Carbon dioxide removal from industrial gases using an indirectly heated and cooled temperature swing adsorption process
In addition to absorption and membrane processes, adsorption processes offer the possibility of capturing and recovering CO2 from fossil fueled power plants. Because of the long heating and cooling times required in a TSA process and because during desorption the recovered component is diluted, an indirectly heated and cooled TSA
process is proposed for CO2 capture. The suitability of the indirectly heated and cooled temperature swing adsorption process for CO2 capture is investigated in this work. The heat transfer characteristics of an adsorbent packed bed with and without convection are investigated, since heat transfer plays a major role in this process. The adsorption characteristics of an indirectly heated and cooled adsorber is also an important topic in this work. A multidimensional mathematical model is derived in order to simulate the indirect heated and cooled temperature swing adsorption process.
The heat conductivity of the solid particle and the wall Biot number are important model parameters and are required for modeling the indirect heated and cooled TSA process. The convective contribution to the radial heat conductivity at low Peclet numbers showed a similar relationship to the relationship that is postulated in the literature for higher Peclet numbers. Contrary to the radial heat conductivity, the convective contribution to the wall heat transfer coefficient shows no clear relationship. The reasons for this are the correlation between the parameters and that the non-convective
contribution dominates over the range of Peclet numbers that are used in this work.
To investigate the adsorption characteristics, an isotherm is scaled by comparing the experimental loading value that is measured with a breakthrough curve and the loading obtained by the isotherm. This is necessary since the same activation procedure cannot be achieved during the in situ activation of the bed. The model is validated and kinetic parameters are calculated. Moreover, the agreement between the model and the measurements is strong, especially for the CO2 concentration. The small discrepancies
between the measured and predicted temperature profiles can be explained on one hand by flow disturbances and on the other hand by the uncertainty caused by scaling the isotherm. The recoveries obtained by the model are within the range of the experimental
error and show the same dependency of the recovery with the regeneration temperature. The average purity of the recovered CO2 is also within the experimental error.
Using the derived model and the determined kinetic parameters, a parametric sweep is conducted in order to see the influence of different parameters on the process. The influence is measured by three different key performance indicators: the average purity of the recovered CO2, the CO2 recovery, and the specific energy required. The numerical study shows that promising results are obtained by reducing the radial thermal resistance. Using optimal parameters, high recoveries and purities can be achieved and specific energy requirements that are lower than the benchmark process (amine wash).

Socio-spatial transformations in the central coastal region of Ecuador by the influence of globalised tourism: Puerto Lopez 1990-2010
Der internationale Tourismus ist in den letzten 50 Jahren einer der dynamischsten und schnell wachsenden Wirtschaftssektoren. Er hat in der Wirtschaftskrise Widerstandsfähigkeit bewiesen und hatte mit einer offiziell unterstützten positiven wirtschaftlichen Prognose ein anhaltendes Wachstum in der ersten Hälfte des 21. Jahrhunderts (Schulte, 2003) (UN-WTO, 2013b); dieser alte, aber gesunde Markt hat positive und nachteilige Auswirkungen auf die Gebiete, in denen sich ausbreitet. Dennoch ist die Natur der sozio-räumlichen Umwandlungen von Landschaften, die sie über Gebiete in den Entwicklungsländern auslöst, weniger rückläufig, sogar untersucht.
Auf der Grundlage des transformationalistischen Ansatzes der Globalisierung (Held & McGrew, 1999 in Murray, 2006) und mit einem starken Glauben an die Fähigkeit der nationalen Regierung und der Gemeinschaften, ihre lokale nachhaltige Entwicklung zu verwalten (Weiss, 2010), versucht die Studie, die Natur der sozio-räumlichen Umwandlungen zu verstehen, die sich in den letzten 20 Jahren in der touristischen Küstenstadt Puerto Lopez vollzogen. Ziel ist es, einen Beitrag zur nachhaltigen Stadtplanung und städtebaulichen Gestaltung dieser Siedlungen zu leisten, die für die Verbesserung der globalen Widerstandsfähigkeit im 21. Jahrhundert strategisch sind.
Durch die Kombination der Stadtbildanalyse (Stadtmorphologie) (Conzen, 2004) und der GIS-Technologie wurde eine Methode entworfen und getestet, um für die Jahre 1990, 2001 und 2010 die Qualität des städtischen Wachstums von Puerto Lopez durch überlappende Formen statistischer und räumlicher Informationen zu beschreiben und zu messen. Sechs Arten von neuen städtischen Formen wurden identifiziert, charakterisiert und hinsichtlich der Nachhaltigkeit bewertet. Die Analyse und der Vergleich von städtischen Formen erlaubten die Beobachtung der sozialen und räumlichen Zersplitterung des städtischen Raums im Jahr 2010 sowie die Bewertung der mangelnden Nachhaltigkeit des unkontrollierten und informellen urbanen Wachstums für den Zeitraum 1990-2010. Das städtische Wachstum von Puerto Lopez wurde durch ein Schema repräsentiert, das auf dem Modell der Badeortentscheidung basiert (Smith, 1991 in Williams, 2009), und Szenarien der städtebaulichen Entwicklung mit und ohne Intervention wurden erarbeitet.International tourism has been one of the most dynamic and growing economic sectors for the last 50 years. With a proved capacity of resilience from the economic crisis and an officially supported positive economic prediction of continuing growth during the first half of the 21st century (Schulte, 2003) (UN-WTO, 2013b), this old but healthy market has produced positive and adverse effects over the territories where it has landed. Nevertheless, the nature of the socio-spatial transformations of landscapes that it triggers over territories in developing countries has been less studied, even minimised.
