Konzeption und prototypische Umsetzung eines mit standortbasierten Informationen arbeitenden Systems für die Nutzer des Philologicums der Ludwig-Maximilians-Universität München
Ziel der Masterarbeit ist es ein, mit standortbasierten Informationen arbeitendes System für die Nutzer des Philologicums der Ludwig-Maximilians-Universität München zu konzipieren und prototypisch umzusetzen. Fundament eines solchen Systems muss eine valide Positionsbestimmung unter Beachtung der gegebenen Rahmenbedingungen sein. Neben der Vermittlung theoretischer Grundlagen, die das Thema „Indoor Positioning System“ tangieren, ist ein wesentlicher Schwerpunkt der Arbeit den aktuellen Stand der Technik, im Hinblick auf die Positionsbestimmung in Innenräumen zu eruieren.
In einer kaskadierenden und durch praktische Analysen untermauerten Evaluation verschiedener Positionierungstechnologien ist schließlich Optical Character Recognition, als Mittel der Wahl bestimmt worden. Basierend auf den Ergebnissen der Anforderungs- und Zielgruppenanalyse sowie den gewonnen Erkenntnissen zu „Location-based Services“ ist ein, in der Form im Bibliothekswesen neuartiges System konzipiert und entwickelt worden.
Die im Rahmen der prototypischen Umsetzung entstandenen Komponenten demonstrieren anschaulich die Funktionsweise und Machbarkeit eines Systems, dass mit standortbasierten Informationen arbeitet. Kernelement ist zum einen die Webanwendung „LMU-Linker“ als Middleware, im Sinne eines verknüpfenden Bindeglieds zwischen Point of Interests und dazugehörigen Inhalten. Zum anderen ist das Front-end, die auf Android basierte App „Philologicum“, zur Bestimmung des Standortes und zur Anzeige der verknüpften Inhalte, essentieller Bestandteil des Prototyps.
Die Masterarbeit soll als Auftragsarbeit die Grundlage für den zukünftigen Produktiv-betrieb des Systems, im entstehenden Philologicum, bilden. Die gewonnen Erkennt-nisse können jedoch gleichermaßen auch für potentiell Interessierte, mit Fokus auf den Themenbereichen „Indoor Positioning“ und „Location-based Services“, aufschlussreich sein
Ein Knowledge Graph für wissenschaftliche Sammlungen : Generierung von Linked Open Data für heterogene museale Sammlungen auf der Basis des ASCH-Modells
Die vorliegende Arbeit liefert eine prototypische Anwendung zur besseren Sichtbarkeit und Verfügbarkeit musealer Sammlungen im Semantic Web.
Es wird ein Verfahren vorgestellt, das die LIDO-XML-Metadaten, die die Sammlungsobjekte der wissenschaftlichen Sammlungen der Georg-August-Universität Göttingen beschreiben, in Linked Open Data (LOD) umwandelt und in einem Triplestore bereitstellt. Der dargestellte Ansatz sowie die Umsetzung eignen sich zur Nachnutzung auch durch kleinere museale Einrichtungen ohne vertiefte Semantic-Web-Erfahrung.
Als Basis für die Generierung der RDF Statements dient das ASCH-Modell, ein unlängst entwickeltes RDF-Datenmodell für heterogene museale Sammlungen, dessen Proof of Concept ein weiteres Ziel dieser Arbeit darstellt.
Für die Datenanalyse werden die Quelldaten in eine XML-Datenbank importiert und mit XPath und XQuery untersucht. Die Konvertierung der Daten in das Serialisierungsformat RDF/XML erfolgt anhand eines XSLT-Stylesheets. Außerdem werden die Daten in das RDF-Format Turtle überführt und in einen mit SPARQL abfragbaren Triplestore geladen.
Die für die Umsetzung notwendigen Komponenten der Abfrage- und Programmiersprachen sowie der Tools werden anhand von Beispielen erläutert.
Die Bereitstellung des Knowledge Graph erfolgt unter Beachtung der Linked Data Principles und der vom World Wide Web Consortium (W3C) veröffentlichten Best Practices for Publishing Linked Data. Ferner finden die besonderen Anforderungen des Kulturerbesektors, insbesondere der wissenschaftlichen Sammlungen, Berücksichtigung.This thesis provides a prototypical application for better visibility and availability of museum collections in the Semantic Web.
