Design, synthesis and structural evaluation of peptidomimetics with a defined secondary structure.
The efficient synthesis of a g-butyrolactone based d-amino acid was realised starting from furan methyl ester.
Its enantioselecive cyclopropanation by copper triflate and a chiral bis-oxazoline led to the formation of an enantiopure cyclopropanated furan product which could be subjected to ozonolysis affording a cyclopropane aldehyde having three chiral centers. A Sakurai allylation was performed efficiently in high d.r. The lactonisation of the allylated cyclopropane alcohol could be realised with barium hydroxide. This reaction could be scaled up using triethylamine in the same conditions. The reductive amination of the resulting
lactonaldehyde could be realised in good yields in mild conditions. The Boc protection of the secondary amine and the PMB cleavage were performed affording the lactone with a protected primary amine. The
allylic double bond could be oxidised by ruthenium chloride and sodium periodate affording the d-amino acid 199 in good yields.
Modification of the lactone by a-substitution was investigated to obtain a better NMR dispersion. The a-methylation with LDA and methyl iodide led to poor diastereoselectivities (d.r. 3/1). The a-fluorination was not diastereoselective as well (d.r. 2/1) and yields were poor. The a-dimethylation of the lactone could be
performed in good yields and the resulting d-amino acid showed an excellent NMR dispersion for further foldamers studies.
The liquid-phase synthesis of the desired tetramer of dimethylated d-amino acid could not be performed, even by using efficient coupling reagents. The synthesis of mixed a-d-peptides with the non-methylated building block was realised. First, an alternated a-d-pentapeptide containing L-Phe and the lactone building block was efficiently synthesised and its secondary structure analysed by various techniques. Despite
overlapping of some protons in NMR spectroscopy, some NOE contacts could be observed and allowed computational studies using NOE constraints. The calculations predicted an extended loop conformation of
the pentapeptide and other analyses such as IR in solution or CD spectroscopy are in favour of intramolecular hydrogen bonding. CD spectra refences missing for that type of alternated peptide, no comparison could be done with similar peptides. The secondary structure could not be determined with
certainty and attemps to crystallise the peptide in a stable form failed.
Secondly, a heptapeptide containing only one d-amino acid was synthesised in order to know whether this building block could induce hairpin secondary structures. IR in solution revealed the presence of
intramolecular hydrogen bonding and CD spectroscopy showed interesting features suggesting the presence of a stable secondary structure although its CD spectrum looked a little disorganised. Its 2D-NMR analysis revealed interstrand contacts and showed that this heptapeptide was forming, among other things, a 13-membered hydrogen-bonded ring. As a consequence, we came to the conclusion that it did not adopt a
hairpin conformation but a disorganised helicoidal structure.
For the second part of this project, the ability of cis-DKP in b-bend ribbon structure was investigated. The trimer of DKP could be synthesised and fully characterised and revealed a partial b-bend ribbon conformation. The tetramer, on the other hand, could not be fully characterised by 2D-NMR, but its CD spectrum showed the presence of a stable secondary structure and comparison with other b-bend ribbon CD
spectra in the literature showed a similar behaviour in terms of shapes and intensities. The CD data collected, in addition to proton-deuterium exchange analysis are in favour of a stable b-bend ribbon secondary structure.
A short cyclic peptide based on a trans-DKP scaffold was successfully synthesised and fully characterised by 2D NMR spectroscopy. The presence of a g-turn in the peptide was detected and confirmed preliminary computational studies realised on a similar peptide. It is very probable that a pentapeptide containing a RGD sequence and the trans-DKP scaffold would as well induce a g-turn, whose presence is primordial for its activity against integrins avb3.
Finally, two potential tripeptidic organocatalysts were synthesised, each of them containing a cis-DKP and being linked by an amide or an ester bond to a proline. Their investigation in organocatalysed aldol reaction and Michael addition is now in progress
Structural determinants of substrate specificity and Cl-role in two invertebrate cotransporters.
The two highly homologous neutral amino acid transporters KAAT1 and CAATCH1, cloned from the midgut epithelium of the larva of the lepidoptera Manduca sexta have been classified as members of the Na+/Cldependent transporter family (SLC6A gene family) (9; 11). These two cotransporters represent an interesting tool for investigating structurefunction relationships concerning ion and substrate selectivity in the SLC6 transporter family. At variance with the strict Na+ dependence of known vertebrate transporters, KAAT1 and CAATCH1 are able to exploit in addition a K+ electrochemical gradient to power up the uptake of neutral amino acids from the intestinal lumen.
