Supplemental irrigation management of rainfed grapevines under drought conditions using the CropSyst model
Aim of study: To determine how much water should be used and when it should be applied in rain-fed grapevine using a cropping system simulation model (CropSyst), and also the economic analysis of supplemental irrigation for rainfed grapevine.Area of study: This study was conducted at the School of Agriculture, Shiraz University, Shiraz, Iran, in 2012, 2013 and 2014.Material and methods: The CropSyst model was calibrated to predict the rainfed yields of ‘Askari’ and ‘Yaghooti’ grapevines in different climates using four amounts of SI: 250 L (I1), 500 L (I2), 1000 L (I3) and 0 (I4), five SI times: single in March (T1), single in April (T2), single in March + single in April (T3), single in May (T4) and single in June (T5).Main results: Treatment T3 increased the average simulated yield of ‘Askari’ by 15% to 40% at regions with P/ETo>0.6, 17% to 61% at 0.2<P/ETO<0.6, and 26% to 61% at P/ETO<0.2, while in ‘Yaghooti’ it increased about 2% to 41% at regions with P/ETo>0.6, 4% to 36% at 0.2<P/ETO<0.6 and 2% to 26% at P/ETO<0.2. By increasing the water price by 30% and 50%, net benefits for the ‘Askari’ decreased by about 31% and 54%, while 6% and 18%, for ‘Yaghooti’ respectively.Research highlights: The CropSyst model can successfully predict soil water content and grapevine yields. Application of SI in May increased significantly the grapevine yield as compared to other SI times
Morphological, molecular and pathogenic characterization of Phytophthora palmivora isolates causing black pod rot of cacao in Colombia
Aim of study: To characterize isolates of Phytophthora sp. causing black pod rot (BPR) of cacao (Theobroma cacao L.).Area of study: Eight cocoa-growing regions in Colombia.Material and methods: Sixty isolates of Phytophthora sp. were obtained from tissues of cacao pods showing symptoms of BPR. Isolates were characterized using the morphology of sporangia and chlamydospores, molecular sequencing of regions of nuclear DNA (rDNA-ITS) and mitochondrial (COX) and virulence in different genotypes of cocoa pods.Main results: A high phenotypic variability between the isolates was determined, being the pedicel length and the length/width ratio (L/W) the most stable characters for species identification. Short pedicels with an average of 3.13 μm ± 0.28 and a length/width ratio of sporangia (L/W) with an average of 1.55 μm ± 0.11 were established as the most consistent morphological characteristics within palmivora species.Research highlights: Phytophthora pamivora was the only species associated to BPR, identified using morphology together with sequence analyses
Genomic differentiation among varieties of Iberian pig
Aim of study: The objective of this study was to identify the autosomal genomic regions associated with genetic differentiation between three commercial strains of Iberian pig.Area of study: Extremadura (Spain).Material and methods: We used the Porcine v2 BeadChip to genotype 349 individuals from three varieties of Iberian pig (EE, Entrepelado; RR, Retinto; and TT, Torbiscal) and their crosses. After standard filtering of the Single Nucleotide Polymorphism (SNP) markers, 47, 67, and 123 haplotypic phases from EE, RR, and TT origins were identified. The allelic frequencies of 31,180 SNP markers were used to calculate the fixation index (FST) that were averaged in sliding windows of 2Mb.Main results: The results confirmed the greater genetic closeness of the EE and RR varieties, and we were able to identify several genomic regions with a divergence greater than expected. The genes present in those genomic regions were used to perform an Overrepresentation Enrichment Analysis (ORA) for the Gene Ontology (GO) terms for biological process. The ORA indicated that several groups of biological processes were overrepresented: a large group involving morphogenesis and development, and others associated with neurogenesis, cellular responses, or metabolic processes. These results were reinforced by the presence of some genes within the genomic regions that had the highest genomic differentiation.Research highlights: The genomic differentiation among varieties of the Iberian pig is heterogeneous along the genome. The genomic regions with the highest differentiation contain an overrepresentation of genes related with morphogenesis and development, neurogenesis, cellular responses and metabolic processes
Effect of antimicrobial peptides and monoterpenes on control of fire blight
