Entwicklung von Filtermedien aus Membran-Gewebe-Kompositen für kontinuierlich betriebene Filteranlagen zur gasdurchsatzlosen Kuchenfiltration
Im Bereich der kuchenbildenden Fest-Flüssig-Trennung wird seit langem die Realisierung einer gasdurchsatzlosen Kuchenfiltration erforscht, da eine Kuchenfiltration ohne Gasdurchsatz bei der Entfeuchtung zu erheblichen Energieeinsparungen führen würde und ebenso zur Reduzierung der mechanisch erreichbaren Restfeuchte in Filterkuchen beitragen könnte. Im Rahmen dieses Projekts wurden daher umfangreiche experimentelle Untersuchungen durchgeführt, welche insbesondere von KMU nicht zu leisten sind, um die Tauglichkeit von Membran-Gewebe-Kompositen für diesen Einsatzzweck zu zeigen. Es wurden insgesamt drei unterschiedliche Fügeverfahren zur Verbindung der Membran auf dem Gewebe untersucht: Die flächige Verschweißung, die punktuelle Verschweißung und die punktuelle Klebung. Hierbei hat sich gezeigt, dass eine Verschweißung der beiden Materialien miteinander höhere Verbindungsfestigkeiten zwischen Membran und Gewebe zur Folge hatte, als dies bei einer Verklebung der Fall war. Jedoch liegt die Herausforderung bei der Verschweißung nach wie vor in der gasdichten Fügung. Durch das Aufschmelzen der Membran wird diese beschädigt, sodass durch offene Stellen erneut Gas durch das Komposit-Material hindurchtreten kann. Daher wird mit Abschluss des Projekts eine Herstellung der Membran-Gewebe-Komposite mittels Punktverklebung favorisiert. Dieses Herstellungsverfahren wird bereits großindustriell von KMU eingesetzt, weshalb ein Übergang zu größeren Maßstäben unproblematisch erscheint und für KMU am einfachsten umzusetzen ist. Anhand von diversen Filtrationsversuchen und μCT-Aufnahmen wurde innerhalb des Projekts die Eignung eines solchen geklebten Membran-Gewebe-Komposits nachgewiesen. Es konnte genauer gezeigt werden, dass der Filtermittelwiderstand nicht signifikant erhöht wird, solange die Klebepunkte auf denen nach obenstehenden Gewebefasern platziert werden. Die Forschungsergebnisse des Projekts sind in zwei Publikationen bereits veröffentlicht worden und stehen somit KMU zur Verfügung, um Ihre eigenen Produkte weiterzuentwickeln und konkurrenzfähig bleiben zu können
Der Systemische Ansatz in der Interaktion mit Personen im Autismus-Spektrum
Autism Spectrum Disorder (ASD) is typically described in deficit-oriented terms, referring to a group of people whose neurological functioning differs from that of the majority. The further development of systems theory—particularly by Niklas Luhmann—provides the foundation for systemic counselling, which involves the guidance and support of individuals or teams by a qualified counsellor.
This master’s thesis explores the research question of what resources the autism spectrum may offer for counselling processes. The study is based on a theoretical literature review, influenced by elements of narrative review methodology. The selection of literature focused on resource-oriented perspectives on the autism spectrum, though such perspectives were not always readily available.
