    Single nucleotide variants in KIF14 gene may have prognostic value in breast cancer

    Introduction Human kinesin 14 (KIF14) is one of the 70 prognostic marker genes (so-called Amsterdam profile) previously identified by the microarray of breast carcinomas, and its high transcript expression in tumor specimens indicates a poor prognosis for patients. We performed a pilot study to explore the prognostic and predictive meaning of KIF14 germline genetic variability in breast cancer patients. Methods KIF14 coding sequence, including 5 ' and 3 ' untranslated regions and overlaps to introns for identification of splicing sites, was analyzed using next-generation sequencing in the testing set of blood DNA samples from 105 breast cancer patients with clinical follow-up. After rigorous evaluation of major allele frequency, haplotype blocks, in silico predicted functional aspects, expression quantitative trait loci, and clinical associations, eight single nucleotide variants were subsequently validated in the evaluation set of 808 patients. Results Carriers of minor alleles G (rs17448931) or T (rs3806362) had significantly shorter overall survival than wild type homozygotes (p = 0.010 and p = 0.023, respectively) thus successfully replicating the results of the testing set. Both associations remained significant in the multivariate Cox regression analysis, including molecular subtype and stage as covariates (hazard ratio, HR = 1.7, 95% confidence interval (CI) = 1.1-2.8 for rs17448931 and HR = 1.9, CI 1.2-3.0 for rs3806362). Discussion In conclusion, our preliminary data suggest that minor alleles in rs17448931 and rs3806362 of KIF14 represent candidate biomarkers of poor prognosis of breast cancer patients.     Objective method for object security evaluation

    The paper deals with designing a method for objectively determining the security level of buildings. Building security design is a crucial element that influences the final implementation. However, this process is highly subjective, as it depends on the experience of the designer, which may not agree with other experts. Therefore, the idea arose to design an objective method based on a mathematical model, the output of which will be a unique value.     Prediction of the fuel spray characteristics in the combustion chamber with methane and TiO2 nanoparticles via numerical modelling

    In this study the methane combustion was analysed with the TiO2 nanoparticles. A series of the simulation runs were performed by varying the fuel inlet velocity. However, the oxidizer and the nanoparticles spray were maintained constant for the entire run. The spray velocity varied from 100 m/s to 200 m/s with titanium dioxide (TiO2) nanoparticles. Using the series of the governing equation and modified Navier Stokes equation the model has been developed with the aid of numerical workbench. Three different domains are generated for fuel, oxidizer and nanoparticles. The velocity of the air and nanoparticles were maintained at constant levels and varying only the spray velocity of the fuel. Based on the findings, the mass fraction of both fuel and formation of the CO2 were dependent on the spray velocity. As the spray velocity increases the turbulence in the combustion chamber increases which ensures the higher mixing of both air-fuel and nanoparticles. From the procured findings 175 m/s and 200 m/s were the ideal range for better combustion efficiency compared to 100 m/s and 150 m/s. The simulation results have ascertained the role of the spray velocity on the emissions and the combustion efficiency of the engine.     Molecular rearrangement of pyrazino[2,3‐c]quinolin‐5(6H)‐ones during their reaction with isocyanic acid

    New tetrahydropyrazino[2,3-c]quinolin-5(6H)-ones were prepared from 3-chloroquinoline-2,4(1H,3H)-diones and ethylene diamine. In their reaction with HNCO, an unprecedented molecular rearrangement produced new types of hydantoin derivatives. All prepared compounds were characterized on the basis of their H-1, C-13, and N-15 NMR and ESI mass spectra and some were authenticated by X-ray analysis of single crystalline material. A proposed mechanism for rearrangement is discussed in this essay.     Evaluation and effect of rice bran and microwave heating on the physicomechanical properties of low-density polyethylene film and packaged milk quality

