Influence de l'environnement prénatal sur le comportement du chien
L'environnement prénatal influence les comportements des animaux et des humains dans le jeune âge avec des répercussions jusqu'à l'âge adulte. Les études sur le sujet se sont essentiellement intéressées aux effets du stress prénatal chez les rongeurs, les carnivores, les animaux de ferme et les humains. Cette étude a suivi 66 chiots issus de cinq chiennes de race Beagle ayant chacune mené trois gestations. Chacune des trois gestations s'est déroulée dans un environnement différent : Base (= groupe contrôle), Enrichi et Appauvri. Le but de cette étude est de mettre en évidence d'éventuelles différences entre ces trois groupes de chiots. Après la naissance, tous les chiots ont vécu dans le même environnement. Les chiots ont été testés jusqu'à l'âge de huit semaines selon quatre critères : l'émotivité, l'attention, l'aptitude à l'apprentissage et la familiarisation à l'homme. Ils ont ensuite été évalués à l'âge adulte au moyen d'un questionnaire soumis au propriétaire visant à révéler d'éventuels problèmes de comportement et d'un test d'aptitude à l'apprentissage. Les chiots du groupe Appauvri montrent, par une réactivité plus grande face à la nouveauté et des vocalisations plus importantes, une émotivité plus forte que les chiots de deux autres groupes dès le plus jeune âge et jusqu'à l'âge de 7 semaines. A 8 semaines, ils sont en outre moins attentifs lors du test d'Attention mais plus performants lors des tests d'Aptitude à l'Apprentissage. Aucune différence significative n'a pu être mise en évidence entre les groupes lors du test de Familiarisation à l'homme. Les chiots ont ensuite été adoptés par des familles auxquelles ont été prodigués des conseils d'éducation. Etude de la réponse de Brucella melitensis 16M aux molécules signal du Quorum Sensing
Quorum Sensing is a bacterial communication system which allows the coordinated gene expression within a population regarding its density and/or environmental properties (diffusion, flow…). In Gram negative bacteria, Quorum Sensing is based on the synthesis of N-acyl-L-homoserine lactones (AHLs) and the detection of these signal molecules by LuxR-type transcriptional regulators. When AHLs concentration reached a threshold (quorum), these molecules bind to the LuxR regulator and lead to conformational changes driving the modification of the regulator activity. Numerous phenomenons are regulated by QS (biofilm formation, fruiting bodies, antibiotics synthesis…), so a competitor able to disturb/inhibit this system could have an advantage on bacteria using Quorum Sensing. The production of inhibitors or enzymes able to degrade signal molecules (AHL-lactonase and acylase) are part of a mechanism called Quorum Quenching.
Brucella melitensis, a Gram negative pathogenic bacterium, possesses a Quorum Sensing system constituted by two LuxR-type regulators (called BabR and VjbR) and an AHL identified as a dodecanoyl-homoserine lactone (C12-HSL). While BabR function is unknown, VjbR is involved in the virulence of this pathogen by regulating the expression of two major virulence factors: a flagellum and a type four secretion system. AHLs have a repressor effect on these two structures, and this effect is mediated by VjbR. In order to investigate the role of Quorum Sensing in B. melitensis, global proteomic and transcriptomic studies were performed on wild type, ∆babR and ∆vjbR mutant strains. These studies demonstrate that Quorum Sensing regulates 5% of Brucella genome in the condition tested, confirming that Quorum Sensing is a global regulation system in B. melitensis. Numerous targets of the two regulators are involved in virulence, stress response, metabolism, cell wall structure… suggesting an important role of Quorum Sensing in the achievement of the infectious cycle of B. melitensis.
