No genome-wide DNA methylation changes found associated with medium-term reduced graphene oxide exposure in human lung epithelial cells
The presence of nanomaterials in our everyday life is ever increasing, and so too are concerns about the possible health consequences of exposure to them. While evidence of their biological activity is growing, there is still scant knowledge of the epigenetic mechanisms that could be at play in these processes. Moreover, the great variability in the chemical and physical structures of these compounds handicaps the study of their possible health risks. Here we have synthesized reduced graphene oxide (rGO) through the thermal exfoliation/reduction of graphite oxide, and characterized the resulting material. We have then made use of Illumina's MethylationEPIC arrays and bisulphite pyrosequencing to analyse the genome-wide and global DNA methylation dynamics associated with the medium-term exposure of human lung epithelial cells to rGO at concentrations of 1 and 10 µg/mL. The results show no genome-wide or global DNA methylation changes associated with either condition. Our observations thus suggest that medium-term rGO exposure does not have significant effects on the DNA methylation patterns of human lung epithelial cells

Improved Cross-Polarization Performance in Reflectarray Antennas by Direct Optimization of the XPD and XPI Parameters
Current satellite missions for communications demand a high polarization purity, with figures of merit such as the crosspolar discrimination (XPD) better than 33 dB. In order to accomplish these values, some kind of optimization must be carried out to improve cross-polarization performance. The most common approach is to minimize the crosspolar component of the radiation pattern in the region of interest. Nevertheless, this strategy produces suboptimal results since the figure of merit for cross-polarization performance is optimized indirectly. Thus, it is proposed to directly optimize the XPD to improve the polarization purity of reflectarrays for satellite missions. For that purpose, the generalized Intersection Approach algorithm is employed to optimize a very large shaped-beam reflectarray for a direct broadcast satellite application with a European coverage. It is shown that directly optimizing the cross-polarization figure of merit provides better results than the usual approach of minimizing the crosspolar pattern. This work was supported in part by the Ministerio de Ciencia, Innovación y Universidades under project TEC2017-86619-R (ARTEINE); by the Ministerio de Economía, Industria y Competitividad under project TEC2016-75103-C2-1-R (MYRADA); by the Gobierno del Principado de Asturias/FEDER under Project GRUPIN-IDI/2018/000191; by the Gobierno del Principado de Asturias through Programa "Clarín" de Ayudas Postdoctorales / Marie Curie-Cofund under project ACA17-09; by Ministerio de Educación, Cultura y Deporte / Programa de Movilidad "Salvador de Madariaga" (Ref. A frequency-tunable nanomembrane mechanical oscillator with embedded quantum dots
Hybrid systems consisting of a quantum emitter coupled to a mechanical oscillator
are receiving increasing attention for fundamental science and potential applications in quantum technologies. In contrast to most of the presented works, in which the oscillator eigenfrequencies are irreversibly determined by the fabrication process, we present here a simple approach to obtain frequency-tunable mechanical resonators based on suspended nanomembranes. The method relies on a micromachined piezoelectric actuator, which we use both to drive resonant oscillations of a suspended Ga(Al)As membrane with embedded quantum dots and to fine tune their mechanical eigenfrequencies. Specifically, we excite oscillations with frequencies of at least 60 MHz by applying an AC voltage to the actuator and tune the eigenfrequencies by at least 25 times their linewidth by continuously varying the elastic stress state in the membranes through a DC voltage. The light emitted by optically excited quantum dots is used as sensitive local strain gauge to monitor the oscillation frequency and amplitude. We expect that our method has the potential to be applicable to other optomechanical systems based on dielectric and semiconductor membranes possibly operating in the quantum regime
El superestrato románico: La huella del español, del francés y del occitano en el siciliano contemporáneo
Sicily has always been a territory under dispute by multiple peoples that eventually managed to mold the culture of the inhabitants of the island. As a result, the Sicilian language absorbed the language of the ensuing settlers, particularly Spanish and French. In view of this, this article addresses the analysis of the Spanish, French and Occitan traces, three Romance variants that, through the semantic superstratum, have had an impact on the linguistic configuration of the Sicilian language. With the focus on the dialectal lexicon used by the Palermitan writer, Giuseppina Torregrossa, in her debut novel, L’Assaggiatrice, this study confirms what previous specialized autho-rities on the field observed in past times: that the Ibero-Romance and Gallo-Romance influence have been, and still are, key to the shaping of the Sicilian language, a linguistic variant that is most frequently used in insular literature regardless of the growing social interest in standard Italian after the unification.
Chromatin regulation by Histone H4 acetylation at Lysine 16 during cell death and differentiation in the myeloid compartment
Histone H4 acetylation at Lysine 16 (H4K16ac) is a key epigenetic mark involved in gene regulation, DNA repair and chromatin remodeling, and though it is known to be essential for embryonic development, its role during adult life is still poorly understood. Here we show that this lysine is massively hyperacetylated in peripheral neutrophils. Genome-wide mapping of H4K16ac in terminally differentiated blood cells, along with functional experiments, supported a role for this histone post-translational modification in the regulation of cell differentiation and apoptosis in the hematopoietic system. Furthermore, in neutrophils, H4K16ac was enriched at specific DNA repeats. These DNA regions presented an accessible chromatin conformation and were associated with the cleavage sites that generate the 50 kb DNA fragments during the first stages of programmed cell death. Our results thus suggest that H4K16ac plays a dual role in myeloid cells as it not only regulates differentiation and apoptosis, but it also exhibits a non-canonical structural role in poising chromatin for cleavage at an early stage of neutrophil cell death
Sequential two-step stereoselective amination of allylic alcohols through combination of laccases and amine transaminases
Artículo de investigación destinado a la obtención de aminas alílicas enantioenriquecidas mediante el desarrollo de un proceso en cascada secuencial multienzimáticaA sequential two-step chemoenzymatic methodology has been described for the stereoselective synthesis of (3E)-4-
(het)arylbut-3-en-2-amines in a highly selective manner and under mild reaction conditions. The approach consists in the oxidation of the corresponding alcohol precursors using the catalytic system composed by the laccase from Trametes versicolor and the oxyradical TEMPO, followed by the asymmetric biotransamination of the corresponding ketone intermediates. Optimisation of the oxidation reaction, exhaustive amine transaminase screening for the biotransaminations and the compatibility of both enzymatic reactions have been deeply studied, searching for the design of a compatible sequential cascade. This synthetic strategy has been successfully achieved, the combination of enzymes displaying a broad substrate scope as 16 chiral amines have been obtained in moderate to good isolated yields (29-75% isolated yield) and excellent enantiomeric excess (94->99). Interestingly, both amine enantiomers can be achieved depending on the selectivity of the amine transaminase employed in the system.Agencia Estatal de Investigación.