    Low-density polyethylene (LDPE) film was developed with a mixture of reference LDPE and rice bran (RB). Three mixtures of 5, 10, and 15% RB were used for the LDPE film preparation. The films were analysed for the physicomechanical properties before microwave heating (MH) and after MH and packaged milk quality was analysed regarding MH and pasteurisation. After MH, the physicomechanical properties of the material change significantly. LDPE film containing 15% RB was created to beat changes in tensile strength, elongation at break, bursting strength, and water vapour transmission rate (WVTR). The total migration of tested packaging materials was within the agreeable limit and after MH it also was within the permissible limit. Four different simulators were used to create food categories. These food simulators included distilled water, 3% acetic acid, 50% alcohol (ethanol), and n-heptane.     The article deals with new possibilities of electronic documentation eHealth (a global concept of electronic health care) and examines the use of information technologies in health care. It describes the issue of keeping medical records in electronic form and then analyzes the method of implementation of these technologies. The electronic form of this information will greatly simplify and make available the transfer of information. Currently, this form is required mainly from workers in the medical and biomedical fields. At the beginning of the article are sections devoted to the history, development, and future possible direction of healthcare. It focuses on the implementation of the information system, ensuring its security and data protection. Followed by general information, characteristics, and elements of healthcare 4.0. Emphasis is placed on the benefits, problems, and possible complications associated with e-health.     The impact of ethical and CSR factors on engineers’ attitudes towards SMEs sustainability

    The aim of this study was to examine the impact of selected ethical factors of business on the perception of defined attributes of decline in the small-and medium-sized enterprise sectors of the V4 countries. The perceptions of ethical business factors between entrepreneurs-engineers and entrepreneurs-non-engineers were evaluated. The study underscores the need for permanent implementation of corporate social responsibility (CSR) analyses from a multilevel perspective that allows the integration of different theoretical frameworks. The results of the study justify the need to carry out deeper structured analyses of the ethical aspects of micro-CSR business at the individual or group level, taking into account multiple categorisation lines. This study also confirms the psychological effects of perceived CSR on a company's decline.     Differences of education level and job position in digital transformation adoption in Vietnam's creative industries

    The aim of this paper is to contribute to understanding the differences in educational level and job titles that affect the adoption of digital transformation in the context of creative industries. The approach is based primarily on the technology-organizational-environment framework, one-way analysis of variance, and multivariate linear regression that were used to test the empirical model. This article describes the difference between the educational level and the job position groups for the adoption of digital transformation in Vietnam's creative industries, as well as an essential difference between them for the predictor variables. Our research has underlined the importance of demographic variables in relation to the adoption of digital transformation. The current study is limited to some fields of creative industries in Vietnam, such as design, creative services, new media, audiovisuals, publishing and printed media, and selected demographic parameters.     Michigan diabetes knowledge test: validation of modified version among Czech students of nursing

    Michigan Diabetes Knowledge Tool (MDKT) was developed to test the level of knowledge in managing Diabetes Mellitus. It is widely used as an original (for patients) and modified tool (for nurses or nursing students) around the world. Objectives: The aim is psychometric testing of the modified MDKT. Methods: This quantitative study uses descriptive statistics, Classical Test Theory (CTT), Item Response Theory (ITT), and reliability estimation within the convenient sample of students of nursing (N = 133). Data collection took place in 2018-2019 at a selected Czech University. Results: Using the MDKT only enables us to differentiate within a narrow spectrum of students whose knowledge is sub-average to average. The analysis has suggested the possibility of reducing the test while maintaining its psychometric parameters. Conclusions: The test is reliable, however too simple for our convenient sample of third-year nursing students, but this result cannot be generalised.     Reprocessed magnetorheological elastomers with reduced carbon footprint and their piezoresistive properties

    Despite the vast amount of studies based on magnetorheological elastomers (MREs), a very limited number of investigations have been initiated on their reprocessing. This paper presents a new type of recyclable MRE which is composed of thermoplastic polyurethane (TPU) and carbonyl iron particles (CI). The chosen TPU can be processed using injection moulding (IM), followed by several reprocessing cycles while preserving its properties. Numerous types of injection moulded and reprocessed MREs have been prepared for various particle concentrations. The effect of thermo-mechanical degradation on the recycled MREs has been investigated while simulating the reprocessing procedure. An apparent decrease in molecular weight was observed for all the examined matrices during the reprocessing cycles. These changes are attributed to the intermolecular bonding between the hydroxyl groups on the surface of the CI particles and the matrix which is studied in depth. The effect of reprocessing and the presence of magnetic particles is evaluated via tensile test, magnetorheology and piezoresistivity.
