Caractérisation du système à deux composants PrlS et PrlR de Brucella melitensis.
The PrlS/PrlR two component signal transduction system (TCS) is highly conserved in sequence and genomic organization in Brucella species, but its function remains completely unknown. TCS regulate genetic expression in response to external or internal environmental signals. In this work PrlS/PrlR mutants were characterized with results suggesting that wt B. melitensis produces aggregates at high osmolarity, contrary to the mutants of the bacterial pathogen Brucella melitensis. Interestingly it could be observed that the matrix of this aggregates contain EPS and that PrlR regulates positively the synthesis and production of EPS. We further investigated the role of prlR on membrane structures such as the flagellum and outer membrane proteins, by studying the regulation of the pfliF promoter in the mutants. Our results suggest that prlR controls both the flagellar system and the regulation of OMPs. B. melitensis mutated for prlR is less virulent compared to the wt suggesting that PrlR may play an important role in virulence. Preliminary transcriptomic comparative analysis of the mRNA profiles of mutant and wild type prlR strains suggests that PrlR regulates approximately 6.41 % of the B. melitensis genome.Le système de transduction de signal à deux composants (SDC) PrlS/PrlR est très conservé en séquences et organisation génomique dans les espéces de Brucella, mais sa fonction est complètement inconnue. Les SDC régulent l‟expression génique en réponse à des signaux environnementaux externes ou internes. Lors de ce travail, nous avons caractérisé des mutants du SDC PrlS/PrlR du pathogène bactérien Brucella melitensis, démontrant que ceux-ci perdent la capacité de la souche sauvage à produire des agrégats en conditions de haute osmolarité. Nous avons observé que la matrice des agrégats produits par B. melitensis contient de l‟exopolysaccharide (EPS) et que PrlR régule positivement la production et la synthèse d‟EPS. Par après, nous avons investigué le rôle que PrlR pourrait exercer sur les structures de surface comme le flagelle et les protéines de membrane externe et ce en étudiant la régulation du promoteur pflF chez les mutants de PrlR. Nos résultats suggèrent que prlR pourrait jouer un rôle dans la régulation de système flagellaire et de certains OMPs. Nous avons également montré qu‟un mutant de prlR de B. melitensis est moins virulent que la souche sauvage, ce qui suggère que PrlR puisse jouer un rôle important dans la virulence. Pour terminer, l‟analyse transcriptomique préliminaire comparative des profils d‟ARNm de la sauvage et d‟un mutant prlR suggère que PrlR régule approximativement 6.41% du génome de B. melitensis.(DOCSC03) -- FUNDP, 201
Etude des kinases activatrices de Cdc2 chez la levure Schizosaccharomyces pombe
Cyclin-dependent kinases (CDK) belong to a group of kinases involved in both cell cycle control and transcription regulation. To be fully active, CDKs require phosphorylation by a Cdk-activating kinase (CAK). The fission yeast Schizosaccharomyces pombe possesses the two types of CAK : trimeric Mcs6-Mcs2-Pmh1 complex and monomeric Csk1 protein. Mcs6 and Csk1 can both phosphorylate and activate Cdc2, the main cell cycle regulator, in vitro. However, strong genetic data indicate that only Mcs6 activates Cdc2 in vivo. Analysis of an analog-sensitive mutant of Mcs6 show that the sole inactivation of Mcs6 is necessary and sufficient to abolish Cdc2 phosphorylation in vivo. Our data also indicate that Csk1, contrary to the related kinase Cak1, is unable to precipitate and phosphorylate Cdc2 from a fission yeast lysate. Here we establish that Mcs6 is the genuine CAK in vivo of Cdc2 in S. pombe.Les kinases dépendantes des cyclines (CDK) appartiennent à un groupe de protéines kinases impliquées dans le contrôle du cycle cellulaire et dans la régulation de la transcription. Pour être actives, les CDK doivent être phosphorylées par une kinase activatrice nommée CAK (Cdk-activating kinase). La levure de fission Schizosaccharomyces pombe possède deux types de CAK : le complexe trimérique Mcs6-Mcs2-Pmh1 et la protéine monomérique Csk1. Mcs6 et Csk1 sont toutes les deux capables de phosphoryler et d’activer Cdc2, la CDK régulant le cycle cellulaire, in vitro mais des données génétiques fortes indiquent que seule Mcs6 active Cdc2 in vivo. L’utilisation d’un mutant de Mcs6 sensible à un analogue d’ATP montre que la seule inactivation de Mcs6 est nécessaire et suffisante pour abolir la phopshorylation de Cdc2 in vivo. Nos données indiquent également que Csk1, contrairement à la kinase apparentée Cak1, est incapable d’immunoprécipiter et de phosphoryler Cdc2 à partir d’un lysat de levure de fission. Study of the 3T3-L1 adipocyte responses to a mild mitochondrial uncoupling : analyses of the modifications affecting the mitochondrial population and the release of adipokines.