Based on the transformationalist approach of globalisation (Held & McGrew, 1999 in Murray, 2006) and with a strong believe in the capacity of the national government and communities to manage their local sustainable development (Weiss, 2010), the study attempts to understand the nature of the socio-spatial transformations experienced for the last 20 years by the tourist coastal town of Puerto Lopez. It aims to contribute to the sustainable urban planning and urban design of these types of settlements which are strategic for the improvement of global resilience in the 21st century.
By the combination of the townscape analysis (urban morphology) (Conzen, 2004) and the GIS technology, a method was designed and tested to describe and measure the quality of the urban growth of Puerto Lopez by overlapping shapes of statistical and spatial information of the years 1990, 2001, and 2010. Six types of new urban forms were identified, characterised, and evaluated regarding sustainability. The analysis and comparison of urban forms allowed observing the social and spatial fragmentation of the urban space in 2010 as well as the lack of sustainability of the uncontrolled and informal urban growth for the period 1990-2010. The urban growth of Puerto Lopez was represented by a scheme based on the model of beach resort formation (Smith, 1991 in Williams, 2009), and scenarios of urban planning intervention and no-intervention were shaped
Application of direct tension indicators in preloaded bolted connections
Das Verfahren mit direkten Kraftanzeigern (engl. Direct Tension Indicator, DTI) ist eines von mehreren nach EN 1090-2 angebotenen und geregelten Anziehverfahren zum Vorspannen von hochfesten Schraubengarnituren. Der Einsatz von direkten Kraftanzeigern ist in Deutschland weitestgehend unbekannt. Aufgrund von mangelnder Anwendungserfahrung wurde das Verfahren über den Nationalen Anhang DIN EN 1993-1-8/NA ausgeschlossen.
Für dieses Anziehverfahren wird im Rahmen der vorliegenden Arbeit ein allgemeines Verständnis über das Last-Verformung-Verhalten des DTI sowie über das Tragverhalten der vorgespannten Verbindung mit DTI gewonnen. Darüber hinaus wird überprüft, ob in einer mittels DTI vorgespannten Verbindung der Erhalt der Vorspannkraft auch bei axialer zentrischer Zugbeanspruchung gewährleistet ist. Eine solche Beanspruchung ist nach EN 1993-1-8 ohne besondere Einschränkungen zugelassen.
Die experimentellen Untersuchungen bestehen aus Drucklastversuchen an einzelnen DTI sowie Anziehversuchen an Garnituren mit DTI, um das Erreichen der Mindestvorspannkraft unter Anwendung der Fühllehre zu überprüfen. Vorgespannte Bauteile, für die die Vorspannkraft mit Hilfe der Fühllehre eingebracht wurde, ermöglichen unter Zugbeanspruchung die Beobachtung der vorhandenen Schraubenkraft und von Vorspannkraftverlusten.
Mit den verfügbaren Last-Verformung-Kurven aus den Drucklastversuchen wird das Verspannungsschaubild für die vorgespannte Schraubenverbindung um die elastischen und plastischen Eigenschaften der vorliegenden DTI erweitert. Damit werden zum einen die durchgeführten Zugversuche auf Grundlage von VDI 2230-1 hinsichtlich der Schraubenzusatzkräfte und Vorspannkraftverluste nachgerechnet, zum anderen die geometrischen Grenzen der vorliegenden Versuche um weitere Varianten mit unterschiedlichen Schrauben und Klemmlängen erweitert.
Das mechanische Modell auf Grundlage des Verspannungsschaubildes hat für die Zugversuche in der Montagekonfiguration mit DTI unter dem Schraubenkopf und gedrehter Mutter eine gute Übereinstimmung zu den Versuchen hinsichtlich der Vorspannkraftverluste aus dem Verhalten des DTI bei Laststeigerung gezeigt. Die Untersuchung von Fallbeispielen anhand von unterschiedlich großen Montagevorspannkräften, Beanspruchungsniveaus und Klemmlängen hat ergeben, dass eine Begrenzung der einwirkenden Zugkraft auf die etwa 0,6- bis 0,7-fache Mindestvorspannkraft die aus dem Verformungsverhalten der untersuchten DTI resultierenden Vorspannkraftverluste auf weniger als 5 % der Mindestvorspannkraft begrenzen kann. Bei einer vorausgesetzten Montagevorspannkraft der 1,1-fachen Mindestvorspannkraft wäre dadurch der Erhalt der Mindestvorspannkraft gegeben.