In fact, KAAT1 and CAATCH1 give rise to specific kinds of current depending on the transported amino acid, the cotransported ion, pH and the membrane voltage. Transportassociated currents with their own characteristics are induced by different organic substrates, which are notably distinct between the two proteins. Differences in amplitude, kinetics and voltagedependence of the transportassociated
currents have been observed, as well as different substrate selectivity patterns, measured by radioactive amino acid uptake assays (9, 4, 23 ) In this work the specificity of KAAT1 was compared to that of CAATCH1, especially with respect to leucine, which in physiological conditions is transported by KAAT1, but not by CAATCH1, where it acts as a blocker. In order to identify the determinants involved in these phenomena, we have mutated the single different residue between the two transporters, the serine 308, that corresponds to that S290 participating in the leucine binding site of LeuTAa, a bacterial member of the SLC6A family of ioncoupled cotransporters (19). This amino acid is conserved only in the members of the family able to transport leucine ( 3), and its substitution in KAAT1 with threonine, blocks the current induced by leucine in sodium, and causes a reduction of about 70% of the current in potassium. The behaviour of CAATCH1 is mimicked by the S308T mutant form of KAAT1, that indicates the participation of this residue in the leucine binding site. The reverse mutation T308S in CAATCH1 conferred to this transporter the ability to transport leucine in presence of K+. The substrate specificity of KAAT1 has been investigated using electrophysiological and radiotracer methods. Competition experiments between different substrates indicate that both transporters bind leucine more strongly than threonine and proline, the difference between KAAT1 and CAATCH1 residing in the incapacity of the latter to complete the transport cycle in presence of leucine. These results may be interpreted by a kinetic scheme in which in presence of Na+ the leucinebound state of the transporter is relatively stable, while in presence of K+ and at negative potentials the progression of the leucinebound form along the cycle is favoured. In this context serine 308 appears to be important in allowing the change to the inwardfacing conformation of the transporter, rather than in determining the specificity of leucine binding. Recent homology modeling work, based on the atomic structure of LeuTAa (19), has pinpointed the possible site of interaction of these proteins with chloride ions (25; 26 ). However, not all transporters in this family are Cldependent, and this difference appears to be due to the presence (or absence) of a critical negative charge in this region. This charge which might be provided either by the intrinsic glutamate side chain in the chlorideindependent transporters, or by a chloride ions in those transporters that have a nonconservative substitution in this position. The different chloride sensitivity observed in KAAT1 and CAATCH1, in spite of an identical sequence in the putative chloride binding site prompted us to re-examine this point. We have then performed electrophysiological experiments in which chloride was replaced either by iodide or by gluconate. Significant transportassociated current reductions were seen with gluconate replacement, while strong potentiation was induced by iodide. These observations indicate that for KAAT1 and CAATCH1 complete chloride dependency can be ruled out
Synthesis of axially chiral molecules and their applications to catalytic enantioselective reactions mediated by transition metals.
Axial chirality is a term used to refer to steroisomerism resulting from the non-planar arrangement of four groups in pairs about a chiral axis, so that the resulting spatial arrangement is not superimposable on its mirror image. It is exemplified by allenes and by atropisomers, i.e. conformers resulting from restricted rotation about single bonds, where the rotational barrier is sufficient to allow isolation of the enantiopure species (see below). While the stereoselective synthesis of compounds containing one or more stereogenic centers has emerged as one of the most important fields in chemistry, axial chirality, by contrast, has often been overlooked or treated as an “academic curiosity”.
This, however, has changed with the recognition that the configuration at a biaryl axis can be a decisive factor in governing the pharmacological properties of a bioactive compound and that axial chirality is the fundamental basis for useful reagents and catalysts in asymmetric synthesis.
In this work, we have developed two main topics:
_ As part of a project aimed at synthesizing new chiral structures to be used as ligands in catalytic asymmetric applications, we decided to investigate the chiral 2-(2-hydroxyaryl)cinnamic amides, which are obtained from the coupling of the corresponding 2-(2-hydroxyaryl) cinnamic acids and a chiral primary amine.1 These molecules were chosen since they possess several potential sites of diversity (R1, R2, R3, Ar), which allow for the fine-tuning of their steric, electronic and conformational properties.
In addition, we were also intrigued by the fact that they were structurally reminiscent of biaryl compounds and might display restricted rotation about the C2-Caryl bond and, depending on the energetic barrier to rotation, be resolved into two diastereomeric atropisomeric forms.
