Tuning Electron‐Transfer Driving Force in Photosynthetic Special Pair Models
Visible‐light excitation of a family of bimetallic ruthenium polypyridines with the formula [RuII(tpy)(bpy)(‐CN)RuII(py)4L]n+ (RuRuLn+), where L=Cl−, NCS−, DMAP and ACN, was used to prepare photoinduced mixed‐valence (PI‐MV) MLCT states as models of the photosynthetic reaction center. Ultrafast transient absorption spectroscopy allowed to monitor photoinduced IVCT bands between 6000 and 11000 cm−1. Mulliken spin densities resulting from DFT and (TD)DFT computations revealed the modulation of the charge density distribution depending on the ligand substitution pattern. Results are consistent with PI‐MV systems ranging from non‐degenerate Class II to degenerate Class III or II/III, with electronic couplings between 1000 and 3500 cm−1. These findings guide the control electron localization‐delocalization in charge‐transfer/charge‐separated excited states, like those involved in the photosynthetic reaction center.In bimetallic {(tpy)Ru‐Ru(L)}, MLCT excited states prepared upon visible light absorption are photoinduced mixed valence systems. This makes them excellent models of the photosynthetic reaction center. In these models, the balance of positive charge density is determined by the tpy radical anion and can be finely controlled by ligand L substitution. Un/met: a mixed-methods study on primary healthcare needs of the poorest population in Khyber Pakhtunkhwa province, Pakistan
Background Access of all people to the healthcare they need, without financial hardship is the goal of Universal Health Coverage (UHC). As UHC initiatives expand, assessing the needs of vulnerable populations can reveal gaps in the system which may be covered by relevant policies. In this study we (i) identify the met and unmet primary healthcare needs of the poorest population of Khyber Pakhtunkhwa province (KP), Pakistan, and (ii) explore why the gaps exist. Methods We used Leveque’s Framework of Patient-centred Access to Healthcare to examine unmet primary healthcare (PHC) needs and their underlying causes for the poorest population in four districts of Khyber Pakhtunkhwa province, Pakistan. Using a triangulation mixed methods design, we analysed data from a quantitative household survey of744 households, 17 focus group discussions with household members and, 11 interviews with healthcare providers. Results Our results show that indicate that despite service utilization, PHC needs were not met, primarily due to prohibitively high costs at each stage of access. Furthermore, gaps in outreach and information (approachability), and varying availability of medicines and diagnostics at facilities (appropriateness) the supply side as well as difficulties in navigating the system (inability to perceive) and adhering to prescriptions (inability to engage) on the demand side, also led to unmet PHC needs. Going beyond utilization, our findings highlight that engagement with care is an important determinant of met needs for vulnerable populations. Conclusion Social health protection policies can contribute to advancing UHC for primary care. A credence-based theory-heavy approach to non-human consciousness
Many different methodological approaches have been proposed to infer the presence of consciousness in non-human systems. In this paper, a version of the theory-heavy approach is defended. Theory-heavy approaches rely heavily on considerations from theories of consciousness to make inferences about non-human consciousness. Recently, the theory-heavy approach has been critiqued in the form of Birch’s (Noûs 56(1):133–153, 2022) dilemma of demandingness and Shevlin’s (Mind Lang 36(2):297–314, 2021) specificity problem. However, both challenges implicitly assume an inapt characterization of the theory-heavy approach. I argue that an alternative characterization of the approach, what I call a credence-based theory-heavy approach, avoids these challenges. Theorists can generate interpretations of their theory, at different levels of generality, and operationalize these into theory-informed markers. These theory-informed markers are assigned a likelihood and are used to assess the probability that a target system is conscious. In providing this characterization, and mapping out the possible ways in which a credence-based theory-heavy approach can be fleshed out, the aim is to situate the theory-heavy approach as a more compelling approach than it is currently being perceived as. Our attention, then, needs to shift towards remaining challenges such as the consensus problem and the problem of calibrating the likelihoods associated with theory-informed markers. Coupling between the photoactivity and CO2 adsorption on rapidly thermal hydrogenated vs. conventionally annealed copper oxides deposited on TiO2 nanotubes
