    CYP450 polymorphisms and clinical pharmacogenetics of ibuprofen after lower third molar extraction

    This study hypothesized that drugs accumulate in the bloodstream of poor-metabolizing patients and may have more adverse effects and different pain perceptions and aimed to investigate the influence of CYP450 polymorphisms on acute postoperative pain, swelling, and trismus controlled by ibuprofen (600 mg) in 200 volunteers after dental extraction. In addition, surgical outcomes can determine pain, edema, and trismus and indicate inflammatory reactions after oral surgeries. Genetic sequencing was performed to identify CYP450 polymorphisms and the surgical parameters evaluated: pre and postoperative swelling, trismus, and temperature; self-reported postoperative pain with visual analog scale (VAS); rescue medication consumed; and severity of adverse reactions. A multiple linear regression model with independent variables [single nucleotide polymorphisms (SNPs), BMI (body mass index), duration, and difficulty of surgery] and dependent variables [postoperative pain by sum of pain intensity difference (SPID), trismus, and swelling] was used for analysis. The duration of surgery was a predictor for pain at 8 h and 96 h after surgery, and BMI was a predictor for both swelling and trismus on the 2nd postoperative day. When evaluating CYP2C8 and C9 genotyped SNPs, it was observed that normal metabolizers showed higher pain levels than the intermediate/poor metabolizers on the postoperative periods as compared with time 0 h. In another analysis, the poor metabolizers for CYP2C8 and C9 presented lower levels of postoperative pain after 8 h and used rescue medication earlier than normal metabolizers. Ibuprofen 600 mg was very effective in controlling inflammatory pain after lower third molar surgeries, without relevant adverse reactions; although in a very subtle way, patients with poor metabolism had higher levels of pain in the first hours, and no longer after 8 h, and used pain relief medication earlier.     Microbial degradation of petroleum is a worldwide issue, which causes physico-chemical changes in its compounds, diminishing its commercial value. Biosurfactants are chemically diverse molecules that can be produced by several microorganisms and can enable microbial access to hydrocarbons. In order to investigate both microbial activities, function-driven screening assays for biosurfactant production and hydrocarbon biodegradation were carried out from a metagenomic fosmid library. It was constructed from the total DNA extracted from aerobic and anaerobic enrichments from a Brazilian biodegraded petroleum sample. A sum of 10 clones were selected in order to evaluate their ability to produce exopolymers (EPS) with emulsifying activity, as well as to characterize the gene sequences, harbored by the fosmid clones, through 454 pyrosequencing. Functional analyses confirmed the ability of some clones to produce surfactant compounds. Regarding hydrocarbon as microbial carbon sources, n-alkane (in mixture or not) and naphthalene were preferentially consumed as substrates. Analysis of sequence data set revealed the presence of genes related to xenobiotics biodegradation and carbohydrate metabolism.     Exponential improving in the activity of Pt/C nanoparticles towards glycerol electrooxidation by Sb ad-atoms deposition

    Tuning the chemical composition of heterogeneous nanocatalysts has emerged as an efficient strategy to improve their electroactivity and selectivity for electrolizers and fuel cells application. The stability of a catalyst is ascribed as the ability of the nanoparticles (NPs) in maintaining their chemical composition and physical structure during recycles of use. By an electrochemical procedure, here we'decorated platinum nanoparticles supported on carbon with different coverage degrees of antimony ad-atom (theta(sb)) to investigate glycerol electrooxidation reaction (GEOR). The successful decoration of Pt/C with Sb was monitored by electrochemical and physicochemical characterizations. theta(sb) is highly dependent on the upper potential applied. Sb atoms are rapidly leached from Pt surface in potentials greater than 0.7 V in acid solution while they remain stable during several cycles in potentials lower than that. We showed that the current density of GEOR exponentially increases with the increase of Sb coverage, reaching 109 times the pseudo stationary current density of Pt/C for high theta(sb). Furthermore, the ad-atom facilitates the reaction by shifting the onset potential towards lower potentials.     Cytotoxicity and inflammatory response of different types of provisional restorative materials