A method is presented that transforms the LIDO-XML metadata, which describe the objects of the academic collections of the Georg-August-University Göttingen, into Linked Open Data (LOD) and publishes it in a triplestore. The illustrated approach and the implementation are suitable for reuse also by smaller museums without deepened Semantic Web experience. The basis for the LOD generation is the ASCH model, a recently developed RDF data model for heterogeneous museum collections. Its proof of concept is another goal of this thesis.
For data analysis, the source data is imported into an XML database and examined with XPath and XQuery. The data conversion into RDF/XML is realized by an XSLT style sheet. In addition, the data is converted into Turtle and published in a triplestore enabling networking with external databases by means of SPARQL.
The components of the query and programming languages and the tools necessary for the implementation are explained by examples.

Electrochemical Activity Determination of Catechol-O-methyl Transferase by Selective Dopamine Detection
For the treatment of Parkinson's disease, as one of the most frequent diseases of the central nervous system, several key enzymes for dopamine metabolism [e. g. catechol-O-methyl transferase (COMT)] are drug targets. For an efficient and long-lasting treatment, the activity of this enzyme should be monitored. In this study, an electrochemical approach using differential pulse voltammetry (DPV) is introduced for the activity determination. The applied electrode material, fluorine-doped tin oxide (FTO), is characterized by a clear discrimination between substrate and product of COMT, a high stability of the dopamine signal during consecutive measurements, and a linear dependency on the dopamine concentration in the range of the maximum reaction rate of COMT. Despite these advantageous results, dopamine detection in the complete activity assay is influenced by each of the added essential assay components, even though none of the added components reveal a current signal at the FTO electrode itself. After adjusting the potential range and the assay composition, these effects can be circumvented. By following the dopamine concentrations during COMT action, it can be shown that the activity of COMT can be detected by using differential pulse voltammetry (DPV) at an FTO electrode and, by analyzing different COMT amounts, quantification can be demonstrated
Evaluierung der Potenz von glykosylierten Impfstoffen am Beispiel des RSV-F Proteins
Trotz intensiver Forschung und immensen Bedarfs steht heutzutage kein RSV-Impfstoff zur Verfügung. Moderne Subunit-Vakzine bedürfen einer Adjuvantierung, um eine ausreichend starke Immunantwort zu generieren. Mit der Veränderung von Glykoslierungsmustern des RSV-F Proteins wird eine kovalent gekoppelte Adjuvantierung hergestellt, die über die Aktivierung des angeborenen Immunsystems eine umfassende Immunantwort generiert. In dieser Arbeit wurden rekombinant hergestellte Varianten des RSV-F Proteins mit defukosylierten oder xylosylierten N-Glykanen auf ihr Potential, das Immunsystem zu aktivieren, untersucht. Dazu wurden Zytokine aus Zellkulturüberständen quantifiziert und Methoden zur Genexpressionsanalyse stimulierter Zellen etabliert. In Vorbereitung auf Stimulationsexperimente mit PBMCs wurde der Einfluss der Kryokonservierung auf Zellen untersucht und die Zytokinexpression auf Gen- und Proteinebene korreliert. Im Rahmen der Stimulationsexperimente mit komplexen Zellgemischen (PBMC und mDC) wurden die stabil exprimierten Referenzgene SDHA, TBP und PPIA für die Etablierung einer validen Genexpressionsanalyse identifiziert. Dabei wurde eine mathematische Konvertierung quantitativer Transkriptzahlen aus dem Sonden-basierten direkten RNA-Nachweis (NanoString®) und der relativen Quantifizierung mittels qPCR entwickelt. Die Zytokinmuster in Zellkulturüberständen aus Langzeitstimulationsexperimenten, welche mit PBMC, mDC und den beiden Impfstoffvarianten in einem komplexen Perfusionsreaktorsystem (HuALN®) durchgeführt worden waren, wurden mittels automatisierter Bead-basierter Multiplexanalytik quantifiziert. Im Zusammenhang mit der Untersuchung umfangreicher Genpanels konnten verstärkte Immunreaktionen durch die glykosidisch veränderten RSV-F Proteine nachgewiesen werden. Die Defukosylierung führt zu einer breiten Aktivierung des zellulären und humoralen adaptiven Immunsystems. Das xylosylierte RSV-F aktiviert zusätzlich Komponenten des angeborenen Immunsystems. Zudem gibt es Hinweise darauf, dass durch den Grad der Defukosylierung das Verhältnis zwischen Th1 und Th2 basierter Antwort modifiziert werden kann. Mit den verwendeten und etablierten Methoden ist es möglich, die Potenz von Impfstoffkandidaten mit