Copula models for complex data: applications to dependent censoring and longitudinal data

    No full text
Throughout the many domains in statistics, traditional models often assume independence of the data. This typically comes with great theoretical simplification, but numerous real-life situations also violate this assumption. In such contexts, copula modelling is a particularly useful tool to enhance model validity. Specifically, copula functions link marginal quantities and capture any interdependence present, while preserving model interpretability due to a conceptual separation of this dependence from the rest of the model. Copula models for two dependence-invoking contexts are considered in this dissertation. We first work in the domain of survival analysis, that is inherently plagued by the phenomenon of censoring: whereas primary interest is in the time T until a specific event occurs, observation of the latter is sometimes precluded by an earlier competing event. Nonetheless, the censoring time C until this alternative event contains relevant information on T. Under independence of T and C, it only contributes to the available information about T by ensuring that T > C for that data subject. Frequently, however, T and C are connected through positive or negative dependence, such that knowing C increases the likeliness of T being in the near or further future, respectively, as compared to the case in which C would not have been observed yet. Such dependent censoring is often handled using copula models. Yet, as T and C are never simultaneously observed for one subject, it is theoretically impossible to distinguish independent from dependent censoring without imposing formal restrictions; this is referred to as nonidentifiability. Literature has therefore, initially, mostly focused on flexible (i.e. nonparametric) marginal modelling, while making the overly strict assumption of a completely known dependence structure. Over the past years, research has shown the possibility of less restrictive copula assumptions – viz. parametric rather than fully known copula functions – at the cost of likewise parametric margins. The first part of this thesis is situated in this area and further explores the boundary between flexibility and nonidentifiabilit

Driving Factors of Household Car Ownership: Development of a National Car Ownership Model for Belgium

    No full text
This study develops a national car ownership model for Belgium, addressing the significant factors influencing household vehicle ownership across various built densities. Using the multinomial logistic regression model and the MONITOR travel survey data from 2016 to 2017, socioeconomic and demographic factors, residential characteristics, mobility resources, and public transport accessibility were examined. Higher income and larger household size strongly correlate with increased car ownership, particularly in Flanders. In contrast, subscriptions to shared mobility services and proximity to train stations reduce the likelihood of owning multiple cars. The results emphasize the importance of regional policies and sustainable transportation options in managing vehicle demand. This study provides valuable insights for policymakers and urban planners to promote ecological mobility and reduce car dependency, offering a basis for research in other high-density, car-centric regions

The influence of the solvent and intermolecular interactions on the physical structure of spray dried amorphous solid dispersions

It was the overall aim of this thesis to improve our comprehension of the influence of two key factors on the physical structure, phase behavior and other properties of spray dried ASDs. The effect of the organic solvent, used during spray drying, and the availability of different API and polymer H-bond donor and acceptor groups was assessed. To this end, a series of ASDs were spray dried, from either different organic solvents, or containing APIs and polymers with varying H-bonding properties. In the introduction, the use and benefits of amorphous solid dispersions were first placed against the background of the current pharmaceutical landscape. The perks, but also the challenges, of oral drug delivery were explained, especially regarding the effect of drug solubility and permeability on the oral bioavailability of the drugs. Lipinski’s rule of five and the different classification systems (like the BCS) using API solubility and permeability were introduced. Considering the increasing amount of poorly soluble compounds in the pipelines of pharmaceutical companies, the need for solubility enhancing strategies was explained. In this regard, the steps in the dissolution process were described and it was highlighted which aspects of this process were impacted by different enabling strategies. The solubility advantage of the amorphous form of an API over the crystalline state was highlighted, and amorphous solid dispersions were introduced as an approach to overcome the thermodynamic instability of pure amorphous drugs. The role of the polymer and its influence on the physical stability and drug release behavior was briefly addressed, and the importance of drug-polymer intermolecular interactions was explained. A theoretical overview of intermolecular interactions was given, followed by further elaborating on the effect of the interactions on the physical stability and dissolution characteristics. As an introduction to spray drying, an overview of the different ASD manufacturing techniques was given, leading up to an explanation of the spray drying process and the different factors that influence it. The introduction was concluded with an overview of the solid-state characterization techniques that were applied in this work, together with the methods used to characterize the stability and pharmaceutical performance of the ASDs