    To evaluate the cytotoxicity and inflammatory response of different types of provisional restorative materials to mice gingival fibroblasts. Cytotoxicity of provisional material discs (thermal-polymerized acrylic resin; auto-polymerized acrylic resin; bisacrylic resin; nano-ceramic resin for CAD/CAM and prefabricated polymer block for CAD/CAM) to Mice (Balb/c) gingival cell were investigated under direct and indirect contact (extracts) at 24, 48 and 72 h, using the MTT and Alamar blue assays. Materials extracts (24 h) were applied to the cell culture (indirect contact) or cells were seeded on discs of provisional materials, and the cytotoxicity and production of IL-6, IL-1β and TNF-α after 24, 48 and 72 h were analyzed through MTT, Alamar Blue® and ELISA. Culture medium was used as control for indirect contact assay (extract) and the surfaces of the wells without discs of provisional materials were used as control for direct contact assay. Results were analysed statistically by ANOVA followed by the Bonferroni-Test correction. Statistically significant differences were considered if P was .05). Auto-polymerized and bisacrylic resins increased IL-6, IL-1β and TNF-α levels mainly at 24 h when compared to the other materials (P < .001). Auto-polymerized and bisacrylic were more cytotoxic to mice gingival fibroblasts.     Prenatal and pubertal testosterone exposure imprint permanent modifications in the prostate that predispose to the development of lesions in old Mongolian gerbils

    The prostate is an accessory sex gland that develops under precise androgenic control. It is known that hormonal imbalance may disrupt its development predisposing this gland to develop diseases during aging. Although the hypothesis regarding earlier origins of prostate diseases was proposed many years ago, the mechanisms underlying this complex phenomenon are poorly understood. Therefore, the aim of this study was to evaluate the prostates of old male gerbils exposed to testosterone during intrauterine and postnatal life using morphological, biometrical, stereological, Kariometric, immunohistochemical, and immunofluorescence analyses. Our findings demonstrate that prenatal and pubertal exposure to testosterone increases the susceptibility to the development of prostate diseases during aging. The presence of a more proliferative gland associated with foci of adenomatous hyperplasia in animals exposed to testosterone during the prenatal and pubertal phase show that the utero life and the pubertal period are important phases for prostatic morphophysiology establishment, which is a determinant for the health of the gland during aging.     DAPT score and the impact of ticagrelor monotherapy during the second year after PCI

    This study assessed the ability of the dual-antiplatelet therapy (DAPT) score in stratifying ischemic and bleeding risk in a contemporary percutaneous coronary intervention (PCI) population. The DAPT score is recommended by guidelines as a tool to stratify ischemic and bleeding risk. Its utility in contemporary PCI is unknown. The study studied patients in GLOBAL LEADERS (A Clinical Study Comparing Two Forms of Anti-platelet Therapy After Stent Implantation) who were free of major ischemic and bleeding events and adhered to antiplatelet strategy during the first year after PCI. The primary ischemic endpoint was the composite of myocardial infarction or stent thrombosis. The primary bleeding endpoint was Bleeding Academic Research Consortium type 3 or 5. Outcomes from 12 to 24 months after PCI were compared according to the DAPT score. Of 11,289 patients that were event-free after the first year, 6,882 and 4,407 patients had low (<2) and high (≥2) DAPT scores, respectively. Compared with a low DAPT score, patients with a high DAPT score had a higher rate of the composites of myocardial infarction or stent thrombosis (0.70% vs. 1.55%; p < 0.0001). The rate of Bleeding Academic Research Consortium type 3 or 5 bleeding was 0.54% and 0.30% in the low and high DAPT score groups, respectively (p = 0.058). The effect of ticagrelor versus aspirin monotherapy on primary ischemic and bleeding endpoints during the second year were no different among the 2 groups. The DAPT score can stratify ischemic but not bleeding risk in a contemporary PCI population during the second year.     Articaine interaction with phospholipid bilayers