geringen strukturellen Veränderungen zu unterscheiden. Um die komplexen Stimulationsexperimente in einem höheren Durchsatz durchführen zu können, wurde an der Entwicklung eines miniaturisierten Bioreaktors mitgewirkt. Die Untersuchungen der Dichtheit des Reaktors mittels Fluoreszenzmikroskopie und der Biokompatibilität der einzelnen Komponenten hatten direkten Einfluss auf das Design des Reaktors, dessen Aufbau- und Verbindungstechnik sowie die Wahl der Materialien. Die Funktionalität konnte in verschiedenen 2D und 3D Kultivierungsansätzen über eine langanhaltende Zellviabilität bestätigt werden. Die verstärkte Modulierung der Immunantwort, hervorgerufen durch modifizierte Glykanstrukturen, zeugt von einer aussichtsreichen Methode, die auf andere Pathogene angewandt werden kann. Das höhere Durchsatzpotential des miniaturisierten Perfusionsreaktors (mehr Zeitpunkte, mehr Replikate) kann zukünftig einen präziseren Einblick in die immunologischen Vorgänge gewähren.Despite intense research and immense demand there is no RSV vaccine available today. Modern subunit vaccines require adjuvants to generate a sufficiently strong immune response. By editing the glycosylation pattern of the RSV-F Protein covalently coupled adjuvants are produced, which generate comprehensive immune responses via the activation of the innate immune system. In this work recombinantly produced variants of the RSV-F protein with defucosylated or xylosylated Nglycans were tested for their potential to activate the immune system. For that, cytokines from cell culture supernatants were quantified and methods for gene expression analysis of stimulated cells were established. In preliminary PBMC stimulation experiments the influence of cryopreservation on cells was examined and the expression of cytokines on gene and protein level was correlated. Within the scope of stimulation experiments with complex cell mixtures (PBMC and DC) the reference genes SDHA, TBP and PPIA were identified as stably expressed in order to establish a valid gene expression analysis. A mathematical conversion of quantitative transcript numbers from a probe-based direct RNA detection assay (NanoString®) and the relative quantification via qPCR was developed. The cytokine pattern in cell culture supernatants from long-term experiments performed with PBMC, mDC and each vaccine candidate in a highly complex perfusion reactor system (HuALN®) were quantified by automated bead-based multiplex analytics. In connection with the analysis of broad gene panels increased immune responses could be detected caused by the glycosidically altered RSV-F proteins. Defucosylation leads to a comprehensive activation of the immune system. Xylosylated RSV-F additionally activates components of the innate immune system. Furthermore, there is evidence that the ratio of the Th1 and Th2 based immune response can be modified by the grade of the defucosylation. The applied and established methods are suited to distinguish the potency of vaccine candidates with minor structural differences. In order to perform stimulation experiments in a higher throughput, this work contributed to the establishment of a miniaturized bioreactor. Leakage testing of the reactor via fluorescence microscopy and the examination of the biocompatibility of all single components directly influenced the reactor design, its assembly and connection techniques as well as the choice of materials. The reactors functionality was confirmed in different 2D and 3D cultivation approaches by means of a long-lasting high cell viability. The stronger modulated immune response evoked by modified glycosidic structures provides a promising method that can be applied to other pathogens, too. By its potential for higher throughput (more time points, more replicas) the miniaturized perfusion reactor could be able to provide a more precise insight into the immunological processes
Implementing Augmented Reality in the Flight Deck for Single Pilot Operations
Single Pilot Operations is a current topic with the potential to significantly affect the future of commercial aviation. While financially attractive for airlines, Single Pilot Operations bring forth important safety concerns, especially regarding the lack of human redundancy in the flight deck, an increased workload for the single pilot, reduced situational awareness and a higher risk of human error.
It is assumed that potential problems affecting Single Pilot Operations could be addressed by implementing an Augmented Reality (AR) device in the flight deck, by presenting additional information and supporting hints within the pilot’s field of view. Concretely, AR could be used to help reduce the single pilot’s workload, improve situational awareness and reduce the risk of human error.