Anhand der experimentellen Untersuchungen und der Modellberechnungen ist ersichtlich, dass der Erhalt der Vorspannkraft unter Zugbeanspruchung stark vom Niveau der Montagevorspannkraft abhängig ist. Da die derzeitigen Gebrauchseignungsprüfungen im Rahmen der normativen Anforderungen nur die Einbringung der einfachen Mindestvorspannkraft gewährleisten, sind die zulässigen Zugbeanspruchungen für vorgespannte Verbindungen mit DTI im Rahmen der Bemessung nach EN 1993-1-8 zu reduzieren. Anderenfalls kann nur generell vom Einsatz des Verfahrens mit DTI in zugbeanspruchten Verbindungen abgeraten werden.The direct tension indicator (DTI) method is one of several tightening methods specified in EN 1090-2 for tightening high-strength structural bolting assemblies to the minimum required preloading force. As the application of direct tension indicators in preloaded bolted connections is largely unknown in Germany and so there is no experience of this method, the use of the DTI method has so far been excluded in Germany by the national annex DIN EN 1993-1-8/NA.
Within the scope of the present study for this tightening method a general understanding of the load-deformation behaviour of the DTI as well as the bearing behaviour of the prestressed connection with DTI is obtained. In addition, it is examined whether the preservation of the preloading force as a result of the DTI method is ensured under external tensile loading. Such a type of loading is permitted according to EN 1993-1-8 without any restrictions.
The experimental investigations consist of compression tests and tightening tests to check the application of a feeler gauge with regard to the achievement of the minimum required preloading force. Tensile tests on preloaded bolted connections give information about the actual bolt force and loss of preload.
The joint diagram according to VDI 2230-1 was extended by the elastic and plastic resiliences of the different DTIs investigated in the compression tests. Herewith on the one hand the experimental tensile tests are simulated. On the other hand the geometrical limitations of the tests are expanded by further variety of the type of bolt and the clamping length.
The mechanical model based on the joint diagram showed a good correlation for the assembly configuration with DTI under the bolt head and rotated nut regarding the loss of preload. The case study on different levels of preload, levels of tensile loading and clamping lengthes showed, that a limitation of the external tensile load to 0.6 to 0.7 times the minimum preloading force may limit the loss of preload induced by the deformation behaviour of the DTI to a maximum of 5 % of the minimum required preloading force. Considering an assembly preload 1.1 times the minimum required preloading force the preservation of the minimum preloading force is ensured.
On the basis of the experimental investigations and the model calculations it can be seen, that under an external tensile loading the preservation of the preloading force mainly depends on the initial level of preloading force. Since the
“suitability test for preloading” only ensures the required minimum preloading force the maximum permissible external tensile loading specified in EN 1993-1-8 should be decreased for the application of the DTI method. Otherwise the DTI method should not be used for preloading structural bolting assemblies which are loaded in tension
Asymmetrische Fluss Feld-Fluss Fraktionierung zur präparativen Aufreinigung funktionalisierter anorganischer Nanopartikel
Im ersten Teil dieser Arbeit wurden Messungen mit verschiedenen Sorten von Nanopartikeln unter wechselnden Bedingungen mittels Asymmetrischer Fluss Feld-Fluss Fraktionierung durchgeführt. Ziel war es, diese Methode als sanfte Alternative zur herkömmlichen Aufreinigung mittels Ultrazentrifugation und Redispergierung durch die Sonotrode zu etablieren. Die dargestellten Ergebnisse sprechen jedoch nicht hinreichend für eine uneingeschränkte Anwendung dieser Methode. Generell wurde durch die Messungen belegt, dass das Ergebnis in erheblichem Maße von den einzustellenden Parametern, wie dem Injektionsfluss, der Injektions- bzw. Fokussierungszeit, dem Fokussierungsfluss und dem Querfluss abhängt. Dies wird auch schon durch die Gleichungen 3.5 bzw. 3.7 zur Berechnung des Diffusionskoeffizienten beschrieben. Weitere Faktoren, die einen starken Einfluss auf das Ergebnis haben sind der zeitliche Verlauf des Querflusses während der Trennungsphase, die Art der Partikel bzw. deren Stabilisierung sowie die Art der Membran. Alle diese Faktoren können mehr oder weniger direkt beeinflusst werden und müssen bei der Bewertung des Ergebnisses berücksichtigt werden. So konnten beispielsweise für CaP/PEI-Nanopartikel mit der RC- und PES-Membran gute Ergebnisse mit einer Rückgewinnungsrate von bis zu 92 % erzielt werden, wie die Messungen 14, 37 und 38 belegen. Auch den positiven Einfluss durch Reduktion der Wechselwirkungen zwischen Partikel und Membran konnte mit den Messungen 39 bis 43 bestätigt werden. Außerdem wurde nachgewiesen, dass ungebundenes Polyethylenimin mittels AF4 von den Partikeln abgetrennt werden kann, wie die Messungen 47 und 48 belegen.
