Isolation and Characterization of Gene Encoded Tonoplast Intrinsic Proteins from Halophyte Plant Salicornia herbacea
Salicornia herbacea is a succulent halophyte plant which grows optimally under 300 mM NaCl. These plants may have acquired specific genes that are essential for tolerating salt. To obtain insight into the comprehensive molecular characteristics related to the salt tolerance mechanism, we performed a screening of salt-inducible genes in S. herbacea shoots by differential display. A comparative analysis of gene expression between control and salt-stressed conditions led to the detection of 6 cDNA clones induced by salt. Sequence analysis and database searching revealed that all of the clones have homology with tonoplast intrinsic proteins. We designated them as ShTIP1 to ShTIP6 (S. herbacea Tonoplast Intrinsic Proteins). One the identified genes, ShTIP6 showed higher induced under salt stress compared to other ShTIP genes. Analysis of Quantitative Structure and Solubility Relationship for Organophospate Active Compounds Linear Model and Cluster Model Approach
Study on quantitative solubility and structure relationship of organophospate pesticide has been conducted. Aim of this research is to analyze relationship between characteristic physical properties (water solubility) of pesticide with molecular structure by a model that can be used to predict physical properties of new compound proposed to be pesticide compound. Analysis was done for 52 organopospate compounds and its water solubility data. The model predicted by 39 fagmental descriptor based on spesific structure of organophospate compound. Analysis used in modeling are cluster analysis, one way analysis of variance (ANOVA) and regression analysis. Result show water solubility of the pesticide compound positive effect by functional group of P=O, N-primer and C=O in the structure. Functional group of S (sulphur), fen (phenil) , C-aromatic, C1-C2, C3-C4, C5-C6, Ortho Aromatic, P=S, C-Cl and log(Mr) decrease water solubility. The model resulted by regression analysis using stepwise and best subset method is : Ln(1+watsol) = 11.0 - 0.352 n C (aromatic) - 5.41 n (P=S) - 1.55 n (C-Cl), F-value = 32.43 (sign. 0,000) , R2 = 0.67 dan S=2.643. Application of Tree Regression in Long Study of University of Jember‘s Student Use R Program
This research aimed to implemate tree regression with one respon and six explanatory variables in R programand apply it to know variables which distinguish long study of University of Jember’ student. Polysulfone Membrane with UV-Photografting Technique and it Application at Soya Milk Filtration Processing
In the separation process of solution containing protein, interaction between membrane surface and protein can cause fouling irreversibely. So, efficiency of filtration process will decline. In this research, to improve the effeciency of filtration was done by modification of membrane using UV-photografting technique. Acrylic acid had been introduced topolysulphone membrane via this technique. The hydrophylic membrane was obtained. The membrane performance was evaluated in soymilk fouling test. The first step of research had obtained PSF membrane optimation about PSF 18%. In second step, the research was focused on the modification and characterization of PSF membrane by UV-Fotografting tehchnique. In this technique, monomer concentration and radiation time was used as variable. As the result, PSF membrane had been modificated succesfully by UV- photografting tehchnique with optimum condition in 15 minutes time radiation and concentration of acrylic acid 5%. Performance of modified membranes (flux and permselectivity) were better than unmodified membranes. The fouling test showed that the modified membranes have reduced fouling degree significantly. X-Ray Diffraction and Microstructure of Tin Dioxide with Addition of Fe
Preparation and characterization of tin dioxide, SnO2, had been carried out by precipitation process of SnCl2.2H2O in ammonia and aqua solutions with comparison of 1 : 7. FeCl3 was added to solution with variation of 0, 0,1, 0.2, 0.5 and 1.0 gram respectively and then sintered at 900°C. The X-ray diffraction (XRD) analysis shows that the intensity of diffraction increases with the addition of Fe. The addition of Fe does not cause the new phase. Generally, the Bragg angle is relative same for all samples that means Fe does not change significantly the cell parameters. Scanning electron microscopy (SEM) image shows the microstructure changed with the addition of Fe. On the small addition, the microstructure tends to oval and on the larger addition to plateau
Study of the Activity of Ni/H5-NZA Catalyst and Co(II)/H5NZA to Catalytic Cracking Methyl Ester Jatropha
