    The interaction between dietary patterns and melanocortin-4 receptor polymorphisms in relation to obesity phenotypes

    Introduction: Data shows that interactions between dietary factors and genetic variants can modulate the association of polymorphisms such as the Melanocortin-4 receptor (MC4R) gene with obesity. Considering the limited data available on this topic we aimed to investigate interactions between dietary patterns (DPs) and MC4R polymorphisms in relation to obesity phenotypes. Methods: This cohort study was performed in the framework of Tehran Lipid and Glucose Study; for eligible participants in this study (n = 3850), the median follow-up was 4 years. DPs were determined using factor analysis. The genotypes of polymorphisms (17782313rs and 12970134rs) were identified and their interaction with DPs were assessed in relation to incidence of obesity phenotypes including central obesity, general obesity and visceral adiposity dysfunction. Results: The mean age of participants (62.5% females) were 37.0 ± 13.7 years. Two main DPs (healthy and unhealthy) were extracted. C-allele carriers of rs17782313 in higher quartiles of the healthy DP score had a significant decrease in the incidence of general obesity, compared to those who had the TT genotype (HR = 0.61, 95% CI = 0.42–0.89, P interaction = 0.01). For rs12970134 A-allele carriers, subjects in the second compared to the first quartile of the healthy DP score, had a significant decrease in the incidence of general obesity (HR = 0.68, 95% CI = 0.46–0.99). There were no significantinteraction between DPs and MC4R variants in relation to other obesity phenotypes. Conclusion: Our results indicate that the healthy DP could interact with rs17782313 in relation to incidence of general obesit

    The efficacy of platelet gel derived from umbilical cord blood on diabetic foot ulcers: A double-blind randomized clinical trial

    Introduction:Type 2 diabetes is one of the most prevalent diseases throughout the world. The foot ulcers areseverecomplicationsofthisdisease.Footulcersinducethehighrateofmorbidity,impairqualityoflife,bringabout extreme costs to health service providers, and give rise to waste oftime. Recently,platelet-rich plasma(PRP) and platelet gel (PG) have been used for the treatment of chronic wounds. In the present randomized,double-blind,placebo-controlledstudy,plateletgelderivedfromumbilicalcordblood(UCB)wasusedtohealthediabeticfoot.Method:Thepatientswererandomlydividedintothreegroups,underthecategoriesofPG,platelet-poorplasma(PPP)andlubricantgel(placebo)(ratio1:1:1).Theprocessesofgelsapplicationwerelaunchedforthesubjectofeachgrouptwiceperweekwith3–4days’interval.Thismechanismprotractedforeightweeks.Aftercompletionof8weeks,thepatientswerefollowedupaftertwoweeksandthencontinuedforninemonthswithone-monthinterval.Result:30 patients underwent statistical analysis. Except for diastolic blood pressure which was significant between groups, there were no statistically significant differences in patients’ baseline demographics. No significant differences were detected between groups at baseline of wounds(P=0.09).Theresultsrevealedthatthereisnostatisticallysignificantinteractionamongthreegroupsduringfollow-uptime.Conclusion:Thepresentstudyprovidesevidencethattherearenosignificantdifferencesinthe size of wound among PG,PPP,andplacebogroups

    Probiotic viability, physicochemical characterization and sensory properties of cornelian cherry (Cornus mas l.) juice supplemented with lactobacillus acidophilus and lactobacillus delbrueckii

    : A large variety of fruits and vegetables and a large number of Lactobacillus strains provide a great opportunity for the development and industrialization of non-dairy beverages. In this study, Lactobacillus acidophilus and Lactobacillus delbrueckii were used for cornelian cherry probiotic juice. Physicochemical factors such as the survival of probiotic bacteria, acidity and the total amount of reducing carbohydrates in probiotic juice were studied. After four weeks, the survival of Lactobacillus delbrueckii was significantly higher than other treatments (P<0.05). After four weeks, no Lactobacillus acidophilus bacteria had survived. In terms of sensory evaluation, there was a significant difference between two treatments of Lactobacillus acidophilus and Lactobacillus delbrueckii, while the treatment with Lactobacillus acidophilus probiotic bacteria was more acceptable than Lactobacillus delbrueckii (P<0.05). Due to the sensory characteristics, the viability of bacteria, and other physicochemical properties, it can be concluded that probiotic juice can be useful for human health if the probiotic bacteria can survive

    Upregulation of Pro-inflammatory Cytokine Genes by Parvovirus B19 in Human Bone Marrow Mesenchymal Stem Cells

    Chronic infammation plays a prominent role in cancer initiation and development. On the other hand, the Infammation can be established by a number of factors such as viral infections. Parvovirus B19 (B19V) is a pathogen with widespread infection, which infects bone marrow erythroid progenitor cells. It has been shown that B19V can also enter human bone marrow mesenchymal stem cells (BM-MSCs). In this study, we hypothesized that BM-MSCs as the main cellular component of bone marrow niche may be induced to secret pro-infammatory cytokines after B19V infection. BM-MSCs were cultured up to passage 3. The cells were then subjected to nucleofection to transfer a plasmid containing B19V genome. After 36 h, total RNA was extracted and the expression levels of IL-1β, IL-6, TNF-α and NF-κB genes were examined using qRT-PCR. Data analysis showed the signifcant increase in expression levels of all studied genes in the B19V-transfected cells (P<0.05). Although further researches are required, our fndings for the frst time suggest the importance of B19V infection to establish an infammatory microenvironment in the bone marrow and its involvement in infammation-related diseases. Finally, based on our results, molecular assay to diagnose B19V infection of BM-MSCs prior to stem cell therapy is strongly recommended

    Epidemiological and Clinical Features of Acute Poisoning in Children in a Referral Teaching Hospital in Iran, 2015 - 2018

    Background: Poisoning is one of the most common medical emergencies in young children. Substantial differences in socioeconomic and cultural situations cause various epidemiological patterns of acute poisoning with different poisonous agents. Objectives: We aimed to determine the extent of pediatric poisoning in a referral teaching hospital in the center of Iran. Methods: The electronic database of Qods Children Hospital in Qazvin, Iran, was searched for children younger than 12 years old and referred for acute poisoning from 2015 to 2018. The cases (n = 184) were classified according to their age, gender, symptoms at the time of admission, the toxins, and their types. Results: The frequency of poisoning in boys was more than girls (M/F = 1.49). The mean age of the patients was 3.4 years, and the average length of stay in hospital was 1.3 days. Children poisoned with petroleum products had the highest average stay (3.10 days). Although more than 65 agents were identified as the poisoning agents, narcotics were the most common category (n = 62, 33.7%), and methadone was the most common agent in our study (n = 46, 25%). Central nervous system symptoms were recorded more than other signs and symptoms on admission or during the stay. Only two patients (1.1%) had been discharged with complications. Conclusions: With predominance for boys, most of the children had toxicity with medications. Methadone, other narcotics, and psychotropic medications were the most common agents that indicate a need for more severe control and education for prevention from poisoning in children