Aim of study: Antimicrobial peptides and monoterpenes are safe compounds that have been used for control of many plant diseases. Herein, the effects of two recombinant antibacterial peptides (AMPs) were compared with two monoterpenes for control of Erwinia amylovora directly or via induction of plant defense enzyme guaiacol peroxidase (GPOD).Area of study: The experiments were performed at the Ferdowsi University of Mashhad (Iran).Material and methods: The central composite design (CCD) method was used to study the effect of mixing the compounds and copper compound (Nordox) in controlling the pathogen. The resistance level was studied on shoots of tolerant (‘Dargazi’) and semi-susceptible (‘Spadona’) pear cultivars treated with the antibacterial compounds.Main results: Thanatin and 1,8-cineole showed the highest and lowest antibacterial effects. All treatments reduced E. amylovora pathogenicity on blossom. The CCD analysis revealed that the best reduction in colony number obtained by mixing Lfc, thanatin, thymol, 1,8-cineole and Nordox at concentrations of 32, 16, 24, 250 and 250 μg/mL. Thymol and 1,8-cineole at 500 μg/mL decreased disease severity significantly compared to that of AMPs. The level of GPOD enzyme in ‘Dargazi’ was higher than in ‘Spadona’. All treatments increased the GPOD levels in both cultivars. Furthermore, resistance level and GPOD ratio were negatively correlated.Research highlights: Antimicrobial peptides showed better effect on growth inhibition of E. amylovora than monoterpenes. Mixing of these peptides and monoterpens at special dosage enhanced their antimicrobial efficacy against E. amylovora; that could represent a new method in control of fire blight disease
Effects of in utero heat stress on subsequent reproduction performance of first-calf Holstein heifers
Aim of study: To determine the reproductive performance of heifers gestated under maternal conditions of heat stress in late gestation.Area of study: Northern Mexico (25° 32’ N, 103° 23’ W).Material and methods: The study included reproductive records of 4976 first-calf Holstein heifers in a hot environment.Main results: Heifers born to cows experiencing no heat stress three months before parturition but with a THI >83 at calving were older (p<0.05) at first calving (743 ± 67 vs. 729 ± 55 days) than heifers gestated under maternal conditions of heat stress. A two-fold increase (p<0.01) in pregnancy rate occurred in heifers gestated under maternal conditions of no heat stress during two or three months before pregnancy and no heat stress at parturition, compared with heifers gestated under maternal conditions of no heat stress. Overall, across in utero heat stress one, two or three months before calving, pregnancy rate to all services was higher (p<0.05) for first-calf heifers gestated under maternal conditions of no heat stress during delivery, compared with heifers gestated under maternal conditions of heat stress (66.7 vs. 51.1%). Median days for getting pregnant was higher (140 d) for heifers whose dams were exposed to THI >83 at calving than heifers whose mothers were exposed to <76 or 76-83 (117 and 114 d) at calving.Research highlights: These data suggest that in utero heat stress during the last three months of gestation negatively affects the reproductive performance of first-calf Holstein heifers
Pollution level and risk assessment of heavy metals in sewage sludge from eight wastewater treatment plants in Wuhu City, China
Aim of study: To investigate the content, contamination levels and potential sources of five heavy metals (Hg, Pb, Cd, Cr, As) in sewage sludge from eight wastewater treatment plants (W1 to W8).Area of study: Wuhu, located in southeastern Anhui Province, southeastern China.Material and methods: The sewage sludge pollution assessment employed the single-factor pollution index, Nemerow’s synthetic pollution index, monomial potential ecological risk coefficient and potential ecological risk index. The potential sources among the five heavy metals were determined using the Pearson’s correlation analysis and principal component analysis (PCA).Main results: The mean concentrations of the heavy metals were 0.27 mg/kg (Hg), 70.78 mg/kg (Pb), 3.48 mg/kg (Cd), 143.65 mg/kg (Cr) and 22.17 mg/kg (As). W1, W5 and W6 sewage sludge samples showed the highest levels of heavy metal contamination, and cadmium had the highest contamination level in the study area. Pearson’s correlation analysis and PCA revealed that Pb and Cd mainly derived from traffic emissions and the manufacturing industry and that As and Cr originated from agricultural discharges.Research highlights: The pollution of cadmium in Wuhu should be controlled preferentially. The heavy metal pollution of W1, W5 and W6 sewage treatment plants is relatively high, they should be key prevention targets
Improvement of drought tolerance in five different cultivars of Vicia faba with foliar application of ascorbic acid or silicon
