    Macrocycle-assembled membranes for high-salinity organic wastewater treatment

    Abstract Membrane processes offer a promising pathway for selectively separating organics and salts to enable water reuse and resource recovery. While polymeric membranes incorporating macrocyclic molecules that feature amphiphilic nature and tunable cavities are well suited for this purpose, traditional macrocycles with limited reactive sites and uncontrolled diffusion are challenging to be assembled into highly interconnected membranes. Here, we introduce tetra-aldehyde appended calixarene (TACA), a macrocyclic monomer featuring three-dimensional cavity and moderate reactivity, for creating loose-structured nanofilms via unidirectional diffusion assisted interfacial polymerization (UDIP). Precise positioning of the lipophilic TACA at the organic phase boundary allows it to polymerize with aqueous-phase diamines on the hydrogel surface, facilitating an undisturbed environment for controlled polymerization. The resultant thin macrocycle-assembled membranes featuring intrinsic water-facilitated through-cavity exhibited high water permeability of 63.8 L m-2 h-1 bar-1, and exceptional dye/salt selectivity and structural robustness, as evidenced by efficient diafiltration of binary dye/salt mixtures and superior operational stability.     Intranasal administration of broad-spectrum macrocyclic peptide inhibitor protects against SARS-CoV-2 Omicron variants

    Abstract Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to cause significant morbidity and mortality despite the end of its pandemic phase. The emergence of highly mutated SARS-CoV-2 variants of concern highlights the requirement of broad-spectrum antiviral countermeasures which possess both prophylactic and therapeutic efficacies. Here, we obtain a macrocyclic peptide, 6L3-3P11K, that effectively inhibits a wide range of SARS-CoV-2 variants and subvariants. Structural studies show that 6L3-3P11K forms homotrimers that lock the spike protein (S) trimer into a “closed” conformation by engaging a conserved non-receptor binding motif (non-RBM) of S. This interaction disrupts the binding between S and ACE2 receptor. Structure-guided modifications result in a thermostable and trypsin-resistant macrocyclic peptide, 6L3-1F3P11hR, that exhibits prophylactic and therapeutic effects against SARS-CoV-2 infection in a male hACE2 transgenic mouse model after intranasal administration. Our results provide a drug candidate for the control and prevention of COVID-19 and may stimulate further research on macrocyclic broad-spectrum anti-coronavirus drug development

    ClpC1-targeting peptide natural products differentially dysregulate the proteome of Mycobacterium tuberculosis

    Abstract Targeting the protein quality control system in Mycobacterium tuberculosis represents a promising and underexplored opportunity for antibiotic development. The ClpC1:ClpP1P2 protease is an essential component of the system that mediates both regulatory and stress-related protein degradation. Several non-ribosomal peptide natural products, including ecumicin, ilamycins (rufomycins) and cyclomarins, have been discovered that bind to the ClpC1 chaperone of the complex and exhibit potent antimycobacterial activity, leading to significant interest in the ClpC1:ClpP1P2 system as a bona fide target for the new tuberculosis drugs. In this study, we combine quantitative proteomics, bioinformatics, transcriptomics, CRISPRi knockdown, and targeted biochemical and biophysical assays to dissect the mechanisms of ecumicin, ilamycin and cyclomarin in clinically relevant Mycobacterium tuberculosis. Strikingly, despite exhibiting similar binding modes to ClpC1, each compound induces distinct effects on protein degradation. Notably, ilamycin and ecumicin do not trigger the ClpC2 rescue mechanism that mitigates cyclomarin-induced mycobacterial toxicity. In addition, we identify a novel interaction between ecumicin and stress-response chaperone Hsp20. The differential disruption of ClpC1 substrates, stress-response chaperones, and distinct reshaping of the Mycobacterium tuberculosis proteome by the three natural products, unveils new opportunities for the development of protein quality control-targeted antimycobacterials

    A stochastic mechanism drives fast substrate translocation in the AAA+ machine ClpB

