Genotypic and phenotypic detection of polyhydroxyalkanoate production in bacterial isolates from food
Polyhydroxyalkanoates (PHAs) are widely used in medical and potentially in other applications due to their biocompatibility and biodegradability. Understanding PHA biosynthetic pathways may lead to the detection of appropriate conditions (substrates) for producing a particular PHA type by a specific microbial strain. The aim of this study was to establish a method enabling potentially interesting PHA bacterial producers to be found. In the study, all four classes of PHA synthases and other genes involved in PHA formation (fabG, phaA, phaB, phaG, and phaJ) were detected by PCR in 64 bacterial collection strains and food isolates. Acinetobacter, Bacillus, Cupriavidus, Escherichia, Klebsiella, Lelliottia, Lysinibacillus, Mammaliicoccus, Oceanobacillus, Pantoea, Peribacillus, Priestia, Pseudomonas, Rahnella, Staphylococcus, and Stenotrophomonas genera were found among these strains. Fructose, glucose, sunflower oil, and propionic acid were utilized as carbon sources and PHA production was detected by Sudan black staining, Nile blue staining, and FTIR methods. The class I synthase and phaA genes were the most frequently found, indicating the strains' ability to synthesize PHA from carbohydrates. Among the tested bacterial strains, the Pseudomonas genus was identified as able to utilize all tested carbon sources. Optical properties of thin tungsten films
In this work optical properties of thin tungsten films deposited by radiofrequency magnetron sputtering close to insulator-conductor transition were investigated for their potential use in optoelectronics. These films were deposited on a V1 mica substrate. An atomic force microscopy topographical maps, optical absorbance measured on a spectrophotometer, and optical transmittance results were reported. © 2022 IEEE
Time profile of the glass-break detector testing scenario based on the accelerometer
This paper is focused on the contact glass-break detectors used in technical security namely in the Intruder Alarm Systems. The glass-break detector is used as the main component of the perimeter layer of protection against the potential intruder. The main aim of this research is to measure and calculate the average time duration of the impact on the glass panel. More specifically, how long does it take to absorb created vibrations by the glass panel and the window frame. This number can also determine the maximal possible detection in one second. The second outcome is the graphical representation of the impact itself in time. The main experiment is done according to the standardization for the contact glass-break detectors and it is done by using the accelerometer placed on the glass panel which measures vibrations created by the impact in real-time. These data are measured and evaluated by the development of the development board called Arduino Nano, and the accelerometer MPU-6050. Interactions between sodium hyaluronate and β-cyclodextrin as seen by transport properties
Knowledge of mass transport parameters, diffusion, and viscosity of hyaluronic acid (HA) in the presence of cyclodextrins is of considerable importance for areas such as food packaging and drug delivery, among others. Despite a number of studies investigating the functionalization of HA or the corresponding sodium salt by cyclodextrins, only a few studies have reported the effect of cyclodextrins on the mass transport of HA in the presence of these oligosaccharides. Here, we report the tracer binary and ternary interdiffusion coefficients of sodium hyaluronate (NaHy) in water and aqueous β-cyclodextrin solutions. The diffusion behavior of sodium hyaluronate was dependent on the reduced viscosity of NaHy, which, in turn, presented a concave dependence on concentration, with a minimum at approximately 2.5 g dm-3. The significant decrease in the limiting diffusion coefficient of NaHy (at most 45%) at NaHy concentrations below 1 g dm-3 in the presence of β-cyclodextrin, taking water as the reference, allowed us to conclude that NaHy strongly interacted with the cyclodextrin.Fundacao para a Ciencia e a Tecnologia (FCT) through COMPETE Programme (Operational Programme for Competitiveness) [UIDB/QUI/00313/2020]; Ministry of Education, Youth and Sports of the Czech Republic DKRVO [RP/CPS/2022/003
Sustainable HRM practices for a stronger employer brand: Leveraging organizational culture
The business of the current era intends to show competitive advantage for a long run in order to get more profits and stay alive in the markets. Getting a competitive advantage is highly related to human resource management (HRM). Companies build a distinct employer brand based on their competitive advantage; however, the role of organizational culture to support the transition of companies to get a powerful employer brand is rarely studied. Consequently, the main purpose of the current study is to determine the mediating role of organizational culture (engagement and productivity) in the relationship between sustainable HRM practices and employer branding. The survey research design was used and data were collected from 420 employees of Azerbaijani organizations using a convenience sample strategy. The findings of the path analysis using AMOS revealed the partial multiple mediating role of organizational culture (engagement and productivity) in the relationship between sustainable HRM practices and employer branding. Moreover, as far as the individual mediating role is concerned, the mediating role of organizational engagement is stronger compared to productivity. Peripheral component interconnect express card for the closed-circuit television application
