Identification and characterization of potential Brucella abortus effectors interacting with Rab GTPases and endoplasmic reticulum exit sites associated proteins
Brucellae are intracellular facultative pathogenic bacteria which are able to invade professional and non-professional phagocytes. Once inside the cell, Brucella resides in a vacuole (BCV, Brucella containing vacuole), which successively interacts with endocytic compartements, i.e. early endosomes, late endosomes and lysosomes. Finally, Brucella reaches endoplasmic reticulum (ER) at particular sites, ER exit sites (ERES), to extensively proliferate in the ER. Type IV secretion system (T4SS) VirB, was shown to be required to sustain interaction with ER and consequently to proliferate. It was suggested that T4SS translocates proteins to hijack membrane trafficking processes such as endocytic and secretory pathways, regulated by Rab GTPases. Up to now, at least three T4SS substrates were identified but their function remains unkown. In order to identify Brucella effectors, we took advantage of the availability of the Brucella ORFeome in our laboratory to perform a genome-wide yeast two-hybrid (Y2H) screening between Brucella predicted proteins and human phagosomal proteins, Rab GTPases and ERES associated proteins.
In the current study, we report the identification of three specific interactions between Rab2 and RicA (Rab2 interacting conserved protein A), Rab34 and PitF (protein interacting with Rab thirty four), and Sec24A and CostA (conserved Sec24A-targeted protein A). We showed that RicA and PitF are translocated during infection of macrophages and these translocations are T4SS-dependent. RicA and CostA are secreted in bacteriological culture and the secretion of these two proteins is kept in the ΔvirB mutant strain, suggesting that another secretion system is functional in these conditions. An in frame deletion was constructed for each Brucella gene coding for a potential effector and mutants were characterized. The ΔricA mutant strain recruits less GTP-locked myc-Rab2 on its BCVs. This mutant has also an altered trafficking and a faster proliferation compared to the B. abortus wild type strain. The ΔpitF mutant has an internalization defect. Finally, the costA mutant is impaired for its association to the Sec23 ERES marker and displayed a particular trafficking, with a lower association to LAMP1 and calnexin at 12 h post-infection in HeLa cells, but its intracellular proliferation kinetics was not affected.
In conclusion, this work allows the identification of two Brucella effectors with a proposed function during a model cell infection. Indeed, RicA could act on trafficking by recruiting Rab2 and PitF plays a role during Brucella internalization, perhaps through its interaction with Rab34.(DOCSC03) -- FUNDP, 201
Combined effects of heat stress and xenobiotic exposure in the European bullhead Cottus gobio. Clues provided by enzymatic and proteomic approaches
Combined exposure to elevated temperature and pollutants is an environmentally relevant scenario that is expected to become more widespread in the future with global warming. Accordingly, the general objective of the present study was to investigate the interactive effects of temperature stress and xenobiotic exposure in the European bullhead, Cottus gobio, a candidate sentinel species. Before studying the combined stresses, we have firstly characterized the toxicity of two ubiquitous contaminants in aquatic environments, i.e., cadmium (Cd) and perfluorooctane sulfonate (PFOS), in C. gobio. Exposure to high pollutants concentrations significantly altered the activity of several enzymes involved in energy metabolism, namely, citrate synthase, cytochrome c oxidase and lactate dehydrogenase, either in liver and/or in gills. Additionally, a proteomic approach was used to further our understanding of toxicity of the selected pollutants in C. gobio. The results showed that most of the identified differentially expressed proteins are associated with central aspects of the evolutionarily conserved cellular stress response and are part of the stress proteome. Secondly, as temperature and Cd are known to induce oxidative stress in aquatic organisms, their combined effects on the oxidative stress status of bullhead were assessed. Overall, we have observed that elevated temperature and Cd exposure independently influenced the antioxidant defence system with clear tissue-specific and stress-specific antioxidant responses. Further, the 20S proteasome activity was increased in response to heat stress in liver and gills. Finally, we have evaluated the effects of temperature and Cd exposure in sequential combination, i.e., fish acclimated to enhanced temperature and subsequently exposed to Cd. A combination of enzyme activity assays and protein expression patterns clearly indicated that increases in water temperature consistent with climate change predictions may modulate the ability of C. gobio to tolerate subsequent exposure to another stressor, such as Cd. Furthermore, the proteomic analysis brings clues to the potential molecular pathways involved in the acclimation process and by which a heat acclimation could interfere with an additional stress to Cd thereby highlighting shared mechanisms of action. As a corollary, the present study demonstrated that an integrative approach using enzymatic and proteomic analyses is worthwhile to better characterize the interactive effects of multiple stressors, such as temperature and metal pollution on fish species, and suggests that additional studies to link the cellular phenotype as observed here to changes at higher levels of biological organization could provide essential information to inform on organisms’ health in their natural environments.L'augmentation de la température moyenne globale et les différents polluants présents dans l’environnement peuvent occasionner des effets néfastes aux organismes aquatiques. Toutefois, l’importance des effets combinés d’une hausse de la température et d’une exposition à un/des xénobiotique(s) reste encore inconnue à ce jour. Dans le présent travail, nous avons premièrement caractérisé la toxicité de deux polluants omniprésents dans les milieux aquatiques, le cadmium (Cd) et le perfluorooctane sulfonate (PFOS), chez le chabot, préalablement aux stress combinés. Des modifications de l'activité de plusieurs enzymes impliquées dans le métabolisme énergétique (la citrate synthétase, la cytochrome c oxydase et la lactate déshydrogénase) ont été observées dans le foie et/ou les branchies après une exposition à de fortes concentrations en polluants. Une analyse protéomique a également été réalisée afin d’approfondir la compréhension des effets toxiques du Cd et du PFOS chez le chabot. Ainsi, nous avons mis en évidence la présence de variations dans l’expression de diverses protéines impliquées dans des voies très diverses et faisant partie du protéome de stress. Deuxièmement, les capacités antioxydantes du chabot ont été étudiées suite à une exposition combinée à la température