The system of atmosphere, land, ice and ocean in the region near the 79N Glacier in northeast Greenland: synthesis and key findings from the Greenland Ice Sheet–Ocean Interaction (GROCE) experiment
The Greenland Ice Sheet has steadily lost mass over the past decades, presently representing the second-largest single contributor to global sea-level rise. In line with the rest of the Greenland Ice Sheet, the glaciers draining the northeast Greenland ice stream have been observed to retreat and thin. Here, we present a comprehensive study of processes affecting and being affected by the mass balance of marine-terminating and peripheral glaciers in northeast (NE) Greenland. Our focus is on the 79N Glacier (79NG), which hosts Greenland’s largest floating ice tongue. We provide new insight into the ice surface melt, the ice mass balance, glacier dynamics, the regional solid Earth response, the ocean-driven basal melt and the consequences of meltwater discharge into the ocean. Our study is based on field observations, remote sensing and simulations with numerical models of different complexity, most of them originating from the Greenland Ice Sheet–Ocean Interaction (GROCE) experiment. We find the overall negative climatic mass balance of 79NG to co-vary with summertime volumes of supraglacial lakes and show that the spatial pattern of the overall negative ice mass balance for NE Greenland is mirrored by the pattern of glacial-isostatic adjustment. We find near-coastal mass losses of both marine-terminating and peripheral glaciers in NE Greenland to be of a similar magnitude in the last decade. In contrast to the neighboring Zachariæ Isstrøm, 79NG – despite experiencing massive thinning of the floating tongue – has resisted an acceleration of ice discharge across the grounding line due to buttressing imposed by lateral friction of the 70 km long ice tongue in the narrow glacial fjord. Observations and models employed in this study are consistent in terms of melt rates occurring below the floating ice tongue. Our results suggest that the multidecadal warming of Atlantic Intermediate Water flowing into the cavity below the ice tongue – supplied by the recirculating branch of the West Spitsbergen Current in Fram Strait – is the main driver of the recent major increase in basal melt rates. We find that the meltwater leaving the cavity toward the ocean at subsurface levels quickly dilutes on the wide shelf. Increased Sensitivity to Ionizing Radiation in a Relevant Subset of Patients with Cancer and Systemic Lupus Erythematosus
It has long been hypothesized that systemic lupus erythematosus (SLE) increases radiosensitivity, but recent studies have yielded mixed results. We studied individual radiosensitivity in 70 individuals with SLE using chromosomal aberrations as biomarkers of radiosensitivity. In total, 33 patients with SLE and 37 patients with SLE and additional oncologic diseases were compared with healthy individuals and with patients with rectal and breast cancer. Individual radiosensitivity was assessed by ex vivo irradiation of G0 blood lymphocytes followed by three-color fluorescence in situ hybridization of chromosomes 1, 2, and 4. SLE patients have slightly higher background rates of chromosomal aberrations than healthy individuals and lower rates than cancer patients. Non-oncologic SLE patients show a rate of chromosomal aberrations similar to that seen in healthy individuals. The outliers in this group, who clearly show increased radiosensitivity, fall between healthy individuals and cancer patients. Patients with SLE and cancer have significantly higher chromosome aberration rates compared to healthy individuals ( p < 0.001) and patients with isolated cancer ( p = 0.007) or isolated SLE ( p = 0.004). The proportion of radiosensitive patients in the oncologic SLE cohort is high, with 45% of patients showing increased radiosensitivity. There is a weak association between anti-Ro-52 autoantibodies and radiosensitivity. Based on the radiosensitivity measurement, radiation dose reduction was recommended in 11 oncological SLE patients and was successfully achieved in 5 patients by up to 21% of the dose per fraction. In the oncologic SLE cohort, a substantial portion of individuals show increased radiosensitivity.This research received no external funding
Comparative Study of Iminodibenzyl and Diphenylamine Derivatives as Hole Transport Materials in Inverted Perovskite Solar Cells
