    Anomalous glucose-responsive rheological changes in a boronic acid-modified hyaluronan

    Herein, we report anomalous glucose (Glc)-responsive gelation/solation in 3-aminophenylboronic acid-modified hyaluronic acid. With 5-20 mM Glc, gelation occurred, resulting in the formation of crosslinks via Glc, which could reversibly bind to the two boronic acid sites.     Cytotoxic Effects of Darinaparsin, a Novel Organic Arsenical, against Human Leukemia Cells

    To explore the molecular mechanisms of action underlying the antileukemia activities of darinaparsin, an organic arsenical approved for the treatment of peripheral T–cell lymphoma in Japan, cytotoxicity of darinaparsin was evaluated in leukemia cell lines NB4, U-937, MOLT-4 and HL-60. Darinaparsin was a more potent cytotoxic than sodium arsenite, and induced apoptosis/necrosis in NB4 and HL-60 cells. In NB4 cells exhibiting the highest susceptibility to darinaparsin, apoptosis induction was accompanied by the activation of caspase-8/-9/-3, a substantial decrease in Bid expression, and was suppressed by Boc-D-FMK, a pancaspase inhibitor, suggesting that darinaparsin triggered a convergence of the extrinsic and intrinsic pathways of apoptosis via Bid truncation. A dramatic increase in the expression level of γH2AX, a DNA damage marker, occurred in parallel with G2/M arrest. Activation of p53 and the inhibition of cdc25C/cyclin B1/cdc2 were concomitantly observed in treated cells. Downregulation of c-Myc, along with inactivation of E2F1 associated with the activation of Rb, was observed, suggesting the critical roles of p53 and c-Myc in darinaparsin-mediated G2/M arrest. Trolox, an antioxidative reagent, suppressed the apoptosis induction but failed to correct G2/M arrest, suggesting that oxidative stress primarily contributed to apoptosis induction. Suppression of Notch1 signaling was also confirmed.     Front propagation and blocking for the competition-diffusion system in a domain of half-lines with a junction

    The two-component Lotka-Volterra competition-diffusion system is well accepted as a model describing the invasion of a superior species into a new habitat. Under a bistable condition, we deal with the system in a domain of half-lines with a single junction and investigate the condition for the invasion from some of the half-lines beyond the junction or blocking the propagation of the superior species. We first give a sufficient condition for the invasion in the whole domain by a subsolution. Then, making use of sub- and supersolutions, we construct a standing front solution blocking the propagation if the number of half-lines occupied by the inferior species is sufficiently larger than that occupied by the superior species

    軟骨細胞におけるグルコサミンのオートファジーに対する効果を検討するため、まずヒト軟骨肉腫由来培養細胞株を用いて、その評価系を構築した。その後、グルコサミンの軟骨細胞におけるオートファジーに対する効果を検討したところ、グルコサミンがオートファジーを活性化する可能性が示唆された。さらに、mammalian target of rapamycin(mTOR)非依存的な経路により活性化されている可能性が示唆された。また、オートファジー活性化の上流に位置していると思われるサーチュイン(SIRT)1の発現も検討したところ、グルコサミンがSIRT1を介してオートファジーを誘導する可能性が示唆された。Glucosamine (GlcN), a naturally occurring amino monosaccharide, has been widely used to treat osteoarthritis (OA) in humans. However, the effects of GlcN on autophagy in chondrocyte are still unknown. In this study, to elucidate the chondroprotective action of GlcN via autophagy, we evaluated the effect of GlcN on the expression of autophagy-related molecules (LC3-II, Beclin-1, ATG5 and ATG7) using a human chondrocyte. GlcN significantly increased protein level of LC3-II and mRNA expression of Beclin-1, ATG5 and ATG7. Moreover, GlcN significantly increased the protein level of SIRT1. Importantly, the GlcN-induced increase of LC3-II protein was inhibited by EX527 (a SIRT1 inhibitor). Moreover, GlcN did not effect the S6K phosphorylation, as a target of mammalian target of rapamycin (mTOR).     The Object of Assault and the Relativity of Concepts of Assult : Using the case law and legislative history of the crime of Disturbance

    わが国の刑法における暴行の概念は、これを 4 分類とする説明が一般化しているが、その区別基準は不統一であるという問題を持つ。そこで、騒乱罪について判例や現行刑法の立法史を参照し、特に暴行の客体の観点と暴行概念の相対性の議論について検討を加える。判例によれば、本罪の暴行はその客体を問わず、「一地方における公共の平穏を害するに足りる程度」であれば足りるとされ、立法史を参照する限り、これは本罪が内乱罪と同質の犯罪として立法されたことを背景とする。攻撃の客体ではなく、保護客体への作用を問題とするという本罪の特徴からその暴行概念の検討し、「客体」は暴行の分類基準としては妥当でなく、保護客体への「作用」の観点から解釈すべきであるとの私見を展開する。The concept of assault in Code Penal of Japan has been generally explained in terms of four categories, but the criteria for distinguishing between these four categories are inconsistent. Therefore, we will discuss the crime of Disturbance by referring to precedents and the legislative history of the current Penal Code, particularly in terms of the object of the assault and the discussion of the relativity of the concept of assault. According to the judicial precedents, it is sufficient that the assault in this crime is “sufficient to disturb public peace in a region,” regardless of its object, and this is derived from the fact that this crime is identical to the riot concept in the crime of Insurrection and is characterized by the fact that the issue is not the object of the attack but the effect on the protected object. From an examination of the assault concept in this crime, “object” is not a valid classification criterion for assault and should be interpreted in terms of “action” on the protected object.論文 / Article

    A study on the evaluation of power production ability in sprint running

    疾走が含まれる競技スポーツ種目では,加速能力を検討する上で,加速中に発揮されたパワーの大きさが重要となる。本研究では,走運動におけるパワー発揮能力およびその評価について検討を行った。その結果,自身が発揮しうる脚伸展パワーを,加速疾走中にどの程度水平方向に発揮できるかというスキルには個人差があることが示唆された。また,加速疾走におけるパワー発揮のトレーニング手段として,傾斜走路を用いる際には,傾斜走路の傾度はパワー発揮特性にも影響することから,練習やトレーニング目的によって,用いる傾度を明確にする必要があることが示唆された。In competitive sports events involving sprinting, the power exerted during acceleration is important in examining acceleration ability. In this study, we examined the ability to exert power during running and its evaluation. The results suggest that there are individual differences in the transference to which a subject can exert leg extension power in the horizontal direction during accelerated sprinting.