This paper sets out to demonstrate two use cases for augmented reality in the flight deck. A system, called Pilot Assist, was developed that allows pilots to conduct checklists interactively with a Microsoft HoloLens. The system also provides a holographic Head-up-Display. Pilot Assist was developed and demonstrated with a fixed base Airbus A320 simulator at the Technical University of Wildau.
With the HoloLens’ spatial mapping capabilities – scanning and recognizing the environment around the user – it was possible to create a system that guides the pilot through the conduction of checklists. This is done by prompting the user towards the location of each checklist item in the cockpit, where information regarding necessary actions is projected. Furthermore, Pilot Assist is integrated with the aircraft systems, making it possible to obtain aircraft status data in real time, thus allowing error-checking of the pilot’s actions as well as automating the progress through checklists.
The holographic Head-up-Display allows the user to look at the surrounding environment while presenting critical flight data within the user’s field of view. The holographic Head-up-Display is intended to contribute to the pilot’s situational awareness.
Experts in the aviation field, including pilots, researchers and engineers had the chance to qualitatively assess the Pilot Assist tool. They pointed to limitations of both Pilot Assist and the HoloLens itself, but shared optimism as to how this technology and similar applications could indeed impact the future of flight operations. Concerns regarding the HoloLens’ weight, comfort and narrow field of view were expressed. However, continued development of head mounted devices (e.g. HoloLens 2) is expected in the coming years.
Further research into augmented reality applications in the flight deck is needed to advance this and other use cases. Nonetheless, the experts agreed Pilot Assist provides beneficial support during single pilot operation considering the current prototypical nature of the system

A Novel Optical Method To Reversibly Control Enzymatic Activity Based On Photoacids
Most biochemical reactions depend on the pH value of the aqueous environment and some are strongly favoured to occur in an acidic environment. A non-invasive control of pH to tightly regulate such reactions with defined start and end points is a highly desirable feature in certain applications, but has proven difficult to achieve so far. We report a novel optical approach to reversibly control a typical biochemical reaction by changing the pH and using acid phosphatase as a model enzyme. The reversible photoacid G-acid functions as a proton donor, changing the pH rapidly and reversibly by using high power UV LEDs as an illumination source in our experimental setup. The reaction can be tightly controlled by simply switching the light on and off and should be applicable to a wide range of other enzymatic reactions, thus enabling miniaturization and parallelization through non-invasive optical means
Quantum Dot-modifizierte TiO2-Strukturen für die Licht-gesteuerte Bioelektrokatalyse
Die funktionale Kopplung von photoaktiven Nanostrukturen mit Enzymen stellt eine neue Strategie zum Aufbau lichtgesteuerter biohybrider Systeme dar. Hier sind Untersuchungen zusammengefasst, welche die effiziente Kontaktierung der FAD-abhängigen Glukosedehydrogenase (FAD-GDH) mit Hilfe eines Osmium-Redoxpolymers (P Os ) an PbS-Quantum Dots (PbS QDs) zeigen, welche direkt auf dreidimensionalen TiO 2 -Elektrodenstrukturen synthetisiert wurden. Diese biohybriden Strukturen erlauben die Licht-induzierte Oxidation von Glukose. Dazu wird zunächst ein Verfahren vorgestellt, bei welchem durch den Aufbau invers-opaler TiO 2 (IO-TiO 2 ) Strukturen hohe Bindungskapazitäten für die Integration von QDs, Redoxpolymer und Enzym erreicht werden. In Folge wird gezeigt wie elektrochemische Signalketten durch Licht gesteuert werden können, indem Ladungsträger in den QDs unter Beleuchtung erzeugt werden. Diese Aktivierung ermöglicht dann die Ausbildung einer Elektrontransferkaskade vom Enzym über das Redoxpolymer zu den QDs und final zur IO-TiO 2 -Elektrode. Die resultierenden anodischen Photoströme können durch das Potential, die Lichtintensität und die Glukosekonzentration moduliert werden. So können in Anwesenheit von Glukose Photoströme von bis zu 207 μA/cm2 und erste Oxidationssignale bereits bei einem Potential von -540 mV vs Ag/AgCl, 1 M KCl erhalten werden. Dies