    Local anesthetics promote analgesia by interacting with excitable membranes. Articaine (ATC) has a unique composition among local anesthetics as it possesses a thiophene instead of the typical phenyl ring. Aiming to characterize the interaction of neutral articaine (nATC) with phospholipid membranes, we have employed a synergistic approach of experimental and computational techniques. Fluorescence measurements supported nATC partitioning into the membranes, since its intrinsic fluorescence anisotropy increased from 0.03 in water to 0.29 in the presence of egg phosphatidylcholine (EPC) liposomes, and the fluorescence of AHBA, a probe that monitors the water-membrane interface, was quenched by nATC. 1H NMR experiments revealed changes in the chemical shifts of articaine and EPC hydrogens after partitioning, and shorter T1 values of nATC hydrogens when inserted into the EPC vesicles. Contacts of nATC and the phospholipid polar head group were inferred from 2D-NOE. Taken together, these results indicate a superficial insertion of the nATC molecules inside EPC bilayers. This conclusion was confirmed by molecular dynamics simulations, which allowed the identification of the key interactions underlying the preferential location of nATC in the bilayer. Um dos objetivos desta pesquisa é verificar a dinâmica das manumissões no decorrer do tempo, jogando luz sobre seus padrões e movimentos, e investigar o perfil dos alforriados e demais pessoas envolvidas em tal processo. Além disso, por meio do estudo de casos, da reconstrução das experiências e estratégias de vida de mulheres e homens escravizados, busca-se compreender em que medida as interações sociais constituídas e mantidas por eles com outros indivíduos da sociedade escravista brasileira contribuíram para a conquista da liberdade de seus filhos recém-nascidos no ato do batismo. Para realizar tais propósitos, recorre-se a tipos documentais diversos, tais como fontes eclesiásticas, censitárias e cartoriais, e ao método de ligação nominativa de fontes. Espera-se, assim, trazer novos aspectos das relações estabelecidas entre a população escravizada e outros sujeitos, avançar na compreensão dos significados atribuídos por eles ao fenômeno da alforria e contribuir com os principais debates em pauta na historiografia que se ocupa do tema da escravidão e liberdade no BrasilAbstract: This work is a study on the baptismal manumissions recorded between 1774 and 1871 in the ecclesiastical books of two different parishes (Nossa Senhora da Conceição e Santa Cruz) from the county of Campinas, São Paulo, and on their historical agents. One of the objectives of this research is to verify the dynamics of the manumissions over time, bringing into light their patterns and movements, and to investigate the profile of freed slaves and other individuals involved in such a process. Furthermore, through case studies and reconstruction of life experiences and strategies of enslaved women and men, this investigation also intends to apprehend to which extent the social interactions established and held by them with other subjects of the Brazilian slave society played a role in their newborn children’s conquering of freedom through baptism. To accomplish these goals, we have resorted to a diverse set of documentary sources, such as ecclesiastical, census, and notarial records, and to the "nominative record linkage" method.     On finite and infinitive verb raising in brazilian portuguese and in colombian spanish: microvariation and cartographies

    O trabalho tem por objetivo investigar a subida do verbo em suas formas finita e infinitiva em perspectiva intralinguística e interlinguística, a partir de um exame de dados de duas línguas intimamente relacionadas, o português brasileiro (PB) e o espanhol colombiano (EC). Para isso, recorre ao framework da Cartografia Sintática (Cinque 1999). Os dados foram obtidos a partir de juízos de gramaticalidade de sentenças a tomarem os advérbios da Hierarquia Universal como diagnósticos para a posição do verbo. Um exame dos dados nos leva a três conclusões interessantes: (i) advérbios são, de fato, bons diagnósticos em estudos sobre variação microparamétrica; (ii) embora, superficialmente, os dados do PB e EC pareçam sugerir convergências em relação à posição do verbo, as derivações envolvidas nas sentenças finitas não são sempre similares e (iii) o PB conta, em sentenças com verbo finito, com um movimento mais restrito que o EC, já que nesse último o verbo pode subir até à última projeção, enquanto em PB ele não sobe para além de AsP122491511The main goal of this paper is to investigate the raising of the verb in its finite and infinitive forms by turning to Brazilian Portuguese (BP) and Colombian Spanish (CS) data. In order to approach the issue of verb raising in a microvariation perspective, it turns to the Syntactic Cartography framework (Cinque 1999). The data were gathered from grammaticality judgments of sentences taking the adverbs from the Universal Hierarchy as diagnostics for the placement of the verb. An examination of the data leads us to three interesting conclusions: (i) adverbs are, in fact, bona fide diagnostics in studies on microparametric variation; (ii) although, superficially, the BP and CS data seem to suggest some convergences with regards to the position of the verb, the derivations involved in finite sentences are not always similar in the two languages considered; and (iii) in sentences with finite verb, BP counts with a more restricted raising if compared with CS.