    Abstract How biological machines harness ATP to drive mechanical work remains a crucial question. Structural studies of protein-translocating AAA+ machines proposed a coupled and sequential translocation process, whereby ATP hydrolysis events lead to short threading steps. Yet, direct real-time observation of these events remains elusive. Here, we employ single-molecule FRET spectroscopy to track substrate translocation through ClpB, a quality control AAA+ machine. We isolate ClpB and its substrate within lipid vesicles and find that translocation events, while dependent on ATP, take milliseconds, much faster than ATP hydrolysis times. Surprisingly, the translocation rate depends weakly on temperature and ATP concentration. Using three-color FRET experiments, we find that translocation events can occur bidirectionally but are not always complete. Replacing ATP with the slowly hydrolysable analog ATPγS abolishes both rapid translocation and directionality. These results indicate a fast, stochastic Brownian-motor-like mechanism, redefining how ATP is coupled with mechanical action in AAA+ machines

    Selective electrosynthesis of urea from nitrate and carbon dioxide with low overpotential

    Abstract Reducing the energy cost and CO2 emission of traditional urea synthesis through alternative strategy is crucial for the sustainable social development. Herein, we report the electrosynthesis of urea from nitrate and CO2 on Ag/CdO heterostructured catalyst with low overpotential. The optimized Ag0.07/CdO catalyst demonstrates a high urea yield rate of 111.6 ± 3.1 mmol gcat. –1 h–1 with 50.0 ± 2.7% Faradaic efficiency at −0.15 V vs. RHE in a flow cell, and can further achieve a urea yield rate of 427.0 ± 30.2 mmol gcat. –1 h–1 in an optimized two-electrode cell with stable response for 1000 h. Operando spectroscopy and theoretical calculations indicate that Ag attracts *NO2 intermediate as well as promotes the reconstruction of CdO into low coordinated CdCO3 that facilitate the activation of CO2, thus benefits *CO/*NO2 adsorption and coupling at low overpotential as well as suppresses the generation of CO and NH3 by-products

    Impact of a single fecal microbiome transplantation in adult women with anorexia nervosa: an open-label feasibility pilot trial

    Abstract Anorexia nervosa (AN) is a severe mental disorder characterized by restrictive eating and disturbance in the way one’s body weight or shape is experienced, often accompanied by depression and anxiety. Current evidence-based treatments for AN have limited efficacy, with less than half of the patients achieving full recovery in long-term follow-up studies. Recent findings have identified gut microbiota (GM) dysbiosis as a potential contributor to AN pathology through the gut-brain axis. This open-label, non-randomized, feasibility trial (Clinicaltrials.gov Identifier: NCT05834010) evaluated the feasibility of utilizing fecal microbiota transplantation (FMT) to modify the GM and GM-associated signaling in females with AN and to examine biological effects following a single FMT procedure. Adult female participants diagnosed with AN were recruited. FMT was administered either orally via capsules or as rectal enema. Stool and blood samples were collected pre- and one week post-FMT to assess GM composition, hormonal changes, and biomarkers. Primary endpoints: Feasibility of FMT in individuals with AN and preferred route of FMT. Secondary endpoints: A single FMT treatment can alter GM composition in individuals with AN short term and relevant gut brain signaling in serum. 18/22 participants (81%) completed FMT and sampling and 19/22 participants chose oral capsules, with no serious adverse effects reported. GM analysis showed significant shifts toward donor composition 1-week post-FMT, with improved stool consistency. No significant changes were observed in psychopathology measures or appetite-related biomarkers. Oral FMT is a feasible intervention for adult women with AN, leading to changes in GM profile. Future studies should focus on placebo-controlled trials to assess the efficacy of repeated oral treatments and explore long-term effects on GM, appetite, body weight, sex hormones, disorder-specific symptoms, and overall well-bein