Perovskite solar cells (PSCs) have recently achieved over 26 % power conversion efficiency, challenging the dominance of silicon‐based alternatives. This progress is significantly driven by innovations in hole transport materials (HTMs), which notably influence the efficiency and stability of PSCs. However, conventional organic HTMs like Spiro‐OMeTAD and PTAA, although highly efficient, suffer from thermal degradation, moisture ingress, and high cost. This study explores the potential of iminodibenzyl, a moiety known for its strong electron‐donating capabilities in pharmaceutical applications, as a novel HTM. A series of fluorene‐based derivatives incorporating iminodibenzyl ( TMF‐2 and TDF‐2 ) and diphenylamine ( TMF‐1 and TDF‐1 ) units were synthesized and characterized. The new HTMs demonstrated commendable optical, electrochemical, and thermal properties, as well as enhanced photostability. Among them, TDF‐2 achieved a power conversion efficiency (PCE) of 19.38 %, the highest of the new materials. Although these efficiencies are slightly lower than the benchmark PTAA (20.20 %), the study underscores the potential of iminodibenzyl to enhance photostability and increase HOMO levels, making it a promising candidate for future HTM development in PSCs.This study introduces novel fluorene‐based hole transport materials (HTMs) for perovskite solar cells (PSCs), integrating iminodibenzyl and diphenylamine moieties. These materials exhibit remarkable photostability, optical, and thermal properties. Among them, TDF‐2 achieved a power conversion efficiency (PCE) of 19.38 %. Comprehensive characterization of cell and tissue responses toward high hydrostatic pressure treatment: Molecular feedback and structural integrity in bone graft processing
In the field of tissue reconstruction, the development and improvement of suitable bone grafts is of increasing importance. The implementation of bone banks enables the international distribution of suitable allografts that can be used for defect reconstruction. Currently used procedures have significant drawbacks, especially regarding biomechanical and structural properties. These can be overcome by using the technique of high hydrostatic pressure (HHP) processing. To date, little is known about the impact of HHP protocol alterations including pressure-transmitting medium or temperature regarding bone graft integrity. Data of the present study show that a low-temperature and medium-pressure treatment using isotonic sodium chloride solution as the pressure-transmitting medium generated devitalized bone tissue with preserved extracellular matrix. Specifically, efficient devitalization of human primary osteoblasts (hOBs) was found starting from 150 MPa with cell death being a complex interaction between different mechanisms. Furthermore, protein denaturation in response to HHP treatment that was predominantly observed at 600 MPa led to non-significant impairment of biomechanical properties. Effects of HHP treatment on the bone tissue did not lead to any noticeable compromise in biocompatibility. Accordingly, the presented protocol may be applied as a medical device to improve the outcome of patients undergoing bone defect reconstruction with allogenic grafts
Auf Dem Weg Zu Einer Wissensallmende Für Objekt-zentrierte Forschungsdaten
This dissertation concerns itself with supporting researchers in their work with one or multiple object-centered datasets.
Modern science relies on an ever-increasing amount of data, a fact that is broadly acknowledged by politics, funding agencies and universities alike. Scientists author, curate, and eventually use or search through varying amounts of data and datasets, which frequently revolve around objects. Some examples of such objects include artifacts of cultural heritage or mathematical structures.
It is well established that datasets should be Findable, Accessible, Interoperable and Reuseable - better known under the FAIR acronym - in order to become useable in a broader and possibly public context. Typical research questions involve more than a single dataset, instead relying on a large group of datasets that effectively act as a single (possibly federated) dataset. Such Research Data Commons accelerate science and prevent duplicate datasets.
Research Questions
Limiting ourselves to FAIR object-centered datasets, this leads to the following research questions:
- How can researchers be enabled to answer their questions using a Research Data Commons?
- What types of software are required for a Research Data Commons? In particular, is it necessary to provide an explicit infrastructure?
- Which costs and benefits are associated with designing, developing, deploying and using a Research Data Commons?
To answer these questions, I explored the design space of Research Data Commons software.
