    Effect of Processing Solid Organic Municipal Wastes on Their Phosphorus Fertilizer Value

    The article processing charge was funded by the Open Access Publication Fund of Humboldt-Universität zu Berlin.In the circular economy framework, municipal wastes are seen as secondary raw materials that can be used to fertilize agricultural soils. This study assessed the effect of different biowaste and green waste treatment schemes on P fertilizer value to learn about the optimal valorization of these feedstocks. The wastes were used either fresh, after composting or anaerobic digestion, or as biochars produced at various pyrolysis conditions. The fertilizer value was determined from the change in soil concentration of plant-available P (PCAL) in incubation experiments with different soils and the temporal dynamics of fertilizer-induced growth and P accumulation of ryegrass in a pot experiment with eight harvests. The mode of waste treatment significantly influenced the P fertilizer value in the incubation and in the pot experiment. In the incubation experiment, the amendment-induced PCAL increase varied between 22% and 33% of applied P on low-P acidic soil and between 55% and 88% of applied P on high-P acidic soil, whereby the amendment effects were mainly determined by their effects on soil pH. In the pot experiment with low-P acidic soil, the apparent P recovery in the plant biomass (APR) varied between 2% of applied P for fresh green waste and 42% for fluid digestate. The amendment effects on APR were not related to soil pH but to the PCAL supply with the amendments and amendment effects on soil P supply.     Towards an interoperable perovskite description or how to keep track of 300 perovskite ions

    Hybrid perovskites are interesting optoelectronic materials. The perovskite ABX3 structure offers a vast compositional space, and we have identified over 300 perovskite ions. This flexibility enables tuneable properties and has significantly contributed to the success of perovskite optoelectronics. However, this diversity also leads to confusion, ambiguity, and inconsistencies causing challenges for data mining and machine learning applications. To address this issue, we propose guidelines and a JSON schema to standardize the reporting of perovskite compositions. The schema adheres to IUPAC recommendations and is designed to make data both human- and machine-readable. It captures key descriptors such as perovskite composition, molecular formula, SMILES representation, IUPAC name, and CAS number for each ion. To facilitate adoption, we have developed utilities to automatically generate comprehensive and standardized perovskite descriptions from standard ion abbreviations and stoichiometric coefficients.     Effect of MACC1 on therapy-induced senescence and its impact on the metastasis formation

