    PERFORMANCE OF PB 330 CLONE RUBBERS

    ABSTRACTKeragaan Klon Karet PB 330 bertujuan untuk mengevaluasi tingkat adaptasi terhadap lingkungan, pertumbuhan tanaman, produktivitas lateks, mutu lateks dan sifat klon karet PB 330, potensi hasil kayu serta ketahanan terhadap hama dan penyakit. Evaluasi adopsi dan keragaan klon karet PB 330 dilakukan melalui pengumpulan data primer, data sekunder dan wawancara dengan peneliti maupun pelaku pengembangannya dengan menggunakan kuesioner terstruktur. Data sekunder diperoleh dari Perusahaan Perkebunan dan Tinjauan Pustaka. Data primer diperoleh melalui kunjungan lapang (field assessment) ke kebun produksi dan kebun pembibitan yang dilakukan pada bulan Oktober 2007. Evaluasi mutu hasil dilakukan melalui studi pustaka dan kunjungan ke pabrik pengolahan. Data hasil survey dianalisis secara deskriptif dan ditampilkan dalam bentuk tabel maupun gambar. Klon PB 330 ternyata memiliki pertumbuhan dan produksi yang sangat baik, dapat dipanen pada umur 4 tahun dengan produktivitas yang langsung tinggi (quick starter or precocious yielder). Dibanding klon PB 260 yang merupakan klon terbaik saat ini, klon PB 330 ternyata memiliki produktivitas sekitar 11% lebih tinggi. Dengan pertumbuhan lebih tegap, maka diperhitungkan PB 330 juga memiliki potensi produksi kayu yang tinggi, mempunyai daya adaptasi cukup baik terhadap lingkungan pertanaman karet di Indonesia dan belum ada laporan kendala gangguan hama dan penyakit yang serius di pertanaman komersial.Kata kunci: PB 330. klon karet unggul, keragaan, produksi

    ABSTRACT Sugarcane (Saccharum officinarum L.) is one of importance crop grown in marginal in Indonesia. Phosphorus (P) is critical to the growth and development of plant in the marginal land. P is stored in plant  as phytic acid (myo-inositolhexakisphosphate).  Phytic acid is hydrolyzed by the activity  of phytases to yield inositol and free phosphate. Genetic transformation of sugarcane with phytases gene holds promise to provide enough P during period of rapid cell division and growth of plant.  Plant transformation mediated by Agrobacterium tumefaciens, has become the most used method for the introduction foreign genes into plant cells and the sub sequens regeneration of transgenic plant. The selection and regeneration of embryogenic callus of transformed plant was done on MS medium containing kanamycin. The main objective of this study were: (1) To find the best kanamycin  concentration for selectable marker; (2) Insert phytase gen into  varieties of sugarcane (cv. PA 175) through Agrobacterium tumefaciens GV 2260 (pBinPI-IIEC); and (3)  To analyze of intregated transgene into genom of sugarcane using PCR  method.  Result of the experiment showed: (1) kanamycin selectable marker lethal doses for transformed sugarcane calli: 100 mg l-1;   (2)  Efficiency transformation of putative transgenic line  was cv. PA 175= 24 %; (3)  The first culture of transformed calli become 24 (Triton cv.), 18 (PSJT 94-41 cv.), and 30 (PA 175 cv.) putative plants; the second sub cullture of putative eksplant regenerate become new plant: 380 (PA 175 cv.) plants.  (4) Analyzed of integrated phytase gene was proven by appearance of 900 bp of PCR band (5) transgenic plants (cv. PA 175)  with highest activities respectively: 45 %; with medium phytase activity: 27 %, and low phytase activity: 27 % from total of sample.  Non transgenic plants, most of sample show low phytase activity respectively:  100 % , noneh show medium - hight activy of phytase.  Key words: Sugarcane, transformation, phytase,  Agrobacterium tumefacien

    PERTUMBUHAN DAN HASIL TANAMAN JAGUNG MANIS (Zea mays saccharata Sturt) PADA BERBAGAI DOSIS PUPUK ORGANIK DAN INTENSITAS PENGOLAHAN TANAH ULTISOLS BANTEN

    ABSTRACT This Research aim to to know sweet maize crop result and growth ( Zea Mays Saccharata Sturt.) At various organic manure dose and intensity processing of soil tillage,This research have been executed in kampong baru petir Banten. Research take place from June until September 2008. Research use Random Device of Group ( factorial RAK) Pattern, consisting of two factor and three restating, first factor is organic manure dose with four level that is, 0; 10; 20; and 30 ha-1 ton. Second factor is intensity processing of soil tillage with three level that is, once, twice, and thrice processing of soil tillage, so that there are 12 treatment combination and 36 set of attempt. Result of research indicate that organic manure dose differ reality, clean weighing of cob at organic manure dose 10 ha-1 ton equal to 1.386,8 g per check, organic manure dose 20 ha-1 ton equal to 1.316,27 g per organic manure and check 30 ha-1 ton equal to 1.393,83 g per check. Intensity Processing of soil tillage show to differ is not real and also  not happened interaction among organic manure dose with intensity processing of soil tillage.Key words: soil tillage, organic manure, Ultisol

