Three Essays in Asset Pricing
This thesis explores how firm relationships and digital infrastructures influence asset prices. The first two
chapters examine customer–supplier links in U.S. equity markets, focusing on known patterns where stock
returns of customer firms predict those of their suppliers. Chapter 1 employs generative AI to classify these
relationships according to product characteristics such as whether goods are tangible or high-value-added,
among other criteria. The results reveal that predictability varies considerably across these classifications.
Only relationships involving high-value-added goods consistently demonstrate strong predictive effects.
Other categories, such as tangible goods, show predictability only in certain periods or under specific
modeling conditions. Chapter 2 uses news data to split customer returns into two components—those linked to observable firm-
specific news and those without identifiable news events. It finds that news-related returns independently
exhibit a lead-lag pattern of comparable magnitude to the overall effect, confirming that explicit news plays
a meaningful role in return predictability. Chapter 3 examines Bitcoin’s high valuation despite major limitations in its original blockchain, including
limited transaction throughput and declining miner incentives. It proposes tokenization—a mechanism
enabling gradual migration of Bitcoins to a more scalable blockchain. The theoretical model illustrates that a
slow transition toward tokenization can naturally occur without legal mandates or centralized coordination,
thus rationalizing Bitcoin’s current market valuation
Loss of the neuronal kinase DCLK3 leads to anxiety-like behaviour and memory deficits.
DCLK3 (Doublecortin-like kinase 3) is a kinase preferentially expressed in neurons. Dclk3 variants are risk factors for psychiatric disorders and brain alterations linked to age-related mild cognitive impairment, suggesting that this kinase could be important to cognition. However, the physiological role of DCLK3 remains unknown. Here, we generated and characterized mice with constitutive or brain region-specific Dclk3 knockout. We found that constitutive pan-deletion of Dclk3 is associated with an anxiety phenotype in males, associated with changes in brain metabolites in the absence of major neuroanatomical alterations, motor deficits or memory deficits. Moreover, virally mediated loco-regional conditional Dclk3 knockout in the dorsal hippocampus of adult mice led to spatial memory deficits and transcriptomic changes, characterized by increased expression of α2, β1 and β2 subunits of the GABAA receptor, downregulation of neuronal activity-regulated genes, such as the immediate early genes (e.g. Egr1, Fos, Per1 and Nr4r1), and transcriptional regulator activity enriched in polycomb-repressive complexes. These new data reveal that Dclk3 modulates behaviour and cognition in association with transcriptomic changes in the hippocampus, providing direct physiological evidence that this kinase is involved in synaptic plasticity, consistent with its crucial role in neurodegenerative and psychiatric diseases

Sous le nom de théorie des textes possibles, on a pu proposer un mode de description qui permette d'inférer de la lettre d’un texte la grammaire des possibles dans laquelle il s’est élaboré. La démarche consiste à envisager chacun des choix de l’auteur, tels qu’on peut les déduire d’une analyse de la composition d’un texte, comme une option parmi d’autres, pour entrer en retour dans l’élaboration de variantes. Ce qui revient à considérer l’œuvre isolée comme un genre à part entière : un système de règles (de fait) dont le texte considéré constituerait la seule réalisation historiquement attestée, mais qui seraient (en droit) susceptibles de donner jour à une nouvelle création, ou qui autoriseraient tout au moins à lui imaginer des variantes. Pushing the boundaries of radiotherapy-immunotherapy combinations: highlights from the 7th immunorad conference.
Over the last decade, the annual Immunorad Conference, held under the joint auspicies of Gustave Roussy (Villejuif, France) and the Weill Cornell Medical College (New-York, USA) has aimed at exploring the latest advancements in the fields of tumor immunology and radiotherapy-immunotherapy combinations for the treatment of cancer. Gathering medical oncologists, radiation oncologists, physicians and researchers with esteemed expertise in these fields, the Immunorad Conference bridges the gap between preclinical outcomes and clinical opportunities. Thus, it paves a promising way toward optimizing radiotherapy-immunotherapy combinations and, from a broader perspective, improving therapeutic strategies for patients with cancer. Herein, we report on the topics developed by key-opinion leaders during the 7 th Immunorad Conference held in Paris-Les Cordeliers (France) from September 27th to 29th 2023, and set the stage for the 8 th edition of Immunorad which will be held at Weill Cornell Medical College (New-York, USA) in October 2024
Gastrostomy tube feeding in children: a single-center experience.