Highly ordered spaced titanium dioxide nanotubes were fabricated via electrochemical anodization and modified with titania nanoparticles and copper oxides. Such materials were rapidly annealed in hydrogen atmosphere or conventionally in a tube furnace in air, in which the temperature slowly increases. Applied synthesis procedure can be considered as simple, cost-effective, and environmentally friendly as it allows for reduction in used materials and enhances sustainable engineering. Manipulating the chemical composition of materials by different thermal treatments resulted in various photoelectrochemical activities and density of CO2 adsorption sites. Rapidly annealed nanotubes decorated by copper oxides exhibit excellent electrochemical properties where one electrode combines both: solar to electricity conversion (photocurrent under visible light 30 µA/cm2) and CO2 adsorption systems (18 times higher current after CO2 saturation). Rapidly thermal hydrogenated TiO2 nanotubes with copper oxides had 17 times higher photocurrent and wider absorption band (380–780 nm) than conventionally annealed ones. Furthermore, the crystal planes such as Cu (111), Cu (220), Cu2O (110), CuO (002) and Cu0, Cu+, Cu2+ oxidation states, and oxygen vacancies were recognized for hydrogenated sample. APOBEC3C‐mediated NF‐κB activation enhances clear cell renal cell carcinoma progression
Renowned as the predominant form of kidney cancer, clear cell renal cell carcinoma (ccRCC) exhibits susceptibility to immunotherapies due to its specific expression profile as well as notable immune cell infiltration. Despite this, effectively treating metastatic ccRCC remains a significant challenge, necessitating a more profound comprehension of the underlying molecular mechanisms governing its progression. Here, we unveil that the enhanced expression of the RNA‐binding protein DNA dC → dU‐editing enzyme APOBEC‐3C (APOBEC3C; also known as A3C) in ccRCC tissue and ccRCC‐derived cell lines serves as a catalyst for tumor growth by amplifying nuclear factor‐kappa B (NF‐κB) activity. By employing RNA‐sequencing and cell‐based assays in ccRCC‐derived cell lines, we determined that A3C is a stress‐responsive factor and crucial for cell survival. Furthermore, we identified that A3C binds and potentially stabilizes messenger RNAs (mRNAs) encoding positive regulators of the NF‐κB pathway. Upon A3C depletion, essential subunits of the NF‐κB family are abnormally restrained in the cytoplasm, leading to deregulation of NF‐κB target genes. Our study illuminates the pivotal role of A3C in promoting ccRCC tumor development, positioning it as a prospective target for future therapeutic strategies.The authors discovered that expression of the RNA‐binding protein APOBEC3C (A3C) promotes clear cell renal cell carcinoma (ccRCC) growth by amplifying nuclear factor‐kappa B (NF‐κB) activity. A3C‐bound mRNAs of NF‐κB regulators are stabilized, which promotes NF‐κB‐subunit shuttling and transcriptional activity. Multischaltbare Nano-Objekte Basierend Auf Photosäuren
Addressing climate change remains a significant global challenge, emphasizing the need for more efficient utilization of renewable energy, with a particular focus on maximizing the potential of solar energy. To fully exhaust all possibilities and the potential of solar energy it is necessary to also design new light-responsive structures, which respond to light in different ways.
In this thesis, a new type of switchable nano-objects is assembled from various photoacid molecules in form of disulfonated naphthol derivates with generation four (G4) poly(amido amine) dendrimers as model polyelectrolytes. Well-defined nanoparticles are formed spontaneously under light exclusion with hydrodynamic radii ranging from 80 nm ≤ RH ≤ 360 nm in aqueous solution. Due to the unique property of photoacids to undergo an excited-state dissociation of the photoacidic hydroxyl group, the assemblies can be switched through irradiation. This leads to a change in size and shape of the self-assembled nano-objects in solution. The structural characterization of the nano-assemblies is studied with dynamic light scattering (DLS), static light scattering (SLS), small angle neutron scattering (SANS) and atomic force microscopy (AFM). While the thermodynamics of the interactions are investigated using isothermal titration calorimetry (ITC) and UV/Vis spectroscopy reveals molecular characteristics, the charges of the assemblies are characterized with ζ-potential measurements.
