    Into space: Statistical inference for stochastic partial differential equations using spatial information

    Diese Arbeit trägt zum Gebiet der Statistik für semi-lineare stochastische partielle Differentialgleichungen (SPDEs) bei. Die Diffusivität und die Stärke des Rauschens sind bekannt, aber die Reaktionsfunktion ist unbekannt und soll basierend auf einer Beobachtung der SPDE geschätzt werden. Da eine Beobachtung der SPDE im Allgemeinen nicht genügend Informationen für die Identifizierung der Reaktionsfunktion enthält, wird analysiert, wie der Informationsgehalt der räumlichen Schnitte über die Reaktionsfunktion erhöht werden kann. Sowohl parametrische als auch nicht-parametrische Methoden zur Schätzung der Reaktionsfunktion werden hergeleitet und analysiert. Die statistische Analyse erfordert das Verständnis der raumzeitlichen Mittelwerte von Transformationen der SPDE. Durch die Nutzung der Clark-Ocone-Formel und der Poincaré-Ungleichung werden neue Konzentrationsresultate, bis hin zu subgaußscher Konzentration, für semi-lineare SPDEs ermöglicht.This thesis contributes to the field of statistical inference for stochastic semi-linear partial differential equations (SPDEs). The diffusivity and the noise level are known constants, whereas the reaction function is unknown and shall be estimated based on one observation of the SPDE. Since one observation of the SPDE does generally not carry sufficient information to identify the reaction function, it is analysed how to increase the information about the reaction function in the spatial sections of the SPDE. Both parametric and non-parametric estimation methods for the reaction function are derived and analysed. The statistical analysis requires the control of spatio-temporal averages of transformations of the SPDE. By using the Clark-Ocone formula und the Poincaré-inequality, novel concentration tools for semi-linear SPDEs are proven. These range from variance bounds to subgaussian concentration for the spatio-temporal and spatial averages

    Inclusive measurement of the ttbar cross-section using proton-proton collisions at √s =13.6 TeV with the ATLAS detector

    Die Messung des inklusiven ttbar-Wirkungsquerschnitts bei 13.6 TeV wird anhand von 29 fb⁻¹ an Daten des ATLAS-Detektors am LHC aus dem Jahr 2022 präsentiert. Für diese Messung sind das Zählen der Ereignisse und die genaue Bestimmung der Luminosität erforderlich, wobei die Luminositätsunsicherheit die größte Unsicherheitsquelle darstellt. Die Luminositätsbestimmung erfolgt nach dem etablierten Verfahren aus Run 2 in drei Schritten: absolute Kalibrierung mit van der Meer Scans, Transfer dieser Kalibrierung auf die physikalischen Datenaufnahmebedingungen und Stabilitätsüberprüfung. Die vorläufige Luminositätsunsicherheit für 2022 beträgt 2.2 %, hauptsächlich bedingt durch den Kalibrierungstransfer und Nicht-Faktorisierbarkeits-Effekte. Der ttbar-Wirkungsquerschnitt wird mit der b-Jet-Zählmethode im Dilepton-Endzustand gemessen und ergibt sigma(ttbar) = 850 ± 3 (stat.) ± 18 (syst.) ± 20 (lumi.) pb, was 1.5 Sigma unter der Standardmodellvorhersage liegt. Das Verhältnis der Z-Produktion zum ttbar-Wirkungsquerschnitt, sensitiv auf das Gluon-zu-Quark-PDF-Verhältnis, beträgt R(ttbar/Z) = 1.145 ± 0.003 (stat.) ± 0.021 (syst.) ± 0.002 (lumi.) und stimmt mit dem Standardmodell überein. Zusätzlich wird das Verhältnis des Wirkungsquerschnitts bei 13.6 TeV zu 13 TeV gemessen: R(13.6/13) = 1.0253 ± 0.0347, 2.4 Sigma von der Vorhersage entfernt. Diese Ergebnisse unterstützen das Standardmodell und unterstreichen die Bedeutung präziser Luminositäts- und Wirkungsquerschnittsmessungen in der Teilchenphysik.The inclusive ttbar cross-section measurement at 13.6 TeV, based on 29 fb⁻¹ of data recorded by ATLAS at the LHC in 2022, is presented. This measurement requires event counting and accurate luminosity determination, with luminosity uncertainty as the largest source of error. The luminosity calibration follows the established Run 2 procedure, involving three stages: absolute calibration via van der Meer scans, calibration transfer from specific scan conditions to physics data-taking conditions, and stability checks. The preliminary luminosity uncertainty for 2022 is 2.2%, mainly due to calibration transfer and non-factorisation effects. The ttbar cross-section is measured using b-jet counting in the dilepton final state, yielding sigma(ttbar) = 850 ± 3 (stat.) ± 18 (syst.) ± 20 (lumi.) pb, which is 1.5 sigma below the Standard Model prediction. The ratio of Z production to the ttbar cross-section, sensitive to the gluon-to-quark PDF ratio, is R(ttbar/Z) = 1.145 ± 0.003 (stat.) ± 0.021 (syst.) ± 0.002 (lumi.), consistent with the Standard Model. Additionally, the cross-section ratio between 13.6 TeV and 13 TeV is R(13.6/13) = 1.0253 ± 0.0347, 2.4 sigma from the predicted value. These results support the Standard Model and emphasize the importance of precise luminosity and cross-section measurements in particle physics