Despite the widespread use of percutaneous endoscopic gastrostomy (PEG) in pediatric populations, there is a paucity of data on the indications and outcomes of this procedure in Switzerland. This manuscript presents our experience with PEG indication, outcomes, and related complications in children.
This single-center retrospective study included patients < 18 years old who underwent PEG placement between 2007 and 2016. We retrieved demographics, PEG indications, associated comorbidities, pre-placement workup, growth parameters up to 12 months, and associated complications.
Eighty-one patients were included, with a median age of 7 years. Common indications included inadequate caloric intake (85%), failure to thrive, and feeding difficulties. Neurological conditions (46%) were the most commonly associated comorbidity. Thirty-six patients (44%) underwent a pH study before PEG placement. There were significant increases in z-scores for weight (p < 0.002) and body mass index (p < 0.001) 12 months after PEG placement. Minor complications were relatively frequent (n = 55, 68%), mainly granulation tissue or local erythema. Two patients had major complications.
PEG is a safe technique for providing long-term enteral nutrition in children, with neurological disease being the most common clinical indication. Our experience demonstrated significant weight gain in children after one year of PEG, with frequent but well-controlled complications
The BARIREF Study: BARIatric Surgery Complications in Patients with Prior REFlux Surgery in a Large National Cohort.
To assess the outcomes of Roux-en-Y gastric bypass (RYGB) and sleeve gastrectomy (SG) in patients with prior fundoplication.
Obesity and gastroesophageal reflux disease (GERD) often coexist, posing clinical challenges. Fundoplication is the standard treatment for GERD refractory to medical therapy. Managing obesity after fundoplication is complex and while RYGB is preferred for addressing both obesity and GERD, its use post-fundoplication requires further evaluation.
Retrospective analysis of a national French database of patients undergoing RYGB and SG between 2013 and 2023. Patients were grouped according to prior fundoplication (PriorF) or no fundoplication (NoF). Baseline demographics and 90-day postoperative outcomes were assessed. Univariate and multivariate analyses identified risk factors; 1:5 matching was applied based on sex, age, body mass index, comorbidities, surgery year, procedure type and center volume.
Among 372,464 patients, 337 (0.1%) had prior fundoplication. PriorF patients had higher rates of severe postoperative complications (Dindo ≥IIIa: 8.9% vs. 3.7%, P < 0.001), longer hospital stays (4.5 ± 4.9 vs. 3.7 ± 2.9 d, P < 0.001), and higher readmission rates (17.8% vs. 10.6%, P < 0.001). Multivariate analysis for RYGB showed no significant association between prior fundoplication and complications (OR 1.71, 95% CI 1.00-2.74, P=0.051). Prior fundoplication was independently associated with increased complications after SG (OR 2.54, 95% CI 1.37-4.32, P < 0.001).
Prior fundoplication increased postoperative complications after SG but not after RYGB. RYGB appears to be the safer bariatric procedure in patients with prior fundoplication
Soil microbiome interventions for carbon sequestration and climate mitigation.
Mitigating climate change in soil ecosystems involves complex plant and microbial processes regulating carbon pools and flows. Here, we advocate for the use of soil microbiome interventions to help increase soil carbon stocks and curb greenhouse gas emissions from managed soils. Direct interventions include the introduction of microbial strains, consortia, phage, and soil transplants, whereas indirect interventions include managing soil conditions or additives to modulate community composition or its activities. Approaches to increase soil carbon stocks using microbially catalyzed processes include increasing carbon inputs from plants, promoting soil organic matter (SOM) formation, and reducing SOM turnover and production of diverse greenhouse gases. Marginal or degraded soils may provide the greatest opportunities for enhancing global soil carbon stocks. Among the many knowledge gaps in this field, crucial gaps include the processes influencing the transformation of plant-derived soil carbon inputs into SOM and the identity of the microbes and microbial activities impacting this transformation. As a critical step forward, we encourage broadening the current widespread screening of potentially beneficial soil microorganisms to encompass functions relevant to stimulating soil carbon stocks. Moreover, in developing these interventions, we must consider the potential ecological ramifications and uncertainties, such as incurred by the widespread introduction of homogenous inoculants and consortia, and the need for site-specificity given the extreme variation among soil habitats. Incentivization and implementation at large spatial scales could effectively harness increases in soil carbon stocks, helping to mitigate the impacts of climate change