Für die CaP/CMC-Nanopartikel konnte jedoch trotz hoher Rückgewinnungsrate keine hinreichende Trennung erzielt werden, wie die Messungen 49 bis 53 zeigen.
Insgesamt hat sich gezeigt, dass sich die RC-Membran offenbar besser für geringe Injektionsvolumina (bis 2 mL) an CaP/PEI-Nanopartikeln eignet, während mit der PES-Membran bessere Ergebnisse mit größeren Mengen (> 5 mL) erzielt wurden. Generell zeigt sich jedoch, dass auch bei idealen Durchflussparametern ein nicht unerheblicher Teil der injizierten Partikel an der Membran adsorbiert, da die Wechselwirkungen zwischen den Partikeln und der Membran auch mit einem Zusatz an NaCl nicht vollständig unterbunden werden konnten.
Dies ist der Hauptgrund, weshalb die AF4 unter den gegebenen Bedingungen nur bedingt geeignet für die präparative Aufreinigung dieser Partikel erscheint.
Ein weiterer wichtiger Aspekt ist die Änderung der Morphologie, die in einigen Messungen beobachtet wurde. Da dies insbesondere bei Messungen mit hohem Querfluss beobachtet wurde, sollte bei zukünftigen Anwendungen hinsichtlich des Querflusses sehr genau zwischen Nutzen und Risiko abgewogen werden.
Für die metallischen Partikel ergibt sich ein weiteres Problem. Diese zeigten unter den gewählten Bedingungen für alle Messungen einen viel stärkeren Trend zur Adsorption an der Membran, als es für die Calciumphosphat-Partikel der Fall war. Dies deutet daraufhin, dass nicht nur die Oberflächenfunktionalisierung, sondern auch der Partikelkern einen Einfluss auf die Wechselwirkung mit der Membran ausübt. Eine weitere Anwendung sollte daher nur in Betracht gezogen werden, wenn die Wechselwirkung zwischen Partikeln und Membran zu einem hohen Anteil unterbunden werden können. Sind biologische Anwendungen mit den gereinigten Partikeln geplant, so scheiden Tenside für diesen Zweck aus. Es ist jedoch eine Oberflächenfunktionalisierung der Membran selbst denkbar.
Auf diese Weise könnte möglicherweise erreicht werden, dass die Partikel weniger stark an der Membran adsorbieren und gleichzeitig nicht durch weitere Zusätze kontaminiert werden.
Auch für die Calciumphosphat-Nanopartikel sollte diese Methode in Betracht gezogen werden, um zunächst die Reproduzierbarkeit zu steigern, bevor eine Anwendung an biologisch aktiven Partikeln untersucht wird.
Im zweiten Teil dieser Arbeit wurden Calciumphosphat-Nanopartikel dargestellt, die über eine kovalente Funktionalisierung den Komplexliganden DOTA tragen. Dieser ermöglicht die Beladung mit Kupfer(II)-Ionen. Die gezeigten Analysen bestätigen, dass sich diese Partikel hinsichtlich Größe und Morphologie kaum von den anderen dargestellten Calciumphosphat-Nanopartikeln unterscheiden. Auch konnten sie mit in einer recht einfachen Methode hergestellt werden. Die Beladungseffizienz mit Kupfer(II)-Ionen liegt dabei in dem literaturbekannten Bereich. Um eine höhere Beladung der Partikel mit Kupfer zu erreichen, wäre es sinnvoll, einen anderen Liganden zu wählen, der von sich aus bereits eine erhöhte Aufnahmefähigkeit für Kupfer besitzt.Within the first part of this thesis, multiple measurements with different kinds of nanoparticles were carried out using various settings concerning asymmetric flow field-flow fractionation. The main goal was to establish this method as soft alternative to the common way of purification, which is ultracentrifugation followed by sonication.