    Dual-fluorescent Molecularly Imprinted Polymers (MIPs) for (Bio)chemical Sensing

    Auf molekular geprägten Polymeren (MIP) basierende Sonden, insbesondere fluoreszierende MIPs, die eine spezifische Erkennung mit Fluoreszenzsignalen kombinieren, wurden aufgrund der vorteilhaften Spezifität und Selektivität von MIPs und der hohen Empfindlichkeit und schnellen Erfassungszeiten der Fluoreszenz für den Nachweis von Analyten aus verschiedenen chemischen Klassen entwickelt. Ziel dieser Arbeit war die Entwicklung von dual-fluoreszierenden Kern-Schale-MIPs als Erkennungselement für den empfindlichen Nachweis von Carbonsäure-haltigen Verbindungen, Chlorogensäure und Sialinsäure-Derivaten, die während des Prägeprozesses als sogenanntes Template dienen und als Zielanalyten in den analytischen Rückbindungsassays fungieren. Zunächst wurden fluoreszierende Siliziumdioxid-Nanopartikel synthetisiert, die Kohlenstoff-Nanopunkte (Carbon Nanodots) und Seltenerdmetall-Ionen enthalten und als emittierender Kern der dual-fluoreszierenden MIPs dienen. Es wurden spektroskopische Untersuchungen über die Wechselwirkung zwischen den Templaten und den fluoreszierenden Monomeren durchgeführt, die in eine dünne fluoreszierende MIP-Schale eingebaut wurden, die auf die Oberfläche der fluoreszierenden Siliziumdioxid-Nanopartikel aufgewachsen wurde und die interne Referenz der dual fluoreszierenden MIPs darstellt. Die zweifach fluoreszierenden MIPs wurden auf ihr optisches Ansprechen, ihre Empfindlichkeit und Selektivität in Küvetten-basierten, durchflusszytometrischen und fluoreszenzmikroskopischen Assays untersucht. Diese Systeme eignen sich für den empfindlichen, schnellen und zuverlässigen (bio)chemischen Nachweis einer potenziell breiten Palette von Analyten. Neben den beiden oxoanionischen Templaten wurde auch ein MIP-System für Triphenylphosphin (TPP) entwickelt, das zeigt, dass der Ansatz der Fluoreszenzmonomer-vermittelten MIP-Detektion auch auf neutrale nukleophile Template ausgeweitet werden kann.Molecularly imprinted polymer (MIP)-based probes, especially fluorescent MIPs, which combine specific recognition with fluorescence signalling, have been developed for the detection of analytes from different chemical classes, due to the favourable specificity and selectivity of MIPs and the high sensitivity and fast acquisition times of fluorescence. This thesis aimed at the development of core-shell dual-fluorescent MIPs as recognition element for the sensitive detection of carboxylate-containing compounds, chlorogenic acid and sialic acid derivatives, which serve as the so-called template during the imprinting process and are the target analytes in the analytical rebinding assays. Fluorescent silica nanoparticles were firstly synthesized, incorporating carbon nanodots and rare-earth metal ions and serving as the emitting core of dual-fluorescent MIPs. Spectroscopic studies were performed on the interaction between the templates and the fluorescent monomers, which were incorporated into a thin fluorescent MIP shell grown from the surface of the fluorescent silica nanoparticles that provide the internal reference for the resulting dual-fluorescent MIPs. The dual-fluorescent MIPs were investigated for their optical response, sensitivity, and selectivity in cuvette-based, flow cytometric and fluorescence microscopic assays. These systems are applicable for sensitive, fast, and reliable (bio)chemical analysis of a potentially wide range of analytes. Besides the oxoanionic templates, a MIP system was also developed for triphenylphosphine (TPP), revealing that the approach of fluorescent monomer-mediated MIP detection can also be expanded to neutral nucleophilic templates

