Growth Characteristics of Arabidopsis thaliana Haploids
We analyzed the morphological and growth characteristics of Arabidopsis thaliana haploids generated from a GFP -ailswap (cenh3-1) × wild-type cross. Haploid seedlings and mature plants were much smaller than those of diploids. For seeds, there was no significant difference in size between haploids and diploids; however, haploid seeds were distorted compared with diploid ones, which were elliptical. Measurements for the root elongation rates indicated that the rate of haploids was suppressed to approximately 50% of that of diploids from four to ten days after sowing. The flowers of haploids were smaller than those of diploids, and the lengths and areas of sepals and petals of haploids were significantly reduced compared with those of diploids. These results suggest that haploidization does not significantly affect growth during embryogenesis, but suppresses overall growth after germination. In addition, the results of flow cytometry analysis conducted on cauline leaves indicated that the haploids underwent higher level of endoreduplication than the diploids. Further studies are needed to elucidate the relationship between the cell cycle and endoreduplication in haploids.原著departmental bulletin pape
