Oncogene-induced senescence and its evasion in a mouse model of thyroid neoplasia

    No full text
Here we describe a conditional doxycycline-dependent mouse model of RET/PTC3 (NCOA4-RET) oncogene-induced thyroid tumorigenesis. In these mice, after 10 days of doxycycline (dox) administration, RET/PTC3 expression induced mitogen activated protein kinase (MAPK) stimulation and a proliferative response which resulted in the formation of hyperplastic thyroid lesions. This was followed, after 2 months, by growth arrest accompanied by typical features of oncogene-induced senescence (OIS), including upregulation of p16INK4A and p21CIP, positivity at the Sudan black B, activation of the DNA damage response (DDR) markers γH2AX and pChk2 T68, and induction of p53 and p19ARF. After 5 months, about half of thyroid lesions escaped OIS and formed tumors that remained dependent on RET/PTC3 expression. This progression was accompanied by activation of AKT-FOXO1/3a pathway and increased serum TSH levels

Monitoring hydrological processes for land and water resources management in a Mediterranean ecosystem: The Alento River Catchment Observatory.

    No full text
In recent years, the critical zone (CZ) of catchments across the Mediterranean region has been influenced by rapid changes in both climate seasonality and land use/land cover. Rural ecosystems in southern Europe are experiencing prolonged droughts, seriously compromising water resources availability and crop yields whilst increasing the risk of wildfire occurrence. Rainy seasons are likely to be characterized by intense storms that trigger floods resulting in increasing damage severity. The negative effects of anthropogenic disturbance on hydrological ecosystem services can be tempered by demand-side adaptation options and appropriate investments to ensure water supply under drought conditions. To shed light on some of the scientific challenges related to these issues, a Critical Zone Observatory (CZO) has being established in the Alento River catchment. Although sampling campaigns and monitoring investigations have been carried out in this area for more than twenty-five years, a more systematic research program was recently started to take comprehensive measurements in representative sub-catchments of the study area. These sites are instrumented with advanced ground-based sensor network platforms that provide hydro-meteorological variables and fluxes in the groundwater-soil-vegetation-atmosphere system. Hydrological models of different complexity exploit the gathered dense information to assess the impact of land use and climate changes on key functions and services of the CZO in the Alento River catchment

MDO framework for university research collaboration: AGILE academy initiatives & outcomes

    No full text
AGILE Project is developing the 3rd generation MDO processes, which will support the development of the next generation aerospace products. The establishment of effective collaborative design methodologies is currently acknowledged as the key enabler for future product development processes. At the same time, the need to introduce collaborative design techniques within educational activities is also well recognized by the Academic, Research and Industrial communities. AGILE project supported by European Commission’s H2020 Programme, is setting the “AGILE Paradigm”, a conceptual framework which contains all the elements to implement a multidisciplinary collaborative design network. The AGILE Academy initiative is conceived to infuse into the Academic organizations and educational environments the “AGILE Paradigm”, and make available all the technologies developed within the AGILE Project, which support the implementation of such a Paradigm. This paper focus is on the inception, approach and results of the AGILE Academy participants from several universities around the world

Minori stranieri non accompagnati: strumenti privatistici per la tutela delle identità culturali

    No full text