By adding another triggerable molecule, it is possible to form multi-responsive supramolecular structures. Therefore, different types of molecules have been combined. First, the dianionic azo dye Acid Yellow 38 (AY38), which exhibits a trans-cis isomerization upon UV-irradiation, directly forms ternary assemblies with a photoacid and the dendrimer. Since the azo dye can be triggered with a different wavelength than the photoacid, a multi-responsive system with different switching possibilities of assembly size and shape is formed. The stability of these nano-assemblies can be understood by the different interaction strength of the cis and trans form of AY38 with the polyelectrolyte. As a further type of triggerable molecule, we choose a hydroxyflavylium molecule. It is known for its network of light-dependent reactions. By combination with the photoacid, two main cycles are possible responding to light irradiation and pH-changes. The reversibility of the switching cycles and structures can be controlled by the component ratios. Lastly, the photoacid is combined with a water-soluble spirooxazine without any polyelectrolyte. Despite the anionic character of both molecules, nano-assemblies are formed directly though dipole interactions. Characteristic ring opening reactions of spirooxazines lead to structural changes due to varying π-π and dipole interaction strength of the isomers.
Overall, this thesis establishes a new concept of using light responsive molecules for supramolecular assembly controlling the shape and size of the resulting nano-objects. The thesis further emphasizes the importance of induced secondary interactions, like hydrogen bonds and π-π interactions, and their fundamental role in self-assembly. Not only are photoacids interesting for multi-switchable assemblies but also for future applications such as switchable hybrid nano-structures
Lungenrundherd Detektion mittels moderner Niederfeld-MRT (0,55 Tesla) im Vergleich zu Computertomographie als Referenz-Standard
Hintergrund und Ziele
Die Lunge stellt mit ihrer geringen Protonendichte und der großen Anzahl an Grenzflächen eine Herausforderung für die Diagnostik mittels Magnetresonanztomographie (MRT) dar. Eine zentrale Aufgabe der radiologischen Untersuchung der Lunge ist die frühzeitige Detektion von potenziell malignen Lungenrundherden. Niederfeldgeräte verfügen über physikalische Besonderheiten, die insbesondere in der Lungenbildgebung von Vorteil sein könnten. Es fehlt jedoch an systematischen Arbeiten mit Magnetresonanztomographen bei niedriger Feldstärke und aktueller technischer Ausstattung. Der Zweck dieser Studie war daher die Untersuchung der Detektionsraten für Lungenrundherde eines modernen Niederfeld-MRTs im direkten Vergleich zum aktuellen Goldstandard, der Computertomographie (CT). Darüber hinaus sollte überprüft werden, ob die im klinischen Alltag gängigen Grenzwerte auch auf Messungen im MRT übertragbar sind. Hierzu erfolgte zusätzlich eine Korrelation der Rundherddiameter zwischen MRT und CT.
Methoden
Zwischen November 2020 und Juli 2021 wurden insgesamt 46 Patienten bei denen mindestens ein Lungenrundherd in einer CT-Untersuchung diagnostiziert wurde zusätzlich mittels 0,55 Tesla (T) MRT untersucht. Es wurden eine protonen-gewichtete transversale Sequenz mit periodically rotated overlapping parallel lines with enhanced reconstruction (PROPELLER/BLADE) und eine T2-gewichtete Sequenz mittels short-tau inversion recovery (STIR) und half-Fourier acquired single-shot turbo spin-echo (HASTE) Technik zur Bilddatenakquisition verwendet. Es erfolgte eine getrennte, verblindete Auswertung der MRT- vor den CT-Datensätzen im Abstand von mindestens sechs Wochen, um Wiedererkennung der Bilddaten zu verhindern. Die Rundherde wurden jeweils in der transversalen Ebene entsprechend der aktuellen Lung-RADS® Empfehlungen des American College of Radiology vermessen. Um die Rundherddetektion in den CT-Daten zu maximieren, wurde der Radiologe hierbei zusätzlich durch Maximumintensitätsprojektionen und eine computerassisiterte Detektion (syngo.CT Lung CAD) unterstützt. Alle durchgeführten Untersuchungen wurden in die Auswertung eingeschlossen. Falsch positive Ergebnisse wurden separat erfasst. Abschließend wurde eine retrospektive Auswertung der im MRT zunächst übersehenen Rundherde durchgeführt. Die statistische Auswertung erfolgte mittels Mann-Whitney U Test, Spearman Korrelationskoeffizienten und Bland-Altman-Analyse.