However, the results presented do not sufficiently indicate the full application of this method. In general, the measurements showed that the results depend to a extensive level on the flow parameters, which are the injection flow, the injection or focusing time, the focus flow and the cross flow. This is also already described by the equations 3.5 and 3.7 for the calculation of the diffusion coefficient. Other factors which have a strong influence on the result are the temporal course of the cross flow during the elution step, the type of the particles or their stabilization as well as the type of the membrane. All these factors can be more or less directly influenced and must be taken into account when evaluating the results. For example, good results with a recovery rate of up to 92 % could be achieved for CaP/PEI nanoparticles with the RC and PES membranes, as shown by measurements 14, 37 and 38. Also the positive influence by reducing the interactions between particles and membrane could be confirmed with the measurements 39 to 43. Moreover, it was shown that unbound polyethylenimine can be separated from the particles by means of AF4, as shown by measurements 47 and 48. For the CaP/CMC nanoparticles, however, a satisfactory separation was not achieved despite a high recovery rate, as shown in the measurements 49 to 53. Overall, the RC membrane has been shown to be better suited for small injection volumes (up to 2 mL) of CaP/PEI nanoparticles, whereas better results with larger amounts (> 5 mL) were achieved with the PES membrane. In general, it is found that even in the case of ideal flow parameters, a not inconsiderable part of the injected particles adsorbs to the membrane, since the interactions between the particles and the membrane could not be completely prevented even with the addition of NaCl. This is the main reason why the AF4 appears to be only of limited applicability for the preparative purification of these particles under the given conditions. Another important aspect is the change in the morphology observed in some measurements. Since this was observed in particular in the case of measurements with a high cross flow, future applications should be weighed very precisely between the risk and the benefit regarding the cross flow. A further problem arises for the metallic particles. Under the selected conditions these showed a much stronger trend for adsorption to the membrane for all measurements as it has been the case for the calcium phosphate particles. This suggests that not only the surface functionalization but also the particle core exerts an influence on the interaction with the membrane. A further application should therefore only be considered if the interaction between particles and membrane can be prevented to a large extent. If biological applications are planned with the purified particles, surfactants are excluded for this purpose.
However, a surface functionalization of the membrane itself is conceivable. In this way, it could possibly be achieved that the particles adsorb less strongly to the membrane and are not contaminated at the same time by further additives.
This method should also be considered for the calcium phosphate nanoparticles in order to first increase the reproducibility before an application to biologically active particles is investigated.
In the second part of this thesis, calcium phosphate nanoparticles were prepared which carry the complex ligand DOTA through a covalent functionalization. This allows loading with copper (II) ions. The performed analyses prove that these particles differ little in terms of size and morphology from the other calcium phosphate nanoparticles shown in this work. Also, they can be prepared by means of a rather easy method. The loading efficiency with copper (II) ions is in the range known from the literature. In order to achieve a higher loading of the particles with copper, it would be advisable to choose another ligand which already has an increased absorption capacity for copper
Investigation of the domain assembly in ligand-gated and voltage-gated ion channels using computational methods
Ionenkanäle sind essentiell für die Erzeugung und Weiterleitung von elektrischen Signalen im menschlichen Organismus. Sie befinden sich hauptsächling in der Zellmembran von Nerven und Muskelzellen, in denen sie spezifische Aufgaben übernehmen und daher spezielle Eigenschaften entwickelt haben, die diesen Ansprüchen entsprechen. Die Konsequenz daraus ist eine Diversifizierung der Ionenkanal Strukturen, die Aufgrund ihrer Eigenschaften wie z.B. die Ionenselektivität oder die Art der Aktivierung, in Klassen und Familien eingeteilt werden können. Die zentrale Rolle der Ionenkanäle birgt wiederum das Risiko ernsthafter Störungen im Organismus, sobald Fehlfunktionen der Kanäle auftreten. Um dem entgegenwirken zu können, wird intensiv an der strukturellen Aufklärung der Ionenkanäle geforscht. Dies ist mit sehr großen Herausforderungen verbunden und bisher sind nur wenige Ionenkanal Strukturen bekannt.
In dieser Promotionsarbeit wurde die korrekte Anordnung von Ionenkanal Domänen zu einem vollständigen Kanal untersucht, und darüber hinaus die Interaktion zwischen transmembranen Helices innerhalb einer Domäne an einem entsprechenden Beispiel untersucht. Es werden Verfahren beschrieben, die zum einen computerbasierte Modelle von Ionenkanälen auf Basis verschiedenster Informationen erzeugen und zum anderen Rückschlüsse auf etwaige Eigenschaften und Funktionen aus den Modellen ableiten können.
Am Beispiel des 5-HT3A Rezeptors wurde die Assoziation der 4 transmembranen Helices zu einer Domäne analysiert. Als Basis dienten Ergebnisse aus Mutagenese Experimenten, womit einzelne, für die Funktion und Stabilität des Kanals wichtige, Residuen identifiziert wurden und als Kriterien für die Erstellung eines Modells genutzt werden konnten. Mit dem Modell wurden in der Folge Molekulardynamik Simulationen und Berechnungen der Bindungsenergien einzelner Residuen durchgeführt, um den Energiebeitrag der einzelnen Aminosäuren zur Assoziation der Helices zu bestimmen und mit den experimentellen Beobachtungen in Relation zu setzen. Die in der Zwischenzeit publizierte 5-HT3A Struktur diente abschließend zur Verifizierung der Ergebnisse.
