    Children plan manual actions similarly in structured tasks and in free play

    Visually guided planning is fundamental for manual actions on objects. Multi-step planning—when only the requirements for the initial action are directly visible in the scene—necessitates initial visual guidance to optimize the subsequent actions. We found that 3- to 5-year-old children (n = 23) who exhibited visually guided, multi-step planning in a structured tool-use task (hammering down a peg) also demonstrated visually guided planning during unstructured free play while interlocking Duplo bricks and Squigz pieces. Children who exhibited visually guided planning in the hammering task also spent more time looking at the to-be-grasped free-play object and at their construction during reach and transport compared with children who did not demonstrate multi-step planning in the hammering task. Moreover, visually guided planning in the Duplo and Squigz tasks was positively correlated, indicating that planning generalizes across contexts. Findings show that visually guided planning in young children generalizes across different manual actions on objects, including structured tool use and unstructured free play

    Penetrant PKCß mutation in ATLL displays a mixed gain-of-function

    Mutations in the T-cell receptor signalling pathway have been identified in patients with adult T-cell leukaemia/lymphoma (ATLL) and one of the most frequently observed targets of these mutations is protein kinase C beta (PKCß). Here we have characterised the most frequent mutation in PKCß (D427N) addressing the issue of gain/loss of function, neomorphic change, assessing the impact of mutation in vivo, in cells, biochemically and structurally. It is concluded that this mutation is a gain-of-function, activating mutation that confers an altered substrate specificity on this protein kinase. In a constitutive knock-in mouse model this activated allele induces splenomegaly associated with extramedullary haematopoiesis. Pharmacologically, the D427N mutant protein displays poor sensitivity to established PKCß inhibitors, necessitating development of bespoke therapeutics for any ATLL intervention through this target. Such efforts could be guided by the availability the D427N mutant-ruboxistaurin structure presented here

    An investigation into network theory and how the SDP logic model facilitates compliance and the safe handling of information

    Social media communications technology (SMCT) helps managers to extend their networks and share information. Information security is essential as regards trust and compliance. This study explains how a secure digital platform (SDP), as an organisational intervention, instils confidence in managers to utilise SMCT and engage in information sharing that facilitates resource utilisation. A theoretical SDP framework was developed by utilising network theory in conjunction with the logic model approach. An online survey (n = 207) was analysed using SEM, AMOS. The results show that a designed-in governance mechanism (DGM) plays a pivotal role in ensuring compliance (DC); at the same time, a DGM positively influences the intention to use an SDP. Also, the subjective norm (SN) positively influences the intention to use an SDP. The deployment of an SDP positively enhances business capability through the use of SMCT. This is the first study that empirically tested how governance establishes the policies and frameworks that ensure compliance, which results in strengthening an SDP and enhances business capability. The finding of this study helps managers and platform developers understand the importance of establishing governance policy and its implementation in digital network platforms to influence/facilitate safe information sharing to enhance business capability

Pour faire face aux défis mondiaux actuels, nous avons besoin de former des biologistes végétaux et d'alerter la population à l'importance des plantes. Ce manifeste détaille et justifie cinq priorités stratégiques pour renforcer l'enseignement en biologie végétale. Les acteurs de la biologie végétale, et au delà, concernés par ce plan vont des éducateurs sur le terrain aux responsables institutionnels, des professionnels des secteurs privé et caritatif aux entrepreneurs et donateurs, des particuliers à leurs représentants législatifs. Il faudra des actions concrètes de tous les acteurs pour renforcer l'enseignement en biologie végétale, ce qui, au final, nous bénéficiera à tous

    Arp2/3-mediated bidirectional actin assembly by SPIN90 dimers

    Branched actin networks nucleated by the Arp2/3 complex play critical roles in various cellular processes, from cell migration to intracellular transport. However, when activated by WISH/DIP/SPIN90 family proteins, Arp2/3 nucleates linear actin filaments. We found that human SPIN90 is a dimer that can nucleate bidirectional actin filaments. To understand the basis for this, we determined a 3 Å resolution structure of human SPIN90-Arp2/3 complex nucleating actin filaments. Our structure shows that SPIN90 dimerises via a 3-helix bundle and interacts with two Arp2/3 complexes. Each SPIN90 molecule binds both Arp2/3 complexes to promote their activation. Our analysis demonstrates that single filament nucleation by Arp2/3 is mechanistically more like branch formation than previously appreciated. The dimerisation domain in SPIN90 orthologues is conserved in metazoans, suggesting that this mode of bidirectional nucleation is a common strategy to generate anti-parallel actin filaments

    Book synopsis: This unique collection of essays and images explores a series of objects in the Royal Collection as a means of assessing the interrelated histories of the British royal family and the Shakespearean afterlife across four centuries. Between the beginning of the eighteenth century and the late twentieth, Shakespeare became entrenched as the English national poet. Over the same period, the monarchy sought repeatedly to demonstrate its centrality to British nationhood. By way of close analysis of a selection of objects from the Royal Collection, this volume argues that the royal family and the Shakespearean afterlife were far more closely interwoven than has previously been realized. The chapters map the mutual development over time of the relationship between members of the British royal family and Shakespeare, demonstrating the extent to which each has gained sustained value from association with the other and showing how members of the royal family have individually and collectively constructed their identities and performed their roles by way of Shakespearean models. Each chapter is inspired by an object in (or formerly in) the Royal Collection and explores two interconnected questions: what has Shakespeare done for the royal family, and what has the royal family done for Shakespeare? The chapters range across the fields of art, theatre history, literary criticism, literary history, court studies and cultural history, showing how the shared history of Shakespeare and the royal family has been cultivated across media and across disciplines

    The mental health impact of repeated COVID-19 enforced lockdowns in England: evidence from the UK Household Longitudinal Study

    Background: Research shows initial COVID-19 lockdowns increased population mental distress. Yet, the mental health impact of repeated lockdowns in England remains unknown. Aims: To: (a) explore changes in population mental health symptoms over the COVID-19 pandemic period (March 2020 to March 2021) in England, comparing this with trends from a decade before (2009–2019) as well as after (2021–2023); (b) compare the mental health impact of each of the three lockdowns in England with periods of eased restrictions, determining who was most affected; (c) examine the impact of demographics and distinct time periods on the prevalence of mental health symptoms. Method: A secondary analysis of a national longitudinal cohort study, utilising data from Waves 1–13 of the UK Household Longitudinal Study and from Waves 1–9 of the COVID-19 Survey. Mental health was assessed using the 12-item General Health Questionnaire. Student t-tests and logistical regressions were conducted. Results: There was a significant increase in the prevalence of self-reported symptoms of mental health during England's pandemic period, encompassing three lockdowns, compared with the average of rates from 10 years before. Rates of reported mental health symptoms were not significantly different across each lockdown, but were significantly higher than pre-pandemic rates, declining with eased restrictions. Rates from the end of lockdown to May 2023 revealed elevated mental health symptoms compared with pre-pandemic. Elevated symptoms were observed for women, people homeworking, those with health conditions, individuals aged 30–45 years and those experiencing loneliness. Conclusion: Repeated lockdowns in England had a substantial impact on mental health, indicating requirements for ongoing mental health support