As obesity results, at least partly, from a chronically positive energy balance,
increasing energy expenditure through mitochondrial uncoupling in adipocytes has been
considered as an attractive approach to fight obesity. Currently, mitochondrial uncoupling in
adipocytes is known to trigger a reduction in triglyceride (TG) content, a phenotype also
found in adipocytes incubated with TNFα (tumor-necrosis factor α), another condition known
to induce the “dedifferentiation” of adipocytes. However, the impact of a mitochondrial
uncoupling on the abundance/composition of mitochondria and its connection with
triglyceride content in adipocytes is largely unknown. In addition, while mitochondrial
activity has been linked to adipokine expression, the effects of mitochondrial uncoupling on
adipokine expression and secretion has been poorly investigated. However, this is a very
important question to address as adipokines (referring to hormones, cytokines and
chemokines released by adipocytes acting on peripheral tissues) are thought to be the
molecular links between obesity and associated diseases such as insulin resistance, type 2
diabetes, cardiovascular diseases and inflammation, brought altogether in the metabolic
syndrome.
In the first part of this study, the effects of a mild mitochondrial uncoupling triggered
by carbonyl cyanide (p-trifluoromethoxy) phenylhydrazone (FCCP) on the mitochondrial
population of 3T3-L1 adipocytes were investigated using both quantitative and qualitative
(biochemical, bioenergetics, genomics, proteomics) approaches. We found that chronic but
mild mitochondrial uncoupling does not stimulate mitochondrial biogenesis in adipocytes, but
induces a subtle adaptive cell response characterized by modifications of mitochondrial
protein content. Subproteomic analysis revealed that the abundance of pyruvate carboxylase
was significantly reduced in mitochondria of TNFα- and FCCP-treated adipocytes. A
functional study showed that overexpression of this key enzyme in lipid metabolism is able to
prevent the TG content reduction in adipocytes exposed to mitochondrial uncoupling or
TNFα. These results provide a new mechanism by which the effects of mitochondrial
uncoupling might limit triglyceride accumulation in adipocytes.
We also found that a mild and chronic mitochondrial uncoupling down-regulates the
expression and the secretion of leptin, Acrp30, and resistin and modifies the abundance of
active PAI-1 in the supernatant of 3T3-L1 adipocytes. In addition, adipocytes treated with
TNFα (a pro-inflammatory cytokine abundantly found in the adipose tissue of obese
individuals) display similar modifications. However and unexpectedly, we observed that
conditioned media from FCCP-treated adipocytes impair insulin-stimulated glucose uptake in
differentiated C2C12 cells while conditioned media from TNFα-treated adipocytes do not
impair insulin-stimulated glucose uptake in these cells. Additionally, we demonstrated, using
a DNA array (“DualChip inflammation” allowing gene expression analysis for 253
inflammatory genes), that mitochondrial uncoupling does not trigger a molecular signature
supporting the initiation of an inflammatory response by adipocytes. Indeed, the expression of
many important pro-inflammatory effectors was not affected in adipocytes exposed to FCCP.