Three studies were identified that suggest individuals on the spectrum may possess enhanced lexical memory, higher fluid intelligence, and increased levels of the protein BDNF, which is associated with cognitive performance. The author evaluated these findings as relevant to systemic counselling, potentially contributing to improved communication skills, a greater capacity for understanding and navigating abstract system structures and processes, and more.Die Masterarbeit untersucht, welche Ressourcen das Autismus-Spektrum für systemische Beratungsprozesse bieten kann. Auf Basis einer theoriebasierten Literaturrecherche mit ressourcenorientiertem Fokus werden drei Studien hervorgehoben, die auf besondere kognitive Potenziale hinweisen: ein ausgeprägtes lexikalisches Gedächtnis, erhöhte fluide Intelligenz und ein gesteigerter BDNF-Spiegel. Diese Merkmale werden als förderlich für systemische Beratung bewertet, insbesondere im Hinblick auf Kommunikation und das Verständnis komplexer Systemstrukturen
Modellierung und Validierung einer Steuerungsarchitektur für autonome Cyber-Physische Matrixproduktionssysteme
CPMPS erhalten als alternative Organisationsform für die industrielle Produktion immer stärkere Beachtung. Aufgrund ihrer gesteigerten Veränderungsfähigkeiten bieten sie Herstellern schnellere Anpassungsmöglichkeiten an externe und interne Ereignisse. Allerdings fehlt es an industriellen Umsetzungsbeispielen, was mit den gesteigerten Freiheitsgraden und der damit verbundenen Steuerungskomplexität begründet wird. Es bedarf einer entscheidungsunterstützenden Steuerungsstruktur, die sich durch eine autonome Optimierung und Rekonfiguration auszeichnet. Wesentliche Anforderungen an die autonome Steuerungsarchitektur sind Selbstorganisation, Anpassungsfähigkeit und Skalierbarkeit. Die entwickelte Architektur wird modellbasiert mithilfe der DACS Methode erstellt. Dies beinhaltet ein Entscheidungsmodell, ein holonisches Agentenmodell und ein Interaktionsmodell. Ergänzt werden diese mit einem MILP-Optimierungsmodell zur Lösung eines erweiterten Flexible Job Shop Scheduling Problems (FJSP). Die Implementierung erfolgt in einem agentenbasierten Simulationsframework mit Optimierungsservice, in dem verschiedene Technologien wie JADE, Tecnomatix-DES, Node-Red Middleware und SQL-Datenbanken enthalten sind. Die Simulationsexperimente als Teil der Validierung zeigen, dass die Steuerungsarchitektur eine skalierbare Auftragsplanung und adaptive Auftragsausführung ermöglicht. Durch serviceorientierte Integration eines Tabu-Search Verfahrens wird ein erweitertes FJSP skalierbar gelöst. Die Ergebnisse bestätigen die Eignung einer holonischen, serviceorientierten Steuerungsarchitektur für eine ganzheitliche autonome Steuerung von CPMPS. Vor allem die Verwendung der holonischen Interaktionsmodelle als Entwurfsmuster wird für die weitere Forschung empfohlen
Contributions to Exact Algorithms for Optimization in Warehousing and Flexible Manufacturing
This thesis advances combinatorial optimization by developing exact solution methods for two NP-hard problems arising in warehousing and flexible manufacturing: the order batching problem (OBP) and the set-union bin packing problem (SUBP). Both are highly relevant to real-world logistics and production systems.
For the OBP, which involves grouping customer orders into capacity-constrained batches to minimize picker travel distance, two problem variants are studied:
single-block and multi-block warehouse configurations.
A new branch-price-and-cut (BPC) method is proposed, combining column generation (CG) with dynamic programming-based pricing using completion bounds, valid inequalities, and tailored branching rules.
Additionally, heuristic approaches are derived from the exact BPC.
The BPC-based methods significantly outperform existing algorithms, improving both runtime and solution quality, and solving many previously open instances.
For the SUBP, a bin packing generalization with overlapping item requirements, a branch-and-price (B&P) algorithm is introduced.
Several pricing strategies are proposed and evaluated, with the most effective combining a greedy heuristic and item-based labeling with completion bounds.
The B&P method solves over 10,000 benchmark instances to optimality and improves many best-known solutions.
Overall, the results demonstrate the effectiveness of structure-aware, CG-based methods for large-scale discrete optimization problems
Struktur-Funktions-Analysen von Glutathion und Glutathion-abhängigen Enzymen
Peroxiredoxine und Glutaredoxine zählen zu den zentralen Enzymen für die Redoxregulation von Cysteinylresten in Proteinen. Dabei stellen Peroxiredoxine die bedeutendsten intrazellulären Hydroperoxidasen dar, können aber auch Zielproteine direkt oxidieren, um deren Aktivität zu regulieren. Während die mechanistischen Klassen der 2-Cys Peroxiredoxine bereits ausführlich charakterisiert wurden, konnten der Katalysemechanismus sowie die physiologischen Rollen von 1-Cys Peroxiredoxinen noch nicht geklärt werden. Glutaredoxine reduzieren glutathionylierte Proteine sowie Proteindisulfide und sind daher neben Thioredoxinen die wichtigsten Enzyme um oxidierte Cysteinylreste zu reduzieren. Zwar konnten verschiedene Reaktionsmechanismen von Glutraredoxinen beschrieben werden, allerdings ist deren Bedeutung für die Substratspezifität und physiologische Funktionen noch häufig ungeklärt.
