    Strong co-nondeterministic lower bounds for NP cannot be proved feasibly

    We show unconditionally that Cook’s theory PV formalizing poly-time reasoning cannot prove, for any non-deterministic poly-time machine M defining a language L(M), that L(M) is inapproximable by co-nondeterministic circuits of sub-exponential size. In fact, our unprovability result holds also for a theory which supports a fragment of Jeřábek’s theory of approximate counting APC1. We also show similar unconditional unprovability results for the conjecture of Rudich about the existence of super-bits

    This short article responds to, and interprets, two epistemic claims made by Mark R. Wynn concerning truth and Christian ethics. The first claim concerns how the body knows something prior to an operation of reason. The second claim concerns the relationship between narrative and metaphysics, particularly when considering the eucharist.     Cities currently harbour more than half of the world’s human population and continued urban expansion replaces natural landscapes and increases habitat fragmentation. The impacts of urbanisation on biodiversity have been extensively studied in some parts of the world, but there is limited information from South Asia, despite the rapid expansion of cities in the region. Here, we present the results of monthly surveys of butterflies in three urban parks in Dhaka city, Bangladesh, over a three-year period (Jan 2014- Dec 2016). We recorded 45% (137 of the 305 species) of the country’s butterfly richness, and 40% of the species detected are listed as nationally threatened. However, butterfly species richness declined rapidly in the three study areas over the three-year period, and the decline appeared to be more severe among threatened species. We developed linear mixed effect models to assess the relationship between climatic variables and butterfly species richness. Overall, species richness was positively associated with maximum temperature and negatively with mean relative humidity and saturation deficit. Our results demonstrate the importance of urban green spaces for nationally threatened butterflies. With rapidly declining urban green spaces in Dhaka and other South Asian cities, we are likely to lose refuges for threatened fauna. There is an urgent need to understand urban biodiversity dynamics in the region, and for proactive management of urban green spaces to protect butterflies in South Asia

    5-HT2C receptor perturbation has bidirectional influence over instrumental vigour and restraint

    The serotonin (5-HT) system, particularly the 5-HT2C receptor, has consistently been implicated in behavioural control. However, while some studies have focused on the role 5-HT2C receptors play in regulating motivation to work for reward, others have highlighted its importance in response restraint. To date, it is unclear how 5-HT transmission at this receptor regulates the balance of response invigoration and restraint in anticipation of future reward. In addition, it remains to be established how 5-HT2C receptors gate the influence of internal versus cue-driven processes over reward-guided actions. To elucidate these issues, we investigated the effects of administering the 5-HT2C receptor antagonist SB242084, both systemically and directly into the nucleus accumbens core (NAcC), in rats performing a Go/No-Go task for small or large rewards. The results were compared to the administration of d-amphetamine into the NAcC, which has previously been shown to promote behavioural activation. Systemic perturbation of 5-HT2C receptors—but crucially not intra-NAcC infusions—consistently boosted rats’ performance and instrumental vigour on Go trials when they were required to act. Concomitantly, systemic administration also reduced their ability to withhold responding for rewards on No-Go trials, particularly late in the holding period. Notably, these effects were often apparent only when the reward on offer was small. By contrast, inducing a hyperdopaminergic state in the NAcC with d-amphetamine strongly impaired response restraint on No-Go trials both early and late in the holding period, as well as speeding action initiation. Together, these findings suggest that 5-HT2C receptor transmission, outside the NAcC, shapes the vigour of ongoing goal-directed action as well as the likelihood of responding as a function of expected reward

    Diagnostic performance of anti-Zika virus IgM, IgAM and IgG ELISAs during co-circulation of Zika, dengue, and chikungunya viruses in Brazil and Venezuela

    Background Serological diagnosis of Zika virus (ZIKV) infection is challenging because of the antibody cross-reactivity among flaviviruses. At the same time, the role of Nucleic Acid Testing (NAT) is limited by the low proportion of symptomatic infections and the low average viral load. Here, we compared the diagnostic performance of commercially available IgM, IgAM, and IgG ELISAs in sequential samples during the ZIKV and chikungunya (CHIKV) epidemics and co-circulation of dengue virus (DENV) in Brazil and Venezuela. Methodology/Principal findings Acute (day of illness 1–5) and follow-up (day of illness ≥ 6) blood samples were collected from nine hundred and seven symptomatic patients enrolled in a prospective multicenter study between June 2012 and August 2016. Acute samples were tested by RT-PCR for ZIKV, DENV, and CHIKV. Acute and follow-up samples were tested for IgM, IgAM, and IgG antibodies to ZIKV using commercially available ELISAs. Among follow-up samples with a RT-PCR confirmed ZIKV infection, anti-ZIKV IgAM sensitivity was 93.5% (43/46), while IgM and IgG exhibited sensitivities of 30.3% (10/33) and 72% (18/25), respectively. An additional 24% (26/109) of ZIKV infections were detected via IgAM seroconversion in ZIKV/DENV/CHIKV RT-PCR negative patients. The specificity of anti-ZIKV IgM was estimated at 93% and that of IgAM at 85%. Conclusions/Significance Our findings exemplify the challenges of the assessment of test performance for ZIKV serological tests in the real-world setting, during co-circulation of DENV, ZIKV, and CHIKV. However, we can also demonstrate that the IgAM immunoassay exhibits superior sensitivity to detect ZIKV RT-PCR confirmed infections compared to IgG and IgM immunoassays. The IgAM assay also proves to be promising for detection of anti-ZIKV seroconversions in sequential samples, both in ZIKV PCR-positive as well as PCR-negative patients, making this a candidate assay for serological monitoring of pregnant women in future ZIKV outbreaks

