    Functional stability despite structural changes in freshwater biofilm communities exposed to an antibiotic and an herbicide - the role of nutrient conditions

    Freshwater autotrophic biofilms play a vital role in primary production and nutrient cycling in freshwater ecosystems but are increas- ingly exposed to chemical stressors such as antibiotics or herbicides. Although nutrient availability may modulate biofilm sensitivity, its impact on biofilm responses to these stressors remains poorly understood. In four independent experiments, we investigated the functional (ash-free dry weight and chlorophyll a, b and c) and structural (16S/18S rRNA metabarcoding) responses of stream- derived biofilms under low- and high-nutrient levels to chronic exposure (14 days) to the antibiotic ciprofloxacin and the herbicide propyzamide in laboratory stream microcosms. High-nutrient levels strongly increased biofilms functional responses and altered the community composition. Chemical exposure led to pronounced shifts in prokaryotic (ciprofloxacin) and eukaryotic (propyzamide) communities, but without significant effects on functional responses, suggesting functional redundancy and ecological buffering ca- pacity of freshwater biofilms. These results highlight the critical role of nutrient supply in biofilm responses and the need for caution when extrapolating laboratory results to field conditions

    Synergy in Sonogashira Cross-Coupling Reactions with a Magnetic Janus-Type Catalyst

    This work describes the straightforward synthesis of a novel heterogeneous palladium catalyst immobilized on magnetic Janus-type silica particles coated with an amphiphilic ionic liquid (IL) layer. The material was prepared via a one-pot process wherein TEOS (tetraethoxysilane) and a bis(triethoxysilane) IL precursor are combined to form hollow shells. The IL motifs are selectively located on the outer surface of the hollow particles and serve as centers for the immobilization of palladium species on the material’s surface. The outer surface also hosts magnetic nanoparticles in close proximity to the palladium sites. Thanks to the uniform coverage of the surface with the amphiphilic IL functionality, the material exhibits a well-balanced wettability with reaction components of different polarities. The catalyst’s activity was tested in the Sonogashira cross-coupling reaction of terminal acetylenes and iodobenzene derivatives in water as the solvent. The results show that the mixed palladium–iron oxide catalyst exhibits higher activity than materials containing either immobilized palladium or iron oxide alone, suggesting a synergistic effect in this reaction. Additionally, the reaction proceeds well in the absence of expensive organic ligands and commonly employed additives such as copper co-catalysts or phase transfer catalysts. Furthermore, the material was also used in the oxidative Sonogashira coupling reaction of phenylboronic acid and phenylacetylene. The catalyst can be easily separated using an external magnet and can be reused several times. The feasibility of producing diphenylacetylene on a gram scale via the Sonogashira cross-coupling reaction was also investigated

    Is the Activation of the Postsynaptic Ligand Gated Glycine- or GABAA Receptors Essential for the Receptor Clustering at Inhibitory Synapses?

    One major challenge in cellular neuroscience is to elucidate how the accurate alignment of presynaptic release sites with postsynaptic densely clustered ligand-gated ion channels at chemical synapses is achieved upon synapse assembly. The clustering of neurotransmitter receptors at postsynaptic sites is a key moment of synaptogenesis and determinant for effective synaptic transmission. The number of the ionotropic neurotransmitter receptors at these postsynaptic sites of both excitatory and inhibitory synapses is variable and is regulated by different mechanisms, thus allowing the modulation of synaptic strength, which is essential to tune neuronal network activity. Several well-regulated processes seem to be involved, including lateral diffusion within the plasma membrane and local anchoring as well as receptor endocytosis and recycling. The molecular mechanisms implicated are numerous and were reviewed recently in great detail. The role of pre-synaptically released neurotransmitters within the complex regulatory apparatus organizing the postsynaptic site underneath presynaptic terminals is not completely understood, even less for inhibitory synapses. In this mini review article, we focus on this aspect of synapse formation, summarizing and contrasting findings on the functional role of the neurotransmitters glycine and γ-aminobutyric acid (GABA) for initiation of postsynaptic receptor clustering and regulation of Cl− channel receptor numbers at inhibitory synapses gathered over the last two decades

    Resilience and tipping points in social-ecological systems of the southwestern Amazon: a participatory systems analysis

