Transcriptome Analysis in Tardigrade Species Reveals Specific Molecular Pathways for Stress Adaptations
No full textTardigrades have unique stress-adaptations that allow them to survive extremes of cold, heat, radiation and vacuum.
To study this, encoded protein clusters and pathways from an ongoing transcriptome study on the tardigrade Milnesium tardigradum were analyzed using bioinformatics tools and compared to expressed sequence tags (ESTs) from Hypsibius dujardini, revealing major pathways involved in resistance against extreme environmental conditions.
ESTs are available on the Tardigrade Workbench along with software and databank updates. Our analysis reveals that RNA stability motifs for M. tardigradum are different from typical motifs known from higher animals. M. tardigradum and H. dujardini protein clusters and conserved domains imply metabolic storage pathways for glycogen, glycolipids and specific secondary metabolism as well as stress response pathways (including heat shock proteins, bmh2, and specific repair pathways). Redox-, DNA-, stress- and protein protection pathways complement specific repair capabilities to achieve the strong robustness of M. tardigradum.
These pathways are partly conserved in other animals and their manipulation could boost stress adaptation even in human cells. However, the unique combination of resistance and repair pathways make tardigrades and M. tardigradum in particular so highly stress resistant
Using a priori knowledge to align sequencing reads to their exact genomic position
No full textThe use of a priori knowledge in the alignment of targeted sequencing data is investigated using computational experiments. Adapting a Needleman–Wunsch algorithm to incorporate the genomic position information from the targeted capture, we demonstrate that alignment can be done to just the target region of interest. When in addition use is made of direct string comparison, an improvement of up to a factor of 8 in alignment speed compared to the fastest conventional aligner (Bowtie) is obtained. This results in a total alignment time in targeted sequencing of around 7 min for aligning approximately 56 million captured reads. For conventional aligners such as Bowtie, BWA or MAQ, alignment to just the target region is not feasible as experiments show that this leads to an additional 88% SNP calls, the vast majority of which are false positives (∼92%)

No full textDer Themenkomplex Klimawandel war traditionell ein Arbeitsgebiet der Naturwissenschaftler. Die verschiedenartigen Anpassungsprozesse, die durch Klimawandel in Gang gesetzt werden, führen allerdings zu starken soziokulturellen Wandlungsprozessen, die ihre Auswirkungen auf die Kulturgemeinschaften weltweit bereits in der Gegenwart haben. Für die Klimaforschung wird eine verstärkt interdisziplinäre Zusammenarbeit immer mehr zur Notwendigkeit, um die komplexen Klimawandelprozesse zu erfassen. In dem vorliegenden Beitrag sollen einige kommunikative Aspekte des Klimawandeldiskurses kurz angeschnitten werden, um aufzuzeigen, dass der durch Klimawandel bedingte Kulturwandel für die Kulturwissenschaft Anknüpfungspunkte bietet und sich zu einem Forschungsfeld der Zukunft entwickeln kann.The complex of topics about climate change has been traditionally the domain of natural scientists. However miscellaneous processes of adaptation caused by climate change lead to deep socio-cultural transformations already impacting cultural communities worldwide. Climate research makes tightened interdisciplinary cooperation necessary in order to capture complex processes of climate change. The present article will address the communicative aspects of the climate change discourse to show that cultural change caused by climate change offers a link for the cultural studies and it may evolve into a field of research in the future

Avoiding gender bias during measurement of individual research output of scientists – adaption of an analytical concept developed for health care research
No full textFor the Framework Research Program 7 funded project »Academic Careers Understood through Measurement and Norms« (ACUMEN), addressing the current discrepancy between the broader social and economic functions of scientific and scholarly research in all fields of the sciences, social sciences and the humanities and the dominant criteria for evaluating performance by researchers, it was necessary to have a concept what allowed to substantiate the gender dimension during the different research activities. The present paper shows the steps adapting a methodological oriented concept, developed systematically for health care studies to the other subject. It summarizes some points identifi ed as possibly relevant and adapted especially for surveys and the qualitative interviews to know more about measuring of scientifi c output of scientists. The deduced procedures and advices are structured with regard to the research concept, the research design/questions, the methods and data collection, the data analysis and interpretation just like title, abstract, diction and visualization of reports as several research project steps. By using the adapted concept were deduced model questions for surveys and interviews, which are planned by ACUMEN partners.Das im 7. Forschungsrahmenprogramm geförderte Projekt »Academic Careers Understood through Measurement and Norms« (ACUMEN) zielt auf die Aufhebung der Diskrepanz zwischen den breiter gewordenen sozialen und wirtschaftlichen Funktionen von Forschung und Lehre in allen Wissenschaftsbereichen und den gegenwärtig genutzten Kriterien für die karriererelevante Leistungsbewertung von Forschern. Um die Geschlechterdimension bei den sehr unterschiedlichen Forschungsaktivitäten aller Arbeitspakete des Projekts zu berücksichtigen, wurde ein methodologisch orientiertes Konzept, welches systematisch für die Gesundheitsforschung entwickelt wurde, in zwei Schritten auf den Forschungsgegenstand »Messung des individuellen wissenschaftlichen Outputs« übertragen. Der vorliegende Artikel fasst die wesentlichen – als relevant identifizierten und angepassten – Leitlinien zusammen. Diese werden mit Bezug auf Forschungskonzept, Forschungsdesign/-fragen, Methoden und Datenerfassung, Datenanalyse und Interpretation sowie Titel, Abstract, Diktion und Ergebnis-Visualisierung als wesentliche Schritte der Projektbearbeitung dargestellt. Unter Nutzung dieser Leitlinien des adaptierten Konzepts werden für die in ACUMEN geplanten Befragungen und Interviews modellhaft Fragen formuliert