Ergebnisse und Beobachtungen
Insgesamt wurden 964 Lungenrundherde in 46 Patienten identifiziert. In der verblindeten Auswertung konnten alle Rundherde ab einer Größe von 6 mm (126/126) korrekt mittels MRT detektiert werden. Die Herdbefunde ab einer Größe von 4 mm bis zu 6 mm wurden zu 80 % (159/200) detektiert. Die schlechteste Sensitivität bestand in der Gruppe unter 4 mm mit einer Detektionsrate von lediglich 23 % (147/638). Der Spearman Korrelationskoeffizient zeigte eine sehr starke, positive Korrelation von ρ = 0,87 (p < 0,001) bei einer mittleren Differenz von lediglich 0,16 ± 0,9 mm.
Schlussfolgerungen
Die Ergebnisse des Methodenvergleichs zeigen die Möglichkeit der Lungenrundherd Detektion mittels moderner Niederfeld-MRT. Dabei war die Sensitivität der MRT für Rundherde mit einem erhöhten Malignitätsrisiko (≥ 6 mm) exzellent. Dem gegenüber blieb die CT der MRT in der Detektion von kleineren Läsionen, insbesondere solcher < 4 mm, überlegen. Die Ergebnisse zeigen zudem eine starke Korrelation der Messwerte, sodass eine Verwendung der bereits etablierten Grenzwerte zum Abschätzen der Malignitätswahrscheinlichkeit einer Läsion möglich scheint
Analysis of the protective effect of Flavonoids in IBD using Nanotechnology
Over the past few years, the incidence rate of inflammatory bowel disease (IBD) has significantly risen all over the world. Most of the patients with IBD suffer from severe symptoms and complications, of which ulcerative colitis (UC) individuals run a high risk of developing colorectal cancer. Being an autoimmune disease, recent studies have shown that the pathogenesis of IBD is related to environmental factors, gut microbiota, excess immunity, and genetic susceptibility. Nevertheless, the precise cause of IBD remains ambiguous.
Up to date, there is no known cure for IBD, and traditional medications have troublesome side-effects. For these reasons, some phytochemicals with more tolerance and less adverse effects capture the interest of medical scientists. In our previous animal experiments, we found that Flavo-Natin, a flavonoid combo mainly consisting of apigenin and epigallocatechin-3-gallate (EGCG), was able to significantly mitigate the levels of inflammation in acute experimental colitis models (oxazolone-mediated). Furthermore, the protective effects of Flavo-Natin in chronic experimental colitis models induced by oxazolone were confirmed. Meanwhile, it was observed that MPO activity and the levels of proinflammatory cytokines in colonic tissue obviously decreased. In contrast, the levels of anti-inflammatory cytokines dramatically increased. However, it was observed that flavonoids exhibited a significant protective effect against colitis only when administered in large doses. In vitro studies, incubating Caco-2 cell monolayers with different varieties of flavonoids, it was found that quercetin, naringenin, and EGCG increased transepithelial electrical resistance (TEER), indicating the protective effect of some flavonoids on the intestinal epithelial barrier function.