In dieser Arbeit wurden die einzelnen Reaktionsschritte des Katalysemechanismus der Glutaredoxine PfGrx und ScGrx sowie der 1-Cys Peroxiredoxine PfAOP und PfPrx6 mittels Stopped-Flow Experimenten, Circulardichroismus-Spektroskopie und Molecular Docking Simulationen untersucht. Dabei konnte gezeigt werden, dass der Dithiol-Mechanismus von Glutaredoxinen deren Spezifität für Glutathion als Reduktionsmittel ermöglicht, und dass glutathionylierte Glutaredoxine mit verschiedenen Thiolen reagieren. Außerdem konnten die molekularen Grundlagen für die effiziente Reaktion von Glutaredoxinen mit Glutathion weiter entschlüsselt werden. Für 1-Cys Peroxiredoxine konnte in dieser Arbeit die Existenz zweier verschiedener mechanistischer Klassen nachgewiesen werden. Promiskuitive 1-Cys Peroxiredoxine, die unspezifisch mit verschiedenen Thiolen reagieren, und protektive 1-Cys Peroxiredoxine, die nur unter spezifischen Bedingungen reduziert werden können. Sowohl für Glutaredoxine als auch für Peroxiredoxine ermöglichen diese Daten ein besseres Verständnis, warum intrazellulär häufig mehrere Isoformen der Glutaredoxine und Peroxiredoxine vorliegen, und wie verschiedene Katalysemechanismen eine Vielzahl an physiologischen Rollen ermöglichen
Factors controlling damage initiation in fatigue loaded unidirectional composites under the influence of different matrix properties
A constituent-based analysis of the fatigue performance and damage initiation of unidirectional reinforced composites is an approach drastically limiting the number of variables to three, but simultaneously ignores other important aspects. Still, the constituent level is the basis for a better understanding of the main mechanisms driving early stages of fatigue damage. This is essential for a high flexibility in the choice of processing methods and the constituents themselves and allows for a targeted testing campaign including associated cost savings. This work investigates how variations in the matrix polymer’s properties affect the fatigue performance under transverse tension-tension and on-axis compression-compression loading. Since it is acknowledged that the fiber-matrix level will only include part of the real composite, a multi-scale approach is perused both experimentally and by Finite Element modelling. Carbon and glass fiber reinforced epoxy and carbon fiber reinforced polycarbonate are investigated. The results identify the relative stiffness and time-dependence as main polymer properties affecting fatigue damage, whereas plastic yielding has a limited influence. As essential additional factors in controlling damage initiation, residual stresses and interface failure could be identified. The concept of an in-situ S N curve could be developed and showed that local variations in fiber-distribution need to be considered to get the intrinsic fatigue strength of any system. Similarly, polymer characterization needs to account for the local stress state within a composite, i.e., constrained conditions and loading sequence
Methodik zur wissensbasierten Unterstützung von Innovationsprozessen in kollaborativen Wertschöpfungsnetzwerken
Klassische domänenspezifische Produkte wandeln sich zunehmend zu komplexen und interdisziplinären Smarten Produkten, die eine erweiterte Funktionalität bzw. einen erhöhten Fokus auf Servitisierung aufweisen. Diese Entwicklung hin zu Systemlösungen, bei denen Dienstleistungen und digitale Services integraler Bestandteil der Wertschöpfung sind, erfordern eine tiefgreifende Transformation von Entwicklungsprozessen. Gleichzeitig erhöhen sie die Komplexität signifikant und erfordern interdisziplinäre Entwicklungsansätze sowie verstärkte Kollaboration über Disziplingrenzen hinweg, um die Komplexität zu beherrschen und die Marktreife des Produkts zu gewährleisten. Die Entwicklung Smarter Produkte erfolgt häufig in dynamisch agierenden Wertschöpfungsnetzwerken, die durch hohe Flexibilität und Anpassungsfähigkeit charakterisiert sind. Sie ermöglichen eine effiziente und beschleunigte Umsetzung der Entwicklungsziele, da sie Integration und Zusammenarbeit