The study of a putative AHL-acylase produced by Brucella demonstrates that this enzyme is an extracellular enzyme able to inactivate AHL signal molecules. Mice infected with an AHL-acylase mutant strain develop a bigger splenomegaly and have a higher bacterial charge in the spleen compared to the wild type strain. These data suggest that Brucella AHL-acylase have also an important role in the infectious cycle of B. melitensis.Le Quorum Sensing est un système de communication permettant à une population bactérienne de coordonner l’expression de gènes cibles en fonction de sa densité et/ou suivant les propriétés du milieu (diffusion, flux, …). Au sein des bactéries Gram négatives, le Quorum Sensing est basé sur la synthèse de N-acyl-L-homosérine lactones (AHLs) et sur la détection de ces molécules signal par des régulateurs transcriptionnels de type LuxR. Lorsque la concentration en AHLs atteint un certain seuil (quorum), ces molécules se fixent au régulateur de type LuxR, entrainant un changement conformationnel et une modification de l’activité du régulateur. De nombreux phénomènes étant régulés par le Quorum Sensing (formation de biofilm, de corps fructifères, production d’antibiotiques, …), un compétiteur capable de perturber/inhiber ce système aurait un avantage certain sur les espèces utilisant le Quorum Sensing. La production d’inhibiteurs ou d’enzymes capables de dégrader les molécules signal (AHL-lactonase et acylase) font partie d’un mécanisme nommé Quorum Quenching.
Brucella melitensis, bactérie pathogène intracellulaire Gram négative, possède un système de Quorum Sensing. Celui-ci repose sur deux régulateurs de type LuxR, nommés BabR et VjbR, et sur une AHL identifiée comme étant une dodécanoyl-homosérine lactone (C12-HSL). Alors que le rôle de BabR est encore inconnu, VjbR est impliqué dans la virulence de B. melitensis via l’activation de l’expression de deux importants facteurs de virulence : un système flagellaire et un système de sécrétion de type IV. Les AHLs ont quant à elles un effet répresseur sur ces deux structures, et cet effet est médié par VjbR. Afin d’aborder plus globalement le rôle du Quorum Sensing chez B. melitensis, des études protéomique et transcriptomique ont été réalisées en comparant la souche sauvage et les mutants ∆babR et ∆vjbR. Nous avons ainsi pu mettre en évidence que le Quorum Sensing permet de réguler 5% du génome de Brucella dans les conditions testées, confirmant que le Quorum Sensing est un système de régulation global chez B. melitensis. De nombreuses cibles des régulateurs sont impliquées dans la virulence mais également dans les réponses aux stress, dans le métabolisme, la structure membranaire, … suggérant un rôle important du Quorum Sensing dans l’accomplissement du cycle infectieux de B. melitensis.
Les analyses effectuées sur une probable AHL-acylase produite par Brucella ont révélé que cette enzyme était capable d’inactiver les AHLs et qu’elle agissait extracellulairement. Des souris infectées par une souche délétée pour cette AHL-acylase présentent une splénomégalie plus importante ainsi qu’une charge bactérienne plus élevée dans la rate comparée aux souris infectées avec la souche sauvage. Ces données nous laissent penser que l’AHL-acylase joue également un rôle important durant le cycle infectieux de B. melitensis.(DOCSC03) -- FUNDP, 200
Selective Growth of CdTe by Molecular Beam Epitaxy on CdTe(211)B Microseeds and Si(100) Nanoseeds Patterned on SiO2
This Ph. D. thesis is dedicated to the study of selective growth by molecular beam epitaxy of CdTe on CdTe(211)B and Si(100) islands patterned on SiO2.
These islands have a micro- and nanometer scale size and act as seeds for the growth of CdTe. These structures are patterned by optical and interferometric lithography.
The selective process is explained by a difference in the physisorption energy of the adsorbed atoms of cadmium and telluride depending on the substrate (CdTe, Si or SiO2).Cette thèse de doctorat étudie la possibilité de faire croître par épitaxie par jets moléculaires du CdTe sélectivement sur des îlots de CdTe(211)B ou de Si(100) qui ont été lithographiés sur un substrat de SiO2.
La création des micro- et nanostructures servant de semence d’amorçage à la croissance sélective du CdTe a été réalisée par lithographie optique et interférométrique.

A new Human Familiarisation and Training Programme for military dog handler teams- Improving Security, Efficiency and Welfare at the Belgian Defence
This project on human-dog interaction was developed after bite incidents occurred involving Military Working Dogs (MWDs).