    Trotz erheblicher Fortschritte in der Krebsforschung bleibt das CRC weltweit eine der Hauptursachen für krebsbedingte Mortalität. Insbesondere die Fernmetastasierung führt zu einer deutlich verringerten Gesamtüberlebensrate und verdeutlicht die Notwendigkeit, die zugrunde liegenden molekularen Mechanismen der Metastasierung zu verstehen. MACC1 hat sich in über 20 Tumorentitäten als prognostischer, prädiktiver und kausaler Biomarker etabliert. In dieser Studie wurde die Rolle von MACC1 auf die Seneszenz und deren nachgelagerte Auswirkungen auf die Metastasierung untersucht. Traditionell wird Seneszenz als Schutzmechanismus gegen maligne Progression betrachtet. Neuere Erkenntnisse deuten jedoch darauf hin, dass seneszente Zellen in bestimmten Fällen mit erhöhter Tumorigenität erneut in den Zellzyklus eintreten können. In dieser Studie wurde gezeigt, dass die Überexpression von MACC1 zu einer signifikanten Verstärkung der therapieinduzierten Seneszenz (TIS) geführt hat. Die Induktion der Seneszenz ging mit einer verstärkten Expression von Stammzell-Markern wie CD44 und LGR5 einher; dieser Effekt war in Zellen mit bereits hohem MACC1-Ausgangsniveau besonders ausgeprägt. Weiterführende mechanistische Untersuchungen ergaben, dass spezifische MACC1-Inhibitoren die von MACC1 induzierte TIS abschwächen können. Die Zellen, die gezielt aus der Seneszenz entlassen wurden, zeigten eine gesteigerte Tumorigenität. In vivo konnten zuvor nicht-metastasierende Zellen nach der Freisetzung aus der Seneszenz Metastasen ausbilden. Darüber hinaus behielten diese post-seneszenten Zellen mehrere TIS-assoziierte Eigenschaften und blieben empfindlich gegenüber senolytischen Substanzen. Zusammenfassend offenbart diese Arbeit eine bisher unbekannte Rolle von MACC1 bei der Induktion von TIS und unterstreicht dessen Einfluss auf die Metastasierung. Diese Erkenntnisse bilden die Grundlage für neue therapeutische Strategien, bei denen MACC1-Inhibitoren zur Einschränkung der TIS-Induktion führen.Despite significant advances in cancer research, colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality worldwide. In particular, distant metastasis leads to a markedly reduced overall survival rate and highlights the need to understand the underlying molecular mechanisms of metastasis. MACC1 has been established as a prognostic, predictive, and causal biomarker in over 20 tumor entities. This study investigated the role of MACC1 in cellular senescence and its downstream effects on metastasis. Traditionally, senescence is considered a protective mechanism against malignant progression. However, recent findings suggest that senescent cells can, in certain cases, re-enter the cell cycle with increased tumorigenicity. This study demonstrated that MACC1 overexpression significantly enhanced therapy-induced senescence (TIS). Senescence induction was accompanied by increased expression of stem cell markers such as CD44 and LGR5; this effect was particularly pronounced in cells with already high baseline levels of MACC1. Further mechanistic investigations revealed that specific MACC1 inhibitors can attenuate MACC1-induced TIS. Cells that were deliberately released from senescence exhibited increased tumorigenicity. In in vivo model, previously non-metastasizing cells were able to form metastases after release from senescence. Moreover, these post-senescent cells retained several TIS-associated properties and remained sensitive to senolytic agents. In summary, this work reveals a previously unknown role of MACC1 in the induction of TIS and underscores its impact on metastasis.     Systemic transcriptional profiling of immune cell heterogeneity in autoimmune disease