All together, these sets of data strongly suggest that insulin resistance is acquired by myocytes
exposed to conditioned media from adipocytes themselves exposed to a mitochondrial
uncoupling and probably triggered by modifications in the set of adipokines (still to be
identified) released by adipocytes in these conditions.
In conclusion, we demonstrated, for the first time, that a mild and chronic
mitochondrial uncoupling affects mitochondrial population without triggering mitochondrial
biogenesis and that pyruvate carboxylase might play a key role in mitochondrial uncouplinginduced
triglyceride content reduction in adipocytes. We also found that mitochondrial
uncoupling affects the expression and the release of several adipokines by adipocytes, which
in turn could trigger insulin resistance in C2C12 myocytes. These results open the way to
further investigation on the role of mitochondrial uncoupling on the secretory function of
adipocytes and bring some highlights on the comprehension of adipocyte cell responses to a
mitochondrial uncoupling. This information is needed before thinking of using controlled and
specifically targeted mitochondrial uncoupling in white adipose tissue as a therapeutic

Conception de matériaux poreux hiérarchisés synthétises au départ de précurseurs moléculaires uniques
With the decline in reserves of certain raw materials, coupled with the ever growing needs of the population, there is increased pressure on industries to devise novel synthetic processes. The main way to improve the efficiency of those chemicals processes is the design of a new generation of inorganic chemistry based catalysts. The Sol-gel process is one of the most promising technologies permitting the development of an amazing new generation of mixed oxide materials featuring porous organizations at the nanometric scale, a controlled stoichiometry and an excellent homogeneity of the active sites. This class of materials could serve as catalysts for the fine chemistry as well as for petroleum cracking processes.
However, the mixed oxide catalytical efficiency is strongly linked to the degree of the active sites dispersion into the silica framework. Usual synthetic methods systematically drive to a phase separation phenomenon, resulting from the different reactivity of the starting independent inorganic precursors used. Synthesized materials are inhomogeneous due to the formation of a poor loaded silica framework and independent metal oxide nanoparticles, characterized by a low catalytical activity. Currently, many efforts are devoted to the improvement of the incorporation of increasing amount of active metal centers into the framework without any phase separation processes.
A very promising strategy consists in the use of single molecular source, instead of a mixture of independent inorganic precursors with different reactivities. The aqueous conversions of these single sources allow the construction of materials featuring unmatched homogeneity and totally controlled stoichiometry. This present work aims in the design of a new generation of homogeneous and porous mixed oxides, prepared from single molecular precursors, for some further catalytical applications.
This lead first to the conception of porous and homogenenous zirconosilicate materials (Si/Zr ~ 4), followed by the design of macro-mesoporous aluminosilicate solids with high loading of tetrahedral aluminium species (1 < Si/Al < 2), by the controlled (pH adjusting step, use of chelating agents, addition of silica co-reactant,…) polymerisation of selected/synthesized single molecular precursors. These new materials have been intensively characterized by a large panel of techniques (NMR, BET, SEM, TEM,…) in order to highlight their higher chemical homogeneity, and large efforts were dedicated to the fine tuning of the multiscaled porous systems. The mechanisms of spontaneous generations of hierarchical macro-mesoporosities within aluminosilicates samples were carefully studied, notably by optical microscopy. Finally, the ionic exchange and thermal stability properties of these new materials were assessed.La raréfaction des matières premières et les besoins sans cesse croissants de l’humanité ont amenés à de nouveaux impératifs au niveau des procédés de synthèse dans l’industrie. Ceux-ci se doivent d’être plus sélectifs, plus spécifiques et plus efficaces tout en préservant l’environnement. De fait, une des grandes et très porteuses tendances actuelles dans le domaine de la chimie des matériaux est la conception de nouveaux oxydes mixtes, tels que des matériaux aluminosilicatés, présentant une ou plusieurs porosités structurées à l’échelle nanométrique, ainsi que des propriétés catalytiques acido-basiques et/ou rédox renforcées. Ce type de matériaux présente un intérêt essentiel dans les secteurs de la chimie fine ainsi que dans les procédés de raffinage du pétrole.