Preliminary assessments (accident reports, questionnaire to the handlers, training programmes) led to the hypothesis that those incidents could find their origin in efficiency, human security and dogs' welfare problems. As a result, a new Human Familiarisation and Training Programme (HFTP) was set up to improve the training methods used by the handlers, increasing handlers' motivation and improving the relationship between the handler and the dog.
A standardised evaluation procedure carried out amongst a Control Group (CG) showed:
(1) efficiency problems through the assessment of obedience and protection work exercises (n=33): only 20% of the dogs released their bite after the first command, only 19% of the dogs carried out the Stand-Off correctly;
(2) security problems through the assessment of a validated aggression test (n=31): in 69.35% of the cases, dogs reacted with fear-related aggression during the aggression test;
(3) no major welfare problems through the behavioural and physiological measurements that were implemented in this project (n=27) as the CG dogs were still able to adapt to environmental challenges (return to baseline of the cortisol levels, a higher posture during the second than during the first challenge). It could not be proven that the welfare of these dogs was hampered, but dog's low posture during obedience exercises indicated is was under threat.
A new HFTP, based on positive and more frequent dog handler (DH) interactions and training, was subsequently set up in order to tackle the existing deficiencies.
An EG was submitted to the HFTP and then underwent the same assessment as the CG. Results revealed:
(1) efficiency increase through a better performance of EG teams during obedience (increase of 34.04%) and protection work exercises (increase of 69.98%) (CG: n=33, EG: n=33)
(2) security enhancement as demonstrated by a decrease of aggressiveness (decrease of 43.78%) during the aggression test (CG: n=31, EG: n=36),
(3) welfare improvements were shown by a significant increase of the EG dogs' posture during the aggression test and during obedience. Nevertheless, EG dogs, used to intense human contact, reacted worse towards kennelling than CG dogs, accustomed to little contact (CG: n=27, EG: n=31).
This project could offer the groundwork of forthcoming research that may offer significant improvements in the way military dogs are raised and/or procured, trained, and employed - not to mention their welfare and the DH relationship(DOCMED01)--FUNDP, 200
Identification of the essential Brucella melitensis Omp2b porin as a modulator of cell death using a genome‐wide functional yeast‐based screening
This thesis is in line with the global context of the identification and characterization of
bacterial effectors responsible for the hijacking of the host cell, and particularly with the
identification of anti-apoptotic effectors of Brucella spp. The introduction of this work is
divided in three chapters, corresponding to the three "themes" that have been approached
during this thesis:
The first chapter of the introduction is centered on the modulation of one cellular
process, apoptosis, by bacteria. The underlying mechanisms leading to apoptosis and the
involvement of this particular cell death during infection will be approached. Our points of
interest considering the modulation of apoptosis by our model pathogenic bacteria, Brucella
spp., will be described.
The second chapter of the introduction is related to yeast-based strategies aiming at
the identification of bacterial effectors, including the strategy that we used to highlight new
bacterial modulators of cell death. This chapter is thus justified by the aim of this thesis,
which was to apply an original strategy to identify candidate anti-apoptotic proteins of B.
melitensis, using the yeast S. cerevisiae as a tool for a genome-wide screening method.
This screen shed light on two candidate proteins, and the one on which we mainly
focused our attention is a porin. Thus, for reasons linked to the understanding of the results
that will follow, the third chapter of our introduction is an annex dedicated to a general
overview of porins, their implication regarding host-pathogen interactions and a summary of
data that have already been published about Brucella porins.
The results obtained in the frame of this thesis are mainly presented in a format that is
convenient for their submission as a manuscript for a scientific journal. This manuscript,
slightly modified in order to fulfill the journal requirements, is currently under submission in
FEBS Letters. An additional part, presented in the results section after the paper manuscript,
contains a description of the screening strategy, more detailed and illustrated than the
corresponding part included in the paper manuscript, as well as supplementary information
and data. The last section of this thesis is a discussion of the results, including suggestions for future

Power, Conflicts, and Subordination
Researchers in the economics of development have been struggling for the past fifty years to find theoretical explanations for the economic backwardness characteristic of an important number of territories around the world. Several countries seem trapped in vicious circles whereby poverty self-sustains under-development. And while the channels linking low wealth to poor economic performances are multiple and highly complex, a recurrent feature of under-developed polities is their weak institutional setting. In the absence of strong and credible institutions, one needs to depart from the neoclassical paradigm to understand how some countries are entrapped in high poverty. An assumption that is both critical for the theoretical reasoning, and dear to economists, gets violated: property rights are not enforced. The consequences of lifting this hypothesis are dramatic since the traditional institution to operate economic transactions, the market, can no more be fully relied upon. Indeed, instead of being secured by a Leviathan or some other third party, transactions and property claims need to be backed by power. As a consequence, rather than integrally allocating scarce resources to production, agents need to divert some production factors to accumulating power.