innerhalb einer heterogenen Stakeholder-Umgebung fördern. In diesen Netzwerken müssen die Akteure schnell auf externe Einflüsse reagieren bzw. neue technologische Entwicklungen und Partner integrieren, um in einer volatilen Marktumgebung bestehen zu können und gleichzeitig resilient sein. Der Erfolg solcher Netzwerke hängt maßgeblich von der Fähigkeit der Unternehmen ab, Wissen effizient zu teilen und zu verwalten. Ein fundierter, frühzeitiger Wissensaustausch zwischen den Akteuren ermöglicht es, kritische Entwicklungsparameter wie Kosten, Zeit und Qualität bereits in frühen Innovationsphasen zu beeinflussen und spätere Anpassungen zu minimieren. Ein weiterer zentraler Aspekt für die erfolgreiche Entwicklung smarter Produkte ist die Strukturierung und Ermöglichung des Wissensaustauschs innerhalb dieser Wertschöpfungsnetzwerke. Hierbei spielen Ontologien eine zentrale Rolle, da sie eine gemeinsame Wissensbasis schafft, die den Austausch von Informationen über Disziplingrenzen hinweg erleichtert.
Die wissenschaftliche Neuheit und vor allem Abgrenzung zu bestehenden Lösungen innerhalb der Methodik, die in dieser Arbeit entwickelt wurde, zielt darauf ab:
• die Innovationsentwicklung smarter Produkte entlang eines Wertschöpfungsnetzwerkes in einem ganzheitlichen Ansatz von Methode, Prozess und Tool zu unterstützen.
• den gesteigerten Kollaborationsaufwand durch eine strukturierte und Ontologie-basierte Wissensvernetzung aufzufangen, die den Wissensaustausch innerhalb des Netzwerks effizienter gestaltet.
• eine mehrstufige dynamische und statische Wissensvernetzung, für die Förderung und Bereitstellungstellung der benötigten Wissenskollaboration und -generation zu erstellen.
Dies ermöglicht es, die hohen Anforderungen an die Komplexitätsbeherrschung zu bewältigen und gleichzeitig eine innovative und wettbewerbsfähige Innovationsentwicklung zu gewährleisten. Diese Wissensvernetzung innerhalb der Wissensbasis und in Kombination mit der Innovationsentwicklungsmethodik fördern und ermöglichen den erhöhten Aufwand der Kollaboration, sodass es zu einer erfolgreichen Innovationsentwicklung kommen kann. Die Instanziierung dieses Ansatzes erfolgt anhand der etablierten Innovation „Wireless Charging“ im Automobilbereich, welche exemplarisch die Anwendung der entwickelten Methodik verdeutlicht.Classical domain-specific products are increasingly transforming into complex and interdisciplinary smart products that exhibit extended functionality or an increased focus on servitization. This shift towards system solutions, where services and digital offerings become an integral part of value creation, requires a profound transformation of development processes. Simultaneously, it significantly increases complexity, necessitating interdisciplinary development approaches and enhanced collaboration across disciplinary boundaries to manage this complexity and ensure the product’s market readiness. The development of smart products often occurs within dynamically operating value creation networks characterized by high flexibility and adaptability. These networks enable the efficient and accelerated realization of development goals by fostering integration and collaboration within a heterogeneous stakeholder environment. In such networks, participants must quickly respond to external influences or integrate new technological developments and partners to remain competitive in a volatile market environment and be resilient at the same time. The success of these networks largely depends on companies' ability to share and manage knowledge efficiently. A solid, early exchange of knowledge between participants allows critical development parameters such as cost, time, and quality to be influenced early in the innovation phase, minimizing later adjustments. Another key aspect of successfully developing smart products is structuring and enabling knowledge exchange within these value creation networks. Ontology plays a central role here, as it creates a common knowledge base that facilitates the exchange of information across disciplinary boundaries.