L’efficacité catalytique de cette gamme d’oxydes mixtes est néanmoins tributaire de l’homogénéité avec laquelle les espèces actives sont dispersées au sein du réseau silicaté poreux. Les méthodes de synthèse conventionnelles mènent systématiquement à des phénomènes de séparation de phases. Ceci découle de la différence de réactivité des précurseurs employés. Ces solides obtenus sont inhomogènes d'un point de vue chimique car constitués d’un réseau silicaté appauvri en sites actifs, et de nanoparticules d’oxyde métallique d’activité catalytique limitée. Un grand nombre d’efforts tendent à contrôler, via le développement de stratégies scientifiques toujours plus efficaces, l’insertion de quantité importante de sites actifs au sein de la charpente silicatée, tout en prévenant ces processus de séparation de phases.
Une stratégie prometteuse consiste en l’emploi de précurseurs moléculaires uniques. La conversion de ces sources moléculaires, se substituant à un mélange de précurseurs indépendants, ayant leur réactivité propre, permet l’obtention d’oxydes mixtes poreux arborant une homogénéité chimique supérieure. L’objectif ce travail est donc la mise au point d’une nouvelle génération de matériaux poreux homogènes synthétisés au départ de précurseurs moléculaires uniques servant potentiellement à des fins catalytiques.
Cette thèse relate premièrement la conception de matériaux zirconosilicatés (Si/Zr ~ 4) homogènes poreux, suivi de la synthèse de solides aluminosilicatés macro-mésoporeux riches en aluminium tétravalent (1 < Si/Al < 2), via l’hydrolyse contrôlée (ajustement du pH, utilisation d’agents chélatants, ajout de co-précurseurs silicatés,…) et la condensation de précurseurs moléculaires uniques, préalablement sélectionnés et/ou synthétisés, en milieux aqueux. Ces solides ont été caractérisés au mieux (RMN, BET, MEB, MET,…) afin de pouvoir mettre en évidence une homogénéité chimique supérieure, tandis qu’un soin tout particulier fut apporté au développement et au contrôle des porosités. La génération spontanée de macro-mésoporosités hiérarchisées présentes au sein des solides aluminosilicatés fit l’objet d’études assidues, notamment par microscopie optique, afin d’en percer les mécanismes de fonctionnement. Chimie de surface des nanotubes de carbone, vers une meilleure dispersion et solubilisation
This thesis is in line with the manipulation and the characterisation of carbon nanotubes. Indeed, these new allotropic forms of carbon, at the center of a lot of studies since their discovery, remain mysterious. With their characteristics of black bodies and their conductor or semi-conductor behaviour, their characterisation can be, at the best, described as delicate. As for their manipulation, it clashes mainly against the problem of its molecular mass which is higher than the million. All this can explain the dispersion and solubilisation problems often found in the literature. These problems of dispersion/solubilisation are also the reason of the difficulties encountered with the incorporation of carbon nanotubes into polymers, in spite of the multiple researches occurring in this field.
In a first time, a preliminary study of the solubilisation of non-functionalized carbon nanotubes in organic solvents was carried out. This study had for target the development of a reliable and reproducible protocol for the solubility measurement. Once the measurement protocol developed, a data-gathering phase could follow. This new step allowed to gather a sufficient number of data’s to correlate the solubility of non-functionalized carbon nanotubes with known physical chemistry parameters. The good correlation between the non-functionalized carbon nanotubes solubility and the Hansen parameters of the used solvents could be demonstrated. Consequently, some applications could be enabled, such as the separation of carbon nanotubes functions of their size or the formation of nanotubes gels.
In a second part, the influence of the surface state of the functionnalized carbon nanotubes on their solubility was studied. After the study of the solubility of the functionnalized carbon nanotubes in several solvents, it could be demonstrated that a strong correlation between the surface state of the nanotubes used and the solubility variations observed was existing. This correlation paves the way for the use of the solubility as a characterisation technique for carbon nanotubes.