The catalytic activity of H5NZA, Ni/H5NZA and Co/H5NZA converting methyl ester jatropha have been studied. The reaction was proceeded in flow fixed bed reactor that operated at the temperature of 450oC for 60 minutes using butanol as the catching-feed. The catalysts were prepared by acidifying of natural zeolite using HF 1%, HCl 6 M and NH4Cl 0.1 M), it was then hydrothermally treated and calcinated with nitrogen. Finally, it is oxidized and impregnated by ion nickel and cobalt (2% w/w). Meanwhile, the methyl ester jatropha as feed stocks was prepared by reacting a jatropha oil with sodium methoxy. The catalytic process was done by placing a catalyst on the flow fixed bed reactor at the temperature of 450oC and flowing a methyl ester and butanol to the reactor for 60 minutes with a fixed flow rate. A liquid fraction have been produced and were then characterized using GC. The maximum percentage conversion of methyl ester was found to be 94.6891% using catalyst of zeolite acid H5NZA which were indicated by a reduction signal of the original methyl ester. The selectivity of this catalyst was performed by the specific product on the retention time of <2.5 minutes. The modified catalysts which were impregnated by the ion Ni and Co shown their selectivity on range of retention time of 2.5-5.0 and 5.0-10 minutes respectively
Design of Multi Syringe Pump for Flow System Analysis
The aims of this work were to research: design of the multi syringe pumps, volume consistency of ISAB and the correlation of the PWM to the flow rate of carrier. Pump speed was set by PWM. Controlling the pump speed and time were performed with a computer using Labview software. It was found that based on the time variation of time then the volume of ISAB has a consistency of the injected volume with Kv 0.25% to 2.53% and the rate of carrier based on multi syringe velocity have a good correlation with the regression value of 0.998 and the value of Kv 0.27% to 1.2%. Flowering Development of Longan (Dimocarpus longan Lour) ‘Diamond river’
‘Diamond river’ is introduction plant that cultivated in Indonesian. Development of longan flower divided into eight stadium during 28 days. First and second stadium is the induction phase that lasts for 8 days, this phase is characterized by a change in color of the leaves become older by using the Munsell color charts for plant tissues indicate the scale of 7.5 GY (4/4) to GY 7.5 scale (3\2). Third stadium is the phase of flower initiation occurred by day eigth. Initiation stage appearance were showed by merristem axilar, which will form part of primordial flower. Fourth to seventh stadium are phase that a differentiation occurred on day 12 to day 24. Differentiation phase showed the development in suitable with the typical angiosperms are sepals, stamens, petals and pistils. Eighth stadium is the phase of anthesis occurred on day 28. Application of Difference Equations Model in Determining Genotype Probability Offspring with Two Different Characteristic
Population genetics is a branch of biology which studies about the gene composition from population and the change of the gene composition is effect from some factors. One of them is lethal gene factor. The change of gene composition will influence the genotype probabilities in the population. In this paper discussed about determining the genotype for the probability of the n-th offspring genotypes in dihybrid mating by observing linkage between the two loci. The mating occurred randomly and without concern ethics in mating. This research was done by making mathematics model to determine allele pair, using difference equation, then from this model will be determined genotypes probability. The result show that the mating happened normally had the same genotype probability of each generation, meanwhile in abnormal mating, the genotype probability whose had lethal gene would decrease and the genotype probability whose did not have lethal gene would increase in each generation.Keywords : Difference equation, dihybrid mating, lethal gene, population genetics, probabilit
Ordinal Regression Model using Bootstrap Approach
The aim of the research content three part, thus a to know misclassification and model discriminant analysis with bootstrap approach, model regression ordinal with bootstrap approach, and model MARS with bootstrap approach. The data used is data of secondary related to matrix variance covariance is same and unequal that is (The data worker standard of living and banking performance). The result of this research shows that in determining distinguishing variable between groups there are difference of variable at each method. This matter because of at each method has specification either from fulfilled of assumption and also estimation its. So also at accuracy of classification between groups there is difference especially at matrix of variance covariance unequal at worker standard of living case. As a whole can be concluded that the problem accuracy of classification bootstrap approach at each method give small mistake of goodness at matrix variance covariance unequal and equal.Keywords: classification, bootstrap, discriminant analysis, ordinal regression, MARS