    Recent reports warn of the imminent surpassing of a tipping point that will transform the Amazon rainforest into a savannah. In the transboundary region shared by Bolivia, Brazil, and Peru (named the MAP region, an acronym for the states of Madre de Dios, Peru; Acre, Brazil; and Pando, Bolivia), this land-use conversion has been triggered by logging and farming, and recently intensified by the construction of a transcontinental highway. As economic incentives grow, so does social and environmental harm. We devised a participatory systems approach to evaluate and characterize the social-ecological system in a site in each of the three countries, and to identify the factors that may lead to the systems’ loss of resilience and eventual trespassing of tipping points. Methodologically, the approach combines stakeholder analysis, network analysis, and systems analysis, and includes transboundary research and locals’ participation as overarching premises adding plausibility and legitimacy to the findings. Results show that the three country-sites, despite their similarities, have evolved divergently into archetypal social-ecological systems, each confronting specific challenges, i.e., in Bolivia, dependence on the volatile commodity Brazil nut, and unstable politics, has weakened a previously stable and functional institutional landscape. In Brazil, the conversion of forests into pastures for cattle ranching remains the major peril, fueled by a steady market demand and, regardless of the party in power, insufficient restraining measures. In Peru, economic diversification has increased the system’s resilience as a whole, however authorities struggle to exert control over the economic activities, some of which are illegal

    Enhancing Interpretable Machine Learning for Earth Observation

    Remote sensing (RS) provides abundant and diverse data for Earth Observation (EO) applications. Machine learning leverages the available data through deep neural networks and specialized architectures. However, increasing model complexity often compromises its interpretability, which is crucial for many EO applications that monitor sensitive human activities or support natural disaster response efforts. This thesis contributes to advancing the interpretability and explainability of complex AI models for various RS applications, with a specific focus on agricultural activities. Our work employs eXplainable AI (XAI) methods to address two main objective for understanding and improving the model predictions within EO applications. First, we focus on XAI for Justification where the model behavior is justified by analyzing how different input features contribute to the outputs. The explanation of individual predictions are leveraged and aggregated to provide a broader understanding of the model’s behavior. We apply and evaluate existing model-agnostic and model-specific methods, while also developing new techniques when necessary. We further explore how multi-task learning can further enhance the explainability of predictions. Second, we apply XAI for Improvement based on insights from our prior model justification results. On one hand, we identify the features that are necessary and sufficient for accurate modeling across different contexts. On the other hand, we focus on optimizing the selection and design of vegetation indices, a key component in EO analysis and modeling. We benchmark our explainability objectives across multiple datasets, covering a range of tasks in EO. We particularly focus on multi-modal datasets, commonly used in EO, to mitigate the research gap regarding the explanation of complex multi-modal networks. The results demonstrate that our approach effectively explains the models by verifying that the model reasoning aligns with expert knowledge. Additionally, our experiments on vegetation indices and the optimization of models through feature reduction yielded promising results, and contributed to enhanced overall model performance and interpretability. Overall, this thesis provides a thorough examination of the interpretability of ML models under complex modeling scenarios. It leverages various explainability tools and objectives to justify model predictions and improve the modeling strategy and performance. This work contributes an important building block towards more transparent and better performing ML models designed for EO applications

    Compliance in einer gemeinnützigen Gesellschaft mit beschränkter Haftung Ausrichtung ambulanter Pflegebereich

    Die Masterthesis beschäftigt sich mit der Frage, ob Compliance ein Thema für eine gemeinnützige GmbH wäre. Ziel der Masterthesis ist es daher, das Thema Compliance in einer gemeinnützigen Gesellschaft mit beschränkter Haftung mit der Ausrichtung im ambulanten Pflegebereich zu untersuchen. Die gemeinnützige Ausrichtung steht bei der Masterthesis im Vordergrund. Die Masterthesis befasst sich mit der rechtlichen Einordnung der ambulanten Pflege sowie deren Tätigkeitsfeld. Es erfolgt eine Abhandlung der spezifischen rechtlichen und steuerrechtlichen Besonderheiten einer gGmbH, mit der Zuordnung der ambulanten Pflege zu den Sphären der gemeinnützigen Zwecke nach der AO. Untersucht wird zudem die Pflicht zur Implementierung eines CMS in einer gGmbH, wobei vor allem die Funktionen von Compliance angeführt werden. Anhand der DIN ISO 37301 erfolgt darüber hinaus eine Beschreibung, wie ein CMS in den Alltag einer gemeinnützigen GmbH eingeführt werden könnte. Dazu werden die wichtigsten Risikobereiche sowie daraus resultierende Sanktionen erläutert. Zudem wird aufgezeigt, wie ein CMS die Haftungsgefahren minimieren könnte und gleichzeitig Schutz für die Verantwortungsträger bietet. Es erfolgt eine Ausführung zur Zertifizierung. Reale Sachverhalte sollen ferner verdeutlichen, weshalb Compliance in den täglichen Arbeitsbereich einer gemeinnützigen GmbH integriert werden sollte