Finally, a third part takes an interest in the interactions between some polyaromatic derivatives and the walls of carbon nanotubes. The adsorption of these compounds made profit for the development of a technique concerning the measure of the specific surface of carbon nanotubes.Cette thèse s’inscrit dans le cadre général de la manipulation et de la caractérisation des nanotubes de carbone. En effet, ces nouvelles formes allotropiques du carbone, au centre de nombreuses recherches depuis leur découverte, demeurent mystérieuses. De par leurs caractéristiques de corps noir et de conducteurs ou semi-conducteurs électriques, leur caractérisation reste décrite comme délicate. Quant à leur manipulation, elle se heurte principalement au problème posé par une masse molaire de loin supérieure au million. Ceci explique les problèmes de dispersion et de solubilisation fréquemment rencontrés dans la littérature. Ces mêmes problèmes de dispersion/solubilisation sont aussi la cause des difficultés rencontrées lors de l’incorporation des nanotubes de carbone dans les polymères et ce, malgré les nombreuses recherches ayant eu lieu dans ce sens.
Dans de travail, une étude préliminaire de la solubilisation des nanotubes des différents types de nanotubes de carbone non-fonctionnalisés dans les solvants organiques a été menée. Cette étude a eu pour but la mise au point d’un protocole fiable et reproductible pour la mesure de leur solubilité. Une phase de collecte de mesures a ensuite été menée afin de récolter suffisamment de données dans le but de corréler la solubilité des nanotubes de carbone non-fonctionnalisés avec des paramètres physico-chimiques tabulés. L’étroite corrélation entre la solubilité des nanotubes de carbone non-fonctionnalisés et les paramètres de Hansen des solvants utilisés a pu être démontrée. Ceci a dès lors permis certaines applications telles que la séparation des nanotubes de carbone en fonction de leur taille ou la formation de gels de nanotubes.
Dans une seconde partie, l’influence de l’état de surface des nanotubes fonctionnalisés sur leur solubilité a été étudiée. Après étude de la solubilité des nanotubes fonctionnalisés dans différents solvants, il a pu être démontré qu’il existait une forte corrélation entre l’état de surface des nanotubes utilisés et les variations de solubilité observées. Cette corrélation ouvre la voie à l’utilisation de la solubilité comme technique de caractérisation pour les nanotubes de carbone.
Enfin, les interactions entre des dérivés polyaromatiques et la paroi des nanotubes de carbone ont été étudiées. L’adsorption de ces composés a été mise a profit pour la mise au point d’une technique de mesure de la surface spécifique des nanotubes de carbone.(DOCSC02) -- FUNDP, 201
Le rôle des microémulsions dans le transport de médicament à travers la peau: application au rétinol et à l'ibuprofène
Microemulsions are easy to prepare and very stable. They can solubilize different types of molecules and are proposed as a permeation enhancer for percutaneous drug delivery. All these characteristics have inspired scientists and they study this type of system as a possible drug vehicle in topical application.
The aim of this work is to find and study a microemulsion system able to be a good drug vehicle for topical application.
In order to fulfil our goal, we have studied different types of microemulsions. When they are characterized, we used these to synthesize nanoparticles by direct precipitation in the water cores of the microemulsions. Nanoparticles have been characterized by electronic microscopy, dynamic light scattering and UV visible techniques.
The faculty of percutaneous permeation of these formulations have been tested by two methods (dialysis and vertical Franz cells system). Other products, including a commercially available formulation, have been tested too to compare the results and evaluate the permeation aptitude of the microemulsion systems.Les microémulsions présentent de nombreux avantages : facilité de préparation, grande stabilité, capacité de solubilisation de diverses molécules ou leur aptitude à faciliter la perméation de la peau. C’est pourquoi, ces dernières décennies, les scientifiques ont montré un intérêt croissant pour les microémulsions comme vecteur potentiel de principe actif en vue d’une administration topique.
L’objectif de cette thèse est dès lors de proposer et d’étudier un système de microémulsion qui pourrait s’avérer être un bon vecteur transcutané de médicaments.
