.6x106) cfu/g and 2% FOS and covered with plastic wraps. The volatile ammonia contents and pH were measured after one hour of standing (0 d) and repeated at 48 h intervals for 6 d. For the dropping slurry study, 300 g of each layer dropping slurry sample were used. Results indicated that 2% Lactobacillus sp or FOS supplementations in the feces and dropping slurry after 1 h up to 6 d reduced the ammonia odor formation, fecal pH, and moisture content. The Lactobacillus sp and FOS treated manure resulted in increasing  Lactobacillus sp count and reducing in E. coli, Salmonella, and Campylobacter in 6 days for both feces and layer dropping slurry. In addition, reducing moisture content was observed in treated manure. It is concluded that Lactobacillus sp and FOS reduced the volatile ammonia formation and pathogenic bacteria from chicken excreta and layer slurry.

    Allelic Frequency of Kappa-Casein Locus (Asp148/Ala) in F1: Simmental (Bos taurus) x Ongole Grade (Bos indicus)

    This study was conducted to detect the genetic variants (single nucleotide polymorphism) of kappa-casein locus (Asp148/Ala) in F1: Simmental (Bos taurus) x Ongole grade (Bos indicus), SIMPO. Genomic DNA was isolated from blood sample of 40 SIMPO (21 males and 19 females). A 780 bp specific fragment of kappa-casein gene spanning from the forth exon region (517 bp) to forth intron (263 bp) was successfully amplified. The result of the PCR-RFLP (Polymerase Chain Reaction - Restriction Fragment Length Polymorphisms) analysis using HindIII enzyme showed that two genotypes (AA and AB) were found at this locus in SIMPO. The frequencies of A and B alleles in SIMPO were 0.79 and 0.21, respectively. The frequency lies between B. taurus (Simmental) and B. indicus group

    Application of Electrical Properties to Differentiate Lard from Tallow and Palm Oil

    This study aimed to differentiate lard from tallow and palm oil based on its electrical properties, namely conductance, impedance and capacitance. These properties were measured at spectra frequencies of 4.20 to 5.00 MHz in room temperature (26-27 oC). Statistic multivariate that consist on principal component analysis (PCA) and cluster analysis (CA) were used to evaluate the data. The results showed that lard and tallow can be differentiated using whole parameters electrical properties of materials. On the other hand, lard and palm oil can only be differentiated using part of the material electrical properties. Good performance of differentiation process was obtained using PCA model at 4.91 to 4.98 MHz. The first two components of PCA, which was derived from conductance, impedance and capacitance, contributed more than 90% of the total variances. CA showed that lard and tallow are different groups based on the Euclidean distance of each electrical properties. This technique can be potentially developed as an electrical sensor for differentiation lard to tallow and palm oil

    Isolation and Characterization of Plantaricin Produced by Lactobacillus plantarum Strains (IIA-1A5, IIA-1B1, IIA-2B2)

    Bacteriocins produced by Indonesian lactic acid bacteria Lactobacillus plantarum IIA-1A5, IIA-1B1, IIA-2B2 were purified and characterized. Plantaricin W gene had been successfully amplified from all strains. This amplicon showed the expected 200 bp size of plantaricin W gene. This bacteriocins purified from L. plantarum IIA-1A5, IIA-1B1, and IIA-2B2 were named plantaricin IIA-1A5, IIA-1B1, and IIA-2B2. Purification by cation exchange chromatography increased the purity (fold) and activity of plantaricins. Purity of plantaricin IIA-1A5 was increased by 3.13 fold with specific activity 13.40 AU/mg. Plantaricin IIA-1B1 had 2.98 fold purity with specific activity 5.12 AU/mg, while purity of plantaricin IIA-2B2 was 1.37 fold with specific activity 7.70 AU/mg. All plantaricins could inhibit the growth of pathogenic bacteria, such as Escherichia coli, Salmonella typhimurium, Bacillus cereus, and Staphylococcus aureus. Plantaricins could be digested by trypsin. Stability of plantaricins at 80 oC for 30 min and at 121 oC for 15 min were affected by type of plantaricin and species of pathogenic bacteria. Generally, plantaricin IIA-1A5 was better as antimicrobial agent than plantaricin IIA-1B1 and plantaricin IIA-2B2

    Histomorphology and Physical Characteristics of Buffalo Meat at Different Sex and Age

    The aim of this experiment was to evaluate histology and physical variables of buffalo meat at different sex and ages. Thirty head of buffaloes were used in this experiment. Muscle fiber diameter, fasciculus diameter, thickness of connective tissue, pH, water holding capacity, Warner-Bratzler shear force and cooking loss were measured in this experiment. Analysis of variance of randomized factorial design was used to analyse the data. The differences were analysed by Tukey test. The result showed that muscle fiber diameter was influenced by ages. The difference muscle fiber diameter among ages indicated that 8-12 mo and 1.5 yr old have smaller diameter compared to three year old (P<0.05), but  there was no differences between  two, three and four years old. The fasciculus diameter, thickness of connective tissue, tenderness, pH value, water holding capacity, and cooking lost  indicated no significant found in all stage of age and sex of samples.