    Was sind die genomischen Fußabdrücke der Immunaktivierung? Diese Dissertation geht dieser Frage nach, indem mRNA- und Omics-Technologien eingesetzt werden – Werkzeuge, die sowohl bekannte Immunantworten analysieren als auch subtile Übergänge aufdecken, welche die immunologische Heterogenität prägen. Ziel ist es, immunologische Signaturen mittels genomweiter Datensätze zu identifizieren und das Verständnis der Immunregulation zu vertiefen. Die Arbeit umfasst drei Studien: zwei experimentelle Datensätze aus der Dörner-Gruppe an der Charité – Universitätsmedizin Berlin und der Romagnani-Gruppe am DRFZ Berlin sowie einen öffentlichen Datensatz. Die erste Studie (PMID: 38102122) untersucht die Genexpression in Immunzellen nach anti-CD3-Stimulation und identifiziert Interferon-stimulierte Gene (ISGs). Die zweite Studie beleuchtet die Reifung natürlicher Killerzellen (NK-Zellen) und deren transkriptionelle Diversität während einer HCMV- Infektion, wobei klonale Expansion und Gedächtnisbildung im Vordergrund stehen (PMID: 36289449). In der dritten Studie wird Einzelzell-RNA-Sequenzierung (scRNA-seq) genutzt, um systemischen Lupus erythematodes (SLE) bei pädiatrischen Patienten zu untersuchen – mit Einblicken in die Rolle von Monozyten und T-Zellen in der Krankheitsdynamik (PMID: 32747814). Diese Studien beleuchten zelltypspezifische Immunaktivierung, Marker autoimmuner Erkrankungen sowie die klinische Relevanz von ISGs, und sie erweitern Einzelzellmethoden zur Identifikation verborgener Akteure der adaptiven Immunität. Ein Schwerpunkt liegt auf der Erfassung dynamischer Immunprozesse durch Genexpressions- Heterogenität. Durch die Abkehr von populationsbasierten Mittelwerten werden feine Regulationsmuster sichtbar, die durch Reifung und Aktivierungszustände geformt sind. Die Studien zeigen, wie transkriptionelle Variabilität die Flexibilität und Reaktionsfähigkeit von Immunzellen in autoimmunen und viralen Kontexten widerspiegelt. Zur Unterstützung wurden rechnergestützte Methoden entwickelt, um Heterogenität zu quantifizieren, Differenzierungspfade zu rekonstruieren und Ligand-Rezeptor-Interaktionen über Immunzelltypen hinweg zu entschlüsseln. Diese Erkenntnisse vertiefen das Verständnis immunologischer Dynamiken und betonen die Rolle zellulärer Diversität in Krankheitsverläufen und Therapieansätzen.What are the genomic footprints of immune activation? This dissertation addresses that question by leveraging mRNA and Omics technologies, tools for dissecting known immune responses and uncovering the subtle transitions that define immune heterogeneity. The primary objective is to identify immunological signatures through genome-scale data and to refine our understanding of immune regulation. It comprises three studies: two experimental datasets from the Dörner group at Charité Universitätsmedizin Berlin and the Romagnani group at DRFZ Berlin, and one public dataset. The first study (PMID: 38102122) investigates gene expression in immune cells after anti-CD3 stimulation, revealing Interferon-stimulated gene (ISG) responses. This analysis provided insights into the functional roles of monocytes and T cells in modulating disease activity and influencing the dynamic trajectory of SLE progression. The corresponding dataset of third study for this investigation was sourced from PMID: 32747814. Collectively, these studies explore immune cell type-specific activation, autoimmune disease markers, and the clinical relevance of ISG, while advancing single-cell methods to uncover hidden mediators of adaptive immunity. Special emphasis is placed on capturing the dynamic immune processes through the lens of gene expression heterogeneity. By moving beyond population-level averages, this work uncovers fine-grained regulatory patterns shaped by immune cell maturation, and activation states. The studies reveal how transcriptional variability reflects the flexibility and responsiveness of immune cells in both autoimmune and viral settings. To support this, computational approaches were developed to quantify heterogeneity, trace differentiation trajectories, and decode ligand-receptor interactions across diverse immune cell types. These insights deepen our understanding of immune response dynamics, emphasizing how cellular diversity drives disease progression and guides therapeutic strategies