    Die vorliegende Masterarbeit untersucht die Transformation von Rettungsdienstorgani- sationen mit dezentraler Aufbauorganisation zu lernenden Organisationen. Angesichts steigender Komplexität im Gesundheitswesen und der Notwendigkeit kontinuierlicher Anpassung stehen Rettungsdienste vor der Herausforderung, ihre organisationale Lern- fähigkeit zu verbessern. Basierend auf einer systematischen Literaturanalyse von 43 wissenschaftlichen Publikationen werden die theoretischen Grundlagen lernender Orga- nisationen dargestellt und ihre Anwendbarkeit auf das deutsche Rettungswesen analy- siert. Die Ergebnisse zeigen, dass etablierte Modelle wie Senges Fünf Disziplinen (Senge, 1990, S. 3-15), das SECI-Modell von Nonaka und Takeuchi (1995, S. 56-94) und Laloux' Teal Organizations-Konzept (2014, S. 43-89) grundsätzlich auf das Ret- tungswesen anwendbar sind, jedoch kontextspezifische Anpassungen erfordern. Die de- zentrale Struktur erweist sich als zweischneidiges Schwert, das sowohl Potenziale für lokale Lernprozesse als auch Herausforderungen für organisationsweite Koordination schafft. Die entwickelten Handlungsempfehlungen umfassen einen vierphasigen Trans- formationsrahmen, konkrete Implementierungsstrategien und Evaluationsmethoden.     This dissertation investigates the advancements in the field of infrared thermography (IRT) for non-destructive testing (NDT) through the implementation of a physics-based one-dimensional N-layer modelling (NLM) approach. This approach provides a robust basis for the development of new IRT post-processing methods and for the improvement of existing methods, covering both theoretical modelling and practical applications. A key focus of this work is the development of Thermal Shock Response Spectrum (TSRS) methods, an innovative approach to IRT data processing. The TSRS is based on an analytical thermal model and represents an approach analogous to the established mechanical Shock Response Spectrum (SRS) method, which is standardized in the international standard ISO 18431-4. The integration of a thermal Q-factor and a characteristic frequency as central control parameters, in conjunction with their algorithmic optimization via Latin Hypercube Sampling (LHS), has resulted in a substantial advancement in the capabilities for detecting and precisely characterizing material defects. The parameters enable targeted adaptation of data processing, which can significantly improve the differentiation between defect-free and defective areas. Another focus is on the use of NLM simulations to generate training data for machine learning (ML), a method that enables the time- and cost-efficient simulation of thermal processes and reduces the dependence on complex experimental data sets. The developed ML models could be successfully used for automated defect detection and material analysis, representing a significant extension of the application spectrum of NLM in IRT. The present work demonstrates that the NLM approach opens up new possibilities for non-destructive testing, both implicitly through optimized IRT methods such as TSRS and explicitly as a basis for training data generation for machine learning algorithms. The results not only offer a significant improvement of existing NDT methods but also provide important impulses for the further development of industrial testing methods and scientific research, especially in the field of inspection and analysis of composite materials.Die vorliegende Dissertation befasst sich mit der Weiterentwicklung im Bereich der Infrarot-Thermografie (IRT) für die zerstörungsfreie Prüfung (NDT) durch die Anwendung eines physikalisch fundierten eindimensionalen N-Layer-Modellierungsansatzes (NLM). Dieser Ansatz bildet eine robuste Grundlage für die Entwicklung neuer IRT-Postprozessing-Methoden und zur Verbesserung bestehender Verfahren, die sowohl theoretische Modellierung als auch praktische Anwendungen abdecken. Ein zentraler Schwerpunkt der vorliegenden Arbeit liegt in der Entwicklung der Thermal Shock Response Spectrum (TSRS)-Verfahren, einem innovativen Verfahren zur IRT-Datenverarbeitung. Das TSRS basiert auf einem analytischen thermischen Modell und repräsentiert einen Ansatz, der in Analogie zu dem etablierten mechanischen Shock Response Spectrum (SRS)-Verfahren steht, welches in der internationalen Norm ISO 18431-4 standardisiert ist. Die Einführung eines thermischen Q-Faktors und einer charakteristischen Frequenz als zentrale Steuerparameter sowie deren algorithmische Optimierung durch das Latin Hypercube Sampling (LHS) ermöglichte eine signifikante Erweiterung der Möglichkeiten zur Detektion und präzisen Charakterisierung von Materialdefekten. Diese Parameter ermöglichen eine gezielte Anpassung der Datenverarbeitung, wodurch die Differenzierung zwischen defektfreien und defektbehafteten Bereichen signifikant verbessert werden kann. Ein weiterer Schwerpunkt liegt auf der Nutzung von NLM-Simulationen zur Generierung der Trainingsdaten für maschinelles Lernen (ML). Diese Methode ermöglicht die zeit- und kosteneffiziente Simulation thermischer Prozesse und reduziert die Abhängigkeit von aufwendigen experimentellen Datensätzen. Die entwickelten ML-Modelle konnten erfolgreich zur automatisierten Defektdetektion und Materialanalyse eingesetzt werden, was eine signifikante Erweiterung des Anwendungsspektrums der NML in IRT darstellt. Die vorliegende Arbeit demonstriert, dass der NLM-Ansatz, sowohl implizit - durch optimierte IRT-Methoden wie TSRS - als auch explizit - als Grundlage zur Trainingsdatenerzeugung für ML-Algorithmen - neue Möglichkeiten für die zerstörungsfreie Prüfung eröffnet. Die Ergebnisse bieten nicht nur eine signifikante Verbesserung der bestehenden NDT-Methoden, sondern liefern auch wichtige Impulse für die Weiterentwicklung industrieller Prüfverfahren und die wissenschaftliche Forschung, insbesondere im Bereich der Inspektion und Analyse von Verbundwerkstoffen