Over 6000 plants in nature can produce pyrrolizidine alkaloids (PAs), which mainly exist in plant seeds. When seeds contaminate tea or herb, PAs can enter the body and generate toxic effects. Flavonoid dietary supplements on the market are often sold in the form of a mixture of various herbal powders, which are easily contaminated by PAs. In order to eliminate potential interference factors in vivo studies, we investigated the enterotoxicity of monocrotaline (MCT) considered as a representative PA. Through oral administration of MCT, we found that enterotoxicity occurred in mice exposed to long-term MCT, mainly affecting upper small intestine. In contrast, changes of the structure in lower small intestine and colon are slight. In addition, MCT did not exacerbate colitis induced by oxazolone.
Currently, the use of flavonoids in IBD is still unsatisfactory due to poor absorption, rapid metabolism, and other reasons, resulting in low bioactivity in vivo. Previous studies conducted in our institute have observed that budesonide loaded nanoparticles (NPs) with pH-sensitive coating greatly improved anti-inflammation activity in experimental models of IBD. Based on these findings, we hypothesized that the bioavailability and bioactivity of flavonoids also can be increased through nanotechnology. Therefore, we fabricated flavonoids loaded NPs encapsulated by polymers (PLGA and Eudragit S100), including apigenin, EGCG, quercetin and naringenin. In vitro study, we found that apigenin and quercetin NPs were able to traverse intestinal epithelial barrier formed by Caco-2 cells, contrastingly, free apigenin and quercetin were unable to penetrate Caco-2 cell monolayer. In vivo study, the anti-inflammatory activities of flavonoid NPs were significantly enhanced in acute oxazolone-induced colitis models, when compared to free flavonoids.
Collectively, our studies demonstrated that flavonoids exert a protective role in oxazolone-induced colitis mouse models. Meanwhile, we found that polymeric nanocarrier can greatly improve the bioactivity of flavonoids in IBD treatment. Our research has also revealed MCT can induce enterotoxicity, which mainly occurs in upper small intestine
MKG-Chirurgie und Oralchirurgie bei Patienten mit gestörter Blutgerinnung - Aktueller Stand
Hintergrund und Ziele
Blutgerinnungsstörungen können angeborene oder erworbene Ursachen haben oder aus einer dauerhaften Medikamenteneinnahme resultieren. Allen betroffenen Patienten ist gemein, dass sie unter einem erhöhten postoperativen Blutungsrisiko leiden, wenn sie sich einem oralchirurgischen Eingriff unterziehen. Deshalb ist ein spezifisches perioperatives Management nötig, um schwere Blutungen und daraus resultierende Komplikationen zu vermeiden. Die AMWF (Arbeitsgemeinschaft der Wissenschaftlichen Medizinischen Fachgesellschaften) veröffentlichte im Jahr 2017 eine S3-Leitlinie „Zahnärztliche Chirurgie unter oraler Antikoagulation / Thrombozytenaggregationshemmung“, welche Empfehlungen und Hinweise zum Umgang mit dem entsprechenden Patientenklientel aussprach. Diese ist jedoch seit 2020 abgelaufen. Ziel dieser Arbeit ist daher, die aktuelle Literatur der vergangenen Jahre zum Umgang mit Patienten mit Gerinnungsstörungen aufzuarbeiten, um die Gültigkeit der in der Leitlinie verfassten Empfehlungen zu überprüfen und weniger beachtete Themen wie angeborene Gerinnungsstörungen näher zu beleuchten.
Material und Methoden
Die einzelnen Kapitel wurden aufgeteilt in einen allgemeinen Teil, welcher den Mechanismus der Gerinnungsstörung oder des oralen Antikoagulantiums erklärt, sowie einen historischen Teil, welche die jeweilige Entwicklung bzw. Entdeckung beschreibt. Anschließend folgt eine Übersicht des aktuellen “State of the Art“, welcher sich auf aktuelle Leit- und Richtlinien deutscher und internationaler Arbeitsgemeinschaften sowie größere Übersichtsstudien und Arbeiten beruft. Im Abschnitt „Aktuelle Literatur“ finden sich aktuelle randomisiert-kontrollierte Studien, Kohortenstudien und Fall-Kontroll-Studien vorrangig aus den Jahren 2022 und 2023. Die Literaturrecherche wurde über die National Library of Medicine, (https://pubmed.ncbi.nlm.nih.gov/) durchgeführt. Teile dieser Arbeit werden im „MKG-Update“ veröffentlicht.