The scientific novelty and, above all, differentiation from existing solutions within the methodology developed in this work is aimed at
• support the innovation development of smart products along a value network in a holistic approach of method, process and tool.
• to absorb the increased collaboration effort through a structured and ontology-based knowledge network that makes the exchange of knowledge within the network more efficient.
• to create a multi-level dynamic and static knowledge network for the promotion and provision of the required knowledge collaboration and generation.
This makes it possible to cope with the high demands of complexity management and at the same time ensure innovative and competitive innovation development. This knowledge networking within the knowledge base and in combination with the innovation development methodology promotes and enables the increased effort of collaboration so that successful innovation development can occur. The instantiation of this approach is based on the established innovation "wireless charging" in the automotive sector, which exemplifies the application of the developed methodology
Development and validation of indices for detecting careless responding in multidimensional forced-choice questionnaires
As an alternative to rating scales, in the multidimensional forced-choice (MFC) format, participants are asked to rank items according to how well the items describe them. However, researchers have not yet determined how to detect careless responding in MFC questionnaires. Careless responding is a response behavior where participants provide an answer without considering part (or any) of the content of the items (e.g., Nichols, 1989). This dissertation provides a thorough investigation of careless responding in the MFC format. We aimed to develop indices that could be used to detect careless responding in MFC questionnaires and that would be suitable for differentiating between thoughtful and careless responses and investigating their validity. To achieve these ends, we conducted three studies.
In Study 1, we developed indices for detecting careless responding in MFC questionnaires and examined their performance and intercorrelations. We conducted an online study with N = 1,169 participants who completed a long survey that included a series of personality questionnaires in the MFC format. We applied the newly developed indices to this data set and computed several analyses to explore their performance and to gain initial insights into careless responding in MFC questionnaires. The correlations among the MFC indices were somewhat lower than those reported in previous studies that used indices to detect careless responding in rating scales. A latent profile analysis indicated that the majority of participants in the online study, approximately 76% to 84%, did not engage in careless responding. However, those who did respond carelessly displayed different levels of careless responding. In addition, we conducted a simulation study to investigate whether the indices worked as conceptually intended. In this study, we simulated three careless responding patterns and different proportions of careless respondents in the sample. With one exception, the indices performed as conceptually intended.
In Study 2, we evaluated the newly developed indices under conditions that are assumed to be related to careless response behaviors, and we explored strategies for responding to MFC questionnaires when feeling tired, listless, or unsure about how to rank items. For this purpose, we conducted a laboratory study including N = 430 participants. We manipulated careless responding and compared its manifestation on 11 careless responding measures. Participants were assigned to one of three conditions: a control condition or two careless responding conditions in which we either instructed the participants to respond carelessly or distracted them to induce careless responding. The survey consisted of five inventories for assessing personality traits in the MFC format and concluded with an oral interview in which we asked participants questions about the response process. We found significant differences between the control condition and the instructed careless responding condition on all of the indices except for survey completion time. However, the control condition and the distracted responding condition were not significantly different from each other on most indices. In the interview, commonly reported strategies when feeling unsure, tired, or listless were reading the items superficially, thinking less about the item content, choosing a random order, or copying the presented order.
In Study 3, we thoroughly investigated the construct validity of the indices for the MFC format in an online study that included three subsamples and a total of N = 2,163 participants. We examined the convergent, discriminant, and criterion-related validity of the indices for the MFC format. A total of n = 1,513 participants completed a self-report survey with several personality questionnaires in the rating scale (RS) and MFC formats and several criteria, such as the motivation to participate, an intelligence measure, and educational level. Moreover, we assessed n = 650 friends or partners of the self-report participants who completed an other-report questionnaire in the RS format about their friend’s or partner’s personality. Except for one index, convergent validity was established for all indices in the MFC format. Moreover, three indices showed higher correlations with their counterparts in the RS format than with other indices in the MFC or RS formats, reflecting discriminant validity. Results for criterion-related validity were mixed. Whereas no support was found for hypotheses on personality traits and educational level, self-reported test motivation and the intelligence measure were related to the indices in the MFC format as hypothesized. The latent profile analysis in Study 3 yielded slightly different results compared with Study 1, identifying one careless class that comprised 13% of the sample. A comparison of the proportions of careless respondents identified by the indices in the RS and MFC formats showed smaller proportions of careless respondents in the MFC format on three indices, whereas no differences were found on three other indices.