    Rethinking Urban Food Systems in Nairobi: Embracing Local Context for Sustainable Transformation

    Urban food systems are crucial for addressing sustainability, equity, and resilience, especially in rapidly growing cities of the Global South. Yet, urban planners have long neglected them. In Nairobi, where up to 60% of residents live in informal settlements and many spend over half their income on food, the food system remains fragmented despite extensive interventions. Drawing on research in Kasarani, a constituency of Nairobi, this article explores how residents navigate and use the social, economic, and infrastructural dynamics of their neighborhoods to secure food and their livelihoods. We show that policy approaches centered on formalization and large‐scale projects often ignore the value of everyday practices and local networks. Informal actors, such as mama mbogas (fresh produce traders), are vital for food security and community resilience. We advocate for urban food governance that embraces the coexistence of on‐ and off‐grid systems and recognizes informal economies as central to urban resilience. By emphasizing residents’ lived experiences, we highlight pathways for more inclusive and transformative urban food planning.Peer Reviewe

    Untersuchungen zur sprachlichen Komplexität von Textvorlagen und Textproduktionen in den Fächern Deutsch und Englisch

    In dieser Dissertation wird untersucht, wie sich sprachliche Anforderungen und Leistungen im Abitur anhand des Konstrukts der sprachlichen Komplexität beschreiben lassen, ob und inwiefern eine Umsetzung von Vorgaben zu sprachlicher Komplexität in den Bildungsstandards für die Allgemeine Hochschulreife stattfindet, und welche Zusammenhänge zwischen sprachlicher Komplexität, Aufgabenmerkmalen, Schüler*innenmerkmalen und Bewertung bestehen. Datengrundlage der drei Studien waren Aufgaben aus den ländergemeinsamen Abituraufgabenpools (2017 bis 2022) sowie originale Abiturarbeiten aus dem Prüfungsjahr 2017. Die sprachliche Komplexität der Texte wurde anhand theoretisch und empirisch ausgewählter Komplexitätsmaße mit Hilfe der Online-Plattform CTAP analysiert. Die Ergebnisse zeigen, dass im Fach Deutsch Textvorlagen zum erhöhten Anforderungsniveau in einzelnen Maßen komplexer waren als Texte zum grundlegenden Niveau. Vor allem inhaltliche und stilistische Aspekte wurden von Lehrkräften als relevant für die Einschätzung der Textschwierigkeit angesehen. Sprachlichen Eigenschaften von Schüler*innentexten im Fach Englisch unterschieden sich vor allem im Zusammenhang mit den Aufgabentypen (Zusammenfassung, Analyse, Argumentation). Außerdem waren besonders Aufgaben, die eine hohe sprachliche Selbstständigkeit erforderten, für die Abbildung der Sprachkompetenz der Schüler*innen geeignet. In ihre Bewertung von Schüler*innentexten im Fach Deutsch ließen Lehrkräfte vor allem bildungssprachliche Aspekte einfließen, wobei Schwierigkeiten in der separaten Bewertung inhaltlicher Aspekte, sprachlicher Leistung und Fehlerfreiheit bestanden. Die Dissertation liefert Erkenntnisse zur validen Operationalisierung und Hinweise für eine Konkretisierung sprachlicher Komplexität in den Bildungsstandards. Sie verdeutlicht zudem Zusammenhänge sprachlicher Komplexität mit Aufgabenmerkmalen, Schüler*innenmerkmalen und Bewertung, die bei der Interpretation sprachlicher Texteigenschaften relevant sind.This dissertation examines how language demands and writing performance can be characterized with regard to linguistic complexity in the context of the German Abitur. It investigates whether specifications regarding linguistic complexity in the German educational standards for upper secondary education are met in practice. Furthermore, the relationships between linguistic complexity, task characteristics, student features, and writing assessment are examined. Abitur examination tasks from the years 2017 to 2022 as well as original student texts written during Abitur examinations in 2017 served as database for three empirical studies. The linguistic complexity of reading materials and student texts was analyzed using the web-based platform CTAP based on theoretically and empirically motivated measure samples. Results showed that reading materials for higher-level exams in the subject German were more complex compared to texts for fundamental-level exams for a subset of measures. Teachers considered content and style aspects as particularly relevant to determine text difficulty. In the subject English as a foreign language, student text features differed primarily by the functional needs of the task types employed (summary, analysis, argumentation). Furthermore, tasks that demanded a high independence in language performance seemed best suited to differentiate students’ language proficiency. In their grading of student essays in the subject German, teachers especially considered academic language features. They faced difficulties in separately grading aspects of language performance, content and accuracy. This dissertation provides insights into the valid operationalization of linguistic complexity and suggestions on how to specify the construct in educational standards. Moreover, it points to relationships between linguistic complexity, task characteristics, student features, and writing assessment that are relevant when interpreting linguistic text characteristics

