    Local hero activities during the corona virus pandemic

    地域キャラクターの一類型であるローカルヒーローは,2019年冬から世界に広がったコロナ禍により大きな影響を被った。従来通りの活動が難しくなる中,新たな活動に挑戦した例もあった。本稿ではコロナ禍において試みられたローカルヒーローの挑戦を大きく 3類型し,具体例を交えつつ概観する。またこうした実践例を参考にしつつ,城西大学経営学部石井ゼミが行った活動について報告する。Local heroes, a type of regional character, have been greatly affected by the coronavirus pandemic that has spread around the world since the winter of 2019. While it became difficult to carry out activities as usual, there were some cases in which participants took on new challenges. This note introduces the challenges of local heroes during the coronavirus pandemic.     In silico model to predict dermal absorption of chemicals in finite dose conditions

    The development of in silico approaches that can estimate the dermal absorption of chemicals exposed in practical conditions is highly anticipated. In the present study, an in silico model to estimate both the dermal absorption rate and dermal permeation profile was developed for the application of chemicals in finite dose conditions. Forty-three chemicals with molecular weights in the range 116–362 and logKo/w in the range 1.1–4.5 were used to develop an in silico model. A gradient boosting tree approach was applied to estimate permeation parameters for diffusion and partition coefficients of the chemicals in skin using physicochemical parameters of the chemicals such as molecular weight, lipophilicity, and the highest and lowest occupied molecular orbitals as the descriptor. In addition, 11 chemicals with different molecular weights and lipophilicities were applied on excised human skin in a finite dose condition, and dermal absorption profiles were obtained. Consideration of donor-solvent evaporation time, saturated concentrations of the chemicals, and donor-solvent coverage area on the skin surface, in addition to estimated skin permeation parameters of the chemicals, showed comparatively good dermal absorption profiles, although some cases of underestimation of dermal absorption were identified. It will be necessary to verify the accuracy of this model through experiments using more chemicals. However, the obtained results suggested that the established model may be valid to estimate the dermal absorption of chemicals in practical conditions

    Survey of the Actual Situation of HRQoL among Patients with Chronic Diseases at Community Pharmacies in Japan and Decision Tree Analysis of Patient Attributes Affecting HRQoL

    Objective: We investigated the health-related quality of life (HRQoL) of patients with chronic diseases in community pharmacies in Japan and examined its relationship to patient attributes, an area that has not been previously studied. Method: We surveyed 1,500 participants with chronic diseases and 1,500 from the general population without chronic diseases (Free-GP) using the EuroQol (5-dimension, 5-level) questionnaire. Patient attributes included the number of drugs used, duration of medication, number of chronic conditions, perceived disease severity, and perceived treatment importance. Logistic regression analyses assessed how these attributes influenced the likelihood of reporting “full health” (no problems on all HRQoL questions) among patients with chronic disease. An exploratory decision tree analysis was performed to identify patient attributes that might decrease HRQoL. Result: The HRQoL score of patients (0.856) was lower than that of Free-GP (0.942). The HRQoL of patients tended to increase with aging. Among patients, the mean HRQoL score was 0.781 for polypharmacy, which decreased with the addition of disease severity perception (even lower for those in their 20s-40). Even in the absence of polypharmacy, the average HRQoL score decreased when patients were in their 20s with multimorbidity. The HRQoL scores of non-polypharmacy patients in their 30s or older were higher than the mean scores of patients with chronic disease. Conclusion: To provide extensive care for patients with reduced HRQoL in community pharmacies, it is necessary to focus on polypharmacy. Reduced HRQoL should be considered in younger patients with multimorbidity, even in the absence of polypharmacy

    Calcium-sensing receptor induces the apoptosis of chondrocytes in cooperation with phosphate transporter

    Excess extracellular inorganic phosphate ions (Pi) and calcium ions (Ca2+) cause apoptosis and subsequent mineralization of chondrocytes. Here, we investigated the mechanism underlying the effect of these minerals. The chondrogenic cell line ATDC5 was treated with 2 mm Pi and/or Ca2+, and apoptosis, mineralization, and intracellular Pi concentrations were determined. Further, Pi- and Ca2+-treated cells were incubated with the Pi transporter (Pit-1) blocker phosphonoformic acid (PFA), the calcium-sensing receptor (CaSR) antagonist NPS-2143, and the CaSR agonist GdCl3. Individual addition of Pi and Ca2+ did not induce apoptosis and mineralization, while combined addition of the minerals induced both. The Pit-1 blocker and the CaSR antagonist completely inhibited the apoptosis induced by combined addition of Pi and Ca2+. Intracellular Pi concentration was remarkably increased by combined addition of Pi and Ca2+ as compared to the findings for individual addition. The Pit-1 blocker and CaSR antagonist completely inhibited the increase in intracellular Pi concentration induced by the combined addition of Pi and Ca2+. The CaSR agonist considerably increased the intracellular Pi concentration. Our results indicate that excess extracellular Ca2+ activates CaSR, which induces the intake of excess extracellular Pi through Pit-1 into ATDC5 cells.