Pour ce faire, nous avons étudié différents systèmes de microémulsion. Une fois, ces systèmes bien caractérisés, nous les avons utilisés pour synthétiser des nanoparticules de rétinol et d’ibuprofène par précipitation directe de ces principes actifs dans les cœurs aqueux des microémulsions. Les nanoparticules synthétisées ont également été caractérisées de diverses manières (microscopie, diffusion dynamique de la lumière, UV).
Le potentiel de transport de médicaments de ces formulations sera évalué par deux techniques : dialyse et perméation sur cellules de Franz. D’autres formulations, dont un produit commercialement disponible, ont également été évaluées vis-à-vis de leur capacité à véhiculer le même principe actif. Une comparaison des résultats obtenus pour les différentes formulations a permis de rationaliser les aptitudes de transports des microémulsions.(DOCSC02) -- FUNDP, 201
Essays on Multivariate Volatility Forecasting
Chapter 1 surveys the most important developments in volatility forecast comparison and model selection, in both the univariate and multivariate frameworks. The chapter reviews a number of evaluation methods and testing procedures for predictive accuracy based on statistical loss functions and recent contributions on the admissible form of loss functions which ensure consistency of the ordering when forecast performances are evaluated with respect to an imperfect volatility proxy.
In chapter 2 we investigate the properties of the ranking between multivariate volatility forecasts with respect to alternative statistical loss functions used to evaluate forecast performances focusing on the multivariate framework and the problems arising due to the latent nature of the conditional variance. We provide conditions on the functional form of the loss function that ensure consistency between the proxy-based ranking and the true, but unobservable one. We identify a large set of loss functions that yield a consistent ranking. We illustrate our findings using artificial data and compare the ordering delivered by both consistent and inconsistent loss functions over different forecast horizons. We discuss the sensitivity of the ranking to the quality of the proxy and the degree of similarity between models. An application to three foreign exchange rates, where we compare the forecasting performance of 24 multivariate GARCH specifications over two forecast horizons, concludes the chapter.
In Chapter 3 we address the question of the selection of multivariate GARCH models in terms of forecast accuracy with a particular focus on relatively large scale problems. We consider 10 assets from NYSE and NASDAQ and compare 125 model based one-step-ahead conditional variance forecasts over a period of 10 years using the MCS and the SPA tests. Model performances are evaluated using four statistical loss functions which account for different types and degrees of asymmetry with respect to over/under predictions. When considering the full sample, MCS results are strongly driven by short periods of high market instability during which multivariate GARCH models appear to be rather inaccurate. Over relatively unstable periods, i.e. dot-com bubble, the set of superior models is composed of more sophisticated specifications such as orthogonal and dynamic conditional correlation, both with leverage effect in the conditional variances. However, unlike the DCC models, our results show that the orthogonal specifications tend to systematically underestimate the conditional variance. Over calm periods, simple assumptions like constant conditional correlation and symmetry in the conditional variances cannot be rejected. Finally, during the 2007-2008 financial crisis, accounting for non-stationarity in the conditional variance process generates superior forecasts. The SPA test suggests that, independently from the period, the best models do not provide significantly better forecasts than the DCC model with leverage effect in the conditional variances of the returns.
In Chapter 4 we derive a class of diffusion approximations based on conditional correlation models. To our knowledge, this chapter represents a first attempt to address the relationship between multivariate discrete and continuous time models, and in particular to conditional correlation models. We consider the consistent DCC (cDCC) model which is particularly appealing because it is based on a representation of the process driving the correlation which preserves the martingale difference property. We point out the existence of a degenerate diffusion limit. The degeneracy of the cDCC diffusion limit is due to the particular structure of the discrete time model in which the noise propagation system of the variances and that of the process driving the correlation are perfectly correlated. More precisely, the diffusion of the variances and that of the diagonal elements of the process driving the correlation are pairwise governed by the same Brownian motion. We also consider, as a special case, the constant conditional correlation model, which can be obtained by imposing suitable parameter restrictions to the cDCC model. In this case, we are able to recover a non-degenerate diffusion. Finally, we propose different sets of conditions regarding the speed of convergence of the parameters of the cDCC model which allow to recover other degenerate diffusion limits, characterized by a stochastic price process while variances and/or correlations remain time varying but deterministic. Reverse Engineering User-Drawn Form-Based Interfaces for Interactive Database Conceptual Analysis
The first step of most database design methodologies consists in eliciting part of the user requirements from various sources such as user interviews and corporate documents. These requirements are formalised into a conceptual schema of the application domain, that has proved difficult to validate, especially since the graphical representations of data models have shown understandability limitations from the end-users standpoint. On the other hand, electronic forms seem to be more natural and intuitive to express data requirements for laymen. Besides, the necessity to associate end-users of a future system with its specification and development steps has long been advocated.