Aim of study: To explore the role of ascorbic acid (AsA) or silicon (Si) in improving drought tolerance in five faba bean cultivars under irrigation water deficit (IWD).Area of study: The experimental farm; 30° 36′ N, 32° 16′ E, Egypt.Material and methods: Three drip irrigation regimes (WW, well-watered, 4000 m3 water ha-1; MD, moderate drought, 3000 m3 water ha-1; and SD, severe drought, 2000 m3 water ha-1) were applied to plants, which were sprayed 25, 40, and 55 days after sowing with 1.5 mM AsA or 2.0 mM Si vs distilled water as a control.Main results: Drought negatively affected physiological attributes (photosynthetic pigments, gas exchange parameters, relative water content, membrane stability index, electrolyte leakage (EL), and lipid peroxidation), which restricted plant growth and yields, and stimulated alterations in both enzymatic and non-enzymatic antioxidant activities. However, AsA or Si application mitigated drought effects on physiological attributes, improving growth, yields and water use efficiency by raising antioxidant activities and suppressing lipid peroxidation and EL in stressful cultivars. The mitigating effects of AsA and Si were more pronounced under MD.Research highlights: ‘Nubaria-2’, ‘Giza-843’, and ‘Sakha-3’ were more tolerant than ‘Giza-716’ and ‘Sakha-4’, suggesting the use of AsA or Si to ameliorate the IWD effects on stressful cultivars. Certain physiological traits exhibited positive association with growth and seed yield, demonstrating their importance in enhancing seed yield under irrigation treatments
Monitoring and bioremediation of organochlorine pesticides in surface water with Enterobacter asburiae
Aim of study: One of the safest techniques regarding the remediation of contaminated water is biological remediation. This study aimed to: (i) monitoring of a collection of organochlorine pesticides (OCPs) in three agricultural drainages (Nashart, no. 9, and El-shoka), located in Kafr El-Sheikh governorate, Egypt; and ii) investigate the biodegradation potential of different bacterial isolates regarding organochlorine pesticides.Material and methods: Analysis of OCPs was carried out by gas chromatography, Enrichment cultures were used for isolation of the bacterial strains capable of OCPs biodegradation and the most efficient isolate was identified based on morphological, biochemical ad molecular characteristics.Main results: The determination of OCPs in water samples by gas chromatography showed varying values of OCPs ranging from 0.0 mg/L (below detection limit) to 0.0385 mg/L. A total of four morphologically different bacterial isolates were obtained, which showed a remarkable capability of OCPs biodegradation detected in mineral salt medium containing 17 OCPs active ingredients by two approaches including the analysis of the OCP residues at the end of the incubation period and measuring the bacterial growth in terms of total viable count and optical density. The bacterial isolate N2 showed the highest degradation capability when the screening process was carried out to select the most efficient isolates, which was identified according to the morphological, biochemical and molecular characterization as Enterobacter asburiae.Research highlights: The biodegradation of OCPs using E. asburiae was proved to be a promising approach for the detoxification and removal of OCPs residues in aqueous systems
A quantitative multivariate methodology for unsupervised class identification in pistachio (Pistacia vera L.) plant leaves size
Aim of study: Genetic diversity of pistachio, can be evaluated by using different descriptors, as adopted in international certification systems. Mainly the descriptors are morphological traits as leaf, which represents an important organ for its sensibility to growth conditions during the expansion phase. This study adopted a rapid and quantitative non-hierarchic clustering classification (k-means), to extract size classes basing on the contemporary combination of different morphological traits (i.e., leaf stalk length, terminal leaf length, terminal leaf width and terminal leaf ratio) of a varietal collection composed by 21 pistachio cultivars.Area of study: Worldwide.Material and methods: The unsupervised non-hierarchic clustering technique was adopted to the entire samples of pistachio leaves from k=2 to k=15 for both four morphological variables (i.e., leaf stalk length, terminal leaf length, terminal leaf width and terminal leaf ratio) and three morphological variables (i.e., terminal leaf length, terminal leaf width and terminal leaf ratio).Main results: A classification model only on the three morphological variables (for results of statistical analysis in which the groups resulted to be more separated and different for all the variables), with k= 5 (five groups), was constructed using a non-linear artificial neural network approach. The percentages of bad prediction in both training and testing resulted equal to 0%. The “terminal leaf length” returned the higher impact (44.89%).Research highlights: The contemporary combination of different morphological leaf traits, allowed to create an automatic classification of size classes of great importance for cultivar identification and comparison