Die Fragestellung beim heterotetrameren Nav1.8 Kanal war, welche Anordnung die 4 untereinander verbundenen Domänen einnehmen, um einen funktionierenden Kanal zu bilden. Neben den in der Literatur diskutierten Varianten, gegen und im Uhrzeigersinn, wurden auch Modelle mit 4 weiteren theoretisch möglichen Varianten mit kreuzweiser Domänenanordnung erzeugt und untersucht. Neben Molekulardynamik Simulationen und Energieberechnungen der Domänen Interfaces, wurde auch die sequenzbasierte Direct Coupling Analyse (DCA) verwendet, wodurch evolutionär miteinander gekoppelte Residuen identifiziert werden können. Mit der Kombination der verschiedenen Ansätze konnte eine klare Tendenz zu einer Anordnung der Domänen im Uhrzeigersinn erkannt werden, die im Einklang mit einer kürzlich veröffentlichten Kalziumkanal Struktur steht

Balance between the innate immune system and adaptive immune responses results in better antiviral immune activation. Not enough or too much activation can lead to impairment of immune responses. In this thesis, we have investigated the role of enforced viral replication and different mechanisms by which adequate innate and adaptive immune responses are generated.
Recently, it was shown that Usp18 dependent viral replication in splenic CD169+ macrophages are important to induce a sufficient type I interferon response resulting in the subsequent activation of adaptive immune responses. Here, in one study, we have shown that the intracellular replication of viral particles is sufficient to generate virus specific CD8 T cell responses. On the other hand, extracellular distribution of viral particles along the splenic conduits was necessary for inducing systemic levels of type I interferon (IFN-I). We have shown that Usp18 is important for viral replication, but that B cell-derived lymphotoxin beta is necessary for the extracellular distribution of virus along the marginal zone in spleen.
In a further study, we have demonstrated a role of CEACAM1 in B cell survival, proliferation and innate immune activation. B cells are important part of immune system because they are in close proximity of blood-borne antigen in splenic circulation, they secrete antibodies and maintain the lymphoid architecture. We have shown that cell intrinsic signalling of CEACAM1 is essential for survival of proliferating B cells. CEACAM1 is involved in B-cell receptor and signals through the Btk/Syk/NF-κB axis. Absence of CEACAM1 signalling leads to reduced number of B-cells in lymphoid organs especially marginal zone B cells and CD169+ macrophages. Absence of CEACAM1 leads to poor anti-viral antibody secretion after systemic infection with cytopathic virus resulting in early death of Ceacam1–/– mice.
In another set of experiment, we investigated the role of virus specific antibodies on adaptive immune activation. With the use of chronic virus strain LCMV-Docile, we determined whether enforced viral replication could occur in presence of virus-specific antibodies or virus-specific CD8 T cells. We found that after systemic recall infection, virus-specific antibodies allow viral replication in splenic marginal zone but controlled in peripheral organs resulting in strong CD8 T cell priming whereas, virus-specific CD8 T cells blunted viral replication in spleen but failed to control the persistent chronic viral infection.
In conclusion, using several mouse models of noncytopathic and cytopathic viral infection, we have elucidated the functional role of lymphotoxin beta, CEACAM1 and virus-specific antibodies in immune function particularly during enforced viral replication in the spleen.Die Aktivierung des adaptiven und angeborenen Immunsystems ist essentiell für die Kontrolle viraler Infektionen. Effects of river restoration on ecosystem metabolism and trophic relationships
Following a long history of human pressures on riverine ecosystems, the European Water Framework Directive (WFD, Directive 2000/60/EC) came into force in 2000 and initiated a new period of river management in Europe. The directive aims at improving the ecological and chemical status of rivers in order to achieve the ‘good status’ of all surface waters until the year 2027. In Europe, degraded hydromorphology has been highlighted as a central impact to the ecological status of the rivers. For instance, in countries such as Germany, the hydromorphology of almost all river sections is affected to an extent that they fail to meet the WFD goals. In response, river hydromorphology is nowadays being restored at an increasing rate. Restoration of river hydromorphology has the potential to affect not only structural ecosystem features, including species composition and diversity, but also functional aspects, such as key ecosystem processes and trophic transfers of energy and nutrients. Despite this, the most-widely used parameters for assessing the success or failure of restoration projects are almost exclusively based on changes in community composition of different biological groups (e.g., fish, benthic invertebrates, and macrophytes). Functional metrics, even though increasingly recognized as a valuable addition to classical assessments, are rarely used to study restoration effects. Consequently, the outcomes of river restoration for key ecosystem processes (e.g., river metabolism) and trophic relationships (e.g., trophic structure of benthic invertebrate communities and trophic connectivity between river and land) remain poorly understood. Against this background, the present thesis focused on the following objectives: (i) The application of stable isotope analysis (δ13C, δ15N) together with quantitative community metrics to characterize changes in the trophic structure of benthic invertebrate communities following restoration. (ii) The characterization of the isotopic composition (δ13C, δ15N) of consumers in aquatic, riparian, and terrestrial habitats to assess restoration effects on the trophic connectivity between river and land. (iii) The assessment of reach-scale restoration effects on hydromorphology, habitat composition and hydrodynamics and the estimation of the corresponding responses of river ecosystem metabolism
Generalized factorial cumulants applied to Coulomb-blockade systems
Tunneling of electrons through a Coulomb-blockade system is a stochastic (i.e., random) process. The number of the transferred electrons per time interval is determined by a probability distribution. The form of this distribution can be characterized by quantities called cumulants. Recently developed electrometers allow for the observation of each electron transported through a Coulomb-blockade system in real time. Therefore, the probability distribution can be directly measured.