et au Cd. Les résultats montrent que les différents systèmes antioxydants enzymatiques sont soit altérés par l’augmentation de la température, soit par le Cd. L’observation des résultats souligne également l’existence d’une réponse variable selon le tissu considéré. De plus, une augmentation de l’activité du protéasome 20S a été mesurée dans le foie et les branchies après un stress thermique. Enfin, nous avons tenté de déterminer si une acclimatation à la chaleur chez le chabot interfère avec une exposition subséquente au Cd. La mesure de l’activité de la lactate déshydrogénase et l’analyse des profils d’expression protéiques ont clairement démontré qu’une acclimatation à une augmentation de la température, conforme aux prévisions du réchauffement climatique, peut modifier la capacité des organismes à faire face à une autre perturbation environnementale comme une exposition à un xénobiotique. Par ailleurs, l'analyse protéomique apporte des indices sur les mécanismes moléculaires possibles mis en place pendant une acclimatation thermique et pouvant interférer avec les effets du Cd. En résumé, ce travail démontre qu'une approche intégrative utilisant des analyses enzymatiques et protéomiques, fournit une meilleure compréhension de l’interaction entre une hausse de la température et une exposition à un polluant chez un organisme aquatique.(DOCSC03) -- FUNDP, 201
Multi-walled carbon nanotubes: decorated with metal nanocrystals and incorporated into metal matrices
Since their discovery, carbon nanotubes have attracted the attention of researchers world wide in the field of nanoscience and nanotechnology, mainly due to their remarkable properties and potential applications. Carbon nanotubes are largely inert and call for surface modification to disperse in many solvents to be further manipulated to decorate them with metal or metal oxide nanoparticles for various applications and incorporate them into metal matrices. In the first part of the work, we have decorated the multi-walled carbon nanotubes with metal and metal oxide nanocrystals. In the present study, we have designed a simple and efficient method to decorate the multi-walled carbon nanotubes with copper and nickel nanocrystals and optimized the conditions to achieve homogeneous decoration of those metals and their metal oxide nanocrystals.
In the second part of the work, we have incorporated the multi-walled carbon nanotubes into metal matrices. In this work, we have explored several methods and a new solvent to efficiently and uniformly incorporate the multi-walled carbon nanotubes into metal matrices. This was achieved by electrodeposition, mainly because it is a cost effective method to obtain the high purity of deposits. In our case, we have demonstrated that it is possible to incorporate multi-walled carbon nanotubes on copper substrate into nickel matrices. We have used different kinds of multi-walled carbon nanotubes, such as pristine, oxidized and amino-functionalized, to incorporate them into nickel matrices in aqueous solutions. Thereafter, we have used oxidized and pristine multi-walled carbon nanotubes as they showed better incorporation into nickel matrices compared to amino-functionalized multi-walled carbon nanotubes. "La politique de l'entre-deux. Relecture de la pensée de l'action chez H. Arendt, en dialogue avec J. Patocka et C. Castoriadis"
XL'hypothèse centrale de cette recherche consiste à interpréter la pensée arendtienne de l'action en termes de coaction, et d'élaborer ainsi ce qui sera progressivement désigné comme « la politique de l'entre-deux ». Il s'agit de montrer comment et pourquoi il est à la fois possible et nécessaire, au départ de la pensée de H. Arendt, de concevoir l'action – et en particulier l'action politique – comme cet événement qui n'advient que dans l'entre-deux de la rencontre des acteurs, initiant ainsi un monde commun dans lequel nous nous distinguons en devenant égaux. Pour ce faire, cette étude se construit en deux grande parties, la première consistant en une relecture du pouvoir de l'action chez H. Arendt en termes d'entre-deux, et la seconde en l'élaboration concrète de cette politique de l'entre-deux mise en évidence dans la pensée arendtienne, en dialogue avec J. Patocka et C. Castoriadis.
La première partie commence par une réinterprétation d'un concept majeur dans l'œuvre de H. Arendt : la pluralité. L'étude de ce concept montre que dès le moment où la pluralité s’avère être condition fondamentale de l'action, celle-ci est à comprendre en termes de coaction, dont l'émergence prend la forme d'un événement. En ce sens, nos rencontres font événement, et c'est au cœur de nos inter-actions que se situe le pouvoir créateur de l'action, qui sera défini par la suite.
L’auteur s'attache ensuite à justifier le choix interprétatif précédent, tant au niveau de la compréhension de l'action arendtienne qu'en ce qui concerne l'action politique de manière plus générale. Penser l'action politique comme cet entre-deux qui fait événement n'est ni utopique ni subjectif : une telle démarche se veut en réalité fidèle à une pratique politique qui ne ruine pas sa condition de possibilité. C'est du moins ce que montre – par la négative – une analyse des Origines du totalitarisme qui met au jour, à travers les figures de l'apatride, de l'homme de la masse et de l'homme du camp d'extermination, que ce qui nous est le plus personnel, à savoir cette initiative sans laquelle il n'y a pas d'action possible, n'apparaît que si l'entre-deux propre à la pluralité est à la fois préservé et actualisé.
La dernière étape de la première partie se propose deux objectifs. D’abord, elle s’emploie à saisir la dimension proprement politique de l'entre-deux telle qu'elle se manifeste dans la pensée arendtienne au départ de quatre expériences socio-historiques précises : l'âge homérique et la polis athénienne, l'âge Romain et la Constitution rédigée par les Pères fondateurs, les conseils et enfin la désobéissance civile. Ensuite, elle pointe les enjeux et difficultés pour qui veut élaborer, au départ de l'entre-deux qui se forme dans nos rencontres, une politique au sens fort du terme : à la formidable puissance de création de la coaction s'associe une grande fragilité qui rend son institutionnalisation extrêmement difficile ; quant à l'originalité de sa spécificité relationnelle elle va de paire avec l'ambiguïté de son statut (s'agit-il de politique de l'entre-deux ou d'un mouvement d'ordre social ?) et exige dès lors de penser l'émergence de l'action politique dans sa dimension incarnée. L’auteur montre que H. Arendt prend peu en considération la question du corps propre et n'aborde jamais de front celle de la création : si c'est au cœur de son œuvre que la politique de l'entre-deux a pu être mise en évidence, répondre aux défis posés par l'élaboration d'une telle politique requiert [donc] d'élargir l'horizon arendtien à d'autres champs de pensée. C'est à cette démarche qu’est consacrée la seconde partie de cette étude, qui se propose de répondre aux défis d’une politique de l’entre-deux.