Experiments and Systems
I conducted three experiments and developed three systems - MathDataHub, the WissKI Distillery and the WissKI Data Viewer - from which the questions could be answered.
The MathDataHub portal allows publishing and semantically searching through mathematical datasets, in particular supporting censuses of discrete (finite) symmetric objects. Objects found in the datasets are grouped by their mathematical meaning as opposed to the dataset in which they first occurred.
The WissKI System is a virtual research environment which allows collecting FAIR data about objects, typically those of cultural heritage, and storing it in a formal graph database. The WissKI Distillery was designed as a cloud solution providing distinct, but jointly managed WissKI installations, enabling the software to act as a base for a "WissKI Commons" supporting creation, curation and connections between datasets. It was deployed as part of the FAUWissKICloud.
The WissKI Data Viewer allows scientists to inspect, re-use, and understand a WissKI dataset directly on their computer without depending on a cost intensive infrastructure or running WissKI system.
Conclusion
Together the experiments demonstrate that there is a disconnect between the costs and benefits of Research Data Commons. Costs need to be payed upfront at the beginning of a research project, whereas the benefits only come into effect much later. The latter are also conditional: They only arise if other researchers invest likewise. To minimize its effect, and enable the formation of Research Data Commons, it becomes necessary to decrease costs (for example by avoiding recurring costs), or to increase benefits (in particular during early project phases).Diese Dissertation befasst sich mit dem Unterstützen von Forschenden bei ihrer Arbeit mit einem oder mehreren objektzentrierten Datensätzen.
Forschende heutzutage sind auf eine immer größere Menge von Daten angewiesen, eine Tatsache, die von Politik, Fördereinrichtungen und Universitäten anerkannt wird. Wissenschaftler erstellen, kuratieren und nutzen eine Vielzahl von Daten und Datensätzen, welche sich häufig auf Objekte fokussieren. Dabei kann es sich beispielsweise um mathematische Strukturen, aber auch um Kulturgüter aus musealen und wissenschaftlichen Sammlungen handeln.
Datensätze sollten auffindbar, zugänglich, interoperabel und wiederverwendbar sein - eine Forderung, die unter dem FAIR-Akronym geläufig ist - um in einem breiteren und potentiell öffentlichen Kontext nutzbar zu werden. Die Beantwortung typischer Forschungsfragen benötigt nicht nur einen einzigen Datensatz, sondern braucht eine Kombination von Datensätzen, die effektiv als ein einziger virtueller (potenziell föderierter) Datensatz agieren. Solche Forschungsdaten-Allmenden (Research Data Commons) führen zur Beschleunigung der Wissenschaft und vermeiden Doppelerhebungen.
Forschungsfragen
Im Kontext von FAIRen und objektzentrierten Datensätzen ergeben sich folgende Forschungsfragen:
- Wie können Forschende mithilfe von Forschungsdaten-Allmenden in die Lage versetzt werden ihre Forschungsfragen zu beantworten?
- Welche Arten von Software werden für eine Forschungsdaten-Allmende benötigt? Ist eine explizite Forschungsdateninfrastruktur erforderlich?
- Welche Kosten und Nutzen fallen bei der Planung, Entwicklung, Implementierung und Nutzung von (Software für) eine Forschungsdaten-Allmende an?
Zur Beantwortung dieser Fragen wurde der Raum von Forschungsdaten-Allmenden Software erkundet.
Experimente und Systeme
Es wurden drei Experimente durchgeführt und drei Systeme - MathDataHub, die WissKI Distillery und der WissKI Data Viewer - entwickelt, aus denen die oben aufgeworfenen Forschungsfragen beantwortet werden konnten.
Das MathDataHub Portal erlaubt es mathematische Datensätze, insbesondere Zensus diskreter endlicher symmetrischer Objekte, zu veröffentlichen und auf semantische Weise zu durchsuchen. Dies wird durch die Gruppierung von Objekten nach Typ und nicht nach Ursprungs-Datensatz ermöglicht.