In this doctoral research, we consequently explore the possible reverse engineering of user-drawn form-based interfaces to perform an interactive database conceptual analysis, and subsequently present the tool-supported RAINBOW approach resulting from this investigation. This user-oriented approach relies on the adaptation and integration of principles and techniques coming from various fields of study, ranging from Database Forward and Reverse Engineering to Prototyping and Participatory Design.La première étape de la plupart des méthodologies de développement de bases de données se concentre notamment sur l'élicitation des besoins des utilisateurs, à partir de sources diverses telles que des interviews et de la documentation existante. Ces besoins sont formalisés par le biais d'un schéma conceptuel représentant le domaine d'application. En pratique, la validation de tels schémas s'avère difficile, étant donné que la compréhension de leur formalisme, même à l'aide de représentations graphiques, est loin d'être triviale. Parallèlement, il apparait que les formulaires électroniques semblent plus naturels et intuitifs pour permettre l'expression de tels besoins par des non experts. Par ailleurs, l'implication des utilisateurs finaux dans la spécification et le développement d'un futur système fait désormais partie des bonnes pratiques de l'Ingénierie Logicielle.
Dans cette recherche doctorale, nous explorons dès lors la possible rétro-ingénierie d'interfaces de type formulaire dessinées par des utilisateurs finaux, afin de permettre une analyse conceptuelle interactive. Le résultat de cette investigation est l'établissement de l'approche RAINBOW et de son support logiciel. Etude des propriétés des hamiltoniens unidimensionnels aléatoires : Application au cas dimère
By scaling the mesoscopic conductance with the system length, the theory of Anderson localisation has predicted insulator transmission regimes in one–dimensional electronic systems for any disorder. Viewed as the origin of destructive interferences, randomness contributes actively to localised states in finite region of space. However by pairing the defect cells, delocalised Bloch-like states and diffusive transport appear in disordered system when transparency conditions of the dimer unit cells occurs. This is the conventional random dimer model.
The re-examination of the pioneer ideas of S. He and J.D. Maynard in the acoustic analogy of mesoscopic systems permitted us to describe the transmission properties of mechanical waves in the classical system of homogenous string charged by masses. By suitably introducing springs, the transmission at the dimer resonance can be enhanced from diffusive to ballistic response. For such ballistic transmission mechanical filters is examined and optimised for disordered host alloys within the random dimer model.
In the same way, the propagation of electromagnetic waves in one-dimensional stratified systems has been examined. By dealing with particular structural parameters, principal standing resonances are added into the transmission spectrum of defect and host unit cells. Ballistic transmission canals can be obtained in binary disordered photonic films at the dimer resonances. Various interesting transmission regimes near the dimer resonances are described.
The electronic transport in disordered carbon nanotubes with chemical defects is also examined. By doping simultaneously with Boron (B) and Nitrogen (N), BN dimer defect cell can be considered when B and N atoms are first closest neighbours. The BN dimer suppresses the localisation signatures induced by the isolated B and N defect states while the transmission for the BN nanodomains depends strongly on both the orientation of the BN nanodefect inside the host structure and the number of B (N) atoms in the nanodomain. By increasing the number of the BN dimers, the transmission in a BN nanodomain is governed by the tunnelling effect. Interesting resonant tunnelling transmissions can be obtained when two BN nanodefects spreading on the whole contour of the nanotube are separated inside the host nanotube according to the concept of the dimer unit cell.