Un premier moment est construit en dialogue avec la phénoménologie du monde de J. Patocka, qui définit l’action comme cet espace en mouvement au travers d'acteurs qui se présentent comme autant de « je peux » qui ne découvrent et ne déploient leurs capacités – parmi lesquelles il faut principalement citer celle de l'autonomie – que pour autant qu'ils agissent ensemble. La coaction se conçoit alors comme un processus d' « intertransformation » qui change le monde tout transformant les acteurs. Néanmoins, alors que la pensée du phénoménologue tchèque, contrairement à la pensée arendtienne, souligne l’inévitable importance du corps pour l’action, elle faillit lorsqu’il s’agit de penser l’ancrage mondain de l’action politique.
Le moment suivant se propose de dépasser cette difficulté en pensant l’action comme ce qui crée un monde, comme cet événement que nous faisons advenir à plusieurs et qui prend une forme concrète au gré de nos promesses mutuelles – ce qui revient à penser la coaction comme cocréation. C'est en dialogue avec C. Castoriadis que cette conception est développée : le penseur grec est effectivement un des rares philosophes qui ait élaboré une interprétation de l’histoire et de l’action en termes de création. La confrontation de l'idée de coaction avec celle d'auto-organisation, telle qu'on la trouve dans les écrits castoriadiens de Socialisme ou Barbarie, permet d'abord de concrétiser le pouvoir créateur de nos « inter-actions » au travers des expériences du mouvement ouvrier. S'élabore ensuite une critique de l’ontologie castoriadienne de l'imaginaire radical, laquelle se trouve au fondement de ses écrits philosophiques. Cette ontologie comporte effectivement le risque de délaisser le lieu véritable de l’action (à savoir : l’entre-deux de la rencontre des acteurs) au profit d'un imaginaire collectif qui ne suffit pas à expliquer l’action collective. Pour éviter cette aporie, la dernière étape de cette recherche suggère de penser l'imaginaire en termes de mémoire vivante, comme ces significations qui sont tout à la fois le fruit de la coaction (ou encore de la cocréation) et ce monde depuis et dans lequel nos actions se forment. Quant à l'action politique, elle ne se conçoit dès lors plus en termes de projet comme le défend Castoriadis tout au long de son œuvre, mais devient à la fois le principe qui anime la coaction et l'attitude que chacun des acteurs peut développer, parce que c'est avec et contre d'autres qu'il agit.
En conclusion, l’auteur dégage la triple portée philosophique, sociopolitique et existentielle de son travail, dont il s’attache à esquisser les contours.(DOCFILO00) -- FUNDP, 201
Identification et caractérisation de deux protéines polaires de Brucella abortus, IfoP et AidB
Bacterial proteins presenting a particular subcellular localization are regularly
described in diverse bacteria, and our model of study Brucella abortus, an
intracellular pathogen, does not depart from the rule. Indeed, in our laboratory, we
recently showed that an essential histidine kinase named PdhS and a fumarate
hydratase called FumC localize at the old pole in B. abortus. These data suggest that
a set of proteins with various functions could be associated with one of both bacterial
poles, old or new. To test this hypothesis, we initiated an exploratory project to
identify on one hand, markers of old and new poles and on the other hand, functions
specifically associated with the poles. The proposed scientific strategy used an
approach on the scale of the Brucella melitensis ORFeome. The detection of the
polar localization of proteins was performed by genetic fusion with the fluorescent
protein YFP (yellow fluorescent protein). Among seven identified candidate proteins,
we were interested in two proteins in particular, a predicted acyl-CoA
dehydrogenase, named AidB, given its homology with the Escherichia coli AidB
protein, and a hypothetical protein called IfoP (for In Front Of PdhS). The functional
characterization of these two candidates, allowed us to demonstrate that the aidB
and ifoP genes are not essential for the viability of B. abortus and that AidB and IfoP
proteins are distributed asymmetrically in B. abortus. AidB localizes systematically at
the new pole and/or at the constriction site in B. abortus. The overexpression of aidB
provokes morphological defects, typically the presence of additional poles. The
genome of B. melitensis encodes 9 other putative acyl-CoA dehydrogenases and two
of them have a diffuse localization in the cytoplasm and do not present particular
morphological phenotype when they are overproduced. Thus, the polar localization of
AidB and the presence of morphological aberrations for the overexpression mutant
seem to be specific characteristics of this acyl-CoA dehydrogenase. These data
suggest that AidB is a marker of new poles and constriction sites, that can be
considered as sites of preparation of new poles for sibling cells. AidB could have a
role in the generation of growing new poles and/or in cell cycle control in B. abortus.