In this thesis, we introduce generalized factorial cumulants as a new tool to analyze the information contained in the probability distribution. For any kind of Coulomb-blockade system, these cumulants can be used as follows:
First, correlations between the tunneling electrons are proven by a certain sign of the cumulants. In the limit of short time intervals, additional criteria indicate correlations, respectively. The cumulants allow for the detection of correlations which cannot be noticed by commonly used quantities such as the current noise. We comment in detail on the necessary ingredients for the presence of correlations in the short-time limit and thereby explain recent experimental observations.
Second, we introduce a mathematical procedure called inverse counting statistics. The procedure reconstructs, solely from a few experimentally measured cumulants, characteristic features of an otherwise unknown Coulomb-blockade system, e.g., a lower bound for the system dimension and the full spectrum of relaxation rates.
Third, the cumulants reveal coherent oscillations or other processes transferring single electrons in a regular manner. The sensitivity for the oscillations increases dramatically in comparison to the commonly used quantities as the waiting times and finite-frequency current noise. The increased sensitivity is especially convenient for the detection of coherent oscillations in already existing experimental set-ups.
Fourth, a violation of detailed balance is indicated by the cumulants. Detailed balance means the absence of net probability currents in the system's state space for the stationary limit. A violation of detailed balance is possible if the state space consist of more than two states connected by tunneling rates in such a way that a closed loop is formed.
We illustrate the application of the generalized factorial cumulants for the transport through single metallic islands and quantum dots tunnel coupled to superconducting, ferromagnetic, or normal metallic electrodes. For all these example systems, it has been demonstrated already in experiments that electron transport can be observed in real time. Thus, our results can be applied immediately to such experimental realizations of a Coulomb-blockade system.Das Tunneln von Elektronen durch ein Coulomb-Blockade-System ist ein stochastischer Prozess bzw. Zufallsprozess. Die Anzahl an transferierten Elektronen pro Zeitintervall ist bestimmt von einer Wahrscheinlichkeitsverteilung. Die Form dieser Verteilung kann durch Größen charakterisiert werden, die Kumulanten genannt werden. Kürzlich entwickelte Elektrometer ermöglichen es, jedes Elektron in Echtzeit zu beobachten, das durch ein Coulomb-Blockade-System transportiert wird. Deshalb kann die Wahrscheinlichkeitsverteilung direkt gemessen werden.
In dieser Dissertation führen wir generalisierte faktorielle Kumulanten als ein neues Instrument ein, um die in der Wahrscheinlichkeitsverteilung enthaltenden Informationen zu analysieren. Für jede Art von Coulomb-Blockade-System können diese Kumulanten wie folgt genutzt werden:
Erstens, Korrelationen zwischen den tunnelnden Elektronen werden durch ein bestimmtes Vorzeichen der Kumulanten nachgewiesen. Im Limes kurzer Zeitintervalle zeigen zusätzliche Kriterien jeweils Korrelationen an. Die Kumulanten ermöglichen das Detektieren von Korrelationen, die von üblicherweise verwendeten Messgrößen wie dem Stromrauschen nicht bemerkt werden. Wir erläutern im Detail die notwendigen Voraussetzungen für das Vorhandensein von Korrelationen im Limes kurzer Zeiten und erklären dabei jüngste experimentelle Beobachtungen.
Zweitens, wir führen eine mathematische Prozedur namens inverse Zählstatistik ein. Die Prozedur rekonstruiert, ausschließlich mittels einiger experimentell gemessener Kumulanten, charakteristische Merkmale eines ansonsten unbekannten Coulomb-Blockade-Systems, z. B. eine untere Grenze für die Dimension des Systems und das volle Spektrum an Relaxationsraten.
Drittens, die Kumulanten offenbaren kohärente Oszillationen oder andere Prozesse, die einzelne Elektronen in einer regelmäßigen Weise transferieren. Die Sensitivität für die Oszillationen steigt dramatisch an im Vergleich zu den im Allgemeinen genutzten Messgrößen wie den waiting times und dem frequenzabhängigen Stromrauschen. Die erhöhte Sensitivität ist insbesondere günstig, um kohärente Oszillationen in bereits existierenden experimentellen Aufbauten zu detektieren.