Taken together, the three studies suggested that careless responding plays an important role in MFC questionnaires and thus should be addressed not only in RS questionnaires but also in the MFC format. The validated indices are publicly available and can easily be applied to questionnaires in the MFC format using triplets. Moreover, the results informed the definition of careless responding in MFC questionnaires
Turbulent Times: Arguments for Advanced Distributional Time Analyses
Since its conception, experimental psychology has been on a quest to unravel the cognitive processes involved in human behavior. Of major interest in this endeavor is not only the identification of said processes, but to also reveal their temporal dynamics. In other words, can we identify at which point in time which processes are at play, and how they correspond to human behavior?
Historically, the fundamental measures when researching the temporal dynamics of human behavior have been, and are to this day, reaction or response time (RT) and accuracy, respectively. In experimental psychology, RT most often refers to the time passed between stimulus presentation and an associated response. Accuracy then evaluates the performance of this action, on a scale from “0 (incorrect)” to “1 (correct)”, depending on the task at hand. Despite the emergence of neuroimaging techniques such as electroencephalography (EEG), magnetoencephalography (MEG), and functional magnetic resonance imaging (fMRI), RT analyses remain a key tool in the field.
The aim of this work is to give a brief overview of the historic developments associated with these measures (Chapter 1; based in parts on Article 1 & 2). Thus, I discuss their properties and application, and the methods created to use these measures as a tool to better our understanding of human cognition and behavior. This reaches from Donders’ subtraction method to Sternberg’s additive factor method. Following this, more recent developments are introduced, focusing on the distributional properties of RT and accuracy, such as quantile plots and Vincentizing. Finally, I make my own case for the analyses of the full distribution of response occurrences (Chapter 2; based on Article 1). To do so, I employ discrete-time event history analysis to various experimental psychology tasks (introduced in Chapter 3; see Articles 1–4). I aim to highlight its advantages, the gains and insights it lends to the field, discuss its shortcomings, and suggest potential developments in the future (Discussion; Articles 1-4). Overall, I show that event history analysis (1) can reveal effects that mean performance measures can conceal, (2) can reveal the temporal dynamics of response behavior, and (3) allows to track performance changes on multiple time scales
Temporal dynamics of the immune response in Astacus astacus (Linnaeus, 1758) challenged with Aphanomyces astaci Schikora, 1906
One of the main drivers of biodiversity loss in freshwater ecosystems are alien invasive species. In Europe, pathogen Aphanomyces astaci Schikora, 1906, is considered as one of the most problematic invasive species, as its introduction caused the severe decimation of the European freshwater crayfish stocks. The most affected are the populations of noble crayfish, keystone species native to European freshwaters. Unfortunately, even after decades of research, we do not understand the temporal dynamics of the noble crayfish immune response during Ap. astaci infection. Here, we studied the changes in the immune response of the noble crayfish during a time course challenge with a highly virulent strain of Ap. astaci. We recorded gross symptoms of the disease, changes in the total haemocyte count (THC), gene expression profiles of putative immune response regulators and pathogen load. Additionally, we conducted a preliminary histological analysis of the pathogen dissemination in host tissues. Based on the occurrence of symptoms we propose three stages in the crayfish plague disease progression: asymptomatic stage, symptomatic stage, and finally death of infected individual. Furthermore, based on the qPCR analysis we could differentiate three Ap. astaci growth phases: initial lag phase, followed by exponential growth phase and finally sporulation phase. We observed that all measured immune response parameters were significantly correlated to the observed increase in the pathogen load (qPCR). Altogether, our results point to the absence of a successful immune response in the noble crayfish to a challenge with a highly virulent strain of Ap. astaci. The noble crayfish immune system was not able to suppress the growth of the intruding pathogen. In general, our observations have to be considered in the context of the specific combination of crayfish plague pathogen virulence and disease resistance of the challenged crayfish population, which defines the temporal dynamics of their interaction