Furthermore, AidB remains polar during a cellular infection and in the presence of an
alkylating agent (EMS). On the other hand, the aidB mutant is more sensitive than
the wild type strain to an EMS treatment, suggesting that the aidB gene would be
involved in the repair or the prevention of alkylating damages. IfoP localizes almost
always at the opposite pole of PdhS, therefore considering IfoP as a marker of new
or "young" pole. Previous studies indicated that B. abortus divides asymmetrically,
following the example of Caulobacter crescentus, to produce a small cell and a large
cell. The use of IfoP as marker of the small cell suggests that this one would be the
privileged state in infection model in epithelial cell, in the non-proliferative phase of
the infection, which means at early time of infection.Des protéines bactériennes présentant une localisation subcellulaire particulière
sont régulièrement décrites chez diverses bactéries, et notre modèle d’étude Brucella
abortus, un pathogène intracellulaire facultatif, ne déroge pas à la règle. En effet, au
sein de notre laboratoire, nous avons montré récemment qu’une histidine kinase
essentielle nommée PdhS et une fumarate hydratase appelée FumC, pour ne citer
qu’elles, localisent au vieux pôle chez B. abortus. Ces données suggèrent qu’une
série de protéines de différentes fonctions pourraient être associées à l’un des deux
pôles bactériens, soit ancien soit nouveau. Pour tester cette hypothèse, nous avons
initié un projet exploratoire afin d’identifier d’une part, des marqueurs de nouveau et
d’ancien pôles et d’autre part, des fonctions associées aux pôles. La stratégie
scientifique proposée utilisait une approche à l’échelle de l’ORFéome de Brucella
melitensis. La mise en évidence de la localisation polaire de protéines fut réalisée
par fusion génétique avec la protéine fluorescente YFP (yellow fluorescent protein).
Parmi les sept protéines candidates identifiées, nous nous sommes intéressés à
deux protéines en particulier, une acyl-CoA déshydrogénase prédite, nommée AidB
suite à son homologie avec la protéine AidB de Escherichia coli, et une protéine
hypothétique baptisée IfoP (pour In Front Of PdhS). La caractérisation fonctionnelle
de ces deux candidats, nous a permis de démontrer que les gènes aidB et ifoP ne
sont pas essentiels à la viabilité de B. abortus et que les protéines AidB et IfoP sont
distribuées asymétriquement au sein de B. abortus. AidB localise systématiquement
au nouveau pôle et/ou au site de constriction chez B. abortus. La surexpression de
aidB provoque des défauts morphologiques, typiquement la présence de pôles
additionnels. Le génome de B. melitensis encode 9 autres acyl-CoA
déshydrogénases putatives et deux d’entre-elles ont une localisation diffuse dans le
cytoplasme et ne présentent pas de phénotype morphologique particulier lorsqu’elles
sont surproduites. Donc, la localisation polaire de AidB et la présence d’aberrations
morphologiques pour le mutant de surexpression semblent être des caractéristiques
spécifiques à cette acyl-CoA déshydrogénase. Ces données suggèrent que AidB est
un marqueur de nouveau pôle et de site de constriction qui peuvent être considérés
comme des sites de préparation de nouveaux pôles pour les cellules filles. AidB
pourrait avoir un rôle dans la génération de nouveaux pôles en croissance et/ou dans
le contrôle temporel du cycle cellulaire chez B. abortus. De plus, AidB reste polaire
au cours d’une infection cellulaire et en présence d’un agent alkylant (l’EMS). Par
contre, le mutant aidB est plus sensible que la souche sauvage à un traitement à
l’EMS, ce qui suggère que le gène aidB serait impliqué dans la réparation ou la
prévention de dommages alkylants. IfoP localise presque toujours au pôle opposé à
PdhS, considérant ainsi IfoP comme un marqueur de nouveau/jeune pôle. Des
études antérieures ont mis en évidence que B. abortus se divise asymétriquement, à
l’instar de Caulobacter crescentus, pour produire une petite et une grande cellules.
L’utilisation de IfoP comme marqueur de la petite cellule suggère que celle-ci serait
la forme privilégiée en modèle d’infection en cellule épithéliale, dans la phase non-
proliférative de l’infection, c’est-à-dire à des temps précoces d’infection.(DOCSC03) -- FUNDP, 201
Rational design, synthesis, molecular modeling and evaluation of -carboline and 5H-indeno[1,2-c]pyridazin-5-one derivatives as potential MAO and IDO inhibitors : Conception rationnelle, synthèse, modélisation moléculaire et évaluation de dérivés -carbolines et 5H-indéno[1,2-c]pyridazin-5-ones comme inhibiteurs potentiels de la MAO et d’IDO
Monoamine oxidase A (MAO-A) and –B (MAO-B) are attractive targets for a broad range of treatments against pathologies including depression, anxiety disorders, Parkinson’s and Alzheimer’s diseases. Most current MAO inhibitors lead to side effects by a lack of affinity and selectivity towards one of the isoforms. Recently, the crystal structures of hMAO-A and –B in complex with inhibitors opened the way towards the discovery of new, more selective and potent inhibitors. Thus, the main objective of this work, was the design of new, more potent, reversible and selective MAO-A or –B inhibitors derived from β-carboline and 5H-indeno[1,2-c]pyridazin-5-one scaffolds respectively, following a classical strategy including experimental (synthesis and biological evaluation) and theoretical (molecular modeling) approaches. The MAO inhibitory potencies showed that the replacement of the methoxy group of harmine by more lipophilic groups increases the inhibition for MAO-A. Studies on 5H-indeno[1,2-c]pyridazin-5-one scaffold bearing lipophilic groups in the 3 and 8-positions showed that the substitution in the 3-position dramatically influences the MAO-B-inhibiting properties.
Furthermore, the involvement through a same metabolic pathway and the similarity in the structural properties of MAO with indoleamine 2,3-dioxygenase (IDO) and lysine specific demethylase 1 (LSD1) respectively, led us to the investigation of the -carboline and 5H-indeno[1,2-c]pyridazin-5-one derivatives as potential IDO and LSD1 inhibitors. However, the two series show no inhibition of those two enzymes and are thus selective of MAO.