The aim of this thesis is to improve our understanding of the allocation of scarce resources under weak institutions. We distinguish two distinct ways to acquire power. Depending on the contexts, an agent can either produce such power himself, or else he may purchase the services of other agents serving his cause. The two first chapters of this manuscript are devoted to the latter situations where an agent (patron) offers a material good to another agent (client) in exchange for power, status, or support. In the last two chapters the purchase of power is ruled out by assumption, but the agents are allowed to devote resources in producing power.
More precisely, the first chapter aims at a better understanding of the conditions under which unequal rank or power positions may get permanently established through asymmetric gift exchange when a gift brings pride to the donor and shame to the subordinated recipient. In this chapter only two players are assumed to interact, the patron and the client. The central result obtained is that an asymmetric gift exchange equilibrium can occur only if the importance attached to social shame by a recipient is smaller than that attached to social esteem by a donor. Moreover, an income transfer is more likely to be traded against social esteem, status, or power when the weight put on these attributes by the donor or patron is higher. In that chapter, co-written with Jean-Philippe Platteau, we also show that the recipient's productivity may take on a rather wide range of values in the domain of feasibility of asymmetric gift exchange, and that, contrary to a commonly prevailing view, it is even possible that his productivity would be identical to that of the donor. Finally, the conditions are spelt out under which the recipient's effort is more likely to be reduced upon entering into asymmetric gift exchange relationships.
In the second chapter, I develop a setting where such power enables the patron to maintain his position and to extract rents. In that setting, instead of directly deriving utility from power and status, autocrats barter political and economic concessions for support to remain in power and extract rents. Contrary to the most of the existing literature, instead of viewing the favors' beneficiaries, i.e. the elites, as an exogenous entity, I allow the ruler to decide whom to coopt provided the subjects are heterogeneous in the potential support - their strength - they could bring to the regime. While the ruler can select the elites on the basis of their personal characteristics, an alternative strategy consists in introducing some uncertainty in the cooptation process. The latter strategy allows the ruler to reduce the clients' cooptation price since in the event of a revolution the likelihood of being included in the future elites is lower. I show that weak rulers are more likely to coopt the society's strongest individuals, while powerful ones diversify the composition of their clientele. Moreover, when agents value more future discounted outcomes, and when power is more dispersed across subjects, the ruler is more likely to randomly coopt subjects.
In the third chapter, co-authored with Giacomo De Luca, patron-client relationships are assumed away. Agents enter in contests for scarce resources by producing power rather than acquiring it through the purchase of support and subordination. Landless subjects intent to obtain land by violent means, while landlords invest resources in defending their property. Our task is to track the effect of land inequality on conflict intensity. A fundamental distinction with the existing literature lies in the nature of inequality under consideration. We investigate how land inequality extit{across landlords only} influences the intensity of the fight against a rebel group constituted by landless individuals. We show that conflict intensity is non-monotonic in land inequality. In particular, the most severe conflicts occur for intermediate land inequality levels. Moreover, a Pareto improving transfer of land from the smaller to the larger landlord may exist.