Das WissKI System ist eine virtuelle Forschungsumgebung, die es ermöglicht Daten über Objekte kulturellen Erbes mittels einer formalen Graphdatenbank in FAIRer Weise zu erfassen. Die WissKI Distillery bildet eine Cloud-Lösung für getrennte aber gemeinsam verwaltete WissKI-Installationen, um WissKI als Basis für eine "WissKI Allmende" nutzbar zu machen. Die Distillery wurde als Teil der FAUWissKICloud implementiert und als Service bereitgestellt.
Der WissKI Data Viewer gestattet es Wissenschaftlern einen WissKI Datensatz unmittelbar auf ihrem Computer zu verwenden bzw. nachzunutzen, ohne von einer kostenintensiven Infrastruktur und insbesondere der Funktionalität einer WissKI Installation abzuhängen.
Fazit
Die Experimente demonstrierten, dass es zwischen Nutzen und Kosten eine Diskrepanz gibt: Kosten für eine Forschungsdaten-Allmende müssen zu Beginn eines Forschungsprojektes getragen werden, während der Nutzen erst im späteren Verlauf entsteht, und das auch nur, wenn andere Wissenschaftler ebenso ihre Datensätze bereitgestellt haben. Um die Entstehung einer Forschungsdaten-Allmende zu begünstigen ist es notwendig, entweder Aufwendungen zu minimieren, beispielsweise durch Vermeidung von wiederauftretenden Kosten, oder den Nutzen zu erhöhen, wenn möglich schon am Anfang der Projektlaufzeit
Multimodale Diagnostik der Aortenisthmusstenose - eine retrospektive Analyse aus dem Herzkatheter
2.1 Hintergrund und Ziele
Die Behandlungsindikation der nativen, residualen – oder rezidivierenden Aortenisthmusstenose wurde in den Leitlinien genau definiert. Koarktation ist eine systemische Erkrankung, die auch bei optimaler Therapie mit einem erhöhten Risiko der arteriellen Hypertonie einhergeht.
Das Ziel dieser Arbeit war festzustellen, ob sich im Herzkatheterlabor zusätzliche Patienten mit der Diagnose einer behandlungsbedürftigen CoA demaskieren, die nach der Angiographie allein keine Behandlungsindikation hätten, indem man neben der Angiographie auch noch eine Messung des Stressblutdruckgradienten mittels Dobutamintestung und eine Ballonmodulation anschließt.
Die kardiale Kernspintomographie hat sich als ein bildgebendes Verfahren in der Diagnostik der nativen, residualen – und rezidivierenden Aortenisthmusstenose bewährt. In dieser Arbeit habe ich retrospektiv überprüft, ob eine im Vorfeld der Herzkatheteruntersuchung durchgeführte kardiale Kernspintomographie hilfreich im Hinblick auf die Stellung der Behandlungsindikation gewesen ist.
2.2 Methoden (Patienten, Material und Untersuchungsmethoden)
87 Patienten, die in den Jahren 2014 bis einschließlich 2016, eine Herzkatheteruntersuchung wegen des Verdachts auf eine native Aortenisthmusstenose oder Re - Stenose erhielten, wurden in die retrospektive Analyse eingeschlossen. Die Gruppe mit Re-Stenosen beinhaltet Patienten, die im Vorfeld entweder interventionell oder operativ oder mit beiden Methoden behandelt wurden. Während der Herzkatheteruntersuchung wurde anhand der durchgeführten Angiographie und der Ruheblutdruckgradientenmessung eine Behandlungsindikation gestellt.
Falls anhand der Morphologie des Aortenbogens – und/oder Aortenisthmusstenose keine eindeutige Behandlungsindikation gestellt werden konnte, wurde bei den Patienten mit einem Ruhegradienten unter 20 mmHg eine Dobutamintestung sowie eine Ballonmodulation mittels Niederdruckballons durchgeführt. Zudem wurde verglichen, ob der Anteil der Patienten mit Behandlungsindikationsstellung nach einer Angiographie und/oder Ruheblutdruckgradientenmessung höher in der Subgruppe der Patienten war, die vor der Herzkatheteruntersuchung einer kardialen Magnetresonanztomographie unterzogen wurden.