The synthesis of several differently substituted chiral amides formally derived from a chiral amine and either E-2-(2-hydroxyphenyl)cinnamic acid or both Eand Z-2-(2-hydroxynaphthyl)cinnamic acid was obtained. These molecules display a restricted rotation about the C2-Caryl bond and, depending on their barrier to rotation, can be isolated in two atropisomeric forms. The barriers to rotation about the C2-Caryl were measured by dynamic 1H-NMR and were found to to vary between 11.8 and 24.5 kcal mol-1, depending on the substitution. In particular, E- 2-(2-hydroxynapthyl)cinnamic amides 2.16 displayed a high barrier to rotation (Gc
‡ > 24.4 Kcal/mol) and could be isolated as both diastereomerically pure forms at room temperature. The X-ray structure of one E-2-(2-hydroxynapthyl)cinnamic amide, (aR)-E-2.16a, was resolved, allowing the determination of the absolute configuration of the chiral axis.
_ As a second project for this thesis work, a small library of chiral monodentate biphenolic and binaphtholic phosphites and phosphoramidites was prepared, and screened in the enantioselective addition of arylboronic acids to aldehydes and Ntosylarylimines for the production of diarylmethanol and diarylmethylamines.2 The ligands were tested either individually or as binary combinations (La/Lb 1:1)
Unfortunately, in the case of arylation of aldehydes (X = O in the scheme above) only a few monodentate ligands showed some catalytic activity albeit with a marginal enantioselectivity.
In the addition to N-tosylbenzaldimine, good enantiomeric excesses were obtained with binaphtholic phosphoramidite ligands containing a bulky chiral amine. The scope of the substrate and the arylboronic acid was also explored. Good to excellent ee’s (76-99%) were also obtained in the arylation of differently substituted Ntosylarylimines, containing either electron-donating or electron-withdrawing substutents. Electron-rich substrates gave generally higher yields, while a slower addition reaction, associated with lower overall yields, occurred with electron-poor substrates. Electron-rich arylboronic acids gave generally good yields in the addition to N-tosylbenzaldimine, associated with moderate/good ee’s
Applicazioni analitiche avanzate della spettrometria di massa con Analizzatori Quadripolari.
Le spettrometrie di massa hanno, negli ultimi anni, aumentato la loro diffusione passando da tecniche avanzate di ricerca a tecniche di “largo consumo” grazie all’enorme avanzamento tecnologico e ai nuovi materiali utilizzati che ne hanno incrementato l’affidabilità e l’applicabilità, accanto ad una migliorata facilità d’uso. Tra gli analizzatori di massa disponibili commercialmente, l’analizzatore quadrupolare offre senza alcun dubbio una grande flessibilità di utilizzo accoppiata a un costo macchina e a dei costi di mantenimento contenuti.
Nel corso dei tre anni di studio del dottorato di ricerca sono stati sviluppati temi di ricerca riguardanti le metodologie analitiche in spettrometria di massa focalizzandosi principalmente sulla risoluzione di problematiche tecnico-strumentali. Le tematiche affrontate sono volutamente molto differenti e hanno riguardato non solo l’aspetto strettamente tecnico-analitico, ma hanno spaziato verso lo sviluppo di sequenze analitiche complete. Tali sequenze comprendono anche l’attenzione verso il campione, verso la definizione dei protocolli di analisi robusti e verso il
trattamento statistico dei dati. Questo processo è stato sviluppato per ottimizzare tutti i parametri legati all’analisi in grado di migliorare l’affidabilità del dato finale e di ottenere la massima informazione in esso celato.
Il primo filone di ricerca del quale mi sono occupato ha riguardato lo sviluppo e l’utilizzo di una metodica completa per la caratterizzazione geochimica di mineralizzazioni cuprifere per lo studio della provenienza di reperti archeologici dell’età del bronzo. La metodologia comprende il trattamento del campione in solido, la digestione acida dello stesso, l’analisi quantitativa delle componenti in traccia, la misura delle abbondanze isotopiche e dei loro rapporti per rame e piombo e il trattamento statistico multivariato dei dati attraverso. Tutte la metodologia ha utilizzato come strumento unico di analisi un ICP-QMS. L’analizzatore quadrupolare si dimostra estremamente valido per l’analisi quali-quantitativa di un gran numero elementi e su un gran numero di campioni in maniera veloce e accurata, ma non trova applicazioni di qualità nel campo dell’analisi isotopica che trova negli strumenti MC-ICP-SFMS la scelta ideale. Le ragioni di questa limitazione sono da ricercare innanzitutto nei forti limiti strumentali, quali l’instabilità del segnale, il mass bias, l’effetto del dead time del rivelatore e, sopra tutte, la non contemporanea rivelazione degli isotopi. Gli ultimi avanzamenti tecnologici hanno tuttavia smussato molto le limitazioni strumentali rendendo più competitivo questo tipo di analizzatore. Ci si è quindi focalizzati nello studio di tutti i parametri che influenzano la precisione e l’accuratezza dela misura ottenibile con un ICP-QMS per giungere ad un miglioramento generale delle prestazioni strumentali. È stata sviluppata una metodologia di bracketing modificata che migliora la produttività delle metodologie già esistenti andando a migliorare anche la qualità analitica del dato. In particolare con il protocollo sviluppato è stato possibile raggiungere un RSD pari a 0.018‰ sui rapporti isotopici 63Cu/65Cu che sono circa tre volte migliori di quelli riportati in letteratura.