The last chapter is perhaps the most abstract piece of the present thesis, making it the most general part of the manuscript as well. In that paper we explore, together with Giacomo De Luca, the existence of deterrence equilibria in a general equilibrium model of guns (power) and butter (goods) production. If fighting entails sufficiently low destruction, war is the unique equilibrium of the game. If, however, conflict generates sufficiently large damages, only mixed strategy equilibria survive, in which players randomize over their deterrence and war strategies. War, therefore, always occurs with positive probability for any positive investment in weapons.(DOCSESG00 ) -- FUNDP, 200
Caractérisation agro-morphologique et moléculaire d’une collection de landraces péruviennes de pigeonpea (Cajanus cajan L. Millsp.) pour l’analyse de sa diversité
Biodiversity used within the context of plant enhancement presupposes an analysis of the locally existing genotypes adapted to particular environments. Within the scope of a pigeon pea enhancement project, we carried out the exploration and collection of pigeon pea landraces from Lambayeque, department located at the north of Peru. Then, we proceeded to the agro-morphological characterization of the maternal families (derived from these landraces) based on descriptors. At the same time, we undertook the molecular characterization of the collection, as well as in some Indian and African accessions, by using microsatellites markers developed by us. We also used microsatellites provided by the ICRISAT. The molecular characterization is an important stage, which requires low-cost precision instruments, efficient and easy to use. One of them are the microsatellites, codominant markers. Thanks to these seven microsatellite markers, we made evident the diversity inside the collection and the proximity between the Peruvian and the African groups. Using molecular techniques such as the microsatellites is important as they are supplementary to that of the descriptors. Besides, they play an essential role in the assessment of promising crops in countries like Peru.L’utilisation de la biodiversité dans le cadre de l’amélioration végétale présuppose une analyse des génotypes existants localement et adaptés à des environnements particuliers. Dans le cadre d’un projet d’amélioration du pois d’Angola ou pigeonpea, nous avons réalisé la prospection et la collecte de landraces du pigeonpea à Lambayeque, département au nord du Pérou. Par la suite, nous avons procédé à la caractérisation agro-morphologique des familles maternelles (issues de ces landraces) sur base des descripteurs. Parallèlement, nous avons effectué la caractérisation moléculaire de la collection ainsi que celles de certaines accessions indiennes et africaines à l’aide de marqueurs microsatellites que nous avons développés. Nous avons également utilisé des microsatellites fournis par l’ICRISAT. La caractérisation moléculaire est une étape importante, elle nécessite des outils précis, au coût peu élevé, performants et facilement utilisables. Un de ceux-ci : les microsatellites, marqueurs co-dominants. Grâce à ces sept marqueurs microsatellites, nous avons pu mettre en évidence la diversité au sein de la collection ainsi que le rapprochement entre le pool péruvien et africain. L’utilisation des techniques moléculaires comme les microsatellites s’avère importante car elle est complémentaire à celle des descripteurs. D’ailleurs son rôle est essentiel dans la valorisation des cultures d’avenir dans des pays comme le Pérou.(DOCSC03 ) -- FUNDP, 200
Techniques de visualisation d'information pour l'analyse de situations coopératives
La thèse présente un caractère multidisciplinaire. Elle fait appel à trois grandes disciplines scientifiques : l'informatique, l'analyse de la coopération et la visualisation d'information (c.-à-d. la représention graphique de données abstraites au moyen de logiciels).
Afin de disposer d'un cadre de référence pour nos réflexions, nous avons conçu un modèle générique de la coopération. Puis, nous avons étudié en détail les données qu'il était utile de visualiser pour aider à analyser une situation de coopération. Nos travaux nous ont amenés à nous intéresser en particulier au concept de conscience de groupe. Dans ce contexte, nous avons proposé une approche conceptuelle qui différencie les informations à visualiser selon leur anonymité et leur granularité. Sur la base de cette classification, nous avons identifié deux cas pertinents en pratique : la conscience de groupe individuelle (Workspace Individual Awareness) et la conscience de groupe globale(Workspace Group Awareness).
L'étude de cette dernière nous a conduit à aborder la question des métriques des activités coopératives. Comment résumer dans un indicateur global la complexité des interactions sous-tendant des activités coopératives ? Alors que la plupart des travaux antérieurs prenaient en compte la quantité d'interactions entre les personnes et éventuellement les ressources pour évaluer le niveau de coopération, nous avons innové en proposant de nouveaux indices qui intègrent, dans une certaine mesure, la qualité de celles-ci en termes de motivation à coopérer.
Assez logiquement, l'étape suivante a visé à représenter graphiquement les indices précédemment définis.