Wie in den vergangenen Jahren waren Promovierende aus allen Teilbereichen des Faches eingeladen, einen Einblick in ihre derzeitigen Arbeiten zu geben und aktuelle fachliche Informationen auszutauschen und zu diskutieren.     A microcontroller-based system for flexible oxygen control in laboratory experiments

    Environmental control systems are important tools for experimental researchers studying animal–environment interactions. Commercial systems for the measurement and regulation of environmental oxygen conditions are relatively expensive and cannot always be adapted to varying experimental applications. Here, I present a low-cost and highly flexible oxygen control system using Arduino microcontrollers in combination with a commercial optical oxygen sensor. Hardware and software examples are provided for three different applications: single-setpoint, sequential and long-term dissolved oxygen (DO) control. All tested control systems created the desired DO conditions with high accuracy and repeatability across trials. The resources provided shown here can be adapted and modified to be used in a variety of experimental contexts.Peer Reviewe

    Super-resolution imaging of native fluorescent photoreceptors in chytrid fungal eyes

    The article processing charge was funded by the Open Access Publication Fund of Humboldt-Universität zu Berlin.Photoorientation in motile fungal zoospores is mediated by rhodopsin guanylyl cyclases (RGCs). In certain chytrids, these photoreceptors form heterodimers consisting of a visible-light-absorbing RGC paired with neorhodopsin (NeoR), a rhodopsin distinguished by its unique spectral properties: far-red absorption and high fluorescence. Leveraging the native fluorescence of NeoR, we detected RGCs in living zoospores of the fungus Rhizoclosmatium globosum . The reversible photoswitching of bistable NeoR enabled super-resolution microscopy, facilitating single-molecule detection and quantification of NeoR proteins within individual zoospores. This approach also revealed the precise localization of RGCs within the rumposome, a chytrid-specific organelle hypothesized to mediate photoreception. Fluorescence tracking across different stages of the chytrid life cycle and the analysis of transcriptomic data confirmed that RGCs are predominantly present during the zoospore stage. Functional assays of recombinantly expressed RGC heterodimers with modified substrate specificity revealed that only one of the two pseudo-symmetric nucleotide-binding sites is catalytically active. Strikingly, disrupting nucleotide binding in the non-catalytic site enhanced light-triggered cyclase activity by up to ninefold, indicating an allosteric regulatory mechanism in heterodimeric RGCs.Fungal zoospores have heterodimeric rhodopsin-guanylyl cyclase complexes for light perception, including highly fluorescent neorhodopsin (NeoR). Native NeoR can be visualized in living cells by means of single-molecule localization microscopy. Bistable neorhodopsin (NeoR) is a mandatory subunit in fungal heterodimeric rhodopsin-guanylyl cylases (RGCs). Intrinsic fluorescence of NeoR enables single-molecule localization microscopy in living zoospores. RGC photoreceptors are hosted within the rumposome, a chytrid specific organelle.     The article processing charge was funded by the Open Access Publication Fund of Humboldt-Universität zu Berlin.This article examines the facilitation of internal mobility as a pillar of regional integration, focusing on the Southern Common Market (Mercosur). Drawing on New and Comparative Regionalism theories, it investigates how bottom-up dynamics at the subnational level interact with supranational objectives of regional free movement in Mercosur. Through document analysis and 26 stakeholder interviews with national and regional institutions, as well as civil society organisations, it reveals challenges in the state-led implementation of the Mercosur free movement framework, including the absence of a right of entry in the agreements, the ineffective implementation of certain agreements, disagreements between member states impeding efforts to strengthen the existing framework and, lastly, discriminatory practices and arbitrary rejections at internal borders. Nevertheless, some of these limitations are addressed and partially mitigated by bottom-up migrant practices and traditional mobility in border regions. Cross-border interactions and mobility in subnational border regions thus represent key dimensions of regional free movement and integration. Thereby, the article underscores the importance of a universal definition of regional free movement that integrates historical particularities and mobility dynamics beyond the European conceptualisation.Peer Reviewe

    Social-ecological justice in cities: a spatial vulnerability approach

    Ecological and social injustices are deeply intertwined, yet their integration into strategic spatial planning is widely lacking. This paper presents a spatial, participatory, multi-criteria approach to assess social-ecological vulnerabilities in the Metropolitan Area of Krakow (MK), Poland. Our approach incorporates co-created insights into urban vulnerabilities, considering social and ecological sensitivities alongside exposure to social and environmental hazards. In collaboration with local planning bodies, the co-creation process identified ten critical vulnerabilities, including to river flooding, to noise pollution, and to drought. A comprehensive data analysis with 47 indicators mapped social and ecological vulnerabilities spatially.