Il secondo filone di ricerca ha riguardato la realizzazione di due reattori a scansione per condurre test catalitici su sistemi planari nano strutturati. La grande disponibilità di metodologie di sintesi controllata di sistemi collettivi nano strutturati ha fatto nascere negli ultimi anni l’esigenza di sviluppare sistemi di caratterizzazione per l’attività catalitica degli stessi. I nuovi reattori devono essere adatti a campioni planari in cui l’area superficiale è molto ridotta così come i tempi di contatto. La realizzazione dell’accoppiamento originale tra il reattore e un sistema di analisi quadrupolare commerciale si è sviluppato in due step attraverso due prototipi.
Il primo prototipo permette di fatto una scansione passo-passo ed è stato utilizzato per lo studio del dimensionamento della camera di reazione, dei flussi gassosi da utilizzare e per individuare le criticità strumentali che l’interfaccia reattore-analizzatore ha generato. Sulla base dei dati raccolti, è stato successivamente disegnato e realizzato il vero prototipo a scansione lineare continua, in grado di testare le proprietà catalitiche di una superficie planare di 28 mm2 di area minima. Il micro reattore è stato realizzato presso la nostra officina meccanica e verrà assemblato e testato nei mesi di dicembre-gennaio. Accanto allo sviluppo strumentale sono state affrontate le problematiche legate alla metodologia di testing delle prestazioni strumentali attraverso la sintesi mirata di campioni ad hoc. I catalizzatori planari utilizzati in questa fase sono stati prodotti presso i laboratori del centro CIMaINa di milano dal gruppo del prof. Paolo Milani. Lo studio di nuovi catalizzatori planari è stata anche affrontata durante un periodo di ricerca all’estero presso la “CHALMERS Tekniska Högskola” di Göteborg (Sverige) sotto la supervisione del Prof. Bengt Kasemo. Presso il CHALMERS ho potuto approfondire varie tecniche di sintesi e caratterizzazione per materiali nano strutturati e ho sintetizzato alcuni campioni planari caratterizzati da una struttura a “nanodiscs array” tramite metodologie litografiche colloidali. Tali catalizzatori sono stati sintetizzati per testare le prestazioni del secondo prototipo di reattore sviluppato.
Presso il CHALMERS ho anche seguito il design e l’assemblaggio di un micro-reattore accoppiatto a un analizzatore di massa quadrupolare per testare l’attività catalitica foto-stimolata di titania modificata. Il reattore è stato assemblato passo-passo andando a risolvere i problemi strumentali di interfaccia del sistema a ultra alto vuoto e il reattore a pressione atmosferica. Il reattore è stato testato con successo. Accanto al lavoro tecnico-pratico è stato sviluppato anche il protocollo di analisi che comprende la preparazione del campione, la sequenza analitica e il trattamento dei dati.

Theoretical aspects of Quantum Accelerator Modes.
This thesis is devoted to the study of some theoretical aspects of the problem of Quantum Accelerator Modes (QAMs), which has received considerable interest in recent years both by experimentalists and theorists in the fields of Quantum Chaos, Quantum Optics and cold-ultracold atomic physics.
QAMs, recently observed in cold atom optics, are formed by exposing cold alkali atoms to periodic kicks in the direction of the gravitational field. The kicks are generated by a pulsed standing wave of light. A QAM is characterized by a momentum transfer, which increases linearly with the number of pulses, to a substantial fraction of the atoms. QAMs arise for values of the pulse period close to a integer multiple of half of a characteristic time TB (the Talbot time), typical of the kind of atoms used.
A detailed theoretical explanation of this quantum effect is presented. The system can be modelled by a variant of the well-known quantum Kicked Rotator, in which the effects of a static force, produced by the earth gravitational field, are taken into account. A pseudo-classical theory has been formulated, in which the role of Planck constant is played by the detuning of the kicking period from the resonant value: in the asymptotic limit for a vanishing detuning, QAMs are described by the stable periodic orbit of a “formally” classical map on the 2-torus. An analysis of parametric dependence of the map is performed perturbatively; the parameter regions where the motion is stable and periodic exhibit Arnol’d tongues-like structures. The ordering of Arnol’d tongues and hence the classification of experimentally observed accelerator modes is provided by a number-theoretic construction known as the Farey Tree.