entspricht einem Potentialgewinn von über 500 mV im Vergleich zu nicht lichtsensitiven Elektroden. Das vorgestellte biohybride System kombiniert Vorteile einer großen Oberfläche (durch IO-TiO 2 -Struktur), die effiziente Ladungsträgergenerierung und -trennung an der QD/TiO 2 -Schnittstelle sowie die effiziente Kontaktierung von FAD-GDH mit den QDs mit Hilfe eines Redoxpolymers. Die Ergebnisse verdeutlichen das Potential dieser leistungsfähigen Photobioanode für die Sensorik und die Erzeugung von Energie aus Licht und Glukose.The combination of photoactive nanostructures with enzymes represents a new strategy for the construction of light-directed biohybrid systems. The study demonstrates the efficient linkage of FAD-dependent glucose dehydrogenase (FAD-GDH) to PbS quantum dot (QD)-sensitized inverse opal TiO 2 (IO-TiO 2 ) electrodes using an osmium redox polymer (P Os ). How these biohybrid structures can oxidize glucose in a light-directed fashion is highlighted in detail. For this purpose, firstly a method for the construction of IO-TiO 2 electrodes is presented, which gives rise to high binding capacities for the integration of QDs, redox polymer and enzyme. It can be shown how electrochemical signal chains can be controlled with light, resulting in a generation of charge carriers within the QDs. The activation of the electron transfer cascade then allows for an electron transfer from the enzyme via the redox polymer to the QDs and finally to the IO-TiO 2 electrode. The resulting anodic photocurrents can be modulated by potential, light intensity and glucose concentration. In the presence of glucose, photocurrents of up to 207 μA/cm 2 can be generated and first electron withdrawal from the biocatalytic
sugar oxidation already proceeds at a potential of -540 mV vs Ag/AgCl (1 M KCl). This corresponds to a potential gain of over 500 mV compared to light-insensitive electrodes. The proposed biohybrid system combines the advantages of a large surface area (IO-TiO 2 ), efficient charge carrier generation (PbS QDs) and separation at the TiO 2 /QDs interface and the efficient linkage of FAD-GDH to the QDs using a redox polymer. This illustrates the potential of this powerful photobioanode for sensing and power supply
Re-Emerging Foci of Visceral Leishmaniasis in Armenia – First Molecular Diagnosis of Clinical Samples
Visceral leishmaniasis (VL) was firstly reported in Armenia in 1913. Following a considerable increase of the number of cases until the mid 1950s, the diseases disappeared after 1969 and re-emerged in 1999. Scientific literature about VL in Armenia is available only in Russian or Armenian. This paper presents a historical overview about leishmaniasis in Armenia based on this literature as well as an epidemiological update since the re-emergence of the disease. In 1999- 2016, 116 indigenous VL cases were recorded mainly in children in 8 of the 11 districts, however VL is underreported because of lack of trained medical personal and diagnostic facilities. The aim of this work was to apply for the first time molecular diagnosis of VL in Armenia. Out of 25 VL suspected patients, 22 were positive by microscopy and PCR. Genotyping using ITS1-PCR-RFLP and sequencing identified the causative agent of VL in Armenia as Leishmania infantum. The present work is an important step towards the inclusion of molecular techniques in the current diagnosis of VL in Armenia and the establishment of local molecular diagnostic facilities
Richterwahl und Richteramt in einem künftigen Multilateralen Investitionsgericht: zentrale Regelungsfragen
Das aktuelle globale Investitionsschutzsystem ist ökonomisch notwendig, aber reformbedürftig. Ein Hauptkritikpunkt am jetzigen Zustand ist die Verfahrensführung durch ad-hoc-Schiedsgerichte, deren Neutralität und Sachkompetenz angezweifelt wird. Deshalb wird u.a. von der Europäischen Union die Etablierung eines ständigen Multilateralen Investitionsgerichts angestrebt. Der Beitrag analysiert zentrale regulatorische Optionen für den Auswahlprozess und die Ausgestaltung der Amtsführung der Richter eines künftigen Multilateralen Investitionsgerichts.The current global system of investment protection is economically necessary but in need of reform. A major critique of the present system is the conduct of proceedings by ad hoc arbitral tribunals, whose neutrality and competence are contested. This is why the European Union, among others, is advocating the establishment of a standing Multilateral Investment Court.