    Der Beitrag entfaltet einen primär syntaktischen Zugang zu den Wortarten des Deutschen und zeigt, wie Lernenden von Beginn an eine funktionale Perspektive auf die Rolle von „Wörtern in Sätzen“ vermittelt werden kann. Mit der sog. Satzmatrix wird ein Modell für den Grundschulunterricht vorgestellt, das die syntaktischen Kontexte definiert, in denen die Wörter einer Klasse typischerweise im Satz auftreten.     Evaluation of the performance of the 5G mobile communications standard for machine tools – An analysis using the example of a digital twin of a numerically controlled milling machine

    Die Herstellung von Werkzeugmaschinen ist ein essentieller Erfolgsfaktor für die deutsche Wirtschaftskraft. Um die globale Marktposition zu sichern oder auszubauen, sind kontinuierliche Innovationen sowohl an der Werkzeugmaschine als auch an deren Steuerungseinheit notwendig. Bestärkt wird dieser Innovationsbedarf durch rasche Fortschritte in der Informations- und Kommunikationstechnologie sowie Informatik, die eine schnelle Adaption und einen kontinuierlichen Wandel innerhalb von Produktionssystemen ermöglichen und notwendig machen. Durch die gestiegene Leistungsfähigkeit kabelloser Kommunikationstechnologien steigt die Verbreitung von Cyber-Physischen Produktionssystemen (CPPS), welche digitale und physische Welten vernetzen und damit eine anpassungsfähige Produktion ermöglichen. Die hierdurch wachsende Komplexität erfordert den Einsatz digitaler Zwillinge (DZ) zur bidirektionalen Interaktion innerhalb des CPPS. Zur Erschließung dieser neuen Möglichkeiten muss die Wandlungsfähigkeit von Werkzeugmaschinen und deren Teilsystemen adressiert werden, wofür neuartige Kommunikationslösungen integriert werden müssen. Insbesondere für automatisierte Werkzeugmaschinen müssen hohe Anforderungen hinsichtlich der Leistungsfähigkeit der Kommunikationslösung sichergestellt werden. Der fünfte Mobilfunkstandard (5G) ermöglicht erstmals die kabellose Steuerung und Regelung von Werkzeugmaschinen, wodurch ein dezentraler, anpassbarer und skalierbarer Betrieb ermöglicht wird. Innerhalb dieser Arbeit werden die Potenziale 5G-basierter Kommunikation für einen DZ einer Werkzeugmaschine untersucht. Das Ziel besteht darin, die Leistungsfähigkeit eines 5G-Campusnetzwerkes in einer industriellen Umgebung zu evaluieren und die Auswirkungen der kabellosen Kommunikation auf die Funktionen des DZ sowie auf die gefertigte Bauteilqualität experimentell zu untersuchen. Hierzu erfolgt zunächst die konzeptionelle Entwicklung eines Rahmenwerks für DZ in Abhängigkeit der Leistungsfähigkeit verschiedener Stufen des 5G-Standardisierungsprozesses. Anschließend wird ein DZ einer Werkzeugmaschine systematisch modelliert, entwickelt und implementiert. Eine Anforderungsanalyse, die Ausarbeitung des strukturellen Aufbaus und Informationsablaufs des Systems sowie die Beschreibung der Funktionseinheiten erfolgen. In Abhängigkeit der Leistungsfähigkeit der 5G-basierten Kommunikation existieren verschiedene Integrationsstufen der Computerized Numerical Control (CNC) der Werkzeugmaschine innerhalb des DZ. Die Funktionalität des Gesamtsystems wird mittels vollfaktoriellen Versuchsreihen untersucht und validiert. Sowohl Leistungskennzahlen der Netzwerkkommunikation wie Latenz, Jitter und Übertragungszuverlässigkeit als auch die Beeinflussung der physischen und digitalen Prozessfähigkeit werden untersucht. Der Einfluss von 5G-basierter Kommunikation auf die Funktionen des DZ sowie die gefertigte Bauteilqualität wird