Quantum decay of the modes from classical stable islands immersed in a chaotic sea is analysed and its relations to the famous Wannier-Stark problem are investigated. Theoretical estimates for the lifetimes of the metastable states as a function of the effective Planck constant are obtained.
QAM dynamics in a Bose-Einstein condensate is then considered. Modifications on transport phenomena, imposed by nonlinearities of Gross-Pitaevskii type, which describe the atomic interactions in a mean-field approach, are examined by computer-assisted analysis.
Finally, the problem of QAMs in the vicinity of a generic quantum resonance, namely for kicking period sufficiently close to any rational multiple of the Talbot time, is studied.
The theoretical framework is nontrivially generalized in terms of spinor dynamics: an ansatz of the Born-Oppenheimer type allows to decouple the dynamics of spinor and orbital degrees of freedom and a description of the orbital motion by means of “formally” classical equations is achieved. New rich families of experimental observable QAMs are predicted
Ruolo degli agenti microbici nella eziopatogenesi delle vasculopatie e del diabete. Studi su cellule endoteliali vascolari e nuovi target terapeutici di intervento.
MODULAZIONE GENICA E FUNZIONALE DI CELLULE ENDOTELIALI ARTERIOSE E VENOSE INFETTATE IN MODO PERSISTENTE DA COXSACKIEVIRUS B: POSSIBILE RUOLO NEI PROCESSI DI TROMBOGENESI E ATEROSCLEROSI
I processi infiammatori svolgono un ruolo di rilievo nella genesi e nella progressione delle lesioni croniche cardiovascolari di natura sia aterosclerotica sia trombotica ed è noto che le alterazioni endoteliali giocano un ruolo centrale in tali processi patologici. In tale ottica deve essere rivalutato il coinvolgimento da tempo ipotizzato degli agenti microbici nella patogenesi dell’aterosclerosi. Tra i microrganismi che sembrano giocare un ruolo nella eziologia dell’aterosclerosi sono compresi sia virus (CMV, HSV, EBV) che batteri (Chlamydia pneumoniae e Helicobacter pylori).
Recentemente è stata evidenziata una possibile correlazione tra aterosclerosi, infarto del miocardio e coxsackievirus B (CVBs), agenti virali appartenenti alla famiglia dei Picornaviridae, coinvolti nella eziologia di processi patologici di tipo cronico degenerativo e di cui abbiamo da tempo dimostrato la capacità di infettare le cellule endoteliali (ECs).
Poiché le ECs presentano caratteristiche differenti a seconda dell’origine, abbiamo indagato gli effetti funzionali dei CVBs su linee di cellule endoteliali venose e arteriose ottenute e caratterizzate nel nostro laboratorio. L’attività di ricerca si è incentrata sullo studio della interazione tra il ceppo cardiotropico CVB3 e le cellule endoteliali umane e sulla caratterizzazione degli effetti dell’infezione sulle funzioni di tali cellule correlate allo sviluppo di infiammazione e alla regolazione della coagulazione. Lo studio è stato effettuato principalmente su un modello di infezione in vitro di cellule endoteliali vascolari umane immortalizzate il cui fenotipo è stato caratterizzato sulla base dell’espressione di antigeni endotelio-specifici. I recettori dei CVBs (CAR e CD55) sono espressi sia in ECs venose che arteriose ed entrambi i tipi cellulari sostengono una infezione persistente produttiva non citopatica da CVB3. Gli effetti dell’infezione sono stati indagati mediante DNA microarray specifici per pathways funzionali endoteliali e confermati mediante RTPCR semiquantitative, test immunoenzimatici e citofluorimetria. I risultati dimostrano che CVB3 stimola nelle ECs un’aumentata espressione delle citochine pro-infiamatorie IL-1, IL-6 e IL-8. L’incremento è maggiore nelle ECs arteriose, nelle quali si osserva inoltre up regolazione delle chemochina MCP-1, della proteina C reattiva (CRP), di INF-1 e di COX-2, un enzima chiave nelle fasi precoci del processo infiammatorio. In entrambi i tipi di ECs l’infezione determina una down-regolazione delle caderine e delle integrine,
