    Time discrimination and change detection could share a common brain network: findings of a task-based fMRI study

    IntroductionOver the past few years, several studies have described the brain activation pattern related to both time discrimination (TD) and change detection processes. We hypothesize that both processes share a common brain network which may play a significant role in more complex cognitive processes. The main goal of this proof-of-concept study is to describe the pattern of brain activity involved in TD and oddball detection (OD) paradigms, and in processes requiring higher cognitive effort. MethodsWe designed an experimental task, including an auditory test tool to assess TD and OD paradigms, which was conducted under functional magnetic resonance imaging (fMRI) in 14 healthy participants. We added a cognitive control component into both paradigms in our test tool. We used the general linear model (GLM) to analyze the individual fMRI data images and the random effects model for group inference. ResultsWe defined the areas of brain activation related to TD and OD paradigms. We performed a conjunction analysis of contrast TD (task > control) and OD (task > control) patterns, finding both similarities and significant differences between them. DiscussionWe conclude that change detection and other cognitive processes requiring an increase in cognitive effort require participation of overlapping functional and neuroanatomical components, suggesting the presence of a common time and change detection network. This is of particular relevance for future research on normal cognitive functioning in the healthy population, as well as for the study of cognitive impairment and clinical manifestations associated with various neuropsychiatric conditions such as schizophrenia

    Transthyretin (TTR) amyloid cardiomyopathy (ATTR-CM) is a life-threatening disease caused by the abnormal production of misfolded TTR protein by liver cells, which is then released systemically. Its amyloid deposition in the heart is linked to cardiac toxicity and progression toward heart failure. A human induced pluripotent stem cell (iPSC) line was generated from peripheral blood mononuclear cells (PBMCs) from a patient suffering familial transthyretin amyloid cardiomyopathy carrying a c.128G>A (p.Ser43Asn) mutation in the TTR gene. This iPSC line offers a useful resource to study the disease pathophysiology and a cell-based model for therapeutic discovery

    Modern readings of the Council of Nicaea (325) show how the homousios should be considered an undue encroachment by Hellenic philosophy into the space of the purity of the biblical datum. In today’s context, however, we can read such a judgement as anachronistic, as the opposition between exegesis and metaphysics is historically unfounded. Plato’s own thought was born as an exegesis of myths, so the council fathers found themselves having to resolve a tension inherent in Origen theology, linked precisely to the relationship between his exegesis and the metaphysical thought inherited from the Greek context. The imperial choice of favouring a mere orthopraxis proved to be unsuccessful and therefore, in the course of the 4th century, the Cappadocian Fathers, in particular Gregory of Nyssa, had to reconcile metaphysics and exegesis with a new orthodox thought, in the light of which the krisis (judgement) operated by the Council of Nicaea acquires a new and potentially very useful value in the post-modern context.      Las lecturas modernas del Concilio de Nicea (325) muestran cómo el homousious debe ser considerado una invasión indebida de la filosofía helénica en el espacio de la pureza del dato bíblico. En el contexto actual, sin embargo, podemos leer tal juicio como anacrónico, ya que la oposición entre exégesis y metafísica carece de fundamento histórico. El propio pensamiento de Platón nació como exégesis de los mitos, por lo que los padres conciliares se vieron en la necesidad de resolver una tensión inherente a la teología de Orígenes, vinculada precisamente a la relación entre su exégesis y el pensamiento metafísico heredado del contexto griego. La opción imperial de favorecer una mera ortopraxis resultó infructuosa y así, en el transcurso del siglo iv, los Padres Capadocios, en particular Gregorio de Nisa, tuvieron que conciliar la metafísica y la exégesis con un nuevo pensamiento ortodoxo, a la luz del cual la krisis (juicio) operada por el Concilio de Nicea adquiere un valor nuevo y potencialmente muy útil en el contexto posmoderno

    The risk of incident depression when assessed with the Lifestyle and Well-Being Index

    Objectives: Novel findings indicate links between unhealthy lifestyles and depression based on active inflammatory processes. Thus, identifying participants with poor habits could reveal differences in trends of incident depression. This study aimed to examine the association between an objective lifestyle assessment, as measured by the Lifestyle and Well-Being Index (LWB-I), and incident depression in healthy participants of a Spanish cohort. Study design: This was a longitudinal analysis of a subsample of 10,063 participants from the Seguimiento Universidad de Navarra cohort study. Methods: Group comparisons and Cox proportional hazard models were conducted using the LWB-I, which categorizes the sample into groups with healthy and unhealthy lifestyles and well-being. The main outcome was incident depression as well as secondary outcomes. Results: Those classified to the transition category of LWB-I were associated with a hazard ratio of 0.67 (95% confidence interval: 0.52e0.87), and those in the excellent category showed a hazard ratio of 0.44 (95% confidence interval: 0.33e0.58), which in both groups reflects a significantly lower risk of incident depression compared with the group including those classified in the poor LWB-I level. Moreover, the available sensitivity analyses concerning time of depression diagnosis or antidepressant treatment further supported the role of nutrition and physical activity on incident depression. Interestingly, throughout the follow-up, incident depression was inversely related to healthier daily habits as measured by the LWB-I.     Speaking without Hurting: Assertiveness and Psychological Safety in Receiving Criticism

    Receiving criticism for improvement can be difficult, especially when that feedback comes from subordinates. One way to make receiving this information easier is to create a team environment that facilitates addressing these sensitive issues without harming personal relationships. This article analyses two factors that can contribute to channelling this type of criticism: assertiveness and psychological safety. To explore these concepts, we carried out a qualitative study based on semi-structured interviews with team managers. The practices that facilitated receiving criticism from subordinates included speaking respectfully, tolerance for errors, and focusing on the problem rather than the person. In general, receiving this feedback was appreciated and served to strengthen the bond with the other team member.Recibir críticas de mejora puede resultar difícil, sobre todo cuando esos comentarios provienen de subordinados. Un modo de facilitar que llegue esta información consiste en crear un clima de trabajo en el equipo que permita abordar estos temas delicados sin dañar la relación personal. El presente artículo analiza dos elementos que contribuyen a dar cauce a este tipo de críticas: la asertividad y la seguridad psicológica. Para ello, se ha realizado una investigación cualitativa basada en entrevistas semiestructuradas a directivos de equipos. Entre las prácticas que facilitaron recibir críticas ascendentes, los participantes destacaron el hablar con respeto, la tolerancia al error y el enfoque al problema en lugar de la persona. En general, el haber recibido este feedback era visto con agradecimiento y servía para reforzar el vínculo con la otra persona

    NatB catalytic subunit depletion disrupts DNA replication initiation leading to senescence in MEFs

    Alpha-aminoterminal acetyltransferase B (NatB) is a critical enzyme responsible for acetylating the aminoterminal end of proteins, thereby modifying approximately 21% of the proteome. This post-translational modification impacts protein folding, structure, stability, and interactions between proteins which, in turn, play a crucial role in modulating several biological functions. NatB has been widely studied for its role in cytoskeleton function and cell cycle regulation in different organisms, from yeast to human tumor cells. In this study, we aimed to understand the biological importance of this modification by inactivating the catalytic subunit of the NatB enzymatic complex, Naa20, in non-transformed mammal cells. Our findings demonstrate that depletion of NAA20 results in decreased cell cycle progression and DNA replication initiation, ultimately leading to the senescence program. Furthermore, we have identified NatB substrates that play a role in cell cycle progression, and their stability is compromised when NatB is inactivated. These results underscore the significance of N-terminal acetylation by NatB in regulating cell cycle progression and DNA replication