2.3 Ergebnisse und Beobachtungen
In 30% (26/87 Patienten) beziehungsweise 15 % (8/52 Patienten) der Fälle in der MRT -Subgruppe wurde allein anhand der Angiographie und/oder der Messung des Ruhegradienten eine Behandlungsindikation gestellt. Bei den restlichen 70% (61/87 Patienten) bzw. 85% (44/52 Patienten) ist die Entscheidung zur Behandlung der Re - CoAo oder nativen CoAo nach der Ballonmodulation und/oder Dobutamintestung gefallen. Letztendlich war in 36% (22/61 Patienten) zum Zeitpunkt der Herzkatheteruntersuchung keine Therapie erforderlich, in der MRT-Subgruppe waren es 29,5% (13/44 Patienten).
2.4 Schlussfolgerungen
Durch die Ergänzung der Herzkatheteruntersuchung um eine Dobutamintestung und eine Ballonmodulation, um die Elastizität der Gefäße zu beurteilen, kann die Behandlungsindikation sicherer gestellt werden. Trotz des in unserer Studie nicht nachgewiesenen Profits von der im Vorfeld durchgeführten kardialen MRT, sollte man auf sie nicht verzichten und die Ergänzung mittels Modulation mit dem Niederdruckballon und/oder Dobutamin – Stress - Test während der Herzkatheteruntersuchung in Erwägung ziehen
Genome‐Wide Association Analyses of HPV16 and HPV18 Seropositivity Identify Susceptibility Loci for Cervical Cancer
ABSTRACT Infection by high‐risk human papillomavirus is known to exacerbate cervical cancer development. The host immune response is crucial in disease regression. Large‐scale genetic association studies for cervical cancer have identified few susceptibility variants, mainly at the human leukocyte antigen locus on chromosome 6. We hypothesized that the host immune response modifies cervical cancer risk and performed three genome‐wide association analyses for HPV16, HPV18 and HPV16/18 seropositivity in 7814, 7924, and 7924 samples from the UK Biobank, followed by validation genotyping in the German Cervigen case‐control series of cervical cancer and dysplasia. In GWAS analyses, we identified two loci associated with HPV16 seropositivity (6p21.32 and 15q26.2), two loci associated with HPV18 seropositivity (5q31.2 and 14q24.3), and one locus for HPV16 and/or HPV18 seropositivity (at 6p21.32). MAGMA gene‐based analysis identified HLA‐DQA1 and HLA‐DQB1 as genome‐wide significant (GWS) genes. In validation genotyping, the genome‐wide significant lead variant at 6p21.32, rs9272293 associated with overall cervical disease (OR = 0.86, p = 0.004, 95% CI = 0.78–0.95, n = 3710) and HPV16 positive invasive cancer (OR = 0.73, p = 0.005, 95% CI = 0.59–0.91, n = 1431). This variant was found to be a robust eQTL for HLA‐DRB1 , HLA‐DQB1‐AS1 , C4B , HLA‐DRB5 , HLA‐DRB6 , HLA‐DQB1 , and HLA‐DPB1 in a series of cervical epithelial tissue samples. We additionally genotyped twenty‐four HPV seropositivity variants below the GWS threshold out of which eleven variants were found to be associated with cervical disease in our cohort, suggesting that further seropositivity variants may determine cervical disease outcome. Our study identifies novel genomic risk loci that associate with HPV type‐specific cervical cancer and dysplasia risk and provides evidence for candidate genes at one of the risk loci.D.R. and T.D. received funding from the H.W. & J. Evaluation des Einsatzes eines neonatalen Kolonisationsscreenings für die empirische Antibiotikatherapie von Sepsis und Pneumonie
Seit 2013 wird auf deutschen neonatologischen Intensivstationen ein wöchentliches Screening auf multiresistente gramnegative (MRGN) Bakterien durchge-führt. Die nationale Leitlinie der Kommission für Krankenhaushygiene und Infektionsprävention (KRINKO) des Robert-Koch Institutes (RKI) empfiehlt neben der wöchentlichen Durchführung des Screenings außerdem die Berücksichtigung der Ergebnisse von Kolonisationsanalysen bei der Festlegung von Antibiotika-Behandlungsschemata im Infektionsfall. Ziel dieser monozentrischen retrospektiven Studie ist es, einen Einblick in die klinische Dichotomie von bakterieller Besiedelung und Infektionsraten sowie eine Evaluation des Nutzens eines neonatalen Kolonisationsscreenings für die empirische Antibiotikatherapie zu ermöglichen. Zusätzlich sollen anhand der Daten Risikofaktoren für den Erwerb resistenter Keime identifiziert werden.