molecole particolarmente importanti in quanto coinvolte nella disregolazione delle interazioni cellula-cellula e cellula-matrice che predispone il tessuto vascolare alla formazione di placche ateromasiche instabili e alla trombosi. A carico delle ECs venose CVB3 altera in senso pro-trombotico le funzioni di regolazione della coagulazione, con riduzione dell’espressione dell’ attivatore tessutale del plasminogeno e down-regolazione dell’ inibitore dell’ attività procoagulativa del fattore tessutale. A carico delle ECs arteriose l’effetto pro-aterosclerotico di CVB3 è stato confermato dall’ induzione di IL-18 e INF-, fattori condizionanti lo sviluppo e l’instabilità delle placche ateromasiche e dall’ aumentata
espressione delle molecole di adesione e attivazione leucocitaria ICAM-1 e VCAM-1. Allo scopo di provare a identificare nuovi target terapeutici di intervento abbiamo sottoposto, in modo preliminare, le nostre ECs a trattamento con statine. La somministrazione di simvastatina determina nelle ECs infettate una riduzione del “virus yield”, evidenziata dal diminuito rilascio di particelle virali infettanti e dalla ridotta presenza di genoma B3 intracellulare. Inoltre si osserva down regolazione di alcuni importanti markers pro infiammatori (IL-1IL-6, INF-, INF-1 e CRP) con effetti più evidenti nel caso di ECs arteriose infettate. I risultati ottenuti dimostrano come CVB3, virus con tropismo cardiovascolare, possa modulare in modo assai complesso le attività funzionali delle ECs e possa rappresentare un modello per analizzare il ruolo dei CVBs, o di altri virus, nello sviluppo di processi infiammatori cronici vascolari ad esito trombotico e/o aterosclerotico e conseguente sviluppo dello studio di nuove prospettive terapeutiche.
MALATTIA PARODONTALE E ATEROSCLEROSI: CORRELAZIONI MICROBIOLOGICHE
Da qualche anno si parla sempre più spesso di una correlazione tra salute orale e alcune importanti malattie sistemiche, infatti numerosi studi epidemiologici condotti negli ultimi dieci anni hanno dimostrato come persone che soffrono di malattie parodontali possano manifestare con maggiore frequenza patologie cardiovascolari, diabete, malattie polmonari e nascite premature. Uno dei potenziali meccanismi alla base dell’associazione tra salute parodontale e malattie sistemiche è rappresentato dalla infezione batterica. Diversi studi hanno suggerito una possibile associazione tra periodontiti, batteri parodonto patogeni e lo sviluppo di malattie cardiovascolari. Studi sperimentali hanno dimostrato l’abilità di batteri parodonto patogeni nell’interagire con le cellule endoteliali, nell’indurre proliferazione delle cellule muscolari lisce vascolari e nel causare un rilascio locale di citochine proinfiammatorie. La presenza di questi batteri a livello delle placche aterosclerotiche potrebbe avere un ruolo di rilievo nelle fasi di iniziazione, sviluppo e progressione del processo aterosclerotico stesso. Fino ad ora non sono state trovate e presentate in letteratura delle evidenze complete e molti studi su questa associazione dovrebbero essere ancora condotti. Nel corso di questo studio sperimentale sono stati reclutati una ventina di pazienti per essere sottoposti ad endoarterioectomia, prelievi parodontali/gengivali e di sangue. Tutti i campioni sono stati adeguatamente trasportati, conservati e processati per essere sottoposti ad estrazione di DNA totale e batterico in base a specifici protocolli ottimizzati. Si è proceduto mediante specifiche indagini di biologia molecolare con un approccio di PCR di tipo Multiplex con cui poter individuare in una singola reazione più frammenti di DNA batterico per ciascun campione attraverso l’uso di coppie di primers specifici per i batteri da noi precedentemente selezionati (Actinobacillus actinomycetemcomitans, Fusobacterium nucleatum, Bacteroides forsythus, Prevotella intermedia, Porphyromonas gingivalis). La valutazione di tutte le analisi PCR Multiplex condotte e soprattutto l’accurata osservazione dei rispettivi gels ad esse associate, ci ha permesso di ottenere una serie di risultati preliminari.