    Three Essays in Macroeconomics and Labor Economics

    Diese Dissertation umfasst drei Essays, welche untersuchen, wie sich Heterogenität, sowohl zwischen Haushalten als auch zwischen Ländern, auf Konjunktur und Arbeitsmarkt auswirkt, mit einem Hauptfokus auf Europa. Der erste Essay untersucht die Zyklizität der Reallöhne anhand von Mikrodaten zu europäischen Haushalten und zeigt eine signifikante Prozyklizität auf individueller Ebene auf, die in aggregierten Daten durch einen Kompositionseffekt maskiert wird. Die Studie stellt erhebliche Unterschiede zwischen den Ländern fest: In den neueren EU-Mitgliedsstaaten und den Ländern außerhalb der Eurozone ist die Lohnflexibilität größer als in den älteren Mitgliedsstaaten und den Ländern innerhalb der Eurozone. Der zweite Essay befasst sich mit der Wirksamkeit von Kurzarbeitsprogrammen, die es Unternehmen ermöglichen, die Arbeitszeit ihrer Beschäftigten in Zeiten eines Konjunkturrückgangs vorübergehend zu reduzieren, während der Staat den daraus resultierenden Lohnverlust teilweise ausgleicht. Eine Analyse von Haushaltsdaten aus Deutschland, kombiniert mit einem heterogenen neukeynesianischen Modell, welches Suchfriktionen sowie Kurzarbeitsentscheidungen berücksichtigt, zeigt, dass Kurzarbeit die Schwankungen der Arbeitslosigkeit mildert, indem sie das Konsumrisiko für Arbeitnehmer senkt. Der dritte Aufsatz untersucht, wie Unterschiede in der Struktur der Arbeitsmärkte zwischen den Ländern die wirtschaftliche Dynamik in einer Währungsunion beeinflussen. In einem neukeynesianischen Zwei-Länder-Modell mit Arbeitslosigkeit und heterogenen Agenten führt vorsorgliches Sparen als Reaktion auf das Risiko von Arbeitslosigkeit zu volatileren Konjunkturzyklen in der gesamten Union. Wenn sich die Arbeitsmärkte erheblich unterscheiden, beispielsweise weil ein Land flexiblere Einstellungs- und Entlassungspraktiken hat als das andere, unterscheidet sich auch das vorsorgliche Sparverhalten, was zu Unterschieden bei den Schwankungen von Inflation und Arbeitslosigkeit beiträgt.This thesis comprises three essays that explore the role of heterogeneity, both across households and countries, in shaping labor market dynamics and macroeconomic outcomes, with a primary focus on Europe. The first essay investigates real wage cyclicality using microdata on European households, revealing significant procyclicality at the individual level, which is masked in aggregate data by a composition effect. The study finds substantial cross-country differences, with newer EU member states and countries outside of the euro area displaying greater wage flexibility than older member states and euro-area countries. The second essay evaluates the effectiveness of short-time work schemes, which allow firms to temporarily reduce employees’ working hours during downturns while the government partially compensates for lost wages. Combining household data from Germany with a Heterogeneous-Agent New Keynesian model, which includes search and matching frictions and short-time work decisions, the essay shows that short-time work mitigates unemployment fluctuations and reduces precautionary savings by lowering consumption risk for workers. The third essay examines how differences in the structure of labor markets across countries influence economic dynamics in a monetary union. Utilizing a two-country Heterogeneous-Agent New Keynesian model with unemployment, the analysis shows that precautionary saving in response to unemployment risk leads to more volatile business cycles across the union. When labor markets differ significantly, for instance, due to one country having more flexible hiring and firing than the other, precautionary savings behavior also differs, contributing to divergent responses of inflation and unemployment to shocks. However, when the model is calibrated to the euro area quantitatively, the relatively moderate asymmetry in the euro area limits its impact on inflation differentials

    Asymptotic self-similar behavior in coupled systems

    Selbstähnliches Verhalten ist ein gut untersuchtes Phänomen in räumlich ausgedehnten Systemen. Der Großteil der Forschung hat sich jedoch auf skalare Probleme, die exakte selbstähnliche Lösungen besitzen, konzentriert, wie beispielsweise die Poröse-Medien-Gleichung. Des Weiteren werden Lösungen meist mit trivialem Verhalten im Unendlichen untersucht, insbesondere bei Betrachtung endlicher Masse oder Energie. In dieser Arbeit wird gezeigt, dass Selbstähnlichkeit auch asymptotisch auftreten kann, wodurch eine qualitative Beschreibung des Langzeitverhaltens ermöglicht wird. Exemplarisch werden wir zwei Systeme untersuchen: Das erste ist ein gekoppeltes Reaktions-Diffusions-System, das das Massenwirkungsgesetz erfüllt. Das zweite enthält die eindimensionalen kompressiblen Euler-Gleichungen mit Dämpfung. Obwohl sich beide Systeme in vielerlei Hinsicht mathematisch unterscheiden, haben sie eines gemeinsam, nämlich ihr asymptotisch selbstähnliches Verhalten. Wir definieren beide Systeme auf dem Ganzraum und statten sie mit nichttrivialen Randdaten im Unendlichen aus. Damit besteht die Hauptaufgabe darin, die Schwierigkeiten des unbeschränkten Gebiets und der unendlichen Masse zu überwinden. Der entscheidende Schritt ist, Raum und Zeit in parabolische Skalierungsvariablen zu transformieren und Energie-Dissipationsabschätzungen für eine relative Entropie herzuleiten. Mit dieser Methode zeigen wir für beide Systeme, dass ihre Lösungen exponentiell zu Ähnlichkeitsprofilen konvergieren, wenn die skalierte Zeit gegen Unendlich geht. In den ursprünglichen Variablen entspricht dies einem asymptotisch selbstähnlichen Verhalten, das das Vermischen der unterschiedlichen Zustände im Unendlichen beschreibt.Self-similar behavior is a well-studied phenomenon in spatially extended systems. However, the majority of research has focused on scalar problems that possess exact self-similar solutions, such as the porous medium equation. Moreover, solutions are usually considered with trivial behavior at infinity, particularly in the case of finite mass or energy. In this thesis, we demonstrate that self-similarity can also occur asymptotically, thus enabling the qualitative description of long-time behavior. As guiding examples, we will mainly examine two systems: First, a coupled reaction-diffusion system with mass-action kinetics, and second, the one-dimensional compressible Euler equations with damping. Even though both systems are mathematically different in many respects, they have one thing in common: their asymptotic self-similar behavior. We consider both systems to be defined on the whole space and prescribed with nontrivial boundary data at infinity. Hence, the main task is to overcome the difficulties of the unbounded domain and infinite mass. The key step in the analysis is rescaling time and space into so-called parabolic scaling variables and deriving energy-dissipation estimates for a relative entropy. With this approach, we show for both systems that their solutions converge exponentially towards similarity profiles if the scaled time goes to infinity. In the original variables, these profiles correspond to asymptotically self-similar behavior describing the phenomenon of mixing the states at infinity