Viertens, eine Verletzung des Prinzips des detaillierten Gleichgewichts wird von den Kumulanten angezeigt. Detailliertes Gleichgewicht bezeichnet die Abwesenheit von netto Wahrscheinlichkeitsströmen im Zustandsraum des Systems für den stationären Limes. Eine Verletzung des detaillierten Gleichgewichts ist möglich, falls der Zustandsraum aus mehr als zwei Zuständen besteht, die über Tunnelraten derart verknüpft sind, dass sich eine geschlossene Schleife bildet.
Wir veranschaulichen die Anwendung der generalisierten faktoriellen Kumulanten für den Transport durch einzelne metallische Inseln oder Quantenpunkte, die an supraleitende, ferromagnetische oder gewöhnliche metallische Elektroden tunnel-gekoppelt sind. Für all diese Beispielsysteme ist bereits in Experimenten demonstriert worden, dass Elektronentransport in Echtzeit beobachtet werden kann. Daher können unsere Resultate sofort auf derartige experimentelle Realisierungen eines Coulomb-Blockade-Systems angewendet werden
Scale-Bridging of Elasto-Plastic Microstructures using Statistically Similar Representative Volume Elements
Die vorliegende Arbeit behandelt die numerische Modellierung des mechanischen Verhaltens mikroheterogener Materialien, wobei das Hauptaugenmerk auf Dualphasenstähle gelegt wird. Ihr makroskopisches Verhalten wird durch die Interaktion der Einzelphasen auf mikrostruktureller Ebene geprägt. Der Einfluss der Morphologie einer realistischen Mikrostruktur kann durch die Verwendung von repräsentativen Volumenelementen (RVEs) unter Anwendung der FE²-Methode direkt in die Materialmodellierung einbezogen werden. Dabei entsteht für RVEs, die als Ausschnitte einer realen Mikrostruktur konstruiert werden, ein enormer Rechenaufwand. Eine Reduzierung des Aufwands ist durch die Verwendung von statistisch ähnlichen RVEs (SSRVEs) möglich. Diese sind durch Ähnlichkeit in Bezug auf bestimmte statistische Maße definiert und liefern gleichzeitig Gleichartigkeit des mechanischen Verhaltens. Die verschiedenen Aspekte der Konstruktion von SSRVEs sind ein Schwerpunkt dieser Arbeit. Es wird gezeigt, dass SSRVEs die mechanischen Eigenschaften der realen Mikrostruktur widerspiegeln und damit ihre Verwendung im Rahmen der FE² -Methode ermöglicht wird. Die Simulation makroskopischer Eigenschaften basierend auf polykristallinen RVEs wird gezeigt. Diese ermöglichen die Beschreibung polykristalliner Materialien, welche von ihrer mikrostrukturellen Textur geprägt werden.The present work deals with the numerical modeling of the mechanical behavior of microheterogeneous materials, with a focus on dual-phase steel. The macroscopic behavior of this material is largely influenced by an interaction of the microstructural constituents. The influence of the morphology of a real microstructure can be included in the material modeling by the application of a suitable representative volume element (RVE) in a direct micro-macro homogenization scheme (also known as FE²-method). However, the use of sections of a real microstructure as an RVE can lead to huge computational costs. A cost reduction can be achieved by the application of statistically similar RVEs (SSRVEs). They are governed by similarities of selected statistical measures with respect to a real microstructure and show a comparable mechanical behavior. The different aspects in the construction method are a main focus of this work. It is shown that SSRVEs can resemble the mechanical behavior of a real DP steel microstructure appropriately, which permits their use in
FE²-simulations instead of real microstructures. Aiming for a description of polycrystalline materials governed by texture, the simulation of macroscopic properties based on polycrystalline RVEs is shown
Structural Reforms and Capital Market Interventions during a Financial Crisis
I study how structural reforms in product and labor markets affect an economy that is going through a financial crisis. Of specific interest is the role of credit intermediation in a crisis and how it is influenced by reforms. I consider three key characteristics of the recent financial crisis that are potentially relevant for policy analysis: First, the crisis was triggered in the financial sector; second, there were spillovers from the financial to the real sector due to credit rationing; third, governments actively intervened in the credit market during the crisis. I construct two dynamic general equilibrium models with financial frictions to address these issues --a closed economy model and a monetary union model. I show that permanent structural reforms have positive effects on aggregate output in both the long and the short run. They affect the capital market positively and stimulate credit intermediation. Contrariwise, reforms that are either implemented temporarily or announced to be implemented in the future have negative consequences for output in the short run. Moreover, reforms that are implemented in one country of a currency union have positive short-run effects on both the reforming country and its foreign counterpart. My results also hold if the central bank is constrained by a lower interest rate bound. I also show that reforms have a qualitatively similar impact as a direct intervention in the credit market. Moreover, credit market interventions are complementary to structural reforms