In our study, the propagation of waves in analogue one-dimensional random systems has been studied within the random dimer model. In addition to the universal diffusive behaviour of the conductance at the conventional dimer resonance, interesting situations improving the transmission properties at the dimer resonance have been found: With a suitable choice of the defect and host unit cells, ballistic transmission canals can be opened providing an additional way to considerably enhance the transmission properties in such particular disordered systems.Le désordre source d’interférences destructives contribue activement au phénomène de la localisation d’Anderson. L’avènement du modèle unidimensionnel du dimère aléatoire comme exception à la règle établie par de la théorie d’échelle de la localisation, a donné encore plus d’importance à ce domaine de recherche: En imposant à un défaut d’apparaitre aléatoirement par paires, le motif dimère (de par sa transparence totale à des énergies particulières) est à l’origine de l’apparition d’états étendus similaires (mais pas identiques) aux états de Bloch. A ces résonances dimère, le mode de transmission est conventionnellement diffusif.
La reprise des idées fondatrices de S. He et J.D. Maynard sur l’analogie acoustique des systèmes mésoscopiques, nous a permis d’examiner les propriétés de transmission d’ondes mécaniques dans un système classique de corde vibrante chargée en masses. L’introduction du ressort sur chaque cellule fournit un moyen pratique pour l’ajustement des paramètres structuraux du milieu de propagation et améliore le régime de transmission à la résonance dimère : Avec l’alignement de la fréquence caractéristique du motif défaut dimère et de la fréquence libre du motif hôte, la transmission balistique est obtenue. Une étude sur les propriétés de transmission optimisées dans les filtres mécaniques balistiques est présentée. La même approche est étendue aux alliages hôtes désordonnés.
De même, en analogie avec le comportement des électrons dans les heterostructures désordonnées, la propagation des ondes électromagnétiques dans les systèmes stratifiés unidimensionnels a été examinée. La prise en considération de paramètres structuraux particuliers assurant l’apparition supplémentaire des résonances stationnaires principales hôte et défaut dans le spectre de transmission, permet d’améliorer les propriétés de transmission de ces structures : Avec l’application de la procédure d’alignement des fréquences caractéristiques des motifs défaut et hôte sur un canal de transmission commun, des transmissions balistiques sont enregistrées à la résonance dimère. Des régimes de transmission distincts proches de la résonance dimère optimisée sont mis en évidence.
La propagation des ondes électroniques dans les nanotubes de carbone à désordre structural a été également étudiée. Le dopage simultané en Bore (B) et Azote (N) par le défaut dimère BN a montré une suppression des effets de la localisation d’Anderson induits par les états défauts B et N respectivement. Par ailleurs, la transmission à travers des monodomaines défaut BN dépend fortement de l’orientation de l’amas BN dans la structure hôte et du nombre d’atomes B (N) qui le composent : la diminution quasi exponentielle de la conductance en fonction du nombre croissant de dimères BN est synonyme d’un effet tunnel. Finalement, dans le cas défauts s’étendant sur tout le contour de nanotube BN, des pics de transmission - synonymes de la présence de l’effet tunnel résonant - peuvent apparaitre dans le spectre de transmission, lorsque le motif dimère conventionnel est considéré.
Notre étude a eu pour vocation globale de décrire l’effet de la corrélation du désordre à courte portée sur la propagation d’ondes dans des systèmes unidimensionnels désordonnés. En plus du caractère diffusif universel de la résonance dimère conventionnelle, nous nous sommes aussi intéressés aux situations pouvant améliorer les propriétés de transmission à la résonance dimère : Avec le choix convenable des motifs hôte et défaut, le régime de transmission balistique peut être obtenu ouvrant ainsi un canal de transmission de premier choix et une voie supplémentaire dans les propriétés de transmission à la résonance dans les systèmes aléatoires à désordre corrélé.(DOCSC01) -- FUNDP, 201
