Stretching the Limits: From Planar-Biaxial Stress-Stretch to Arterial Pressure-Diameter
Understanding the physiological condition of the vascular system is critical to explain, treat, and manage vascular disease. Numerous experimental and computational studies characterize the mechanical behavior of arterial tissue under controlled laboratory conditions. However, translating this knowledge into physiologically realistic conditions remains challenging. Key difficulties include selecting suitable and relevant test methods, minimizing uncertainty, and ensuring robust model validation. Here, we present a novel integrative approach to translate laboratory experiments on arterial samples into clinically relevant pressure–diameter behavior. We perform controlled planar-biaxial tests on carotid arteries under three stretch ratios and generate axial and circumferential stress–stretch data to calibrate a fiber-reinforced soft tissue model. Using an analytical thick-walled cylindrical model, we predict subject-specific pressure–diameter behavior, informed by arterial prestretches from ring opening experiments. We systematically compare predictions against extension-inflation experiments on tubes from the same artery by applying controlled pairs of axial stretch and inner pressure, while recording outer diameter. We quantify prediction error in absolute and relative stretch regimes and evaluate the importance of the load-free reference dimensions. Results show how planar-biaxial tests probe different stretch regimes compared to extension-inflation deformations, leading to extrapolation of model predictions. We demonstrate how the constitutive material parameters can be fitted to different biomechanical loading conditions, and we assess the sensitivity of the simulations to axial stretch and circumferential prestretch. Population pharmacokinetics of edoxaban in geriatric patients with atrial fibrillation
BACKGROUND: Atrial fibrillation affects nearly half of geriatric inpatients, with edoxaban frequently used for stroke prevention. However, geriatric patients are underrepresented in drug trials with edoxaban. The limited data and high pharmacokinetic (PK) variability result in using this high-risk medication without a full understanding of its exposure in this population. OBJECTIVES: To characterize the dose-exposure relationship and predict the sufficiency of current fixed-dose guideline for geriatric patients. METHODS: We conducted a single-center prospective PK study in geriatric inpatients. We built a population PK model and used it to simulate various dosing strategies (n = 1000 simulations). RESULTS: Seventeen frail geriatric patients (median age, 87 years; IQR, 83-90) receiving routine edoxaban contributed a total of 85 steady-state PK samples. A 2-compartment population PK model best described the edoxaban and M4 metabolite concentrations, with proton pump inhibitors (PPIs) reducing relative bioavailability by 24%. Key parameters included an absorption rate constant of 0.104 per hour, clearance of 9.02 L/h, and central and peripheral volumes of 47.4 L and 42.7 L, respectively. Edoxaban concentrations correlated with factor Xa inhibitor concentrations (ρ = .966). Nearly half of the patients had minimum concentrations > 43 ng/mL, with no difference between PPI and non-PPI groups. Simulations showed a 6.75% probability of overexposure with 15 mg, 42.4% with 30 mg, and 84.25% with 60 mg. CONCLUSION: Estimated PK parameters in geriatric patients differed from those reported in pivotal trial populations, suggesting increased exposure, thereby potentially influencing bleeding risk. The impact of PPIs was not clinically relevant. UNLABELLED: A patient previously diagnosed with moderate obstructive sleep apnea and treated with continuous positive airway pressure was seen for her yearly routine follow-up. A continuous positive airway pressure adherence report of the previous year revealed periodic usage with almost stable monthly fluctuation between proper and poor adherence. The patient declared consistent usage when sleeping. The patient's sleep habits, such as usual bedtime and start of continuous positive airway pressure treatment, had not changed during the monthly variations. Dopering van platina voor hoogefficiënte spin-orbit torque magnetoweerstand random-access geheugen
Spin-orbit torque magnetoresistive random-access memory (SOT-MRAM) is a promising candidate for replacing non-volatile SRAM. Platinum (Pt) is a favourable SOT material due to its low resistivity and compatibility with back-end-of-line (BEOL) processes in CMOS technology. However, the relatively high write current remains a significant challenge for SOT-MRAM applications. Enhancing the charge-to-spin conversion efficiency of SOT materials while maintaining perpendicular magnetization and BEOL compatibility is therefore crucial. This study explores Pt doping as a strategy to improve SOT efficiency. A comparative analysis of local and global doping reveals that, depending on dopant solubility, Pt alloys (global doping) achieve higher SOT efficiency while preserving sufficient perpendicular magnetic anisotropy (PMA) in the magnetic layer. This improvement is attributed to the formation of binary Pt alloys, which retain intrinsic spin Hall contributions while dopants enhance extrinsic scattering effects. Additionally, these Pt alloys serve as effective seed layers for PtCo alloy formation after BEOL annealing. The most promising Pt-based material, PtCu alloys, was used to fabricate state-of-the-art perpendicular magnetic tunnel junctions (pMTJs) with strong free-layer PMA and stable tunneling magnetoresistance. When patterned into nanoscale SOT-MRAM devices, PtCu alloys reduced the required write current by half, demonstrating their potential for improving SOT-MRAM efficiency through Pt doping.status: Publishe