Für die retrospektive Untersuchung wurden mikrobiologische Daten aller zwischen Januar 2013 und Dezember 2021 in einem Level 1 Perinatalzentrum behandelten Neugeborenen analysiert. Hierfür erfolgte ein Vergleich zwischen der Besiedelung mit multiresistenten Erregern (MRE, umfasst MRGN, MRSA und VRE) und Serratia marcescens (SERMA) und den jeweiligen mikrobiologischen Befunden bei Sepsis und Pneumonie.
Insgesamt wurden 917 Blut- und 1.799 Trachealsekretproben analysiert. Nach Anwendung der Kriterien des Krankenhaus-Infektions-Surveillance-Systems für Neugeborene (NEO-KISS) wurden 52 Fälle von Sepsis und 55 Fälle von Pneumonie in die Studie eingeschlossen. Eine Besiedelung mit MRE oder SERMA konnte bei 19,2% der Sepsispatienten und 34,5% der Pneumoniepatienten festgestellt werden. Unter den untersuchten Sepsispatienten konnte kein Fall auf MRE und nur ein Fall auf SERMA zurückgeführt werden, während bei den Pneumonien in zehn Fällen MRE und in einem Fall SERMA für die Infektion ursächlich waren. Als Risikofaktoren für den Erwerb einer Kolonisation mit oder einer Infektion durch MRE oder SERMA konnten wir die Late-Onset Pneu-monie (LOP) sowie die Geburt per Kaiserschnitt identifizieren.
Das Kolonisationsscreening mag als nützliches Instrument für die frühzeitige Erkennung von nosokomialen Übertragungen und die zeitnahe Umsetzung von präventiven Hygienemaßnahmen dienen. Unsere Daten deuten jedoch darauf hin, dass die Berücksichtigung einer Kolonisierung mit MRE/SERMA den unnötigen Einsatz von Reserveantibiotika bei Neugeborenen fördern könnte und daher die Abdeckung des Kolonisationskeims bei der Wahl des Antibiotikums im Falle einer Infektion nicht uneingeschränkt zu empfehlen ist
In silico characterization of the interaction of AKR1B1 with the deacetylase activation domain (DAD)
The aldo-keto reductase family 1 member B1 (AKR1B1) plays a key role in cancer progression by competing with histone deacetylase 3 to bind to the deacetylase activation domain (DAD) of the nuclear receptor corepressor SMRT. Previous studies showed that the L289A mutation in AKR1B1 disrupts its ability to form a dimer with DAD but further details of this interaction remain uncharacterized. This study aimed to model the DAD/AKR1B1 dimer by molecular docking and characterize the complex using molecular dynamics simulations. We identified a new model with increased structural stability for AKR1B1, reduced disruption of secondary structures of DAD, and lower ΔG than a previously reported one. In silico mutagenesis of AKR1B1 assessed the contributions from individual residues. We identified six hotspot residues that mediate the complex interface. Those residues are located in the α8 and H2 alpha helices of AKR1B1 and include the experimentally determined L289. These results propose new hypotheses regarding the interaction between DAD and AKR1B1, guiding future experimental approaches