evaluiert. Die Ergebnisse zeigen, dass 5G hinreichend leistungsfähig für die kabellose Überwachung und Steuerung von Werkzeugmaschinen ist. Für die Virtualisierung und kabellose Integration der CNC ist insbesondere eine deterministische Kommunikation erforderlich, welche entsprechend der Spezifikationen des 5G-Standards künftig erreicht werden sollte. In Summe zeigt 5G ein hohes Potenzial für die kabellose industrielle Vernetzung und die Umsetzung von DZ von Werkzeugmaschinen. Demnach ermöglicht 5G-basierte Kommunikation kabellose Anwendungen für Werkzeugmaschinen, welche zuvor auf eine kabelgebundene Verbindung angewiesen waren.The production of machine tools is a significant contributor to Germany's economic strength. In order to secure or expand its global market position, continuous innovation is required both in the machine tool and in its control unit. This need for innovation is reinforced by rapid advances in information and communication technology and computer science, which enable rapid adaptation and continuous change within production systems. In particular, the increased performance of wireless communication technologies enables the adoption of cyber-physical production systems (CPPS), which interconnect the digital and physical worlds thereby enabling adaptable production and more effectively meeting individual market requirements. The resulting increase in complexity requires the use of digital twins (DTs) for bidirectional interactions within the CPPS. Automated machine tools in particular must meet high requirements in terms of the performance of the communication solution. The fifth mobile communications standard (5G) enables wireless control of machine tools, leading to decentralized, flexible and scalable operation. In this thesis, the potential of 5G-based communication for DTs of machine tools is investigated. The objective is to evaluate the performance of a 5G campus network in an industrial environment and to experimentally investigate the effects of 5G-based communication on the functionality of the DT and on the manufactured component quality. For this purpose, a framework for DTs depending on the performance characteristics of different stages of the 5G standardization process is developed. Then, the DT of the machine tool is systematically modelled and implemented. A requirements analysis is conducted, the structural design and information flows of the system are specified, and the functional units are described and specified. Depending on the performance of 5G, different levels of integration of the computerized numerical control (CNC) within the DT of the machine tool can be achieved. The functionality of the system is analyzed and validated with multiple full-factorial test series. Different variables such as latency, jitter, and transmission reliability are investigated. Moreover, the influence of 5G on the functions of the DT and the manufactured part quality are evaluated. The results show that 5G is sufficiently powerful for the wireless monitoring and control of machine tools. However, time-deterministic communication is required for virtualization and wireless integration of the CNC. This requirement can be met in the future, as defined in the specifications of the 5G standard. To sum up, 5G shows potential for wireless industrial communication and the realization of DTs of machine tools. Accordingly, 5G-based communication enables wireless applications for machine tools that previously relied on wired connections