Dei 20 tamponi parodontali testati tutti infatti sono risultati positivi per Fusobacterium nucleatum (100%), il 70% ha mostrato positività per Prevotella intermedia, il 65% per Porphyromonas gingivalis, mentre il 30% per Actinobacillus actinomycetemcomitans e infine il 10% per Bacteroides forsythus. Delle 20 placche analizzate il 35% sono risultate positive per Fusobacterium nucleatum, il 15% per Prevotella intermedia e il 5% per Porphyromonas gingivalis così come per Actinobacillus actinomycetemcomitans. Le indagini condotte su campioni di sangue intero hanno rilevato una batteriemia in circa il 35% dei casi e di cui il 20% ad opera di Fusobacterium nucleatum, il 5% di Prevotella intermedia così come di Porphyromonas gingivalis. In seguito ad analisi globale e comparativa di tutti i risultati ottenuti nel caso dei tre tipi di campioni indagati, abbiamo evidenziato come in tre pazienti vi sia del DNA batterico in tutti i campioni da loro prelevati (tampone parodontale, placca aterosclerotica e sangue) e in particolar modo come Fusobacterium nucleatum sia generalmente il battere più rappresentato. In modo preliminare abbiamo quindi ottenuto delle prime indicazioni con cui poter affermare che le placche siano state colonizzate da batteri parodonto patogeni entrati in circolo e che il DNA in esse ritrovato non sia dovuto a semplici batteriemie transienti o a contaminazioni tecnico/operative. Ulteriori indagini hanno evidenziato la presenza del genoma di Chlamydia pneumoniae nel 54% degli ateromi. Il genoma di Enterovirus è stato isolato nel 14% delle placche aterosclerotiche. Tre pazienti hanno rilevato a livello delle loro placche aterosclerotiche un evidente “pathogen burden” (Porphyromonas gingivalis+Fusobacterium nucleatum+Chlamydia pneumoniae+enterovirus) correlato positivamente con una maggiore condizione infiammatoria della placca come dimostrato dalla elevata espressione di citochine e chemochine pro-infiammatorie (IL-6, IL- 1, IL-8, MCP-1) e della proteina C reattiva (CRP), a conferma del ruolo, sempre più evidente, che i microrganismi giocano nella eziopatogenesi dell’aterosclerosi.
I COXSACKIEVIRUS B POSSONO INDURRE DIABETE ATTRAVERSO L’INFEZIONE DELLE CELLULE ENDOTELIALI DELLE ISOLE DI LANGERHANS DEL PANCREAS
Il diabete di tipo 1 (TD1) è dovuto ad un danno delle cellule produttrici di insulina per lo più conseguente all’attivazione di processi cronici di natura infiammatoria e immunologica e mediato dal rilascio di citochine nelle isole di Langerhans. I virus (in particolare Coxsackievirus B4, CVB4) sono ritenuti possibili cause di diabete, ma i meccanismi patogenetici non sono stati finora chiariti. Gli enterovirus causano frequentemente viremia e diffusione a vari organi viscerali tra cui anche il pancreas. Dati siero epidemiologici suggeriscono che molti pazienti con diabete di tipo 1 sono positivi per Coxsackievirus, in particolar modo per CVB4. Studi attuali hanno dimostrato che pazienti con TD1 recente mostrano titoli virali e CVB-IgM più elevati, soprattutto per il sierotipo B4, rispetto a soggetti non diabetici.
Il meccanismo attraverso il quale gli enterovirus contribuiscono alla distruzione delle cellule _ pancreatiche rimane ancora da stabilire e per ora possono essere formulate solo delle ipotesi. Le cellule del microcircolo (MVECs) svolgono un ruolo chiave nei processi fisiopatologici insulari che portano al diabete. La capacità delle MVECs di presentare antigeni insulari ai linfociti T citotossici specifici (GAD65, proinsulina e tirosina fosfatasi IA-2), di esprimere molecole di adesione (ICAM-1, VCAM-1, integrine) e di produrre tutta una serie citochine e chemochine, le rende protagoniste nei processi di distruzione autoimmunitaria delle cellule beta pancreatiche.