Technical security became very popular and convenient in many sectors where physical security is not a sufficient solution. Many companies use the Closed-Circuit Television (CCTV) application for security applications. The aim of this paper is to design the extended Peripheral Component Interconnect Express (PCI-E) card for the CCTV application for extensive companies. The card has three independent Ethernet terminal where IP cameras can be connected. This solution is much more cost-effective and easy to use solution than traditional Digital Video Recorder (DVR). The card communicated via the Universal Serial Bus (USB) interface which is implemented in the PCI-E standard. Physicochemical and sensory properties of Czech lager beers with increasing original wort extract values during cold storage
The scope of the study was the evaluation of the selected physicochemical (O-2 and CO2 contents, bitterness, color, total polyphenol content (TPC), turbidity, foaming stability) and sensory properties of Czech lager beer with different original wort extract (OWE) values (OWE of 10.0; 11.0; 11.5; 12.0% w/w) during a cold storage period of 6 months (4 +/- 2 degrees C). The length of the cold storage period did not influence the values of dissolved O-2 and CO2 , bitterness, color and foam stability of the samples. Contrarily, the TPC, turbidity, and sensory attributes of the samples were affected by the course of cold storage. The OWE values did not affect the development of the parameters tested. All beer samples stored until the 5th month presented "very good" sensory characteristics. Functional and quality profile evaluation of butters, spreadable fats, and shortenings available from Czech market
The aim of this study was to assess the functional properties of butters, spreadable fats, and shortenings, collected from the Czech market, in correlation with their nutritional values declared by the producers. Various methods were applied to determine relevant parameters of the products. Using penetration tests, samples were characterized by specific textural attributes according to their composition and processing type, particularly for the presence of milk/vegetable fats. Using differential scanning calorimetry (DSC), thermal peaks corresponding to medium- and high-melting triacylglycerol fractions were detected in the ranges 15-16 degrees C and 31.5-34.5 degrees C, respectively. Rheological analysis revealed that the viscoelasticity of samples was related to frequency behavior of the fat structure, characterized by the dominance of elastic modulus (G') over viscous modulus (G '') up to the frequency of 10 Hz. This indicated good emulsion stability of the products in the region of linear viscoelasticity. For spreadable fats, the structure was resistant to phase separation in the whole frequency range under study (0.1-100 Hz). Role of chicken fat waste and hydrogen energy ratio as the potential alternate fuel with nano-additives: Insights into resources and atmospheric remediation process
The main focus of the study was to witness the effects of chicken waste-based biodiesel blends along with constant hydrogen injection in a modified diesel engine. Furthermore, the nanoparticle multiwall carbon nanotubes (MWCNT) effects on the engine efficiency were also examined. A series of tests was conducted in the single cylinder, water cooled engine fuelled with diesel, CB100N, CB10N, CB30N, and CB50N. Throughout the entire run, constant hydrogen injection of 5 LPM has been maintained. The parameters such as brake thermal efficiency, brake specific fuel consumption, heat release rate and the emissions of different pollutants were determined for a variety of engine speeds. ASTM standards were applied to measure the viscosity, density and calorific value. From the reported findings, it was clear that the addition of the chicken waste biodiesel could be a sustainable substitute for the existing fossil fuels. Although the emission of the pollutants was dropped significantly, there was a massive drop in the BTE values. To compensate such shortage of power, the biodiesel was dispersed with MWCNT at the concentration of 80 ppm. Compared to the regular biodiesel, MWCNT inclusion increased the BTE by 14%. Further, the consumption of the fuel was also reduced marginally. Considering the pollutants, the catalytic activity of the MWCNT reduced the emissions of CO, NOx, and HC at various engine speeds. Besides, 10% reduction in NOx had been reported at lower engine speeds and was reduced to 8% at higher speed regimes. Compiling all together, increasing the concentration of the biodiesel blends obviously reduced the performance values and however, there was a great advantage in terms of the emission magnitudes irrespective of the engine operating conditions. © 2022 Elsevier Inc.King Saud University, KSUKing Saud University, Riyadh, Saudi Arabia [RSP-2022/257]; Van Lang University, Vietna
Verification of functional prototypes of bearings in experimental equipment ability to simulate real operation
The reliability and service life of dynamically stressed machine components are most often determined by the time of operation and the probability of achieving it. With the trend of increasing the power of machinery and the goal of minimizing the dimensions, the density of transmitted power increases, and thus the demands on the bearing capacity and service life increase significantly. One of the ways to increase the reliability of bearings, to reduce the risk of their sudden failure, in addition to their monitoring, is the use of so-called self-balancing bearings. The article deals with the experimental verification of the functionality of a newly developed self-equalizing bearing and its ability to operate in real conditions. Three types of experimental devices were used during testing while the temperature of tilting segments, the uniformity of temperatures around the perimeter, force distribution, and deflection were monitored. The results showed that the tested prototypes of bearings have met all requirements based on which the newly developed bearings have been successfully implemented in real practice. © 2022 American Institute of Physics Inc..