Notre étude de l'état de l'art sur les techniques de visualisation de données associées à un contexte coopératif a mis en évidence le besoin d'une meilleure structuration de ce domaine au niveau conceptuel. Nous avons donc entrepris l'élaboration d'une taxonomie que nous avons nommée IVF. Elle identifie un ensemble de propriétés permettant de décrire une situation coopérative, les aspects de celle-ci qu'il peut être utile de visualiser et les techniques utilisées pour représenter graphiquement ces derniers.
Constatant que dans un nombre significatif de cas, les données que nous cherchions à visualiser se présentaient sous la forme de structures hiérarchiques, nous avons ensuite entrepris l'étude des travaux antérieurs en matière de visualisation de données de ce type.
L'étude de la théorie 'Gestalt' issue de la psychologie cognitive nous a inspiré et nous avons voulu intégrer ses lois dans notre démarche de conception d'une nouvelle technique de visualisation d'information. Dans un premier temps, nous avons confronté la technique des 'treemaps', connue pour son approche originale de représentation de hiérarchies, avec la théorie 'Gestalt'. Cette confrontation, assez inhabituelle, nous a incité à proposer une nouvelle approche, nommée 'ellimaps', qui nous semblait plus conforme aux principes de la théorie 'Gestalt'. Cette nouvelle technique de visualisation fit l'objet diverses évolutions afin de résoudre ou d'atténuer ses faiblesses.
Au cours du processus de conception et de développement du prototype logiciel qui implémentait le concept d'ellimaps, plusieurs évaluations formatives furent réalisées afin de confronter nos résultats partiels avec des utilisateurs potentiels. Nous avons également décidé de comparer formellement l'approche ellimaps avec la technique des 'treemaps' dont elle est conceptuellement issue. Nous avons étudié en détail la problématique de l'évaluation de logiciels et plus spécifiquement celle d'interfaces homme-machine. Sur cette base, nous avons identifié les biais potentiels dans la conception et la mise en oeuvre d'une telle étude comparative. Pour répondre à certains d'entre eux, nous avons conçu et développé un environnement logiciel spécifique (appelé FESTIV) qui nous a permis de contrôler précisément les variables indépendantes de notre évaluation et de collecter des mesures les plus précises et significatives possibles.
Nous avons ensuite confronté notre intuition initiale et les résultats exérimentaux pour déterminer notamment si, dans les conditions expérimentales de notre étude, les ellimaps améliorent la perception de la structure des données par rapport aux treemaps.(DOCSC06)--FUNDP, 200
La subsidiarite procedurale dans le système africain de protection des droits de l'homme
L'analyse du système africain des droits de l'homme dans sa dimension procédurale constitue une matière en friche, les quelques chercheurs qui s'y sont intéressés n'ayant abordé que la dimension substantielle et historique de la Charte africaine. Rares sont les travaux réalisés sur l'étude de la jurisprudence de la Commission africaine. D'une manière générale, l'étude de la jurisprudence d'un organe de protection des droits de l'homme, à l'instar de la Commission africaine, se fait de manière comparée. On souligne les points de convergence et de divergence avec les autres mécanismes internationaux, voire régionaux ou universel. Cette comparaison présente un intérêt indéniable dans la mesure où elle contribue à enrichir chacun des divers systèmes des méthodes et des raisonnements appliqués dans l'autre. En prenant appui sur cette démarche comparative, notre étude permettra :
- de déterminer d'abord la mise en oeuvre de la subsidiarité procédurale dans le système africain des droits de l'homme ;
- de mesurer ensuite le rapport entre cette mise en oeuvre et l'effectivité de la Charte africaine des droits de l'homme et des peuples ;
- Elle permettra enfin de proposer des solutions de nature à concilier la subsidiarité procédurale et l'effectivité des droits de l'homme inscrits dans cette Charte.
Les développements relatifs à notre étude permettront de mettre en lumière certaines particularités du mécanisme de contrôle institué par la Charte africaine des droits de l'homme notamment au regard de la nature des recours admis, la composition, la procédure et l'exécution des décisions de l'organe de recours.(DOCDROI00)--FUNDP, 200