    The politics of seeking and avoiding discourse in parliament

    When do politicians debate each other in parliament, and when do they prefer to avoid discourse? While existing research has shown MPs to unilaterally leverage the dialogical nature of legislative debates to their advantage, the circumstances facilitating actual discursive interaction have so far received less attention. We introduce a new framework to study the emergence of discourse in political debates. Applying this framework, we expect ideological differences and government–opposition dynamics to shape politicians' choices about seeking or avoiding discourse. To test these hypotheses, we draw on an original dataset of all 14,595 attempted and successful interventions ( Zwischenfragen ) – extraordinary, voluntary discursive exchanges between speakers and MPs in the audience – in the German Bundestag (1990–2020), extracted using an annotation pipeline developed specifically for this study. We find that MPs separated by diverging preferences seek discourse with one another more often than their ideologically aligned counterparts. At the same time, these exact attempts do less frequently result in discursive interactions. When considering government–opposition dynamics in this process, we observe very similar patterns: Attempts to initiate discourse are particularly common among opposition MPs facing government speakers, and we find tentative evidence suggesting that government actors are most likely to avoid these invitations to discursive interaction.     Introduction: Theorizing Transtextual Characters in Ancient and Medieval Literature

    This introduction maps the field of transtextual characters: what are such characters, how have they been approached in scholarship, especially narrative theory and media studies, and why can a perspective on premodern material in particular give new impulses to their analysis? We discuss, first, the relevance of distinguishing between text and work. In premodern contexts, texts are often unstable and transmitted in various witnesses, versions, and redactions, calling into question the notion of a ›work‹. This has an impact also on how transtextual characters function. The proximity and distance of single texts to one another are decisive for the investigation of transtextual characters, and too great a correspondence does not create transtextual characters. We then consider the pragmatic contexts of reception in antiquity and the Middle Ages. These are a further challenge because fictionality, fictivity, and factuality were construed differently, based on the ancient distinction between historia , argumentum , and fabula . What is more, the transtextuality of characters is also dependent on genre. The generic dimension of transtextual characters relies on narrative worlds of high stability, which are clearly separated from one another, and can be amplified by seriality. We then chart four theoretical approaches to determine and describe the identity of literary characters in different works: (1) the idea of an iconic core, which refers to a stable, nuclear set of properties that are not modified; (2) the concept of persona , which is based on the ancient idea of characters as masks for text-external persons; (3) the prototype model, which is based on the presence or absence of key character traits across time; and (4) typological writing, that is, the use of prefiguration as a method to build character.Peer Reviewe