Finally, starting from the same strategy used for MAO, we are also interested in the synthesis of two new 3-substituted--carboline derivatives with amino groups and directly derived from 3-butyl--carboline, a known IDO inhibitor. Indeed, these two compounds display a positive charge at physiological pH which might establish an additional coulomb interaction with 7-propionate of the heme compared to 3-butyl--carboline. Howerver, first results tend to demonstrate that the introduction of a positive charge abolishes the inhibition of IDO.La monoamine oxydase A (MAO-A) et –B (MAO-B) sont des cibles intéressantes pour une large gamme de thérapies contre des pathologies telles que la dépression, l’anxiété et les maladies de Parkinson et d’Alzheimer. La plupart des inhibiteurs actuels mènent à des effets secondaires par un manque d’affinité et de sélectivité envers une des isoformes. Récemment, les structures cristallographiques de hMAO-A et –B en complexe avec des inhibiteurs ont ouvert le chemin vers la découverte de nouveaux inhibiteurs plus sélectifs et plus puissants. En conséquence, l’objectif principal de ce travail, fut la conception de nouveaux inhibiteurs plus puissants, réversibles et sélectifs de MAO-A ou –B à partir des motifs β-carbolines et 5H-indéno[1,2-c]pyridazin-5-ones respectivement, en suivant une stratégie classique comprenant une approche expérimentale (synthèse et évaluation biologique) et théorique (modélisation moléculaire). Les pouvoirs d’inhibition sur la MAO ont montré que la substitution du groupe méthoxy de l’harmine par des groupes plus lipophiles augmente l’inhibition de MAO-A. Des études sur les 5H-indéno[1,2-c]pyridazin-5-ones substituées à la fois aux positions 3 et 8 par des groupes lipophiles ont montré que la substitution en position 3 influence significativement les propriétés d’inhibition de la MAO-B.
De plus, l’implication à travers une même voie métabolique et la similarité dans les propriétés structurales de la MAO avec l’indoléamine 2,3-dioxygénase (IDO) et la lysine spécifique déméthylase 1 (LSD1) respectivement, nous ont amené à l’investigation des -carbolines et 5H-indéno[1,2-c]pyridazin-5-ones comme inhibiteurs potentiels d’IDO et de LSD1. Cependant, les deux series n’inhibent pas ces deux systèmes enzymatiques et sont donc sélectives de MAO.
Finalement, en partant de la même stratégie utilisée pour la MAO, nous nous sommes aussi intéressés à la synthèse de deux nouvelles -carbolines substitutées en position 3 par des groupements aminés et directement dérivées du 3-butyl--carboline, un inhibiteur connu d’IDO. Ces deux composés présentent en effet une charge positive à pH physiologique laquelle pourrait établir une interaction coulombienne supplémentaire avec le 7-propionate de l’hème comparé au 3-butyl--carboline. Cependant, les premiers résultats tendent à démontrer que l’introduction d’une charge positive abolit l’inhibition d’IDO.(DOCSC02) -- FUNDP, 201
ECOLOGIE ALIMENTAIRE COMPAREE DE LIMNOTHRISSA MIODON ET DE LAMPRICHTHYS TANGANICANUS AU LAC KIVU (AFRIQUE DE L'EST)
Limnothrissa miodon, a clupeid endemic to Lake Tanganyika, was voluntarily introduced in Lake Kivu between 1958-1959 and enabled the development of a flourishing pelagic fishery. Recently, Lamprichthys tanganicanus, a poeciliid also endemic to Lake Tanganyika, was accidentally introduced into Lake Kivu. Because potential preys for both species are scarce in the lake, L. tanganicanus could compete with L. miodon and could constitute a threat to pelagic fisheries which rely mainly on L. miodon. The objective of this work was to study the feeding ecology of these two fish species using several methods: analysis of stomach contents, identification of dietary patterns, analysis of stable isotopes of C and N and analysis of fatty acids in fishes and in the main components of plankton. The results showed a strong diet overlap between these two fish species, indicating some level of interspecific competition. However, their differing feeding activities patterns and differing use of terrestrial, littoral and pelagic food resources could reduce this competition. Indeed, although both species occupy a similar trophic position, L. miodon is dependent much more on pelagic food resources than L. tanganicanus which has a stronger link with the littoral food web. The profiles of fatty acids in fishes and plankton components indicate that the fatty acids compositions of both species are very close, and that the nutritional quality of zooplankton varies relatively little during the year. As for the productivity of zooplankton, it undergoes significant seasonal variations, which are dependent on both quantity and composition of phytoplankton. Finally, fatty acid profiles also revealed that bacteria do contribute significantly to zooplankton and fish biomass, highlighting the contribution of the microbial food web to consumer production.Limnothrissa miodon, Clupéidé endémique au lac Tanganyika, a été volontairement introduit au lac Kivu entre 1958-1959 et y a permis le développement d’une pêcherie florissante. Comme les proies potentielles de ces deux espèces sont peu abondantes dans le lac, L. tanganicanus pourrait être en compétition avec L. miodon et constituerait alors une menace pour la pêche en milieu pélagique, qui repose principalement sur L. miodon. Ce travail a consisté à étudier l’écologie alimentaire de ces deux espèces par différentes méthodes: analyse des contenus stomacaux, détermination des rythmes alimentaires, mesure des isotopes stables du C et du N et analyses des acides gras dans les poissons et les principaux composants du plancton. Les résultats ont révélé un fort recouvrement du régime alimentaire entre ces deux poissons, signe d’une probable compétition interspécifique. Cette compétition serait cependant atténuée par leurs rythmes alimentaires différents ainsi que par leur utilisation différente des ressources littorales, terrestres et pélagiques. En effet, bien que les deux espèces occupent pratiquement la même position trophique, L. miodon est beaucoup plus dépendant des ressources alimentaires pélagiques que L. tanganicanus, qui s’insère plus dans le réseau trophique littoral. Les profils des acides gras dans les poissons et les composants du plancton indiquent que les deux espèces de poissons ont des compositions en acides gras très proches et que la qualité nutritionnelle du zooplancton varie relativement peu au cours de l’année. Quant à la productivité du zooplancton, elle subit des variations saisonnières importantes, qui sont dépendantes à la fois de la quantité et de la composition du phytoplancton. Enfin, les profils d’acides gras ont aussi révélé que les bactéries contribuent significativement à la biomasse du zooplancton et des poissons, ce qui démontre la contribution de la boucle microbienne à la production des consommateurs.(DOCSC03) -- FUNDP, 201