    ABSTRACTBanten bay coastal zone is a unique ecosystem which has a variety potencies and problems of using some natural resources, especially in the trade off between economic growth and ecological preservation. Based on these conditions, this research aimed to identify the management feasibility of Banten Bay coastal resources. An extended cost-benefit analisys which integrated the external and/or environmental benefits, damage cost and cost of society of environmental protection measures was used as methods of this research. The result of this research showed that all of management options of Banten Bay coastal resources were feasible to be developed. Based on comparative performance index, it was known that sustainable mangrove management, sustainable harvest and beach protected areas were the most feasible management option to be implemented in the management of Banten Bay coastal resources.   Key words:  cost-benefit analysis, sustainable management, coastal and marine resources

    ABSTRACTResearch about the effect of multiple doses of gamma irradiation to improved the appearance of the chrysanthemum plant has been carried out on chrysanthemum varieties Gold van Langen-doses of gamma irradiation: 0 Gy, 5 Gy, 10 Gy, and 15 Gy.  Gamma irradiation has given at 2-week-old seedlings, which already have roots. Seeds that have been irradiated maintained up to 16 WAP. The results showed there are no all doses causes death in up to 16 WAP chrysanthemum seedlings but with a dose of 15 Gy seedlings did not grow at all. Dose of 5 Gy encourage the formation of lateral shoots on chrysanthemum. The higher the dose given causing the growth of seedlings of chrysanthemum hampered.Keywords:  Chrysanthemum, Mutation Induction, irradiation

    FORMULASI BAHAN DASAR LENGKAP LARUTAN PENGAWET BUNGA POTONG KRISAN (Dendranthema indicum)

    ABSTRACTThe quality of cut flowers can be improved by soaking them in a preservative solution, containing three basic compounds, sucrose, citric acid, and Lysol (called the basic formula). A research was conducted to find out an exact formulation which can give an optimal result by mixing the basic formula with cytocinine as a growth substances or anti-ethylene molecule as an completing agent (called the complete basic formula) for chrysanthemum cut flower. This research was done in the plant physiology laboratorium at Faculty of Agriculture of Bandung Raya University, Bandung on July to August 2004 by using univariat Randomized Block Design method consists five treatments, which are basic formula, basic formula + cynetine, basic formula + BAP, basic formula + AOA, and basic formula + STS. The result shows that the quality of chrysanthemum cut flowers can be kept by soaking it in a basic formulation preservative solution only, which containts 48 x 103 mgL-1 sucrose + 241 mgL-1 citric acid + 800 mgL-1 lysol in 14 hours. Soaking the cut flowers in a preservative solution, the decreasing of fresh weight is also minimalized and relative 2-days water uptake is maximalized. The number of ethylene that synthesized and respiration activity of chrysanthemum cut flowers can be pushed by adding the cytocinine into basic formula. Key Words:  Preservative Solution, Quality Of Chrysanthemum Flowers, Growth Substances Cytocinine, Anti-Ethylen

    ABSTRACTTo understand the function of KNAT1 gene Arabidopsis in the forming and developing of Indonesian origin orchid shoot Phalaenopsis amabilis through transform p35S::KNAT1 and pGreen to protocorm like bodies (plb) of the orchid mediated by Agrobacterium tumafaciens LBA4404. The plb transformants were grown on New Phalaenopsis selection medium containing 5 mg/l BAP, 0.15 mg/l NAA, 15 mg.l Kanamycin and addition of 300 mg/l Cefotaxim to eliminate the overgrowth of Agrobacterium. The analysis of positive transformant use Polimerase Chain Reaction (PCR) with the specific oligonucleotide primers for KNAT1 gene: KNAT1F1R1 and universal primers for pGreen: 35SO and Tnos. The result shows that 3 shoots of 1850 transformants positively carry out the 35S::KNAT1 construct (frequency of transformation was 0.16 %) while 5 shoot of 1850 transformants also positively carry out the pGreen vector, with the frequency of transformation was 0.27 %. The phenotype analysis of 35S::KNAT1 transformants show multiplication on forming of the leaf from a plb to + 10 shoots and forming of the leaf shape which has terompet like shapes and rectangular shape.Key words: Arabidopsis, KNAT1 gene, Phalaenopsis amabilis, shoo

    ABSTRACTA research was carried out to know response duku seedling on soil from various location to NPK fertilizer. The research was conducted in green house of BPSB TPH IX from January until April and arranged in Randomized Completely Block Design with Split Plot Design and to know NPK absorbtion to leaf meristem obtained through laboratory analysis. The result of research showed that response of duku seedling on soil from various locations to NPK fertilizer was tested not significant to all of parameters observed except to plant height accretion of NPK fertilizer.Key words: Duku (Lansium domesticum Correa), Seedling, NP