Poiché il nostro gruppo ha dimostrato che i CVBs possono infettare in modo persistente le cellule endoteliali con attivazione di processi infiammatori cronici, abbiamo indagato la suscettibilità delle MVECs insulari a CVB4 e gli eventuali effetti pro-diabetogeni indotti dall’infezione. A tale fine abbiamo allestito colture primarie di MVECs insulari da cui abbiamo sviluppato linee immortalizzate con SV40 fenotipicamente omogenee, stabili, a rapida espansione, con morfologia analoga alla coltura primaria e con importanti caratteristiche biologiche tipiche delle cellule parentali. Le colture immortalizzate mostrano inoltre somiglianza a quelle primarie da un punto di vista funzionale nell’espressione delle molecole di adesione, nell’aderenza dei linfomonociti e nelle funzioni di barriera. Le MVECs insulari sono state caratterizzate in base all’espressione di markers endoteliali specifici e ne è stato valutato il profilo immunologico. Entrambi i tipi di coltura se messi su substrato di matrigel formano strutture simili a cordoni capillari. Le MVECs insulari mostrano una elevata espressione basale per ICAM-1, una moderata espressione di E-selectina e una non rilevabile espressione di VCAM-1. Il trattamento con TNF-up-regola l’espressone di ICAM- 1 e E-selectina e induce neo espressione di VCAM-1. Il trattamento con INF-induce l’espressione di HLA-DR (molecola HLA di classe II). Le prove di infezione hanno dimostrato CVB4 replica in modo persistente nelle MVECs insulari con rilascio di virus infettante, ma in assenza di un effetto citopatico rilevante. A carico delle MVECs insulari infettate sono stati riscontrati effetti pro-infiammatori persistenti in termini sia di iper espressione di molecole di adesione (ICAM-1, VCAM-1 e integrine 53) sia di produzione di citochine/chemochine (IL-1, IL-6, IL-8, INF-, INF-, TNF-, linfotactina). E’ da rilevare che tali mediatori non solo svolgono un ruolo decisivo nella patogenesi immuno-mediata del diabete, ma possono anche danneggiare in modo diretto le cellule . Nelle fasi iniziali dell’infezione è stato rilevato rilascio di IL-18, citochina inducente INF-, altro importante mediatore del danno insulare. Dato di particolare interesse è stato il riscontro dell’espressione nelle MVECs insulari infettate di Fas-L, fattore decisivo nei processi di apoptosi a carico delle cellule . E’ possibile pertanto concludere che l’infezione delle MVECs insulari da parte di CVB4 determina in modo sia indiretto che diretto l’attivazione di meccanismi di danno delle cellule produttrici di insulina. I risultati e le osservazioni, condotti in questi tre anni di ricerca, hanno reso le cellule endoteliali vascolari ECs interessanti e sicure protagoniste nella patogenesi delle vasculopatie e del diabete di tipo 1 in corso di infezioni acute e/o persistenti sia di natura virale che batterica. I dati fino ad ora ottenuti potrebbero indurre tutta una serie di studi finalizzati ad esplorare nuovi approcci terapeutici e proprio l’analisi dei cambiamenti molecolari delle ECs durante i processi di infezione/infiammazione potrà aiutare a identificare i nuovi target di intervento per malattie diffusissime come l’aterosclerosi e il diabete
Molecular and functional characterization of ANKRD1, a candidate gene for total anomalous pulmonary venous return.
Total Anomalous Pulmonary Venous Return (TAPVR, OMIM %106700) is a rare congenital heart defect in which the pulmonary veins fail to enter the left atrium and drain instead into the right atrium or one of its venous tributaries. Previously, a TAPVR patient bearing a t(10;21) de novo balanced translocation was found in our laboratory and the ANKRD1/CARP gene, mapping on chromosome 10 about 130 Kb proximally to the breakpoint, was defined as a good TAPVR candidate gene. Significantly, we found highly increased ANKRD1 expression levels in lymphoblastoid cell lines derived from both the translocation-bearing proband and a second independent sporadic TAPVR patient, suggesting that disruption of the normal ANKRD1 expression pattern is associated with TAPVR. Moreover, an ANKRD1 non-conservative missense mutation was found in a third sporadic TAPVR patient. Such mutation was subsequently found to confer an increased stability to the ANKRD1 protein in vitro and to affect its function as a repressor of cardiac-specific gene expression. The aim of this doctoral project was to characterize at the molecular and functional level the ANKRD1 gene, in order to provide new evidences for its involvement in TAPVR pathogenesis. First, ANKRD1 function as a negative regulator of cardiac genes was further investigated, and its role as a repressor of Nkx2.5 and Pitx2cmediated activation of the cardiac-specific ANF promoter was demonstrated. Moreover, in situ hybridization assays revealed that the expression pattern of the murine Ankrd1 gene strongly overlaps with that of both Nkx2.5 and Pitx2 genes, supporting both the occurrence of a functional interaction between these genes and a role for ANKRD1 in TAPVR pathogenesis. Co-immunoprecipitation assays added a further support for this hypothesis, by showing a physical interaction between the ANKRD1 and Nkx2.5 proteins. The intracellular pathways involved in ANKRD1 turnover were also investigated. ANKRD1 was shown to behave as a short-lived protein in vivo, with the PEST motif playing a crucial role in its turnover rate. Moreover, intracellular degradation of ANKRD1 was found to be carried out mainly by the ubiquitin proteasome system, through several independent degron elements. These data defined for the first time both the intracellular pathway responsible for ANKRD1 degradation and the functional relevance of the PEST motif in this process. Finally, ANKRD1-overexpressing transgenic mice were generated in the attempt to develop an animal model for TAPVR.
