    ctl: A package for assessing agreement based on clinical tolerance limits

    The ctl command implements a recently proposed statistical methodology to assess the level of agreement or interchangeability between two quantitative measurement methods. It is based on tolerance limits that are specified by the user. The methodology requires repeated measurements by at least one of the two measurement methods. It accommodates heteroskedastic measurement errors and often performs well even when the user has only one measurement by one of the two measurement methods and at least five repeated measurements from the other. It provides a more direct assessment of the agreement level than the Bland-Altman limits of agreement method and circumvents some of its deficiencies

    The Swiss canton of Neuchâtel was able to develop and become one of the economic poles of the country – particularly in the export of watches – thanks to the movement of emigrants to Europe and European immigrants to Switzerland. At the same time, missionaries, explorers and businessmen left Neuchâtel to travel to the colonial world. Historiography has never linked these two migratory phenomena. This article aims to fill this gap. On one side, it will provide an overview of the existing historiography. On the other, through some case studies, it will propose evidence and new research approaches. The result will be an invitation to de-Europeanise the history of Neuchâtel in order to inscribe it in a global history in the colonial era

    Joint hypermobility is a common feature in children and adolescents, often physiological and benign, but it is also a manifestation of hereditary connective tissue disorders. In pediatrics, there is a real need to differentiate Ehlers-Danlos syndrome (EDS) hypermobile type (hEDS) from benign hypermobility, in the absence of a genetic marker specific to this form of EDS. In 2023, the International Ehlers-Danlos Consortium revised the diagnostic criteria for children and adolescents, introducing the concept of hypermobility spectrum disorder. In line with these recommendations, the clinical criteria for hEDS established for adults do not apply in paediatrics, and hEDS should no longer be diagnosed before the age of biological maturity. At the end of the child's growth, a specialized reassessment is required, applying the 2017 hEDS criteria

    Co-targeting CDK4/6 enhances anti-cancer activity and alleviates immune-related adverse events of anti-PD-1 antibody for breast cancer

    Early-onset breast cancer (EOBC) increases annually with unclear mechanisms and shows poor therapeutic responses and survival rates. We identify environmental exposure to the common plasticizer di-2-ethylhexyl phthalate (DEHP) as a contributing factor to breast tumor initiation by impairing cancer immune surveillance. DEHP exposure upregulates the immune checkpoint molecule programmed death-ligand 1 (PD-L1) through estrogen receptor beta (ERβ) activation in human breast cancer cells and mouse models. Transcriptomic analysis reveals cyclin-dependent kinase 4 (CDK4) as a key mediator of DEHP-induced immunosuppressive signaling. The CDK4/6 inhibitor palbociclib suppresses ERβ, programmed cell death protein 1 (PD-1), and PD-L1 expression, enhances anti-tumor responses, and reduces immune-related adverse events caused by anti-PD-1 antibody treatment in mice. While the strongest immune alterations appear in DEHP-exposed EOBC patients, the mechanisms extend to DEHP-associated breast cancer regardless of age. This study highlights immune dysregulation as a hallmark of DEHP-related breast carcinogenesis and supports co-targeting CDK4/6 and PD-L1 as a therapeutic strategy. Copyright © 2025 The Authors.     Une étude plurielle des réseaux de personnages : narrations interactives, jeux vidéo et adaptations

    Si les réseaux de personnages sont couramment utilisés dans les études littéraires depuis le début du siècle, leur déclinaison à des narrations moins linéaires ou plurielles pose encore de nombreux défis. Le présent travail s'emploie à adapter cet outil à deux terrains nouveaux : les narrations interactives, et les études comparatives d'œuvres cross-médiatiques. Le premier axe propose un modèle théorique appelé "flux narratif", qui transforme des œuvres pensées de manière interactive en une structure permettant d'y appliquer des outils de distant reading, jusqu'ici réservés aux narrations linéaires. Ce modèle est ensuite mis en pratique à l'aide de réseaux de personnages, sur la franchise de jeux vidéo Life is Strange. Le deuxième axe explore plusieurs approches de comparaison de réseaux, dans l'objectif de mettre en parallèle deux œuvres cross-médiatiques grâce à leur réseau de personnages respectif. La partie pratique présente une dizaine d'adaptations et étudie en détail les résultats obtenus pour chaque paire d'œuvres, proposant ainsi une base de travail pour des recherches ultérieures

    CDK4 inactivation inhibits apoptosis via mitochondria-ER contact remodeling in triple-negative breast cancer.

    The energetic demands of proliferating cells during tumorigenesis require close coordination between the cell cycle and metabolism. While CDK4 is known for its role in cell proliferation, its metabolic function in cancer, particularly in triple-negative breast cancer (TNBC), remains unclear. Our study, using genetic and pharmacological approaches, reveals that CDK4 inactivation only modestly impacts TNBC cell proliferation and tumor formation. Notably, CDK4 depletion or long-term CDK4/6 inhibition confers resistance to apoptosis in TNBC cells. Mechanistically, CDK4 enhances mitochondria-endoplasmic reticulum contact (MERCs) formation, promoting mitochondrial fission and ER-mitochondrial calcium signaling, which are crucial for TNBC metabolic flexibility. Phosphoproteomic analysis identified CDK4's role in regulating PKA activity at MERCs. In this work, we highlight CDK4's role in mitochondrial apoptosis inhibition and suggest that targeting MERCs-associated metabolic shifts could enhance TNBC therapy

    Ce document s’intègre dans une série d’études qui, avec une perspective de droit comparé, visent à faire une présentation du principe de protection de l’environnement dans différents ordres juridiques. Après avoir expliqué le droit positif et la jurisprudence d’application, le contenu, les limites et la possible évolution de ce principe sont examinés. La présente étude a pour objet le cas de la Suisse. La protection de l’environnement s’est construite de manière sectorielle, depuis la première moitié du XXe siècle. En complément aux législations spécifiques à la protection de la nature, de l’eau, des forêts, des animaux, ou à la législation sur la chasse et la pêche, la loi sur la protection de l’environnement du 7 octobre 1983, entrée en vigueur le 1er janvier 1985, vient non seulement combler les lacunes relativement à la protection de l’homme et de l’environnement en matière d’atteintes nuisibles ou incommodantes, mais également poser des principes généraux, applicables à l’ensemble de la matière environnementale. Ces principes d’anticipation des atteintes (prévention/précaution), de causalité des coûts (pollueur-payeur) et de durabilité des ressources sont désormais exprimés dans la Constitution fédérale de 1999 aux art. 73 et 74 Cst. et constituent le pivot de la matière. Le droit suisse n’a pas consacré de droit à un environnement sain. Cependant, les principes évoqués ci-dessus sont, dans une certaine mesure, justiciables, soit directement, soit par leur portée interprétative, de telle sorte qu’ils permettent une harmonisation de la mise en œuvre des textes infraconstitutionnels. Par ailleurs, dans les domaines qui touchent à la sphère de protection des individus, le droit infraconstitutionnel donne des droits invocables en justice : droit d’exiger l’absence d’atteintes nuisibles ou incommodantes, droit d’exiger le contrôle des actes de surveillance d’installations dangereuses (dans le domaine nucléaire, notamment)