    The implications of interrelated assumptions on estimates of divergence times and rates of diversification

    Phylogenies are increasingly being used as a basis to provide insight into macroevolutionary history. Here, we use simulation experiments and empirical analyses to evaluate methods that use phylogenies as a basis to make estimates of divergence times and rates of diversification. This is the first study to present a comprehensive assessment of the key variables that underpin analyses in this field—including substitution rates, speciation rates, and extinction, plus character sampling and taxon sampling. We show that in unrealistically simplistic cases (where substitution rates and speciation rates are constant, and where there is no extinction), increased character and taxon sampling lead to more accurate and precise parameter estimates. By contrast, in more complex but realistic cases (where substitution rates, speciation rates, and extinction rates vary), gains in accuracy and precision from increased character and taxon sampling are far more limited. The lack of accuracy and precision even occurs when using methods that are designed to account for more complex cases, such as relaxed clocks, fossil calibrations, and models that allow speciation rates and extinction rates to vary. The problem also persists when analyzing genomic scale data sets. These results suggest two interrelated problems that occur when the processes that generated the data are more complex. First, methodological assumptions are more likely to be violated.     Are more GPs associated with a reduction in emergency hospital admissions? A quantitative study on GP referral in England

    Background Recent studies have found an association between access to primary care and accident and emergency attendances, with better access associated with fewer attendances. Analyses of an association with emergency admissions, however, have produced conflicting findings. Aim This study investigated whether emergency admission rates in an area are associated with 1) the number of GPs, and 2) mean size of GP practice. Design and setting Analysis was conducted utilising Hospital Episode Statistics, the numbers of GPs and GP practices, Office for National Statistics population data, Quality and Outcomes Framework prevalence data, and Index of Multiple Deprivation data, from 2004/2005 to 2011/2012, for all practices in England. Method Regression analysis of panel data with fixed effects to address 1) a potential two-way relationship between the numbers of GPs and emergency admissions, and 2) unobservable characteristics of GP practices. Results There is not a statistically significant relationship between the number of GPs in a primary care trust area and the number of emergency admissions, when analysing all areas. In deprived areas, however, a higher number of GPs is associated with lower emergency admissions. There is also a lower emergency admission rate in areas in which practices are on average larger, holding GP supply constant. Conclusion An increase in GPs was found to reduce emergency admissions in deprived areas, but not elsewhere. Areas in which GPs are concentrated into larger practices showed reduced levels of emergency admissions, all else being equal

    Structures of the Plasmodium falciparum heat-shock protein 70-x ATPase domain in complex with chemical fragments identify conserved and unique binding sites

    Plasmodium falciparum invades erythrocytes and extensively modifies them in a manner that increases the virulence of this malaria parasite. A single heat-shock 70 kDa-type chaperone, PfHsp70-x, is among the parasite proteins exported to the host cell. PfHsp70-x assists in the formation of a key protein complex that underpins parasite virulence and supports parasite growth during febrile episodes. Previous work resolved the crystallographic structures of the PfHsp70-x ATPase and substrate-binding domains, and showed them to be highly similar to those of their human counterparts. Here, 233 chemical fragments were screened for binding to the PfHsp70-x ATPase domain, resulting in three crystallographic structures of this domain in complex with ligands. Two binding sites were identified, with most ligands binding proximal to the ATPase nucleotide-binding pocket. Although amino acids participating in direct ligand interactions are conserved between the parasite and human erythrocytic chaperones, one nonconserved residue is also present near the ligand. This work suggests that PfHsp70-x features binding sites that may be exploitable by small-molecule ligands towards the specific inhibition of the parasite chaperone

    Drivers behind the summer 2010 wave train leading to Russian heat wave and Pakistan flooding

    Summer 2010 saw two simultaneous extremes linked by an atmospheric wave train: a record-breaking heatwave in Russia and severe floods in Pakistan. Here, we study this wave event using a large ensemble climate model experiment. First, we show that the circulation in 2010 reflected a recurrent wave train connecting the heatwave and flooding events. Second, we show that the occurrence of the wave train is favored by three drivers: (1) 2010 sea surface temperature anomalies increase the probability of this wave train by a factor 2-to-4 relative to the model’s climatology, (2) early-summer soil moisture deficit in Russia not only increases the probability of local heatwaves, but also enhances rainfall extremes over Pakistan by forcing an atmospheric wave response, and (3) high-latitude land warming favors wave-train occurrence and therefore rainfall and heat extremes. These findings highlight the complexity and synergistic interactions between different drivers, reconciling some seemingly contradictory results from previous studies