Five-membered heterocycles as novel molecular scaffolds for targeting triplehydrogen bonding interactions
Due to its prominent directionality and strength, H-bonds are ones of the most widely used non-covalent interactions in supramolecular chemistry. Despite its relative high strength (energy of an H-bond in the gas phase typically ranges between 0−5 Kcal mol−1) in comparison with other non-covalent interactions, association of two molecules by means of a single H-bond leads to complexes displaying low thermodynamical stabilities, thus limiting their exploitation in the non-covalent synthesis of functional materials for real-world applications. Thereby, when stronger interactions are required, the general engineering approach focuses on the covalent synthesis of rigid planar molecular scaffolding in which several H-bonding donating (D) and accepting (A) moieties are arranged into a so-called ‘H-bonding array’. Due to the selective recognition processes and to the tunability of their association strength, multiple H-bonding arrays have become an indispensable molecular module in the tool-box of supramolecular chemists, allowing, through selective self-assembly and/or self-organization processes, the bottom-up preparation of functional materials such as liquid crystals, patterned surfaces and supramolecular polymers. In principle, the stability of H-bonded supramolecular complexes could be modulated in an indefinite number of ways. For example, when stronger interactions (e.g., higher association constant values) are required, the increase of the number of the H-bonding sites represents one of the efficient strategy to reinforce the stability of the ultimate assembly. Nevertheless, a strong Ka value is not always requested. In fact, whilst highly stable complexes are required in the field of supramolecular polymers, whose properties at the molecular level (such as degree of polymerization, Dp, and viscosity) result linearly correlated to the Ka values, these may instead be detrimental for the construction of more sophisticated hierarchized nano-architectures, arising from a delicate interplay between internal (e.g. ii stacking, solvophobic/solvophilic interactions) and external (e.g. time, temperature, concentration, etc.) factors.
The aim of this thesis is to design and synthesize novel triple H-bonding arrays (DAD, ADD and DDD) based on five-membered heteroaromatic rings. The proposed use of thiolyl, oxolyl, azolyl, and triazolyl scaffoldings for recognition systems, it is intended as a mean to better achieve the control on the binding properties and selectivity of triple H-bondind recognition arrays, allowing an easy tunability of the binding motifs. With the variation of the substituents and the heteroatom onto the hetero-aromatic rings, it has been intended to create a selection of versatile, structurally similar, host-guest pairs complexes that display different association constants (Ka) in order to better match the requirements of different supramolecular applications.
Focusing on the most relevant factors that influence the association constants of hydrogen bonded complexes, in the first part of Chapter 1 the reader is introduced on how specific H-bonding arrays, featuring wide ranges of Ka values (spanning among eight orders of magnitude) can be designed. Subsequently, the second part is focused on the physical and chemical properties of a large variety of H-bonding assembled molecular modules that upon self-assembly and self-organization processes opened new ways towards novel fascinating applications.

Chapter 2 deals with the description of the synthetic efforts undertaken towards the preparation of the DAD and DDD H-bonding arrays. The first two subsections (2.1-2) describe the rethrosynthetic approaches and the results of the unsuccessful methodological routes (through Buchwald-Hartwig amidation cross-coupling reactions, reduction of azido-derivatives and nucleophilic addition of organo-metallic reagents to isocyanate derivatives as produced through Curtius rearrangement) tackled to introduce amidic and/or ureidic functions at the 2-position of five-membered heteroaromatic rings.
Several DAD H-bonding arrays based on thiolyl scaffolding were successfully synthesized (sections 2.3).

In section 2.4 are presented the synthetic step undertaken in the attempt to generate DAD arrays based on oxalyl derivatives. Unfortunately the introduction of electron-donating groups such as amidic or carbamic functions to the ring led to very unstable intermediates, and thus the amido-oxolyl derivatives capable of recognition mediated by triple H bonding were never isolated.

The synthetic strategies towards the synthesis of DDD arrays based on of azolyl scaffolding are described in section 2.5. Protected azolyl module 182 (see Figure 4) was synthesized in thirteen steps starting from the pyrrole module. Unfortunately, due to the complications encountered in the cleavage of the N-azolyl protecting group, the synthesis of the azolyl DDD H-bonding arrays based could not be finally accomplished.

Section 2.6 presents the synthesis of newly designed self-adapting ADD/DDD H-bonding array based on ureido-triazolyl scaffoldings. Exploiting the prototropic equilibrium of the triazole nucleus the modules synthesized are expected to show an ADD or a DDD arrangement of the binding sites depending on the H-bonding functionalities of the complementary guest used for the complexation.