Ergebnisse
Im Bereich der angeborenen Gerinnungsstörungen finden sich in der aktuellen Literatur zu oralchirurgischen Eingriffen wenig neue Entwicklungen. Bei Von-Willebrand-Jürgens-Syndrom Typ I sowie milder bis moderater Form von Hämophilie A kann präoperativ neben lokalen hämostyptischen Maßnahmen Desmopressin intranasal oder intravenös zum Einsatz kommen. Liegt ein vWJS Typ II oder III oder eine ausgeprägte Form der Hämophilie vor, sollen entsprechende Faktoren-Konzentrate zum Einsatz kommen. Für Hämophilie A ist außerdem seit 2018 der monoklonale Antikörper Emicizumab zur Blutungsprophylaxe zugelassen, die aktuellen Studien bestätigen geringere postoperative Blutungskomplikationen. Bei FVL muss insbesondere bei mikrovaskulären Eingriffen auf eine präoperative Diagnostik zur Erkennung sowie auf eine ausreichende und suffiziente Thromboseprophylaxe geachtet werden.
Aktuelle Literatur zu erworbenen Funktionsstörungen im Zusammengang mit oralchirurgischen Eingriffen findet sich kaum. Im Zentrum steht hier die prätherapeutische Diagnostik der entsprechenden Organfunktionsstörung sowie die Substitution entsprechender Faktoren bei verstärkter Blutung und Faktorenmangel. Die enge Zusammenarbeit mit dem behandelnden Internisten ist unabdingbar.
Die Einnahme von Thrombozytenaggregationshemmern soll gemäß der aktuellen Literatur vor oralchirurgischen Eingriffen trotz erhöhtem postoperativen Blutungsrisiko nicht pausiert werden. Planbare Operationen sollen bei dualer Plättchchenhemmung, wenn möglich, verschoben werden. Lokal-topische hämostatische Maßnahmen sollen maximal ausgeschöpft werden.
Keine eindeutigen Empfehlungen ergeben sich aus der aktuellen Literatur zum perioperativen Management bei oraler Antikoagulationstherapie mit Vitamin-K-Antagonisten. Bridging kann das Blutungsrisiko gegenüber der fortgesetzten Einnahme von VKA verringern, geht aber oftmals mit einem erhöhten thromboembolischen Risiko einher. Eine aktuelle Studie zeigt, dass unter modifiziertem Bridging-Schema die postoperative Blutungsrate unter gleichbleibendem thromboembolischen Risiko deutlich reduziert werden konnte.
Bei fortgesetzter DOAK-Therapie ist das Blutungsrisiko postoperativ signifikant erhöht. Die aktuelle Literatur beschäftigt sich vor allem mit der Frage, ob und in welcher Form die Fortsetzung oder Pausierung der DOAK-Therapie sinnvoll ist. Bei kleineren Eingriffen ist das Blutungsrisiko vergleichsweise gering, hier wird meist die Fortsetzung der DOAK-Einnahme empfohlen. Bei größeren geplanten Eingriffen kann in Abhängigkeit des thromboembolischen Risikos erwogen werden, die Einnahme für eine Dosis oder 24 Stunden präoperativ auszusetzen.