    A neutral cyclic aluminium (I) trimer

    Abstract As part of the quest to develop metal-based redox chemistry beyond the d-block, low oxidation state aluminium complexes have gained wide recognition as discrete and versatile 2-electron reductants. Despite reports of monomeric, dimeric and tetrameric neutral structures, as well as a range of charged aluminyl compounds, neutral trimeric structures have remained notably absent. Furthermore, trimeric nuclearity has previously not been considered when investigating reaction mechanisms. Here, we report two neutral AlI trimers, cyclotrialumanes. The molecules are extensively characterised using both experimental and computational techniques, with the Al–Al bonds described as principally covalent in nature and the trimeric structure shown to be retained in solution. The cyclotrialumanes are highly reactive, activating a range of small molecules and unsaturated substrates (e.g. H2, alkyne, benzene). Most significantly, through a series of extraordinary reactions with ethylene, the cyclotrialumanes are shown to react directly as trimers, forming 5- and 7-membered Al–C ring systems

    تبیین ابعاد و مؤلفه‌های هم‌افزایی فرهنگی در کلان‌شهرهای چندقومیتی ایران: مطالعه‌ای داده‌بنیاد در بندرعباس

    این پژوهش با هدف تبیین ابعاد و سازوکارهای هم‌افزایی فرهنگی در یک کلان‌شهر چندقومیتی ایرانی انجام شد؛ شهری مثل بندرعباس که از یک‌سو به‌دلیل موقعیت بندری، مهاجرت و تنوع قومی ناگزیر از زیستِ کنارِ هم است و از سوی دیگر، برای عبور از تنش‌های پنهان و شکاف‌های اجتماعی به یک مدل بومی برای هم‌افزایی نیاز دارد. پژوهش از نوع کیفی و با راهبرد نظریه‌ی داده‌بنیاد (نسخه‌ی نظام‌مند) انجام شد. داده‌ها از طریق مصاحبه‌های نیمه‌ساختاریافته با ۱۸ نفر از نخبگان فرهنگی، جامعه‌شناسان شهری، فعالان سازمان‌های مردم‌نهاد و شهروندان آگاه از بافت فرهنگی بندرعباس گردآوری و در سه گام کدگذاری باز، محوری و انتخابی تحلیل شد. تحلیل‌ها به استخراج یک مدل سه‌بُعدی انجامید که شامل: بُعد زمینه‌ای (شرایط تاریخی و اجتماعیِ امکان هم‌افزایی مانند تنوع قومی، تجربه‌ی زیسته‌ی هم‌زیستی و حضور نهادهای فرهنگ‌ساز)، بُعد تعاملی (سازوکارهای واقعیِ تماس و گفت‌وگو مانند فضاهای عمومیِ مشترک، میانجی‌گری نهادی، آموزش میان‌فرهنگی و رویدادهای نمادین مشترک) و بُعد پیامدی (ترجمه‌ی تعاملات به توسعه‌ی فرهنگی و اجتماعی، افزایش اعتماد میان‌گروهی و مشارکت شهری) است. یافته‌ها نشان دادند که در بندرعباس، هم‌افزایی فرهنگی نه با توصیه‌های کلان، بلکه با طراحیِ موقعیت‌های تعاملِ عادلانه و حمایت نهادی از صداهای ضعیف‌تر محقق می‌شود و بُعد تعاملی نقش حلقه‌ی واسط میان ظرفیت‌های فرهنگی و توسعه‌ی اجتماعی را ایفا می‌کند. این مدل می‌تواند مبنایی برای سیاست‌گذاری فرهنگی و مدیریت شهری در شهرهای جنوبی و بندری ایران قرار گیرد