Due to solubility limitations in common organic solvents (e.g., CDCl3 and CD2Cl2), the molecular recognition ability in solution could not be studied and further modifications of the molecular structural properties are required.Grâce à leur directionnalité proéminente ainsi qu’à leur force, les ponts hydrogène sont l’une des interactions non-covalentes les plus exploitées en chimie supramoléculaire. Malgré leur force relativement élevée en comparaison avec d’autres interactions non-covalentes (l’énergie d’un pont hydrogène en phase gazeuse se situe typiquement entre 0 et 5 Kcal mol-1), l’association de deux molécules au moyen d’un pont hydrogène simple mène à des complexes présentant des stabilités thermodynamiques faibles, limitant ainsi leur exploitation dans la synthèse non-covalente de matériaux fonctionnels menant à de réelles applications. Dès lors, lorsque des interactions plus fortes sont nécessaires, l’approche d’ingénierie habituelle se concentre sur la synthèse covalente d’échafaudages moléculaires rigides et planaires dans lesquels plusieurs fonctionnalités donneurs (D) et accepteurs (A) de ponts hydrogènes sont arrangées dans ce que l’on appelle un ‘réseau de ponts hydrogène’. Grâce aux procédés de reconnaissances spécifiques et au contrôle de leur force d’association, les réseaux de ponts hydrogène multiples sont devenus des modules moléculaires indispensables dans la boîte à outils des chimistes supramoléculaires, permettant, via des procédés sélectifs d’auto-assemblage et/ou auto-organisation, la préparation bottom-up de matériaux fonctionnels tels que des cristaux liquides, des surfaces à motifs et des polymères supramoléculaires. En principe, la stabilité des complexes supramoléculaires à base de ponts hydrogène peut être modulée d’un nombre infini de manières. Par exemple, lorsque des interactions plus fortes (e.g., des valeurs de constante d’association élevées) sont requises, augmenter le nombre de sites de ponts hydrogène représente une stratégie efficace pour renforcer la stabilité de l’assemblage final. Néanmoins, une valeur élevée de Ka n’est pas toujours nécessaire. En effet, alors que des complexes hautement stabilisés sont requis dans le domaine des polymères supramoléculaires, dont les propriétés à l’échelle moléculaire (tel que le degré de polymérisation Dp, et la viscosité) corrèlent linéairement aux valeurs des Ka, ces dernières peuvent également être néfastes à la construction de nano-architectures hiérarchisées plus sophistiquées, résultant d’une interaction délicate entre des facteurs internes (comme par exemple du π−π stacking, des interactions solvophobiques/solvophiliques) et externes (tels que le temps, la température, la concentration etc.).
L’objectif de cette thèse est l’élaboration et la synthèse de nouveaux réseaux de ponts hydrogène (DAD, ADD, DDD) basés sur des cycles hétéroatomiques à cinq chaînons. Nous proposons l’utilisation d’échafaudages thiolyl, oxolyl, azolyl, et triazolyl comme systèmes de reconnaissance, afin de réaliser un meilleur contrôle des propriétés de liaison et de la sélectivité des réseaux de reconnaissance à ponts hydrogène triples, permettant ainsi une transformation aisée des motifs de liaison. La variation des substituants et de l’hétéroatome sur les cycles hétéroaromatiques permettrait de créer une sélection de complexes de paires host-guest versatiles et de structure similaire, présentant ainsi différentes constantes d’association (Ka) pour mieux répondre aux différentes demandes d’applications supramoléculaires.
Se concentrant sur les facteurs les plus pertinents qui influencent les constantes d’association des complexes à base de ponts hydrogène, le lecteur est introduit dans la première partie du Chapitre 1 sur la façon dont peuvent être élaborés des réseaux spécifiques à base de ponts hydrogène présentant une large gamme de valeurs de Ka (s’étendant sur un ordre de huit de magnitude). Ensuite, la deuxième partie traite des propriétés physiques et chimiques d’une grande variété d’assemblages par ponts hydrogène de modules moléculaires qui après auto-assemblage et auto-organisation ont ouvert de nouvelles voies à l’élaboration de nouvelles applications fascinantes.

Le Chapitre 2 présente la description des efforts synthétiques réalisés pour la préparation des réseaux de ponts hydrogène DAD et DDD. Les deux premières sous-sections (2.1-2) décrivent les approches rétrosynthétiques et les résultats des voies méthodologiques non fructueuses (via des réactions de cross-coupling d’amidation de Buchwald-Hartwig, réduction de dérivés azido et addition nucléophile de réactifs organométalliques à des dérivés isocyanate comme produit par le réarrangement de Curtius) entreprises pour introduire les fonctions amidiques et/ou uréidiques en position 2 des cycles hétéroaromatiques à cinq chaînons.
Plusieurs réseaux de ponts hydrogène DAD à base de thiolyl ont été synthétisés avec succès (section 2.3).

Dans la section 2.4 sont présentées les étapes synthétiques réalisées pour générer les réseaux DAD à base de dérivés oxalyl. Malheureusement, l’introduction sur le cycle de groupements électrodonneurs tels que des fonctions amidiques ou carbamiques a mené à des intermédiaires très instables. Dès lors, les dérivés amido-oxolyl capables de reconnaissance via des ponts hydrogène triples n’ont jamais été isolés.

Les stratégies synthétiques menant à la synthèse des réseaux DDD à base d’azolyl sont décrites dans la section 2.5. Le module protégé par le groupemement azolyl 182 (voir Figure 4) a été synthétisé en treize étapes à partir du module pyrrole. Malheureusement, à cause de complications rencontrées lors du clivage du groupement protecteur N-azolyl, la synthèse des réseaux de ponts hydrogènes DDD protégés par l’azolyl n’a finalement pu être réalisée.

La Section 2.6 présente la synthèse de réseaux de ponts hydrogène auto-adaptant ADD/DDD, nouvellement élaborés à partir d’échafaudages ureido-triazolyl. Grâce à l’équilibre prototropique du noyau triazole, nous nous attendons à ce que les modules synthétisés présentent un arrangement des sites de liaisons ADD ou DDD, selon les fonctionnalités ponts hydrogène des guest complémentaires utilisés pour la complexation.

Regard institutionnel sur la transposition didactique du Théorème de Lagrange en mathématiques et en économie
Because of its many uses, the constrained optimization problem is presented in most calculus courses for undergraduate mathematics and economics students. Since the Theorem of Lagrange and the consequential method of Lagrange multipliers (named after Joseph-Louis Lagrange (1736-1813)) provide an appealing strategy for finding the maxima and minima of a function subject to equality constraints, we are interested in studying the teaching of this theorem in both branches of study, mathematics and economics. In particular, this thesis aims to answer the following questions:
- What are the main characteristics of the mathematical organizations around Lagrange's Theorem?
- What important part does the proof of Lagrange's Theorem play in the teaching and understanding of Lagrange's Theorem?
