Role of estrogen receptors, P450 aromatase, PCNA and p53 in high-fat-induced impairment of spermatogenesis in rats.
Obesity and overweight are frequently associated with male subfertility. To address new
findings on the players involved in the obesity-induced impairment of spermatogenesis,
we used a high-fat diet-induced overweight-rat model. Following four weeks of high-fat
diet, the organization of seminiferous epithelium was affected, and tubules lumen showed
immature/degenerated cells, typical signs of hormonal imbalance and testicular damage.
Real-time PCR analysis allowed us to detect increased levels of ERa and decreased levels of
aromatase CYP19 transcripts in testis, suggesting an increase in circulating estrogens
derived from the accumulating adipose tissue rather than the induction of testicular
estrogen synthesis. Moreover, in situ hybridization analysis showed an increased
susceptibility towards estrogens in testis from high-fat fed rats, being ERs expressed
not only in spermatogonia, as in control testis, but also in spermatids. Western blot and
immunohistochemical analyses revealed an increase in the amount of p53 and PCNA,
together with a change in their immunodetection, being the proteins localised on germ
cells at different stages of maturation. Differences in p53 and PCNA expression may give
evidence and be part of a cellular response to stress conditions and damage caused by the
excessive intake of saturated fatty acids
A promising medium-term follow-up of pediatric sclerosing cholangitis: Mild phenotype or early diagnosis?
AIM:
Sclerosing cholangitis (SC) is a chronic cholestatic liver disease that is being increasingly diagnosed in childhood. The long-term course and prognosis of pediatric SC are poorly described.
METHODS:
We reviewed data of pediatric SC patients, followed in two referral centers, during a period of up to 20 years. We aimed to evaluate long-term outcomes according to SC phenotype.
RESULTS:
Among 45 patients (median age, 10.4 years; male patients, 73.4%) 29 (64.4%) were asymptomatic at presentation. Twenty patients (44%) had a concomitant inflammatory bowel disease (SC/IBD). Autoimmune features were found in 20 patients (44%). Liver biopsy showed severe fibrosis or cirrhosis in 32% of cases. Patients with SC alone had a higher rate of interface hepatitis at liver biopsy than SC/IBD patients. All children received ursodeoxycholic acid at diagnosis, and 17 received steroids and/or azathioprine. After a mean follow-up of 8.7 ± 5.6 years, all patients were alive and seven developed at least one liver-related complication. At the end of follow-up, 10 patients stopped immunosuppressants and two had no therapy. Only two patients underwent liver transplantation. Complication-free survival did not differ between SC/IBD and SC patients, but survival was longer in patients without autoimmune features.
CONCLUSIONS:
In our early diagnosed cohort, the 9-year survival with native liver was better than that reported in other studies. Approximately 15% of patients developed liver-related disease complications, less than previously reported. The long-term course of SC was negatively influenced by the presence of autoimmune features, but not by concomitant IBD

Automatic grading of Acne vulgaris using deep learning
In this study, we trained a neural network to
perform automatic grading of digital images of acne patients with
reliabilities comparable or superior to those of expert physicians.
A dedicated device was employed to acquire images of 479
patients belonging to three different ethnic groups. A
convolutional neural network trained with features extracted
from local patches extracted from the facial skin showed an
accuracy of 0.85 and a correlation between manual evaluation
and automatized IGA of r=0.96. This is the first work where a
neural network was able to directly classify acne patients
according to an ordinal scale with no human intervention and no
need to count lesions

Attualmente i porti, come nodo del sistema logistico a supporto dello scambio commerciale internazionale, rappresentano una realtà economica determinante per lo sviluppo economico del territorio e della nazione (Ministero delle Infrastrutture e dei Trasporti, 2013), che determinano impatti significativi sulla città sia influenzando le condizioni di benessere e qualità della vita che comportando numerosi conflitti spaziali e ambientali. In particolare, gli obiettivi 11, 12 e 14 dei Sustainable Development Goals (SDGs, Agenda 2030) mirano a conservare e valorizzare in modo sostenibile mari e risorse marine, attraverso la costruzione di modelli sostenibili di produzione e di consumo, per rendere operativo il concetto di città sostenibile.
In questo senso la Direttiva europea 2014/89 del 23 luglio 2014 individua la pianificazione dello spazio marittimo come strumento utile alle autorità pubbliche e alle parti interessate per realizzare un approccio integrato ed ecosistemico, nell’intento di promuovere lo sviluppo delle economie marittime e costiere e l’uso sostenibile delle risorse. How to assess the volume of a DIEP flap using a free online calculator: The DIEP V (volume) method
Among the few methods available, none is able to determine accurately the volume of a DIEP flap. Specimen weight is commonly used to assess the amount of flap needed to reconstruct a breast, but the density of breast is different from that of abdominal tissues; therefore, the volume should be used as a unique unit of measure. The purpose of this study was to provide a simple method to calculate the predicted volume of a DIEP flap in order to match the volume of the breast being reconstructed

Deiodinases and stem cells: an intimate relationship
Thyroid hormone is a major determinant of tissue functions in vivo. The deiodinase family controls the tissue-specific activation or inactivation of intracellular thyroid hormones. Precise control of the T3-dependent transcriptional program is required by multiple cell systems, including the stem cell. In this context, the identification of a close connection between thyroid hormones and different signal pathways involved in the control of stem cell functions suggested that the deiodinases may play a role in the definition of stem cell biology and physiology. Stem cells have an unlimited self-renewal capacity and the potential to differentiate into different types of mature cells. Deciphering how all these events are achieved, how the T3 signal is controlled and integrated in stem cells and their niches, and how it can impact on them is essentially unknown and represents a challenge for coming years. In this review, I will explore the role played by the deiodinases in the modulation of the TH signal in stem cells of adult tissues, namely muscle and intestine, and how their actions control the delicate balance among self-renewal, proliferation and differentiation. Elucidation of the molecular mechanisms presiding thyroid hormone action in stem cells may reveal therapeutic potential, for example in the fields of regenerative diseases and cancer