    ارزیابی دسترسی افراد معلول به خدمات آتش‌نشانی در سطح شهر اردبیل

    عدالت فضایی در دسترسی گروه‌های آسیب‌پذیر به خدمات اضطراری، از جمله خدمات آتش‌نشانی، یکی از ارکان اساسی ایمنی و شهروندی فراگیر در شهرهای معاصر محسوب می‌شود. این پژوهش با رویکرد توصیفی- تحلیلی و با هدف ارزیابی دسترسی فضایی افراد دارای معلولیت به ایستگاه‌های آتش‌نشانی در شهر اردبیل انجام شده است. داده‌های پژوهش شامل موقعیت ۶۵۸۶ فرد معلول ثبت‌شده در اداره بهزیستی استان، هفت ایستگاه آتش‌نشانی فعال و شبکه معابر شهری است که با استفاده از روش‌های تحلیل شبکه، محدوده خدماتی (Service Area) و مدل تخصیص مکان (Location-Allocation) در محیط GIS تحلیل شدند. یافته‌ها نشان می‌دهد که توزیع جمعیت معلول در سطح شهر به‌صورت خوشه‌ای و نامتوازن است؛ به‌طوری که منطقه ۴ بیشترین (۱۹۶۷ نفر) و منطقه ۳ کمترین (۶۱۷ نفر) تعداد را داراست. همچنین، هیچ ایستگاه آتش‌نشانی در منطقه ۵ یکی از مناطق حاشیه‌ای و آسیب‌پذیرمستقر نیست. با وجود این، بیش از ۹۹٫۸ درصد افراد معلول در شعاع پاسخ‌دهی ۷ دقیقه‌ای قرار دارند. با این‌حال، تحلیل‌های فضایی هشدار می‌دهند که دسترسی ظاهری لزوماً معادل دسترسی واقعی نیست، زیرا موانع کالبدی، ارتباطی و نهادی می‌توانند مؤثر باشند. پیشنهادهایی با رویکرد تحقق اهداف توسعه پایدار (SDG) و اصول کنوانسیون حقوق افراد دارای معلولیت سازمان ملل متحد برای مکان‌یابی بهینه ایستگاه جدید به‌ویژه در منطقه ۵، بازنگری در معیارهای توزیع خدمات و تلفیق متغیرهای جمعیتی آسیب‌پذیر در برنامه‌ریزی بحران می‌تواند دسترسی بهینه به خدمات آتش‌نشانی را برای کلیه افراد به‌ویژه معلولین تسهیل نماید

    عشایر دشت مغان از دیرباز در این منطقه به صورت چادر نشینی زندگی کرده و شغل اصلی آنها دامداری بوده است از این رو این قوم به یکجا نشینی عادت نداشته و برای تأمین علوفه احشام خود دایما کوچ می کردند و اسکان مشخصی نداشتند. در اواخر دوره قاجار که برخی شهرهای ایران در اختیار روسیه قرار گرفت، بخشی از دشت مغان نیز به روسیه ملحق شد و عشایر شاهسون از رفتن به آن سوی مرز منع شدند؛ اما آنها دایما به سرزمین‌های تازه اشغال شده تاخت و تاز کرده و گاهی دست به غارت می زدند از این رو دولتین ایران و روس سیاست‌هایی اتخاذ کردند که دست این افراد را کوتاه کنند. در این مقاله با هدف شناخت تاثیر سیاست گذاری های داخلی و خارجی دولت ایران بر زندگی عشایر شاهسون به بررسی اوضاع دشت مغان و ایلات شاهسون پرداخته و به این نتیجه رسدیم که در اثر تصمیمات دولت ایران در اواخر قاجار و دوران پهلوی مبنی بر خلع سلاح عشای، ملی شدن مرتع ها، اصلاحات ارضی، اسکان عشایر، به کار گیری عشایر در بخش کشاورزی و صنعت، زندگی ایلات شاهسون دچار تغییرات بسیاری شده و مجبور به اسکان و تغییر شغل دامداری به کشاورزی شدند و به دلیل کمبود زمین کشاورزی و نتایجی که از اصلاحات ارضی به بار آمد برخی مجبور به مهاجرت به شهر ها شدند