- What kind of difficulties do mathematics and economics students encounter when being taught Lagrange's Theorem?
Didactic transposition and the Anthropological Theory of Didactics are our main theoretical frames and guide most of our analysis.L'optimisation joue un rôle important en recherche opérationnelle, dans les mathématiques appliquées, en analyse et en analyse numérique, en statistique, dans la recherche de stratégies optimales en théorie des jeux ou encore en théorie du contrôle et de la commande. Par ses applications, elle est un sujet central non seulement en mathématiques, mais aussi en économie. La recherche d'extremum sous contraintes d'égalité est présentée à l'université dans la plupart des cours de calcul différentiel et intégral destinés à des étudiants non seulement en mathématiques mais aussi en économie. En tant qu'enseignant face aux étudiants en sciences mathématiques mais aussi en sciences économiques, nous avons souhaité élargir le champ des recherches en didactique des mathématiques vers l'étude du Théorème de Lagrange. Nos travaux visent à apporter des éléments de réponse aux questions suivantes:
- Quelles formes d'enseignement du Théorème de Lagrange rencontre-t-on?
- Quel rôle joue la preuve du Théorème de Lagrange dans l'enseignement mais aussi dans l'apprentissage du théorème de Lagrange?
- À quelles difficultés un étudiant en mathématiques ou en économie est-il confronté lorsque le Théorème de Lagrange lui est enseigné?

Essays on Family Farms and Inheritance
This collection of essays, entirely based on first hand data, brings some new contributions to the fascinating question of the evolution of family structure in poor agrarian societies. Family is a rather complex concept because its composition and role vary from one place to another and, above all, because it gathers human beings who interact in many ways. Relationships between family members are of various kinds. Belonging to a family of any type implies emotional and affective links as well as relations of interest and power, between persons of the same status and also among members from different hierarchical levels.
Families are micro societies that vehicle values and social norms. “One of the fundamental roles played by parents is that of transmitting distinct cultural traits to their children. These traits include elements of preferences such as the degree of altruism, risk aversion, attitude towards fertility and labour force participation, religious traits, etc.” (La Ferrara, 2011). Another important role played by older generations is the transmission of wealth, so that the division of assets also influences the intergenerational transmission of inequality. Furthermore, inheritance patterns are crucial elements in contexts where access to land in severely restricted or where land market is not or hardly developed. The questions of how and when parental wealth is transferred to children are, therefore, of high interest since it can shape the evolution of the future generations.
The first two chapters tackle the issue of land inheritance patterns using first hand data collected in the Peruvian highlands. Andean peasants are among the poorest in Latin America, they mainly live on land which they access through inheritance. These communities are also characterized by a high migration rate and a very low level of education. In this area, the sense of belonging to a family is closely linked to the relation people have with their home land (the “Pacha Mama”), with their native community. A better knowledge of inheritance practices is of importance in the study area where rural land market barely exists and which suffers from severe problems of land fragmentation.
In Chapter 1, the land division pattern is analyzed through the estimation of an inheritance function. One of the main results is that a child’s caring attitude towards his/her parents has a positive effect on the access to land bequest, especially for migrant children. We also argue that potential heirs play an active role in the determination of the inheritance outcomes.
In Chapter 2, we explore, theoretically and empirically, the influence of migration and parental land endowment on the probability for children to obtain their inheritance in the form of inter vivos transfers, post-mortem bequests, or a combination of both. First, we find that migrant children tend to receive their entire land inheritance post-mortem. Second, we observe that (remaining) children from more wealthy families have a higher probability to obtain their inheritance in the forms of both inter vivos transfers and bequests, while (remaining) children from poorer families tend to obtain it post-mortem.
In the third and last chapter, we study another aspect of the family structure in a completely different society. On the basis of first hand data collected in the San-Koutiala-Sikasso region in Mali, we compare land productivity between collective fields and plots that are individually cultivated by male or female members of the household. We do not focus on the inheritance pattern anymore, yet the individual plots can somehow be considered as inter vivos gifts of land.
During our field work in the old cotton zone in Mali, we were puzzled by the co-existence of various farm-cum-family structures within a same village. We could find large traditional extended families co-existing with smaller ‘nuclear’ families, on the one hand and, purely collective farms co-existing with mixed farms (farms in which there exist collectively cultivated fields as well as individually cultivated plots), on the other hand.
Inspired by these observations, many research ideas came up and three companion research papers have been written (until now). First, Guirkinger and Platteau (2011a) developed a theoretical model to explain the evolution of large purely collective farms into mixed farms and/or into smaller collective farms (split). Assuming that collective production is plagued with moral-hazard-in-team problem, they show that “as land scarcity increases, or as exit options available to family members improve, the pure collective farm will unavoidably become inferior to alternative farm structures from the standpoint of the family head who draws his entire income from a share of the collectively produced harvest. The intuition is that, when land becomes scarcer, the head has to give more weight to efficiency considerations compared to his rent-capturing ability. This is because he has to satisfy the members' participation constraints under harsher conditions than before”.In a second paper, the same authors (2011b) empirically confirmed one of the predictions of their theoretical model: “increasing land scarcity prompt household heads to give individual plots of land to (male) members”.
In a third paper (which happens to be the third chapter of this thesis), we compare land productivity between plots controlled by different members of the household by using detailed information on input and output data. Firstly, we find that land yields are significantly larger on (male) private plots than on common plots with similar characteristics planted to the same crop in the same year after all appropriate controls have been included. And secondly, we bring strong suggestive evidence that a moral-hazard-in-team problem exists on the collective fields (yet only with regard to care-intensive crops) that could explain their relatively poor performance. This last result tends to confirm the rightfulness of the assumptions used in the theoretical model.
Despite data limitation, some of the results presented in this thesis offer new insights into the modus operandi of families in two different poor areas. The understanding of the way family members interact is crucial so as to design appropriate poverty reduction policies.(DOCSESG00) -- FUNDP, 201
