Effect of Nattokinase in D-galactose- and Aluminum Chloride-induced Alzheimer’s Disease Model of Rat
Background/Aim: Alzheimer’s disease (AD) is the most common form of dementia worldwide. Nattokinase is a serine protease extracellularly produced by natto, a fermented product of Bacillus subtilis var. natto. In this study, we investigated the therapeutic effects of nattokinase in a rat model of AD induced by aluminum and D-galactose. Materials and Methods: Forty Wistar rats were randomly divided into four groups: normal, vehicle, and orally administered nattokinase (NK65 and NK130 groups). Except for the normal group, all groups were treated with AlCl3 and D-galactose for 10 weeks. The NK65 and NK130 groups additionally received 65 mg/kg/day and 130 mg/kg/day nattokinase, respectively. We analyzed β-amyloid levels in the cerebrospinal fluid (CSF), and the spatial reference test was evaluated using the Morris water maze test. After the Morris water maze test, rats of all groups were subjected to micro-computed tomography (μCT) to assess constructional changes in the brain. Aluminum concentration and β-amyloid levels were analyzed by histochemical staining in all brain tissues. Results: Oral administration of nattokinase in the AD rat model increased free-form β-amyloid levels in the CSF and improved aluminum and amyloid plaque accumulation in the brain. Brain μCT images showed enhanced brain volume with fewer constructional changes after treatment with nattokinase. In the behavioral tests, both the escape latency time in the spatial reference test and the time taken to cross the platform area in the spatial probe test improved partially. The Commencement of Execution in Special Fraud Card Switching Theft Cases: ― Analysis of the Supreme Court Decision(Third Petty Bench) of February 14, 2022 (Keishū Vol. 76, No. 2, p. 101)―
近時,特殊詐欺の一種である「すり替え窃盗」が社会問題化している。この問題に関連して,最三決令和 4 年 2 月 14 日刑集 76 巻 2 号 101 頁は,被害者宅到達前の段階における窃盗罪の実行の着手を認める判断を示した。すり替え窃盗は,複数の行為によって構成される犯罪類型であることから,一連の行為論による分析が重要となる。具体的には,「架け子によるうそ」と「被害者宅での窃取行為」を一連の行為として評価したうえで,第 1 行為である電話でのうそに抽象的危険性が認められる場合には,その時点で実行の着手を肯定することが可能となる。しかし,実行の着手と未遂犯の成立とを区別して考察する立場からは,窃盗未遂罪の成立には,法益に対する具体的危険性が発生する被害者宅到着時点まで要すると解される。したがって,すり替え窃盗の事案においては,すり替え担当者が被害者宅へ到着していない段階では,法益に対する具体的危険性が発生していないため,未遂犯の成立を肯定することは困難であるといえよう。Recently, "card switching theft," a type of special fraud scheme, has gained significant attention as a pressing social issue in Japan. In connection with this issue, the Supreme Court Decision (Third Petty Bench) of February 14, 2022 (Keishū Vol. 76, No. 2, p. 101) recognized the commencement of execution for theft before the perpetrator's arrival at the victim's residence. Card switching theft is understood as a crime that is accomplished through multiple acts, necessitating analysis from the perspective of continuous act theory. Specifically, when viewing the "fraudulent communication by the caller" and "the act of theft at the victim's residence" as continuous acts, the commencement of execution can be recognized at the point when abstract danger is present in the first act. However, from the perspective that distinguishes between the commencement of execution and attempted crime, the establishment of attempted theft requires reaching the stage of arrival at the victim's residence, where concrete danger to the protected legal interest arises. Therefore, in the case of the 2022 Supreme Court Decision, since concrete danger to the protected legal interest had not yet materialized when the card switcher had not arrived at the victim's residence, it is difficult to establish the crime of attempted theft at that stage.論
Collagen-derived dipeptide prolyl-hydroxyproline cooperates with Foxg1 to activate the PGC-1α promoter and induce brown adipocytelike phenotype in rosiglitazone-treated C3H10T1/2 cells
Background: The global obesity epidemic is a significant public health issue, often leading to metabolic disorders such as diabetes and cardiovascular diseases. Collagen peptides (CP) and their bioactive component, Prolyl-hydroxyproline (Pro-Hyp), have shown potential in reducing adipocyte size, with unclear mechanisms concerning brown adipocyte differentiation. Methods: We investigated the effects of Pro-Hyp on the differentiation of brown adipocytes in C3H10T1/2 mesenchymal stem cells, focusing on its impact on adipocyte size, gene expression related to brown fat function, and mitochondrial activity. Results: Pro-Hyp treatment decreased adipocyte size and upregulated brown fat-specific genes, including C/EBPα, PGC-1α, and UCP-1. Remarkably, it did not alter PPARγ expression. Pro-Hyp also elevated mitochondrial activity, suggesting enhanced brown adipocyte functionality. A Pro-Hyp responsive element was identified in the PGC-1α gene promoter, which facilitated the binding of the Foxg1 transcription factor, indicating a novel regulatory mechanism. Conclusion: Pro-Hyp promotes brown adipocyte differentiation, potentially offering a therapeutic strategy for obesity management. This study provides a molecular basis for the anti-obesity effects of CP, although further in vivo studies are needed to confirm these findings and to investigate the potential impact on beige adipocyte differentiation.This is an open-access article distributed under the terms of the Creative Commons Attribution License (CC BY).