Schlussfolgerungen
Für die angeborenen Gerinnungsstörungen existieren keine einheitlichen Therapieempfehlungen oder Leitlinien, diese sollten im Rahmen der Überarbeitung der AWMF-Leitlinie mit abgebildet werden. Ebenfalls nicht eindeutig definiert ist der perioperative Umgang mit DOAK, hier sollte nach der aktuellen Literatur und dem Vorbild der Leitlinie der SDCEP aus Schottland eine eindeutige Empfehlung zum Pausieren oder Fortsetzen der DOAK-Therapie in Abhängigkeit des Blutungsrisikos festgelegt werden. Insbesondere im Umgang mit Tranexamsäure herrscht in der Literatur Uneinigkeit, hier bedarf es weiterer Studien zur Wirksamkeit. Lokalplastische Maßnahmen sollten bei allen Gerinnungsstörungen maximal ausgeschöpft werden
Funktionsselektive- und ortspezifische Liganden für den α2A-Adrenozeptor und den NK1 Rezeptor als potenzielle Schmerzmittel
In dieser Arbeit wurden neuartige niedermolekulare Verbindungen als vielversprechende Kandidaten für die Schmerztherapie identifiziert. Es wurden α2A-Adrenozeptor-Agonisten synthetisiert und charakterisiert, die sich durch einen völlig neuen Chemotyp zur Aktivierung des α2A-Adrenozeptor auszeichneten. Diese Stoffe konnten durch funktionelle Selektivität und den damit verbundenen neuen pharmakologischen Eigenschaften in in vivo Studien eine effektive Analgesie ohne Sedierungseigenschaften hervorrufen. Dies machte diese neue Substanzklasse dem bereits bekannten α2A-Agonisten und klinisch eingesetzten Dexmedetomidin deutlich überlegen. Die identifizierten Agonisten zeigten effektive Analgesie, keine Sedierung, orale Bioverfügbarkeit sowie eine beträchtliche metabolische Stabilität. Eine weitere Strategie war es den NK1-Rezeptor (NK1R), welcher stark in das Schmerzgeschehen involviert ist, in endosomalen Kompartimenten zu adressieren. Hierfür wurde ein bereits klinisch erfolgreicher NK1R-Antagonist physiko-chemisch modifiziert, um das analgetische Potential dieser Substanzklasse zu erwecken. Die neuartigen Antagonisten zeichneten sich durch ihre hohe Persistenz am Rezeptor aus und waren effektiv in der Inhibierung endosomaler Signalkaskaden. Folglich wurden diese neuartigen NK1-Antagonisten in in vivo Schmerzmodellen getestet und zeigten dort ausgezeichnete Wirkeigenschaften. Zusammenfassend konnten vielversprechende Wirkstoffkandidaten zur Bekämpfung von Schmerzen mit erheblich reduzierten Nebenwirkungspotential identifiziert werden. Die Liganden zeichneten sich aus durch ihre hervorragende Analgesie in in vivo Studien zusammen mit neuartigen pharmakologischen Eigenschaften in in vitro Studien. Die gefundenen Moleküle sind für die zukünftige Entwicklung neuartiger nicht-opioider und damit sicherer Schmerzmedikamente von großem Wert. Noch immer benötigen wir weltweit schmerzlindernde Medikamente, um zukünftige Krisen, die durch das hohe Missbrauchspotential vieler etablierter Analgetika gegeben ist, zu verhindern.In this work, novel small molecules were identified as promising candidates for pain therapy. α2A-adrenoceptor agonists were synthesized and characterized. The novel agonists were found to differentiate with an entirely new chemotype from known α2A-adrenoceptor agonists. These potential drug candidates were able to induce effective analgesia without sedation in in vivo studies due to functional selectivity and the associated new pharmacological properties. This made this new substance class clearly superior to the already known α2A agonist and clinically used dexmedetomidine. The identified agonists showed effective analgesia, no sedation, oral bioavailability, and considerable metabolic stability. Another strategy was to target the NK1 receptor (NK1R), which is highly involved in pain signaling, in endosomal compartments. For this purpose, an already clinically successful NK1R antagonist was physico-chemically modified to awaken the analgesic potential of this substance class. The novel antagonists showed high persistence at the receptor and were effective in inhibiting endosomal signaling cascades. Consequently, these novel NK1 antagonists were tested in in vivo pain models and demonstrated excellent properties. In summary, promising drug candidates to face pain with significantly reduced side effects were identified. The ligands were characterized by their excellent analgesia in in vivo studies together with novel pharmacological properties in in vitro studies. The potential drug candidates identified are of great value for the future development of novel non-opioid and thus safe analgesic drugs. We still need analgesic drugs worldwide to prevent future crises caused by the high abuse potential of many established analgesics
